1.Non-coding RNAs in castration-resistant prostate cancer.

Non-coding RNAs (ncRNAs) are a large class of RNA molecules that do not encode proteins, regulate gene expressions multifacetedly, and influence the metabolism, proliferation, differentiation and apoptosis of cells as well as the occurrence and progression of tumors. Some of the ncRNAs act as cancer genes, such as miR-19a, miR-125b, miR-616, miR-7, miR-221, MALAT-1, and PRNCR1, which are upregulated in castration-resistant prostate cancer (CRPC) tissues or cell lines, and promote the development and progression of CRPC, some act as tumor suppressor genes, including miR-185, miR-342, miR-15, miR-16, and miR-146, which are downregulated in CRPC tissues or cell lines and inhibit or delay the occurrence of CRPC, and still others, such as miR-7, miR-19a, miR-125b, miR-221, and MALAT-1, are differentially expressed in the serum or tissue and can be used as potential biomarkers for the early diagnosis and prognosis of CRPC. This article presents an overview on the roles of ncRNAs in the occurrence, progression, diagnosis, and prognosis of CRPC and advances in their studies.

2.Analysis of X-ray Findings of Diaphragm and Other Related Features in Hyperthyroid Cardiopathy

Objective To analyze X-ray findings of diaphragm and other related features in the diagnosis of hyperthyroid cardiopathy.Methods Forty cases without any other disease were taken chest plain film before therapy.The X-ray findings were analyzed.Results In all of X-ray features,low and flat diaphragm;barrel chest;increased and wide lung marking,which presented as network-like structure with mottling in bilateral inferior lung were closelv related to pathologic changes.Pneumonemia;pulmonary stasis;prominence of pulmonary artery segment;dilatation of right inferior pulmonary artery were related to othe change of heart and pulmonary circulation.Conclusion X-ray findings are closely related to the severe degree of pathologic changes in hyperthyroid cardiopathy.It is valuable for clinical diagnosis and treatment to recongnize these X-ray findings accurately.
3.Preliminary experience in the treatment of chronic voiding dysfunction by sacral nerve neuromodulation (report of 2 cases)

50%) in the objective findings and/or subjective symptoms in both patients.In case 1 and case 2,the frequency of daily urination decreased by 7.3% and 34.3%,the volume of urination increased by 118.2% and 65.7%,the degree of urgent urination declined by 72.0% and 68.1%,respectively.Then they received permanent electrode and neurostimulator implantation and the improvement remained significant.They were followed up for 26 and 17 months,respectively,and no significant complications were found. Conclusions SNN is a micro-invasive,effective,and safe therapy for chronic voiding dysfunction.It may also be effective for some neurogenic dysfunctions of the bladder and urethra.
4. Rapamycin promotes differentiation and proliferation of mouse CD4 +CD25+ regulatory T cells in vitro

Objective: To observe the influence of rapamycin on the differentiation and proliferation of CD4+ CD25+ Treg cells, so as to lay a foundation for further studying its role in inducing immune tolerance. Methods: The spleens were taken from C57BL/6 mice under sterile condition and the mononuclear cells were isolated. CD4+ T cells were isolated by immunomagnetic beads (negative) and were divide into normal control group, rapamycin (0.1 μmol/L, for 7 days) group and cyclosporine (0.5 μmol/L, for 7 days) group. Flow cytometry was used to determine the proportions of CD4 +CD25+ Treg cells, and RT-PCR was used to examine the expression of Foxp3 mRNA. Results: Compared with the control group, the proportion of CD4+CD25+ Treg cells in the CD4+ T cells was significantly decreased ([7.42±0.82]% vs [3.72±0.74]%, P<0.01) in the cyclosporine group and was significantly increased ([7.42±0.82]% vs [11.47±1.08]%, P<0.01) in the rapamycin group. Expression of Foxp3 mRNA in rapamycin group was significantly higher than that in the other 2 groups (P<0.01), expression of Foxp3 mRNA in the cyclosporine group was significantly lower than that in the control group (P<0.05). Conclusion: Rapamycin can promote the proliferation and growth of CD4 + CD25+ Foxp3+ Treg cells in vitro, and facilitate the development of immune tolerance. It has a different immune suppression mechanism with cyclosporine A.
5.Study on Extraction Process of Volatile Oil and Inclusion Preparation of Hydroxypropyl-β- cyclodextrin from CompoundJinling Sini Siwu Shixiaosan Granules

Objective To optimize the extraction process of volatile oil and inclusion preparation of hydroxypropyl-β-cyclodextrin from compoundJinling Sini Siwu Shixiaosan Granules.Methods The orthogonal approach of L9 (34) was applied to investigate the volume of volatile oil extraction by 3 factors to obtain the optimum extraction conditions. With inclusion rate of volatile oil and yield of inclusion compound as comprehensive evaluation indexes, the optimum preparation conditions were discussed. The characterization of this prepared inclusion was analyzed by TLC.Results The optimum extraction conditions were as follows:the crude medicine with medicinal powder was shattered into 20 meshes;8 times the amount of water was added and was extracted for 8 h. The best inclusion conditions were as follows:the proportion of volatile oil to HP-β-CD was 1∶6, with inclusion temperature of 30℃, grinding time of 2 h. Analysis results of TLC showed that stable inclusion compound was formed from volatile oil and HP-β-CD.Conclusion The optimized extraction process and inclusion preparation are with stable quality and high feasibility.
6.Effect of cyclophosphamide on proliferation and apoptosis of rat mesangial cells in vitro

