1.Endoscopic treatment of septated chronic subdural hematoma With a report of 45 cases.

Objective To evaluate the therapeutic results of neuroendoscopy for the treatmentof of patients with septated Chronic Subdural Hematoma(SCHS). Methods 45 patients with SCSH were operated on under endoscope through a burrhole approach.neomembranes were resected by small microscissors.A closed drainage system was applied temporarily to ensure the efflux of the remaining hematoma. Results Forty-five cases were treated successfully.All patients had a sufficient or complete hematoma evacuation whithout recurrence of subdural hematoma.There was no subdural infection,bleeding,brainedema and cranial aerocele. Conclusions Neuroendoscopy is minimally invasive technique and have the advantages ot simple,effective,safe,less complication,less expensive and shorter hospitalization in the treatment of SCSH.

3.Virtual Screening ofα-glycosidase Inhibitors in Cortex mori and Folium mori

OBJECTIVE:To virtually screen potential α-glycosidase inhibitor ingredients from C. mori and F. mori,and to pro-vide reference for finding out new typeα-glycosidase inhibitor ingredient. METHODS:Surflex-Dock module of Sybyl-x 2.0 molecu-lar simulation software was used to perform the docking of small molecule compound,which was from the ingredients of C. mori and F. mori as ligand stated in literatures,with α-glycosidase. Total score of affinity scoring function was equal to 7 as the thresh-old value,to judge potential α-glycosidase inhibitor ingredient in C. mori and F. mori. RESULTS:After 70 small molecule com-pounds docked with α-glycosidase, 10 compounds showed binding activity (Total score≥7.00). Among them, moracin M-3′-O-β-D-glucopyranoside,5,7,2′-trihydroxyflavanone-4′-O-β-D-glucoside,mulberroside A,resveratrol-4,3′-di-O-β-D-gluco-pyranoside and 1,4-dideoxy-1,4-imino-(2-O-β-D-glucopyranosyl)-D-arabinitol had higher binding activity with α-glycosidase(Total score>8.00). CONCLUSIONS:Multi-constituents of C. mori and F. Mori show potential α-glycosidase inhibitory activity. The method is a kind of highly targeted,rapid and efficient approach to discover α-glycosidase inhibitor from traditional Chinese medi-cine.
4.Identification of Original Plants of Guanzhong (Dryopteris Species) by Chloroplast psbA-trnH Intergenic Region

This article was aimed to study the species identification capability of psbA-trnH among Guanzhong herbs from Dryopteri. The nucleotide sequence information of psbA-trnH region was abstracted using standardized manners from 9 Dryopteri species (19 samples). The identification efficiency of psbA-trnH was analyzed based on TaxonGap among both tested materials (9 species, 19 samples) and tested materials plus GenBank data (17 species, 44 samples in total). The results showed that with the expanded species range, the discriminative efficiency of psbA-trnH de-clined from 100% (9/9) to 82.4% (14/17), while the proportion of within-specific heterogeneity larger than between-specific separability increased from 11.1% (1/9) to 52.9% (9/17). It was concluded that psbA-trnH can be recom-mended as the valuable barcode for homonym distinguishment among Guanzhong herbs, with attention of adequate sampling within species.
5.Clinical study on thirty-eight cases of percutaneous microwave ablation in the treatment of hypersplenism

