1.Optimization of the urine trace albumin and glycosylated hemoglobin measurements by response surface methodology

Objective To optimize the measurements of glycosylated hemoglobin and urine trace albumin based on the response surface methodology .Methods 60 cases of patients with diabetes were enrolled .All the patients were collected 3 mL fasting blood and 6 mL urine in the morning .The simple factors experiment was taken in different maintain temperatures and times .Based on the Box-Behnken design principles ,the temperatures and times were as the factors influencing .The response surface methodology was proceeding centered on the condition that the maintain temperature was 4 ℃ and the maintain time was 3 d .Results The best tem-perature for save urine and blood was 4 ℃ and the best time was 1 d .In this condition ,the measured value of glycosylated hemoglo-bin and urine trace albumin were mostly closed to the ideal value .Conclusion Using the response surface methodology ,the measure condition of glycosylated hemoglobin and urine trace albumin can be optimized .
2.Effect of Jiedu Quyu Formula for Psoriasis Vulgaris and Its Influence on Plasma Endothelin

Objective To study the therapeutic effect of Jiedu Quyu Formula (Formula for Removing Toxins and Blood Stasis) on psoriasis vulgaris of progressive stage and its mechanism. Methods The 70 patients of progressive psoriasis vulgaris were randomized into treatment group (35 cases), treated by Jiedu Quyu Formula, and control group (35 cases), treated by Compound Qingdai Capsule. Healthy control group was of 30 healthy subjects. The contents of plasma endothelin (ET) were determined before and after treatment respectively, and the therapeutic effect of both treated groups were observed. Objective To determine the concents of palmatine hydrochloride and berberine hydrochloride in the preparation of Fu Yan Xiao Lotion(FYXL).Methods The HPLC method was established for the determination of palmatine hydrochloride and berberine hydrochloride in the preparation of FYXL.The chromatographic column is Agilent Extend C18(4.6mm? 150 mm,5 ? m),mobile phase consists of water-acetonitrile(40 ∶ 60,3.5 g potassium dihydrogen phosphate and 1.5 g sodium dodecyl sufate per 1 000 mL),column tempreture was 30 ℃,and the flow rate was 1.0 mL/min.The detection wavelength was 345 nm.Results A good linearity is obtained in the range of 0.062 5 ? g to 1.250 ? g of berberine hydrochloride,and 0.068 6~ 1.372 ? g of palmatine hydrochloride.The average recovery of berberine was 98.63 %,RSD being 2.01 %.The average recovery of palmatine was 102.34 %,RSD being 1.67 %.Conclusion This method is simple and quick,and can be used for the quality control of FYXL.
4.Monitoring and nursing care of toxicity reactions in 32 patients treated with cetuximab combined chemotherapy

This paper introduce the observation and nursing care of toxicity reactions in thirty-two advanced cancer patients treated with cetuximab combined chemotherapy. The nursing measures focused on pre-treatment psychological nursing,strictly abiding by the cetuximab combined chemotherapy protocols,as well as close monitoring and nursing interventions of toxicity re-actions. All the thirty-two patients accomplished the cetuximab combined chemotherapy. During the drug therapy,two patients suf-fered from allergic reaction and three patients with grade Ⅲ skin rash. All the toxicity reactions were alleviated after treatment.
5.Ultrasound densitometry analysis of human carotid plaque components

Objective To correlate the ultrasound imaging of atherosclerotic carotid in vivo and in vitro with the histological analysis, and to evaluate the usefulness of gray scale ultrasound densitometry analysis of ultrasound images. Methods Twenty-two carotid plaque lesions of 20 patients were examined by high resolution B-mode ultrasound pre- and post operatively, and the images of carotid plaque were quantitatively analyzed by 256 gray scale ultrasound densitometry. The transverse pathological sections of the plaques were made, and matched and compared with the images of ultrasound. Results Ultrasound density measurement of calcium, fibrous tissue, hemorrhage / thrombus and lipid deposits was 89?12, 53?8, 37?6, and 39?3 respectively in vivo, and was 168?11, 136?12, 85?12 and 89?10 respectively in vitro. Besides the comparison between hemorrhage and lipid deposits, there were significant statistic differences of density measurement between different components. Conclusions Gray scale ultrasound densitometry analysis can provide quantitative measurement of atherosclerotic carotid plaque, and is useful for evaluate the stability of the plaque.

