1.The biological effect and metabolic characteristics of hydroxytyrosol

Hydroxytyrosol ( 3 , 4-dihydroxyphenyle-thanol) presents in olive leaves and fruits with an ex-ceptional antioxidative activity. As a promising func-tional food ingredient, hydroxytyrosol shows many bio-logical activities, including preventing cardiovascular diseases, protecting retinal pigment epithelial cells, preventing osteoporosis, and improving the cognitive a-bility. According to the research progress in recent years,we have summarized the biological effect and the metabolism of hydroxytyrosol in this review.
2.Mitochondria:new intracellular pharmacological target

Mitochondria not only acts a key role in en ergy generating process within the cell, but also is intimately associated with some important even ts in cells, such as apoptosis, generation of reactive oxygen species, lipid met abolism etc. For its special structure and function, mitochondria, as the pharma cological target for many agents, interacts with drugs extensively. Learning the interaction between mitochondria and drugs will greatly contribute to the under standing of the mechanism of the drugs, the design of new agents and the avoidan ce of the drug toxicity and side effect. This review is to present the related p roperties of mitochondria and its interactions with classical mitochondrial-tar geted drugs.
3.Influence of zinc on expression of zinc transporter mRNA in Caco2 cells

BACKGROUND: Decreasing of absorption of zinc from small intestine can induce dermatitis, alopecia, growth and developmental disorder and so on. It is not very clear that how to keep homeostasis when there is low zinc concentration. The discoveries of zinc transporters and relative researches provide new study direction.OBJECTIVE: To study the effect of different zinc concentration on the expressions of divalent metal transporter 1 (DMT1) and human zinc-regulated transporter (ZRT), iron-regulated transporter (IRT)-like protein (ZIP) 4mRNA, and analyze the potential pathway of absorption of zinc from small intestine in low zinc concentration.DESIGN: Blank-controlled experiment.SETTING: Department of Military Hygiene, Navy Faculty, Second Military Medical University of Chinese PLA.MATERIALS: The experiment was accomplished in Department of Military Hygiene, Navy Faculty of Second Military Medical University of Chinese PLA between October 2004 and May 2005. Materials were human colon adenocarcinoma cells Caco2 that were purchased from Shanghai Institute of Cell of Chinese Academy of Sciences.Time-dependent effect: The expression of DMT1 and ZIP4 mRNA was detected with reverse transcription-polymerase chain reaction (RT-PCR) at 0,dependent effect: The expression of DMT1 and ZIP4 mRNA were measured after the 0, 2.5, 5, 7.5 and 10 μmol/L TPEN exposure, respectively, by RT-PCR.MAIN OUTCOME MEASURES: Time-and dose-dependent effect of zinc on the expression of DMT1 and ZIP4 mRNA in Caco2 cells.RESULTS: ①Time-dependent effect: Compared with 0 hour, the expression of DMT1 mRNA obviously increased at 6, 8 and 10 hours (P < 0.05 ), but there was no significant change at 2 and 4 hours. The expression of ZIP4 mRNA markedly increased in all timing, and ZIP4 mRNA level at the 6th hour reached the peak with the prolongation of low zinc duration,which was 2.1 times ofthat at 0 hour. ②Dose-dependent effect: The expression of DMT1 mRNA distinctly increased at the concentration of 7.5 and 10 μmol/L TPEN exposure as compared with that of the control group (P < 0.05), but there was no significant change at 2.5 and 5 μmol/L. The ZIP4 mRNA expression increased with the increasing of the concentration of TPEN, the expression of ZIP4 mRNA was 2.7 times of that at 0 μmol/L. CONCLUSION: Zinc can regulate the expression of DMT1 and ZIP4 mRNA in Caco2 cells, and there is time-and dose-dependent effect. But the mRNA expression of ZIP4 is more sensitive and prompt than DMT1. Cells can upregulate the expression of DMT1 and ZIP4 mRNA to keep the homeostasis in low zinc condition.
4.S100A7:Biological properties and implications in diseases