Objective To investigate the efficacy and safety of percutaneous microwave ablation in the treatment of hypersplenism.Methods From March 2007 to May 2011,38 patients with hepatitis B virus caused liver cirrhosis and complicated with hypersplenism received percutaneous microwave ablation treatment for several times.Before percutaneous microwave ablation treatment,the volume of spleen was calculated according to 3D computed tomography (CT) scan.Ultrasound,blood routine,urine routine,liver function test,kidney function test,serum amylase and lipase were also tested.For the first time,1/3 volume of spleen was ablated.If no complication were observed in one week after ablation,then another 1/3 volume of spleen was ablated.Blood routine and liver function test were checked on the 1st,3rd and 5th day after microwave ablation.Blood routine,liver function test and ultrasound were examined on 7th and 14th day after microwave ablation.On the 30th day CT examination was conducted.Ultrasound,blood routine,urine routine,liver function test,kidney function test,serum amylase and lipase were detected at one month,three month and every three month after microwave ablation.The follow up duration was over two years.The t-test was performed for clinical data comparison.Results The mean ratio of ablated spleen was (47 ±5)% (range 41% to 57%).Preoperative white blood cell count was (2.46±0.78)× 109/L,which gradually increased after operation and peaked on the 3rd day after operation ((5.34 ± 2.10) × 109/L).Then gradually decreased,which was (3.16 ± 1.02) × 109/L at 24 month and the difference was statistically significant compared with that of preoperation (t=-3.349,P<0.01).Preoperative platelet count was (46.58 ± 17.30) × 109/L,which gradually decreased after operation and was lowest on the 3rd day after operation.Then gradually increased,which peaked at 30 days after operation ((101.79 ± 25.80) × 109/L) and then gradually decreased,which was (61.97 ± 15.09) × 109/L at 24 month and the difference was statistically significant compared with that of preoperation (t=-4.135,P<0.01).The inner diameter of portal vein was (14.66±0.88) mm preoperation,which was (13.22±0.64) mm at three month after operation and the difference was statistically significant compared with that of preoperation (t=8.145,P<0.01).It was (14.64±0.81) mm at six month after operation and the difference was not statistically significant compared with that of preoperation (P> 0.05).The major adverse effects were fever,left upper abdominal pain,left shoulder pain,pleural effusion,intraperitoneal hemorrhage and temporary hemoglobinuria which all recovered after symptomatic treatment.No severe complication such as uncontrollable bleeding,splenic abscess,spleen rupture and the surrounding organ injury and treatment related death were observed.Conclusion Percutaneous microwave ablation for several times could safely destroy suitable volume of spleen,increase platelet and white blood cell count,improve portal hypertension and with rare complications,which might be a minimally invasive techniques with clinical application value in the treatment of the hypersplenism.
6.Platelet α and β secretase activities in mild cognitive impairment and Alzheimer's disease

Objective To investigate platelet α and β secretase activities and the amounts of platelet soluble fragment of APP (sAPPα) produced by α-secretase in patients with mild cognitive impairment (MCI) and Alzheimer's disease (AD).Methods The neurological functions of 48 nondemented patients,42 MCI and 40 AD patients were evaluated by neuropsychological examinations.The platelet α and β secretase activities and sAPPα production in each group were measured by fluorescence and Western blotting analysis respectively.Results The α secretase activities in non-demented,MCI and AD group were 100.0% ± 10.6%,78.2% ± 9.4% and 61.8% ± 7.2% respectively.As compared with nondemented group,the α secretase activities in MCI and AD group were decreased (F =22.935,P =0.001).The α secretase activity in AD group was significantly lower than MCI group.The β secretase activities in non-demented,MCI and AD group were 100.0% ± 11.2%,145.8% ± 12.7% and 189.8% ± 14.2%respectively.The β secretase activities in MCI and AD group were significantly higher than that in nondemented group (F =16.368,P =0.001).The β secretase activity in AD group was significantly decreased as compared with MCI group.The sAPPα amounts in MCI group and AD group were all decreased as compared with that in control group; the sAPPo amount in AD patients was significantly decreased as compared with that in MCI group.Conclusions The platelet α secretase activity and its production sAPPα in MCI and AD patients are decreased,while β secretase activity is increased,as compared with that in control group; the altered α and β secretase activities may participate in the pathogenesis of MCI and AD patients and may have diagnostic potential for them.
7.Screening lentiviral vectors carrying effective siRNA of the ROCK2 gene.

OBJECTIVETo screen the lentiviral vector carrying siRNA and capable of significantly suppressing the ROCK2 gene expression in the corpus cavernosum smooth muscle cells of spontaneously hypertensive rats (SHR).
METHODSWe designed and synthesized 4 siRNA fragments targeting the ROCK2 gene and packaged them into lentiviral vectors. We collected corpus cavernosum smooth muscle cell samples from 5 male SHRs and randomly divided them into groups A (non-transfection control), B (GFP lentiviral transfection), C, D, E, and F (lentiviral transfection with siRNA fragments 1 -4 targeting the ROCK2 gene). Each group consisted of 5 samples and each sample 3 x 10(4) cells. At 48 hours after transfecting MOI = 80 into the SHR corpus cavernosum smooth muscle cells, we detected the expression of GFP under the fluorescent microscope and the mRNA expression of the ROCK2 gene by RT-PCR.
RESULTSThe transfection efficiency of the SHR corpus cavernosum smooth muscle cells was > 50%. Compared with group A, the expression of ROCK2 mRNA in the corpus cavernosum smooth muscle cells showed no remarkable change in group B (P > 0.05) but was inhibited very significantly in C ([43.91 +/- 8.19]%), D ([47.15 +/- 6.64]%), and F ([25.7 +/- 6.03]%) (P < 0.01), and significantly in E ([16.81 +/- 5.94]%, P < 0.05).
CONCLUSIONWe successfully constructed 4 lentiviral vectors carrying siRNA targeting the ROCK2 gene, all of which can significantly suppress the ROCK2 expression in the SHR corpus cavernosum smooth muscle cells, and one has a highly strong inhibitory effect.