AIM: To investigate the effect of cyclophosphamide(CTX) on proliferation and apoptosis of mesangial cells(GMC) of rat in vitro. METHODS: GMC proliferation was detected by MTT method, GMC apoptosis was examined by inverted microscopy for phase-contract and fluoroscopy and flow cytometry analysis. The levels of Fas and Bcl-2 were also detected by immunohistology. RESULTS: The proliferation of GMC were inhibited by CTX, methylprednisolone(MP), low molecular weight heparin(LMWH). Apoptosis of GMC was induced by CTX, the apoptosis rate of GMC was 8.2%, and the Fas level was increased. CONCLUSION: CTX could inhibit proliferation and induce apoptosis of GMC possibly by enhancing the Fas level.
7.Establishment of bone marrow failure mouse model and detection of FGF2 expression

Objective Previous study found that ifbroblast growth factor-2(FGF2) expression was at a low level in bone marrow of children with aplastic anemia. In this study, we established a bone marrow failure animal model to investigate whether FGF2 is involved in bone marrow failure. Methods BALB/c mice were irradiated by 5.0 Gy ray, and then infused with 1×106 lymphocytes from allogeneic mice lymph node. Peripheral blood cells and bone marrow mononuclear cells counting and bone marrow pathology were done. FGF2 protein in bone marrow mononuclear cells was measured by ELISA. Results Compared with the control group, the counting of hemoglobin, white blood cell, platelet and bone marrow mononuclear cell in aplastic anemia mouse model were signiifcantly deceased (P<0.05). Moreover, FGF2 expression in aplastic anemia mouse model were signiifcantly reduced (P<0.05). Conclusions 5.0 Gy ray irradiation and then 1×106 lymphocyte infusion in mice can induce bone marrow failure similar to the features of aplastic anemia. The low expression of FGF2 in bone marrow of aplastic anemia patients may play an important role in the pathogenesis of aplastic anemia.
8.Study on the Lactagogue Effect of Rutong Granule on Rat with Postpartum Hypogalactia

OBJECTIVE: To study the lactagogue effect of Rutong granule on rat with postpartum hypogalactia. METHODS: Female rats which had kittening behavior within 24 h were selected to induce rat model by perfusing bromocriptinum. Meanwhile, model rats were given Rutong granule via i.g. gtt. Milk yield of female rats and body weight of young rats were determined everyday from the second day. Rats were sacrificed on the 8th day. The level of PRL and histological change of breast were observed. RESULTS: Rutong granule can increase the serum PRL level gradually to promote the increase of acinar number and dilatation of gland ducts. Rutong granules can also increase the milk yield of female rats and weight of young rats. CONCLUSION: Rutong granule can promote secretion of milk by means of increase of PRL level and expediting of secretion channels.
9.Experience in 42 cases of laparoscopic splenectomy

Objective To explore the surgical skills and clinical efficacy of laparoscopic splenectomy (LS). Methods The operative duration, perioperative blood loss and postoperative complications were determined in 42 patients receiving LS in our hospital from October 2006 to May 2008 were retrospectively analyzed. Of the 42 patients, 12 suffered from splenomegaly due to blood disease, 1 from splenic tuberculosis, 1 from splenic cyst, 4 from traumatic splenic rupture,1 from malignant lymphoma and 23 from hypersplenism due to portal hypertension. Results LS was successfully performed in all the 42 patients. The mean perioperative blood loss was (300±110.60)ml, average operative duration (170±45.65) min (60-260 min) and mean postoperative inhospitalization duration (8.10±3.52)d. There was no severe complication after the operation. Conclusion For most types of splenic diseases, LS is safe and feasible. It is crucial to manage the splenic pedicle according to disease type, size, morphology and splenic hilus.
10.Clinical experiences of LC treatment for acute calculous cholecystitis(120 cases)

Objective To explore the operative main points and clinical effects of LC treatment for acute calculous cholecystitis. Methods Between August 2006 and June 2008, LC treatment for acute calculous cholecystitis in 120 cases as performed, the data of which were analyzed retrospectively. Of these patients, there were gallbladder with adherent omentum (72 cases), gallbladder with adherent hepatic flexture of the colon (9 cases), gallbladder with duodenum (5 cases), obstruction of the cystic duet by a gllstone (68 cases), and Mirizzi syndrome(2 cases). Results All the 120 cases performed LC successfully. There were no converting to open operation, and no biliary leakage. The mean operating time was (45. 0 ± 13. 1) minutes. Conclusions Acute calculous cholecystitis was not a contraindication for LC. Rich experience and skilled technique were key points for the success of operation. It shows advantages of less pain, less complications and more rapid recovery.
