1.Study on Quality Standard of Jingwu Oral Liquid

OBJECTIVE:To improve the quality standard for Jingwu oral liquid.METHODS:The qualitation of Radix Polygoni Multiflori preparata and solomonseal rhizome in Jingwu oral liquid were identified by TLC and the assaying of 2,3,5,4’—tetra—hydroxyl diphenyl ethylene—2—O—?—D—glucoside in the oral liquid were determined by HPLC.RESULTS:In the TLCs of Radix Polygoni Multiflori preparata and solomonseal rhizome,the color of speckles was the same as that of the control articles in the corresponding places.The linear range of 2,3,5,4’—tetra—hydroxyl diphenyl ethylene—2—O—?—D—glucoside was 0.0 562?g~0.6 744?g(r=0.9 996);The average recovery was 100.1%(RSD=1.89%).CONCLUSION:The method is simple,fast,and reproducible and of high recovery rate,which can be used as quality control for Jingwu oral liquid.
3.Observation of the effect of card other tromethamine for postpartum hemorrhage

Objective To observe the clinical effect of card other tromethamine for postpartum hemorrhage. Methods 80 cases with postpartum hemorrhage were randomly divided into the observation group and control group, 40 cases in each group.The control group was given oxytocin treatment.While the observation group was given oxyto-cin and card other tromethamine treatment.The clinical curative effect,bleeding time after treatment,postpartum 2h haemorrhage amount and adverse reaction,etc.were observed and compared.Results The total effective rate of the observation group was 95.0%,which was significantly higher than 75.0% of the control group,the difference was sta-tistically significant(χ2 =15.68,P <0.05).The haemorrhage amount of postpartum 2h of the observation group was (273.7 ±10.5)mL,which was significantly lower than (547.1 ±16.4)mL of the control group,the difference was statistically significant(t =88.79,P <0.05).The bleeding time of the observation group was (14.8 ±3.4)min,which was significantly shorter than (34.9 ±8.4)min of the control group,the difference was statistically significant (t =14.02,P <0.05).The incidence rate of adverse reactions of the observation group was 7.5%,that of the control group was 5.0%,there was no statistically significant difference between the two groups(χ2 =0.53,P >0.05 ). Conclusion Card other tromethamine in the treatment of postpartum hemorrhage has good clinical curative effect and high safety.
4.The changes of the serum ferritin,fibrinogen and D-dimer levels in patients with small vessel disease and its significance

Objective To explore the correlation of serum ferritin (SF),fibrinogen(FIB) and D-dimer levels in patients with small vessel disease(SVD).Methods 359 patients with SVD were selected,including 186 patients with lacunar infarction ( LI) and 173 patients with leukoaraiosis ( LA) .176 healthy controls were enrolled in this study as control group.The SF,FIB and D-dimer concentrations were measured by radioimmunoassay method and Clauss method respectively .Results The serum levels of SF , FIB and D-dimer in SVD patients were significantly higher than those in the controls [(171.82 ±156.28)μg/L vs (100.37 ±70.11)μg/L,(3.27 ±0.93) g/L vs (2.97 ± 0.88)g/L,(1.10 ±1.06)mg/L vs (0.83 ±0.55)mg/L](all P<0.05).Compared with LI subgroup,the levels of SF,FIB and D-dimer were higher in LA subgroup [(185.22 ±108.55)μg/L vs (159.36 ±89.69)μg/L,(3.32 ± 0.86)g/L vs (3.23 ±0.99)g/L,(1.29 ±1.17)mg/L vs (1.03 ±0.80)mg/L],but the differences were not signifi-cant(all P<0.05).Multivariate regression analysis revealed that by comparison to the reference quartile ,the odds ra-tio (95%CI) for SVD patients in the higher quartile (SF>151.16μg/L,D-Dimer>1.02 mg/L) were 3.444(95%CI=2.097-5.158,P<0.001) and 1.767(95%CI=1.114-2.803,P=0.016),respectively after multivariable ad-justment.Conclusion The increased levels of SF ,FIB and D-dimer are closely related with SVD .FIB and D-dimer might come to be attractive approaches to prevent the progress of SVD .
5.Neoadjuvant systemic therapy for breast cancer

Neoadjuvant systemic therapy for breast cancer patients means systemic treatments before operations.Combination chemotherapy,endocrine therapy and targeted therapy before operation can degrade the tumor staging and increase the breast-conserving rates.Neoadjuvant chemotherapy has been accepted as one of the standard therapies for patients with locally advanced breast cancers.In the neoadjuvant setting,the studies about the use of aromatase inhibitors and trastuzumab in selected patients have also made great progresses.
6.Analysis on Composing Principles of Prescriptions for Lijie Disease Based on The Prescription of Traditional Chinese Medicine Dictionary

