1.Differentiation into oligodendrocytes of bone marrow stromal cells transplanted to the injured spinal cord

Objective To investigate the feasibility of transplanting bone marrow stromal cells (BMSCs) in the injured spinal cord for differentiation into functional oligodendrocytes in rats. Methods The ultra-structure of transplanted BMSCs in the injured rat spinal cord was observed at 1,3,5 weeks after transplantation by immuno-electron microscopy and low temperature embedding.The expressions of myelin basic protein (MBP) and proteolipid protein (PLP) in the transplanted BMSCs in the injured spinal cord were observed at 1,3,5 weeks after transplantation by immunofluorescence staining and laser confocal scanningmicroscopy. Results The organelles in the BMSCs migrated into the white matter of the spinal cord anddeveloped at 1 week after transplantation,showing the ultra-structure features of oligodendrocytes.The BMSCs migrating into the white matter of the spinal cord showed mature ultra-structure features of oligodendrocytes and formed myelin-like structures at 3 and 5 weeks after transplantation.The BMSCs migrating into the white matter of the spinal cord expressed the MBP and PLP at one week after transplantation,and continued to express the MBP and PLP at 3 and 5 weeks after transplantation. Conclusion The BMSCs transplanted into the injured spinal cord nay partly differentiate into functional oligodendrocytes.
2.Clinical significance of The changes of C-reactive protein in acute phase of chronic obstructive pulmonary disease

Objective To investigate the changes of CRP in diagnosis of the acute exacerbation of COPD and its significance.Method CRP and WBC were measured both when the patients just before and after the treatment with effective antibiotics to compare their change.Results The positive rate of CRP is 85.40%.and the difference Was obvious(P<0.01).At the same time,the CRP's difference between before and after the use of antibiotics was also obvious(P<0.01).Conclusion CRP is a sensitive and specific index to the acute exacerbation of COPD,and is helpful to discover this acute disease early as well as a good guide to estimate the serious of pneumonia sand to respond the efficacy of antibiotics.
3.Effects of inflammation after spinal cord injury on the survival and immigration of bone marrow mesenchymal stem cells injected intravenously

Objective To determine the optimal time for intravenous injection of bone marrow mesenchymal stem cells(BMSCs)by observing the effects of inflammation after spinal cord injury on the survival and immigration of BMSCs injected intravenously.Methods Forty SD rats were randomized into 1 non-injury group and 7 spinal cord injury(SCI)groups in which the injured spinal cords were harvested at 0 h, 6 h, 12 h, 24 h, 3d, 5 d, and 7 d respectively.Hematoxylin and eosin(HE)staining was used to e-valuate the histopathologic changes.Myoleperoxidase(MPO)activity was measured to study the duration of polymorphonuclear leukocyte(PMNL)infiltration in the spinal cord after injury.Another 40 SD rats were also randomly divided into 1 non-injury group and 7 transplantation groups in which intravenous injection of BMSCs was performed at 0 h, 6 h, 12 h, 24 h, 3d, 5 d, and 7 d after SCI.The survival counts and immigration distance of BMSCs in the spinal cord in all the groups were measured.Results The PMNL infiltration into the injured spinal cord and the MPO activity started from 6 h after SCI.There were a large number of infiltrating PMNL in the spinal cord and the MPO activity reached its peak value at 24 h.The infiltrating PMNL in the injured spinal cord began to decrease and the MPO activity began to weaken from 3 d.Proliferation of glial cells and formation of empty holes in the spinal cord started from 5 d.The survival counts of BMSCs were smaller but the immigration distance of BMSCs was longer in the 0 h, 6 h, 12 h, and 24 h transplantation groups.The survival counts of BMSCs were larger and the immigration distance of BMSCs was longer in the 3 d transplantation group.The survival counts of BMSCs were smaller but the immigration distance of BMSCs was shorter in the 5 d and 7 d transplantation groups.Conclusion The optimal time for intravenous injection of BMSCs may be 3 days after SCI.
4.Clinical comparison of different blood collection methods in routine blood test results

Objective To compare clinical value of peripheral blood collection and venous blood collection methods in blood tests.Methods 467 patients who received blood test were selected.The venous blood and peripher-al blood were collected,the indicators of blood test were detected,and the test results were analyzed.Results The white blood cell count (WBC),red blood cell count (RBC),platelet count (PLT),red blood cell hematocrit (HCT),hemoglobin (HB)of venous blood were (5.7 ±1.2)×109 /L,(3.72 ±0.48)×109 /L,(218.0 ±57.5)× 109 /L,(41.8 ±19.8)g/L,(115.7 ±13.8)g/L respectively,those of peripheral blood were (6.7 ±3.4)×109 /L, (3.62 ±0.72)×109 /L,(204.0 ±87.7)×109 /L,(32.7 ±20.7)g/L,(103.6 ±19.6),the differences were statis-tically significant (t =5.994,2.497,2.885,6.865,10.908,all P <0.05).The mean red cell hemoglobin concentra-tion (MCHC),mean red cell volume (MCV),mean erythrocyte hemoglobin (MCH)and other indexes had no signifi-cant differences(all P >0.05).Conclusion The test results in patients with venous and peripheral blood was quite different,venous blood test results obtained with greater value in clinical use,it is recommended that routine blood test used venous blood.
5.Study about effects of BCG treatment on expression of stem cell factor in asthmatic mice

