1.Pax6 expression in rat whisker hair follicle

Objective To investigate the expression pattern of transcription factor,paired homebox 6(Pax6) in rat whisker hair follicle.Methods Whisker hair follicle was collected from 5 7-day-old SD rats.Immunohistochemical assay was carried out to detect Pax6 in the follicle.Pax6 mRNA expressions and its splicosome,Pax6-5a were detected by RT-PCR.Whisker hair follicle was primarily cultured to harvest rat hair follicle bulge cells.Then expressions of Pax6,CD34,?6-integrin,and Ki67 were detected in the obtained cells by immunofluorescent method.Results Pax6 was positively expressed in rat whisker hair follicle.The mRNA expression of Pax6-5a was stronger than that of Pax6.The primarily cultured cells expressed CD34 and ?6-integrin in the membrane.Pax6 was mainly located in the nuclei of the cells with Ki67,and little in the cytoplasm.The proliferative ability of the cells was correlated with the location of Pax6 protein.Conclusion Pax6 expresses in rat whisker hair follicle.Its expression patterns indicate that it may be involved in the formation and function of whisker hair follicle.
2.Changes in near vision and stereopsis after orthokeratology

AlM:To provide a proper assessment of the clinical use of orthokeratology by observing and analyzing the ocular biometric changes of the eyes and the stereopsis of the myopia.
METHODS:Sixty eyes from 30 myopia ( from 8 to 17 years old) were fitted with orthokeratology. Stereopsis, visual acuity, near visual acuity, central corneal thickness, anterior chamber depth, average anterior corneal refractive power ( K value ) , and intraocular pressure were measured before the orthokeratology treatment and 3mo after it. Refraction was expressed as spherical equivalent ( SE) , and the subjects were divided into 3 groups according to refraction: low myopia group (SE<-3.00D), moderate myopia group (-3. 00D≤SE<-6. 00D), and high myopia group (SE≥-6. 00D).
RESULTS:All subjects had significant improvements in visual acuity and near visual acuity 3mo after the orthokeratology treatment (P<0. 01). ln the comparisons of any pair of the groups, there was no significant difference in the near visual acuity ( P>0. 05 ). Three months after the orthokeratology treatment, Naked eye near stereoacuity values of all subjects were decreased (P<0. 01). There was no significant difference in the comparisons of the differences of stereopsis between any pair of the groups ( P> 0. 05 ). There was significant correlation in stereopsis and anisometropia ( Pearson coefficient r = 0. 778, P < 0. 01 ). And with greater anisometropia, the stereopsis was higher. All subjects had significantly lower K values than before 3mo after the orthokeratology treatment ( P< 0. 01 ). There was no significant difference in central corneal thickness, anterior chamber depth, and intraocular pressure (P>0. 05).
CONCLUSlON:Orthokeratology could lower K value in a short time and change the corneal curvature to correct myopia, to improve visual acuity and near visual acuity. lt also has an influence on improving stereopsis. There are no obviously changes in patients' central corneal thickness, anterior chamber depth or intraocular pressure after the orthokeratology treatment, making it a safe and effective treatment for adolescent.

6.Application of LMA in patients with acute severe asthma emergency

Objective To investigate the clinical value of laryngeal mask airway (LMA) in patients with a-cute severe asthma(ASA). Methods 32 patients with ASA treated with LIMA or mouth-nose mask during 2002 -2009 in our hospital were retrospectively analyzed. Those treated with laryngeal mask airway was taken as observation group and those with Mouth-nose mask as control group. Results The period to oxygen saturation in arterial blood, the time to remove ventilator, and the time to disease improvement in the observation group (389.63±32.82)s, (19.31±2.26) hours,(16.22±3.85) hours were different from that in control group (467.36±41.15) s, (25.18±3.73) hours,(23.66±2.38) hours (P<0.01). After non-invasive positive pressure ventilation, PaCO_2 decreased, PaO_2 and pH increased at 3 and 12 hours in the observation group (P<0.05 or 0.01) from that before treatment. PaCO_2 and pH at 3 hours in the control group were no significant difference before and after treatment (P > 0.05),with an exception of PaO_2 (P < 0.05). PaCO_2, PaO_2 and pH were significantly different (P < 0.05) at 12 hours after treatment from those before treatment. Conclusions LMA should be considered in the selection of non-invasive positive-pressure ventilation (NIPPV) in patients with ASA, for a better improvement of ventilation ef-fectivenoss and accelerating the mitigation of clinical manifestations.
7.Optimization of Lipase Production Conditions by Geotrichum candidum Y162 Using Single Factor-response Surface Methodology