S100 A7 is a small calcium-binding protein that has an important role to play in the differentiation of human kerati -nocytes.It also helps build the chemical defense system of the skin and responds to oxidative stress and inflammation .The expression of S100 A7 can be induced by physical , chemical and biological stimulations .S100 A7 has many ligands and is related to the reactive oxygen species (ROS) via specific signaling pathways.As an important antimicrobial peptide, S100A7 helps the skin fight against main pathogens .S100 A7 is also a key indicator for the state of inflammation and cell differentiation;and its up-regulation has been cor-roborated in various skin diseases .In the brain of patients with Alzheimer′s disease, the up-regulation of S100A7 in primary cortico-hippocampal neurons can inhibit the generation of β-amyloid peptides .Moreover , S100 A7 is also up-regulated in different types of cancer and closely related to tumor malignancy and progression .In this review, we introduce biological properties of S 100A7 and high-light its expression characteristic in related diseases .This should have implication for the discovery of potential biomarkers and targets for the treatment of S100A7-related diseases.
5.Effects of different zinc intakes on brain and testis expression of zinc transporter 3 mRNA in weaned mice

Objective:To observe the growth and development of the weaned mice fed with different levels of dietary zinc and to explore the expression of zinc transporter 3(ZnT3) mRNA induced by different dietary zinc intakes. Methods: Twenty male weaned mice (postnatal day 21) were divided into 4 groups: zinc deficient (ZD), zinc adequate(ZA), zinc supplemental (ZS) and pair fed(PF). Mice were fed with different levels of dietary zinc for 3 weeks (from postnatal day 21 to postnatal day 42) ;the zinc contents of ZD, ZA, ZS and PF group were 1 mg/kg, 30 mg/kg, 180 mg/kg and 180 mg/kg respectively. From postnatal day 21 to postnatal day 42, the diet intakes and weight of the mice were measured everyday. On postnatal day 42, the mice were sacrificed and tissues were immediately isolated and frozen lor RNA extraction. The serum zinc concentrations were measured by AAS and the expression of ZnT3 mRNA was determined by semiquantitative RT-PCR. 6.Study of zinc element and ZnT3 mRNA expression in mouse brain

AIM:To explore the interaction and function between ZnT3 mRNA expression and zinc elemental distribution in brain slice of mouse.METHODS:Zinc distribution was determined by SRXRF and ZnT3 mRNA expression in tissue was examined by RT-PCR method.RESULTS:Zinc content in cerebral cortex and hippocampus was significantly higher than that in other positions.The highest expression of ZnT3 mRNA was observed in cerebrum,hippocampus and testis.However,the ZnT3 mRNA was not detected in heart,liver,lung,spleen,kidney,intestine,olfactory bulb and cerebellum.CONCLUSION:ZnT3 facilitates the accumulation of zinc in synaptic vesicles and may play important roles in structuring of vesicular zinc pool.
7.Clinical Observation of Heat-sensitive Moxibustion Combined with Modified Pipa Qingfei Decoction in the Treatment of Acne Vulgaris

OBJECTIVE:To observe the clinical efficacy and safety of of heat-sensitive moxibustion combined with modified Pipa qingfei decoction for acne vulgaris.METHODS:A total of 120 patients with acne vulgaris were randomly divided into control group and observation group,with 60 cases in each group.Control group was given Minocycline hydrochloride capsules 50 mg/d,bid.Observation group was given heat-sensitive moxibustion (every other day) combined with modified Pipa qingfei decoction (one dose every day,decocting to 300 mL,morning and night).Both groups were treated for 42 d.Skin lesion score and serum testosterone level were observed in 2 groups before treatment,1st and 90th day after the end of treatment.Clinical efficacies were evaluated in 2 groups on 1st and 90th day after the end of treatment,and the occurrence of ADR was observed in 2 groups during treatment.RESULTS:Before observation,there was no significant difference in these indexes.On 1st day after the end of treatment,the scores of skin lesion items and serum testosterone level in 2 groups were all decreased significantly compared to before treatment,and the scorns of skin lesion items in observation group were significantly lower than control group (P<0.05);there was no significant difference in the serum testosterone level between 2 groups (P>0.05).On 90th day after the end of treatment,the scores of skin lesion items and serum testosterone level in control group had no statistical significance compared to before treatment (P>0.05).The scores of skin lesion items and serum testosterone level in observation group were decreased significantly compared before treatment,and were significantly lower than control group at corresponding period (P<0.05).On 1st and 90th day after the end of treatment,total response rates of observation group were 85.00% and 73.33%,which were significantly higher than 73.33% and 55.00% of control group,with statistical isgnificance (P<0.05).There was no statistical significance in the incidence of ADR between 2 groups (P>0.05).CONCLUSIONS:The heat-sensitive moxibustion combined with modified Pipa qingfei decoction can significantly reduce the scores of skin lesion and serum testosterone levels,improves the symptoms of skin lesions,and shows good safety.