ABRO1 (Abraxas Brother 1), known also as KIAA0157 or FAM175B, is an important component of BRISC deubiquitinating enzyme ( DUB) complex which specifically shears K-63 linked polyubiquitin chains .This article reviews the research progress in ABRO 1 in terms of its structural characteristics , BRISC complex composition , the regulation of DUB activity and interferon response , oxidative stress response and anti-myocardial ischemia and provides new ideas on fur-ther study of the physiological function of ABRO 1 and its association with immune and cardiovascular diseases .
8.The Activity Study of Aminodeoxychorismate Synthase of Differernt Corynebacterium glutamicum

The two pabAB genes encoding aminodeoxychorismate synthase(ADC synthase)from a L-serine producing strain Corynebacterium glutamicum SYPS-062 and model strain Corynebacterium glutamicum ATCC 13032 were ampilified by PCR.The result of nucleotide sequence analysis showed that both pabAB fragments were 1863bp,encoding 620 amino acids.16 bases differences that resulted in the changes of 7 amino acids were found in the pabAB of SYPS-062.The two pabAB were inserted into pET-28a to yield the recombinant expression vector pET-28a-pabAB and then transfromed into BL21(DE3).Upon IPTG induction,soluble ADC synthase was over-produced by E.coli BL21(DE3)harboring the expression construct.Recombinant ADC synthase purified by Ni-NTA affinity chromatography showed a single band about 67kDa on SDS-PAGE gel,and activity of aminodeoxychorismate synthase analysis show that the enzyme specific activity of SYPS-062 is 46.6% lower than ATCC 13032.
9.Expression of Anti-Tumor-Associated Glycoprotein-72 Single Chain Variable Fragment in Human Breast Cancer Cells

Objective To investigate the expression of tumor associated glycoprotein-72 (TAG-72) in human breast cancer cells. Methods Two breast cancer cell lines MCF-7 and Bcap-37 were cultured and prepared. TAG-72 expressions in MCF-7 and Bcap-37 were detected immunochemically with anti-TAG-72 single chain variable fragment (scFv). Results TAG-72 was positively expressed in MCF-7 cells but negatively expressed in Bcap-37 cells. Conclusion Tumor associated antigen TAG-72 is expressed in certain human breast cancer cells, which indicate TAG-72 may be used as a tumor maker and anti-TAG-72 scFv may play a role in the diagnosis and treatment of breast cancer.
10.Construction and Expression of Hepatocellular Carcinoma-Specific Expressing Eukaryotic Vector for Anti-Angiogenesis Therapy

Objective To construct a mammalian vector encoding angiostatin kringle 5 (K5) under the control of ?-fetoprotein (AFP) enhancer and albumin promoter, and to observe the expression of angiostatin by introducting angiostatin gene into hepatocellular carcinoma cells through gene transfection. Methods Angiostatin cDNA was amplified from normal human eukaryotic cells by using RT-PCR. Meanwhile, AFP enhancer and albumin promoter sequences were directed cloned and were inserted into vector pcDNA3.1. The recombinant vector of pcDNA3.1-AFAB-angiostatin K5-His was constructed, which contained the angiostatin K5 cDNA sequence that was under the control of the AFP enhancer and promoter. Angiostatin K5 cDNA was introduced into human AFP positive hepatocellular carcinoma cell lines with the transfected cultured cells that were mediated with Lipofectamine 2000. The expression of angiostatin K5 was analyzed by Western blot and the protein was dectected with anti-His antibody. Results The 500-base pair of angiostatin K5 was in accordance with the expected sequence and the recombinant vector of pcDNA3.1-AFAB-angiostatin K5-His was also confirmed as the anticipated sequence. The expression of angiostatin K5 in AFP positive hepatocellular carcinoma cells was detected both by SDS-PAGE and Western blot. Conclusion Efficient construction and expression of angiostatin K5 to AFP positive cells make it possible for antiangiogenesis therapy of human hepatocellular carcinomas, which may provide a promising approach.