8.In vivo high-resolution magnetic resonance imaging in severe intracranial stenosis

Objective To assess the feasibility and clinical value of high-resolution magnetic resonance imaging (HRMRI) in patients with symptomatic severe intracranial stenosis (SSIS).Methods HRMRl wasperformed with a 3.0 T MR scanner on 5 patients with symptomatic middle cerebral(n=3) or basilar (n=2) arterial stenosis of≥70% confirmed bv DSA.Image diagnosis Was made on the basis of HRMRI findings of vessel wall at the stenotic segment by 2 neuroradiologists blinded t0 patient's status.Results Three of the five patients were diagnosed to have advanced intracranial atherosclerosis based on the presence of a complex eccentric atherosclerotic plaque containing a large lipid-rich necrotic core with a heterogeneous post-contrast enhancement and with signs of ruptured fibrous cap.Two other patients were likely to suffer from non-atherosclerotic lesion.HRMRI revealed an iso-signal septum in the arterial lumen attaching to the slighfly thickened arterial wall that was iso-signal with a homogeneous post-contrast enhancement in one patient and an obviously concentrically thickened arterial wall with hypo-intense signal on T1 WI and slightly high signal on T2 WI and PDWI and without any post-contrast enhancement in the remaining patient.Conclusions In vivo HRMRI in patients with SSIS is technically feasible.It provides detailed information of intracranial arterial wall at the stenotic segment.
9.The Progress of Recombinant Escherichia coli for Production of Succinic Acid

Succinic acid is regarded as C4 platform chemical, which has a huge potential market with wide applications in biopolymer, food, medicine, and so on. Production of succinic acid by microorganism fer-mentation has attracted much attention in recent times. By fermentation, succinic acid could be produced from renewable resources in place of petroleum with low cost and low pollution; especially CO2 could be fixed by microorganism to form the product, which is a promising way to deal with the green gas. Some E. coli have been chosen as good succinic acid producing strains because of their clear genetic backgrounds, convenience to be modified and controlled, and good growth properties with low nutrients requirements. The progress of genetic engineering strategy and metabolic engineering technology for construction of succinic aicd producing recombinant E. coli has been introduced in this paper with the discussion of the future re-search in this area.
10.Three-dimensional EH composite artificial template for repairing cranial defect in 53 cases

OBJECTIVE: To observe effects of three-dimensional EH composite template on repairing cranial bone defect. METHODS: A total of 53 inpatients were analyzed retrospectively at the Department of Neurosurgery, Taixing People's Hospital from July 2004 to May 2007, comprising 31 males and 22 females, aged 19-68 years. They were in accordance with cranial repairing indication. Reasons for skull defect: traumatic brain injury (41 cases), hypertension (7 cases), intracranial cancer (3 cases), intracranial aneurysm (2 cases). Thirteen patients developed bilateral cranial defect. The defect region: frontal (5 slices, comprising 2 with severe orbital part defect), temporal (9 slices), frontotemporal (42 slices), temporoparietal (3 slices), parietoocipital (4 slices),frontoparietal (3 slices). The smallest single-chip defect size was 4 cm × 6 cm, and the largest one was 12 cm×18 cm. All the patients were mended by three-dimensional EH composite template using spiral CT (≤5 mm thickness) scanning, CAD three dimensional reconstruction, quickly modeling technique. Infection of incisional wound, rejection and complications were observed. RESULTS: A total of 53 patients were followed up, with an average follow-up period of 18 months, and all of them were satisfied with the shapes (100%). Mild collection of the fluid under the scalp was found in 2 cases 4 days and 1 week following surgery, and disappeared after suction and pressure dressing. No other frequent complications were observed after cranioplasty. CONCLUSION: Three-dimensional EH composite template is an ideal material for cranial bone defect, because of its good biocompatibility, easy operation, good postoperative shape and less complications.
