A novel method was developed for the direct analysis of testosterone, androstenedione, methyltestosterone and methenolone in serum samples by fully automated online turbulent flow solid phase extraction coupled with high performance liquid chromatography-tandem mass spectrometry. An aliquot of 50 μL serum sample was preconcentrated directly on a Turboflow SPE column after centrifugation. Turboflow SPE C18-P could be used to remove serum matrix effectively. The optimum loading flow rate and elution time were 4. 0 mL/min and 1. 0 min, respectively. The linearity ranges were from 1. 0 μg/L to 100. 0 μg/L for four target compounds. The method limits of detection ( LODs) were in the range of 0. 2-0. 3 μg/L. The relative standard deviations (RSDs) ranged from 2. 9% to 14. 1% (n=5). The time for one sample analysis including extraction, separation and determination was 32 min. The proposed method has been successfully applied for the analysis of serum samples.

Objective To explore the generalization effect of a semantic navigation strategy(SNS)on naming in patients with aphasia. Methods Fourteen patients with aphasia were randomly divided into an SNS group (n =7)and an unrelated semantic(US)group(n =7).One hundred and twenty-two words chosen from Chinese word association norms were used in this study. The presentation sequence of words was generated automatically with network analysis techniques for the SNS group and randomly without network analysis techniques for the US group. After baseline tests using the western aphasia battery(WAB)and the mini-mental status examination(MMSE),all patients received language training once a day for 10 consecutive days. The amount of correct responses was recorded every day in order to draw the learning curve. The generalization effect was evaluated with 46 untrained items before and after training with all patients. Results The average learning curve of the SNS group was steeper than that of the US group.Correct responses on trained items were significantly more frequent in the SNS group than in the US group after the 6th day of training. After 10 days the frequency of correct responses on untrained items in the SNS group was significantly higher than it had been before training, and higher than the frequency after training in the US group. The frequency of no response on untrained items in the SNS group had declined, and it had declined more than in the US group. Conclusion A semantic navigation strategy may promote significant generalization while improving the learning curve in naming among patients with aphasia.

Objective To evaluate the efficacy of percutaneous transluminal septal myocardial ablation (PTSMA) and septal myectomy (SM) for treatment of hypertrophic obstructive cardiomyopathy. Methods We searched all clinical controlled trials of PTSMA and SM in treatment of hypertrophic obstructive cardiomyopathy in Cochrane Library,PubMed,Web of Science,Wanfang Database and CNKI update to August,2012.The selected studies in Chinese and English were used for Meta-analysis with software RevMan 5.0.Results Five studies including 286 cases (PTSMA:149,SM:137) were included in the study.Left ventricular outflow tract pressure was significantly reduced by SM treatment compared with PTSMA (OR =9.07,95% CI:0.96-17.18,P =0.030).There were no significant differences in interventricular septum thickness (OR =1.15,95% CI:-0.79-3.10,P =0.240),left atrial diameter (OR =1.95,95% CI:-0.16-4.07,P =0.070),left ventricular end-diastolic (OR =0.70,95% CI:-0.79-2.19,P =0.360),end systolic diameter (OR =1.14,95 % CI:-1.65-3.93,P =0.420),degree of symptom improvement (chest pain:OR =1.07,95 % CI:0.35-3.23,P =0.910;syncope:OR =0.32,95% CI:0.08-1.23,P =0.100),and postoperative mortality (OR =1.63,95%CI:0.46-5.76,P =0.450) between PTSMA and SM treatment; but the rates of pacemaker implantation (OR =5.91,95% CI:2.04-17.11,P =0.001) and reintervention treatment (OR =6.62,95%CI:1.20-36.62,P=0.030) in patients with PTMSA were significantly higher than those with SM.Conclusions There is no evidence to indicate that PTSMA can completely replace SM,as an alternative choice the therapy can be used according to left ventricular out flow tract pressure, symptoms, anatomy of target vessel and other conditions.

Idiopathic pulmonary fibrosis(IPF), with unknown pathogeny, is an interstitial lung disease. The pathological features are diffuse epithelial-cell lesion, fibroblast proliferation, myofibroblast differentiation and excessive extracellular matrix deposition.CXCR4 is the predominant chemokine receptor on fibrocytes;CXCL12 is the only ligand of CXCR4.A large number of studies have shown that CXCL12/CXCR4 biological axis plays an important role in the pathogenesis of idiopathic pulmonary fibrosis.Under the regulation of hypoxia, HIF-1α and PI3K-Akt-mTOR path, CXCL12/CXCR4 biological axis promotes lung fibroblast proliferation, myofibroblast differentiation and extracellular matrix deposition, resulting in development and progression of IPF.

BACKGROUND: Nano-hydroxyapatite has been widely used as reconstructive and prosthetic material for osseous tissue, owing to its excellent biocompatibility and bioactivity. But, its brittleness is one of the most serious obstacles for its wider applications as load-bearing implants. Therefore, hydroxyapatite/chitosan composites have been widely studied as bone substitute, due to their excellent biocompatibUity and suitable mechanical properties.OBJECTIVE: To review various preparation methods of hydroxya patite/chitosan composites and the development trend in this field.METHODS: Articles were searched in Medline Database (1995-01/2009-03), VIP Database (1995-01/2009-03) and "qinghuatongfang" Database (1995-01/2009-03) with the keywords of "nano-hydroxyapatite, chitosan, preparation, development trend".RESULTS AND CONCLUSION: Total 2 034 related articles were collected, including 1 634 in Chinese and 670 in English. Following excluding earilier published, duplicate, and similar studies, we brought into 37 sub-standard literatures. Hydroxyapatite/chitosan composite in nanometer size, with similar structure as natural bone, has better biological function compared with corresponding micrometer composite. Simultaneously, optimized material composition, structure and technology will obtain more suitable bone substitute in mechanical property compared with natural bone. This paper reviews preparation methods at home and abroad in recent years, with the rapid development of biological materials, hydroxyapatite compositematerials have been wisely used in bone tissue repair and replacement surgery, but its poor mechanical properties limits its clinical application.

Aim To study the inhibitory effects and its mechanisms of oridonin on human pancreas adenocarcinoma SW1990 cells. Methods Cell growth inhibition mediated by oridonin on SW1990cells was measured by MTT assay. The morphological changes were observed by Hoechst33258 fluorochrome staining and electron microscope.Cell cycle and apoptosis rate were analyzed by flow cytometry. The molecular mechanisms involved in the effects of oridonin on SW1990 cells were studied by RT-PCR.Results The growth of humen pancreas adenocarcinoma SW1990 cells was significantly inhibited by oridonin.Apoptosis morphological changes about chromatic agglutination and nuclear condensation were detected by Hoechst 33258 fluorochrome staining and electron microscope in oridonin treated SW1990 cells."Sub-G_1" phase peak and G_2/M growth arrest werer found with flow cytometry. The upregulating mRNA expression of p21 and downregulating mRNA expression of survivin were detected by RT-PCR.Conclusion The inhibitory effect of oridonin on human pancreas adenocarcinoma SW1990 cells through induced apoptosis and G_2/M growth arrest and the mechanisms may be through surviving-p21 co-regluation pathway.