Objective To analyze the composing principles of the prescriptions for Lijie disease by using The Prescription of Traditional Chinese Medicine Dictionary. Methods The prescriptions for Lijie disease in the book of The Prescription of Traditional Chinese Medicine Dictionary were collected. Database was established by using TCM Inheritance Support System (V1.1). Medicine using frequency, composing principles, and composition of new medicine were analyzed through association rules and entropy clustering method. Results Based on the analysis on 187 cases of prescriptions, 42 Chinese herbal medicines were with the frequency of 42 times and Angelicae Sinensis Radix was the core herbal medicine. 69 core combinations with three to four herbal medicines and 6 new prescriptions were obtained through evolution. Conclusion According to the analysis of composing principles and clustering new prescriptions of Lijie disease, the treatment for Lijie disease is toning liver and spleen, and nourishing liver and kidney.
7.The clinical value of liquid tumor biomarker detection for breast cancer

Circulating tumor markers have been paid more attention in the application of the treatment for breast cancer, the level of which has extended from protein to gene, including traditional tumor markers, HER-2 extracellular domain, circulating tumor cells, circulating tumor DNA (ctDNA), circulating RNA (ctRNA) and so on. As “liquid detection”, the detection of circulating tumor markers with real-time dynamic, easy operation, good reproducibility and other advantages are widely used in aiding early diagnosis, determining prognosis, prospectively predicting response or resistance to speciifc therapies, surveillance after primary surgery, and monitoring therapy in patients with advanced disease, The further study of circulating tumor markers may contribute to patient’s individual treatment.
9.Construction of combined lentiviral vectors for rat Slit2 gene RNA interfering and its down-regulation effects on Slit2 gene in rat retinal pigment epithelial cells

Background Age-related macular degeneration (AMD) is the leading cause of blindness in people over 50 years old,of which 90% cases are caused by choroidal neovascularization (CNV).Current treatments on AMD have gained great achievements,but there are still some drawbacks.So we need to search for new targets to cure CNV.Objective This study was to construct two combined lentiviral vectors for rat Slit2 gene RNA interference (RNAi) and verify its interfering effects on Slit2 gene in rat retinal pigment epithelial (RPE) cells.Methods Two specific siRNA sequences targeting towards rat Slit2 gene were designed and were annealed to DNA sequences.The DNA sequences and GV248-enhanced green flourescent protein (EGFP) vectors were combined together as recombinant vectors and then were identified.The GV248-EGFP vector,helper 1.0 and helper 2.0 were transfected together into 293T cells and the two combined lentiviral vectors for rat Slit2 RNAi were gained from the cell supernatant after 72 hours of transfection.The titers of the combined lentiviral vectors were measured.The cells were divided into blank control group,Lv-EGFP vector group,Lv-rSlit2-siRNA1 group and Lv-rSlit2-siRNA2 group.The interference efficacy of the combined lentiviral vectors targeting to rat Slit2 gene were identified by real-time fluorescence quantitative PCR and Western blot.The sequence with higher interference efficacy was transfected to rat RPE cells again.The transfected and nontransfected rat RPE cells were treated with 0,100,200 and 400 μmol/L CoCl2 for the preparation of hypoxia models.The expression of vascular endothelial growth factor-A (VEGFA) mRNA in rat RPE cells was finally measured by real-time fluorescence quantitative PCR and the concentration of VEGFA protein in cell supernatant was assayed by ELISA.Results The recombined lentiviral vectors for rat Slit2 gene R NAi were successfully constructed.The titers of the two reconbinant sequences were 5× 108 TU/ml and 3×l08 TU/ml,with the transfected rate ≥70%.The relative expression levels of Slit2 mRNA in the Lv-rSlit2-siRNA1 group and LvrSlit2-siRNA2 group were 0.67±0.09 and 0.23±0.11,respectively,which were lower than 1.03±0.31 and 0.92± 0.07 in the blank control group and Lv-EGFP vector group (all at P<0.01).The expression levels of Slit2 protein in the Lv-rSlit2-siRNA1 group and Lv-rSlit2-siRNA2 group were 0.62±0.07 and 0.49±0.02,respectively,which were lower than 1.00±0.10 in blank control group and 0.95±0.11 in Lv-EGFP vector group (all at P<0.01).Significant differences were found in the expression of VEGFA mRNA and protein in RPE cells among different concentrations of CoC12 groups (mRNA:F tration =127.998,P<0.01;Fgroup =69.663,P<0.01.Protein:F ion =17.059,P< 0.01;Fgroup =91.791,P<0.01),and the expression of VEGFA mRNA and the concentration of VEGFA protein were evidently lower in the Lv-rSlit2-siRNA2 group than those in the blank control group after being treated by 100,200 and 400 μmol/L CoCl2 (all at P<0.01).Conclusions Recombined lentiviral vector for rat Slit2 gene RNAi is successfully constructed,which can effectively knockdown rat Slit2 gene and inhibit the expression of VEGFA in rat RPE cells.
10.Sulfotransferase and breast cancer

Sulfotransferase (SULT) enzymes play important roles in the biosynthesis of estrogen.These enzymes catalyze the sulfation of estrogens to form biologically inactive molecules. Several studies have shown that Arg213His polymorphism in SULT1A1 gene may be a risk factor for breast cancer. In addition, a low mRNA expression of SULT has been significantly associated with reduced recurrence and improved overall survival in patients with estrogen receptor (ER)-positive tumors. Furthermore, SULT may be a crucial factor in the response to tamoxifen treatment. As an important part of estrogen synthetic pathways, SULTs are closely re lated to breast cancer.