Objective To study the effects of Bacillus Calmette-Guerin (BCG) intervention on the expression of stem cell factor (SCF) in asthma. Methods Kunming mice were randomly divided into asthmatic group, BCG group and control group of ten mice each group. Mice were sensitized and challenged with ovalbumin (OVA) to establish asthmatic model. After twenty-four hours of last challenge, eosinophils (EOS) were counted in the lung tissue and bronchoaveolar lavage fluid (BALF). The level of SCF in BALF was determined by enzyme-linked immunosorbent assay. The expression of SCF protein in lung tis-sue was measured by immunohistochemistry technique and computerized image analysis system. Results The number of EOS in lung tissue was (10.67±1.94)/HP, (6.40±1.55)/HP and (0.37±0.33)/HP in asthmatic, BCG and control group respectively. The number of EOS in BALF was (7.58 ± 1.30) × 107/L, (3.78 ± 1.15) × 107/L and 0 in asthmatic, BCG and control group respectively. The difference between each group was statistically significant (P<0.01). The immunohistochemical index of SCF protein in lung was (24787.97±7214.12), (20509.50±4775.27) and (12261.66±3277.65) in asthmatic, BCG and control group respectively. The SCF level of BALF was (280.25 ± 14.20) pg/ml, (266.77 ± 31.15) pg/ml and (223.59 ± 15.61) pg/ml in asthmatic, BCG and control group respectively. The SCF level in BALF in asthmatic and BCG group was significantly different from that in control group (P<0.05). There was no significant difference of SCF level in BALF between BCG and asthmatic group (P>0.05). In asth-matic group, the expression of SCF in lung was positively correlated with the number of EOS (P<0.05). Conclusions BCG treat-ment can markedly decrease the airway inflammation in asthmatic mice. BCG cannot inhibit the expression of SCF in asthma.
6.EVALUATION OF THE DIAGNOSIS OF INSULINOMA

The article presents an analysis of clinical signs, pathological findings and results of laboratory examinations of 10 cases of insulinoma. Diagnostic procedures such as blood sugar and serum insulin determinations, elective arteriography and CT scanning are evaluated. Besides, the relationship between the clinical signs and pathological findings were studied. The results of the study showed that the ratio of blood sugar to insulin and elective ateriography were valuable significant in diagnosing insulinoma, and that the diagnostic efficiency of CT left much to be desired. They also showed that duration of the disease had remarkable effects on patients with insulinoma.
7.Hand-assisted laparoscopic liver resection without hand-port devices: Report of 8 cases

Objective To investigate the feasibility of hand-assisted laparoscopic liver resection without hand-port devices. Methods Eight patients were selected into the study, including 5 cases of primary liver cancer located at the segment Ⅳ or Ⅴ or the left lateral lobe, 2 cases of liver spongy hemangioma and 1 case of hepatic focal nodular hyperplasia. After the laparoscopic devices were set up, an oblique incision below the right costal margin was made. The length of the incision was the same as the operator’s wrist. In the absence of hand-port or “blue disc”, the operator’s left hand directly entered the abdominal cavity through the incision. With the assistance of operator’s left hand, the laparoscopic liver resection was conducted, with the ligaments surrounding the liver dissociated, the liver transected and the cut surface properly managed. Results Laparoscopic liver resection was performed successfully in all the 8 patients. The operation time ranged 90~180 min (mean, 118 min), and the blood loss was 50~ 300 ml (mean, 160 ml). There was no uncontrolled bleeding during the operation and no complication happened. Conclusions Hand-assisted laparoscopic liver resection without hand-port devices is a safe and feasible procedure. It may not only reach the purpose of hand assistance but also lower the surgical costs.
8.Exploration on the Ethical Problems in the Implementation of the Two-Child Policy in China

The implementation of two-child policy has great significance.It can rationalize the population structure,diversify the family model,easethe pension problem significantly,and rationalize parent-child relationships.At the same time,there exist ethical problems in some aspects of society,such as the attribution of female reproductive fights,the anxiety the only child suffered from the second child,the continuing imbalance of gender,the occupational predicament of female reproduction,the effects on women health and family economy.Therefore,the country and society should respect and maintain women's reproductive rights,guarantee women's occupational stability,pay attention to the education of family ethics,promote the traditional values of respecting the old and cherishing the young,and develop the social old-age security system,thus to ensure the effective implementation of the two-child policy and build a harmonious family and society.
10.Compared with conventional electrophysiological mapping of radiofrequency catheter ablation of the prema-ture ventricular contractions in the ventricular outflow tract

Objective To assess the efficacy of radiofrequency catheter ablation ( RFCA) in premature ven-tricular contractions ( PVCs) by electrophysiological mapping.Methods 60 patients with symptomatic PVCs original from the right ventricular outflow tract underwent RFCA guided by pace mapping and activation mapping,ablation was performed by a catheter with temperature control.The patients were divided into 3 groups,20 cases in each group.The first group ( the combination group ) was the combination with pacing mapping and activation mapping underwent RFCA.The second group ( the activation mapping group) was guided by activation mapping underwent RFCA.The third group( the pacing mapping group) was guided by pacing mapping underwent RFCA.Successful ablation immedi-ate end was that PVCs disappeared and could not be induced by isoproterenol.The success rate,time and composition ratio of the three groups were observed.Results The immediate success rate of ablation of either the combination group or the activation mapping group was higher than that of the pacing mapping group.But the ablation time of the combination group was the shortest.There was no statistical difference between the baseline results of each test group. The cardiac arrhythmia originated from the septum was usual (45/60,75.0%),and the arrhythmia originated from the free wall (15/60,25.0%) was less.The success rate of PVCs ablation of right ventricle was high,and the total success rate was 91.7%(55/60).There was no significant difference in the success rate of the combined mapping and the activation mapping (χ2 =0.084,P>0.05),but were significantly higher than those in the pacing mapping group (χ2 =0.032,0.047,all P<0.05).The ablation time of the pacing mapping group and the combination group were significantly lower than that of the activation mapping group(all P<0.05).Conclusion The effect of the com-bination with pacing mapping and activation mapping is the best.Pacing mapping is better than activation mapping during RFCA of PVCs.
