1.Primary culture and identification of mouse PASMCs and effects of hy-poxia on proliferation and apoptosis of PASMCs

AIM:To establish a fast , accurate and economical technique for culturing mouse pulmonary arte-riolar smooth muscle cells ( PASMCs ) , and to explore the effects of hypoxia on the proliferation and apoptosis of the PASMCs.METHODS:In sterile condition, the pulmonary artery was isolated from the male BALB/c mice by digesting with collagenase I, and the cells were cultured in fetal bovine serum-coated flask.Centrifugal procedure was not used dur-ing the cell passage .The cell morphology was observed under an inverted phase-contrast microscope .α-Smooth muscle ac-tin was identified by immunocytochemistry and immunofluorescence .The effects of hypoxia on the proliferation and apopto-sis of the PASMCs were detected by CCK-8 assay and TUNEL assay .RESULTS:PASMCs were identified by the methods of immunocytochemistry , immunofluorescence staining and observation of morphology .Unlike the rat PASMCs with typical subcultured peak-vally pattern, the mouse PASMCs showed a lot different without a peak-vally pattern.The cells could be subcultured after 5 d to 7 d and there was 3 to 5 generations depending on the activity of the cells .CCK-8 assay demonstra-ted that the A values of PASMCs exposed to hypoxia increased after 24 h ( P<0.05) as compared with normoxia .TUNEL result showed that the apoptotic index of the PASMCs in hypoxia decreased after 24 h (P<0.05).CONCLUSION:This technique for obtaining cultured mouse PASMCs is simple , fast, accurate and economical .The digestion time is easy to control.Hypoxia promotes the proliferation and inhibits the apoptosis of PASMCs .
2.To investigate the immunity characteristics of a conserved sequence on the C terminal region of the human papillomavirus major protein L 1

Objective To investigate and detect the immunity characteristics of a conserved sequence including 30 amino residues which is located in the C terminal of HPVL1 .Methods The immune model of mouse was establish with a polypeptide synthetized based on this sequence .The amount of CD3+ ,CD3+CD4+ or CD3+CD8+ lymphocyte of the mouse spleen was detected by Flow cy-tometry ,then the value of CD4+ /CD8+ were calculated .The cell proliferation was detected by MTT assay .Sample was took from the cell supernatant to detect the content of IL-4 and IFN-γby double antibody sandwich ELISA .Results (1)The amount of CD3+CD8+ lymphocytes and the value of CD4+ /CD8+ were significantly increased than control group(P<0 .05) .(2)The result of T cell proliferation showed no significant difference (P>0 .05) .(3)The level of IL 4 were significantly higher than that of control group (P<0 .01) .The content of IFN-γlevel showed no significant difference compared with control group (P>0 .05) .Conclusion The results showed that it was affirmative that the polypeptides induced humoral immunity ,which was worth to be furtherly studied as a preventive vaccine .But its cell immunogenicity is weak ,and the attenmpt to induce the response related to the cell immunogenicity was failed ,and the immunogenicity of the peptide remains to be improved .
3.Design and application of performance salary management system in public hospitals based on job value

Scientific performance-based salary system is of great significance for hospitals to implement effective internal management, improve the enthusiasm of medical staff, and promote the high-quality development of public hospitals. Based on the requirements of the national salary system reform, a tertiary general hospital has been designing and implementing a performance-based salary system since 2016. The hospital has established a job evaluation index system to stratify various job sequences in the hospital, and determined the job value coefficients for different sequences and levels. It has also established a performance salary project system that covered job performance salary, specific performance salary, and innovative performance salary. In addition, the hospital has established a performance-based salary management system that covered salary standard formulation, performance salary accounting and distribution, and dynamic adjustment of the performance-based salary management system. The application of this performance-based compensation management system has achieved the matching of employee value, job hierarchy, and medical services. From 2016 to 2020, employees′ overall satisfaction with performance-based salary exceeded 85%. At the same time, the system could enhance the operational efficiency and quality of the hospital, drive technological development and scientific research innovation, playing a positive incentive role in the high-quality development of the hospital.
4.The role of hydrogen sulfide and cystathionine-gamma-lyase in allergic rhinitis guinea pigs.

OBJECTIVE:
To study the change of endogenous hydrogen sulfide (hydrogen sulfide, H2S) and its rate-limiting enzyme Cystathionine-gamma-lyase (CSE) in allergic rhinitis through guinea pigs with intervention treatment.
METHOD:
Twenty-four guinea pigs were divide into 4 groups at random, one group were models of allergic rhinitis (AR) which were established by using ovalbumin, the second group were treated with NaHS after sensitized, the third group were treated with Propargylglycine (PPG) which was suppression of CSE after sensitized, and the last group were treated with saline for control. The concentration of eotaxin of nasal lavage and H2S in plasma were recorded, and then the expression of CSE in nasal mucosa was determined by real-time fluorescence RT-PCR.
RESULT:
The concentration of eotaxin in nasal lavage of sensitized group were higher than those of control (P < 0.01), and concentration of H2S in plasma and expression of CSE in nasal mucosa were lower than control (P < 0.05). The concentration of eotaxin decreased when treated with NaHS and increased when treated with PGG (P < 0.05). Level of H2S in plasma and expression of CSE increased when treated with NaHS and decreased when treated with PGG (P < 0.05), and the level of H2S was positive linear correlate with the expression of CSE.
CONCLUSION
Endogenous H2S perhaps plays a significant role in the pathogenesis of allergic rhinitis, and it was mainly regulated by CSE.