The fermentation conditions of lipase production by Geotrichum candidum Y162 were optimized. Initially, the most suitable carbon olive oil, nitrogen source soybean flour and NH4Cl, salt BaCl2 and MgCl2 were selected according to single factorial experiments respectively. Based on the result, screening methodology Plackett-Burman design was used to evaluate the effects of twelve factors related to lipase production and three statistically significant factors olive oil, BaCl2 and NH4Cl were selected. The path of steepest ascent was used to approach the optimal region of lipase production subsequently. Then, the optimal combined concentration for maximum enzyme activity were further optimized by response surface methodology and determined as follows: olive oil 2.35%, BaCl2 0.36%,and NH4Cl 4.69%.The optimization of culture conditions of G.candidum Y162 led to a 2.25-fold increase in lipase production relative to initial result 14.16 U/ml, which indicate that single factor in combination with response surface methodology is an effective method for optimization of lipase production conditions by G.candidum Y162.
8.Optimization of Fermentation Conditions of Lipase Produced by Pseudomonas cepacia PCL-3

The fermentation conditions of alkaline lipase producing by Pseudomonas cepacia PCL-3 were optimized.Based on the analysis of single factorial experiments,dextrin was the most suitable carbon source,peptone and urea were the suitable compound nitrogen sources among the examined materials.Three significant factors(urea,inoculum and initial pH) were selected from the eight factors related to lipase production by Plaekett-Burman method,and were further optimized with response surface analysis.And then,steepest ascent procedures were applied to define the optimal response region of the three factors.The obtained optimal conditions were urea 0.15%,inoculum 3.05% and initial pH 8.38,under which conditions,the enzyme activity was improved from 25.37 U/ml to 48.88 U/ml,enhanced 1.93 folds.Starting from the flask conditions,the highest lipase activity of 47.69U/ml was achieved by batch fermentation in a 10 L fermentor after 52 h of the cultivation.
9.Use of a saline-coupled bipolar sealer (Aquamantys(R)) in liver resection for primary hepatic malignancy: a retrospective cohort study

Objective To evaluate the efficacy and outcomes with the use of a saline-coupled bipolar sealer during liver resection.Methods 101 patients underwent liver resection for primary hepatic carcinoma at the People's Hospital Affiliated to Zhengzhou University from August 2015 to December 2016.The patients were divided into two groups according to whether the Aquamantys(R) system was used or not.In group A (n =62) the clamp crushing technique was used for liver parenchymal transection.In group B (n =39) the Aquamantys(R) system was used.The intraoperative and postoperative complication rates were compared.Results The operation time in group B was significantly longer than group A (216.4 min vs.253.5 min,P < 0.05).The intraoperaitve blood loss in group B was significantly less than group A (381.1 ml vs.257.2 ml,P < 0.05),and less blood transfusion was required (211.3 ml vs.90.9 ml,P < 0.05).The volume of abdominal drainage fluid in group B in the first and the 5th day was significantly less than group A (242.6 ml vs.199.2 ml,P<0.05;84.3 ml vs.70.4 ml,P<0.05,respectively).The drainage tube in group B was taken off earlier than in group A (8.1 d vs.7.0 d,P < 0.05).The average hospitalization stay after surgery in group B was also significantly shorter (13.4 d vs.11.6 d,P < 0.05).There was no significant difference in the overall postoperative complication rate (P > 0.05) between the 2 groups,and no death was observed.Conclusion The use of a saline-coupled bipolar sealer (Aquamantys(R)) in liver resection for hepatocellular carcinoma was associated with less intraoperative blood loss and was better for the patients' postoperative recovery.
10.Comparison of early clinical effects between Activ C cervical disc replacement and anterior cervical discectomy and fusion for single-level cervical spondylosis.

OBJECTIVETo compare the early clinical effects of Activ C cervical disc replacement (ACDR) and anterior cervical discectomy and fusion (ACDF) in treating single-level cervical spondylosis.
METHODSThe clinical data of 76 patients with single-level cervical spondylosis underwent surgery from July 2009 to September 2012 were retrospectively analyzed. Among them, 28 patients were treated with ACDR (ACDR group), including 18 males and 10 females, aged from 32 to 62 years old with an average of (45.2±6.2) years; and 48 patients were treated with ACDF (ACDF group), including 28 males and 20 females, aged from 33 to 60 years old with an average of (45.8±6.4) years. Visual analogue scale (VAS), Japanese Orthopedics Association (JOA) score, Short Form-36 (SF-36), imaging data were used to assess the clinical effects after operation.
RESULTSA total of 76 patients were followed up from 6 to 24 months with an average of 13.2 months. VAS of neck pain and brachialgia were improved in all patients after operation (P<0.05), there was no significant difference between two group (P>0.05). Somato-score and psycho-score of SF-36 of two groups were obviously increased (P<0.05), ACDR group was better than that of ACDF group (P<0.05). In ACDR group, there was no significant difference in the range of motion of surgical segments and adjacent segments between preoperative and postoperative (P>0.05); heterotopic ossification around the edge of vertebral body occurred in 1 case on the 6th month after operation, no fusion was found on the 1st year after operation. In ACDF group, the adjacent vertebral disease occurred in 1 case and the patient underwent the reoperation.
CONCLUSIONActiv C cervical disc replacement can reduce the degeneration of adjacent segments and its early outcomes for the treatment of single-level cervical spondylosis are satisfactory, but the long-term effects still need study.
