Journal of International Oncology 2011;38(8):624-626
Primitive neuroectodermal tumor (PNET) is a kind of malignant tumor, which most often affects children and adolescents. The incidence of PNET has risen recently. PNET has low early diagnosis rate and poor treatment effect. With the application of RT-PCR and FISH in pathological diagnosis, and MRI and PET-CT in imageological diagnosis, as well as the implementation of target treatment and multidisciplinary integrated treatment concept in the treatment, the pathogenesis, diagnosis technology and treatment means of of PNET have made remarkable progress.
2.Platelets and erectile dysfunction.
National Journal of Andrology 2015;21(9):771-774
Platelets, small pieces of cytoplasm with biological activity, split and fall off the megakaryocytes and mature from the bone marrow. After stimulated, platelets produce nitric oxide to inhibit their own activation and aggregation. Pathologically, the injury of endothelial cells activates platelets and changes their functions. The release of inflammatory mediators and cytokines induces and enhances the development and progression of atherosclerosis, and thereby promotes the occurrence of erectile dysfunction. Besides, platelets and their related functional parameters may serve as important indicators in the diagnosis and treatment of erectile dysfunction.

ObjectiveTo investigate genetic polymorphism of 3 STR loci in Bouyei ethnic group in Guizhou. Methods The DNAs of 101 healthy unrelated Bouyei individuals in Guizhou were extracted using chelex-100 method and were multiplex amplificated by PCR technique and the denatured PAGE was used, the PCR product was typied using silver stain method. The data was statistically analysed by modified-powerstat software package. Results A total of 21 alleles and 50 genotypes were observed in the 3 loci and the distributions of these genotypes were consistent with the law of Hardy-Weinberg equilibrium. The heterozygosity(H) of CSF1PO, TPOX and TH01 loci were 0.7287,0.5423 and 0.6904; the polymorphism information content(PIC) were 0.6805,0.4479 and 0.6426; the discrimination power(DP) were 0.8782,0.7361 and 0.8563 and the probabilities of paternity exclusion (PPE)were 0.6148、0.3643 and 0.5737 respectively. Conclution CSF1PO and THOX are highly genetic polymolphism, which is valuable for population genetic research and forensic individual identification. The distrbution of gene frequencies and their area of ethnic groups have parallel relation.

Journal of Applied Clinical Pediatrics 2006;0(17):-
Objective To evaluate the feasibility of cell free fetal DNA(cffDNA)-based noninvasive prenatal diagnosis,we developed a precise technique for fetal sex determining region of Y chromosome(SRY)gene detection using size-fractionated cell-free DNA in maternal plasma. Methods Peripheral blood samples were collected form 117 pregnant women.cffDNA was extracted based on a column absorbent method and isolated by agarose gel electrophoresis.A dulex-polymerase chain reaction(PCR)was used to detected SRY gene and glycerol-dehyde-phosphate dehydrogenase(GAPDH)gene. Results Both SRY and GAPDH gene were detected in 86 cffDNA samples from women bearing male fetuses. And only GAPDH gene was detected in 71 cffDNA samples from women bearing female fetuses. These results had a coincidence whit those of villus or amniotic fluid samples. The specificity and sensitivity reached 100%(117/117)and 100%(66/66),respectively.Conclusion By agarose gel electrophoresis,re-extratedand and dulex PCR,size-fractionated cell-free fetal DNA in maternal plasma can be selective enriched and used to noninvasive prenatal diagnosis of sex-linked disorders and single gene disorders.

Objective To discuss the clinical curative effects of Kaihoujian spray in the treatment of acute suppurative tonsillitis in children. Methods 120 cases of pediatric patients with acute suppurative tonsillitis were selected and randomly divided into the observation group and the control group (60 cases in each group); the control group were given conventional western medicine treatment, while the observation group received Kaihoujian spray on the basis of the control group treatment; clinical symptoms and curative effects of the two groups were evaluated and compared. Results In the observation group, sore throat and fever disappearance time, purulent secretion, tonsil swelling disappearance time and hemogram recovery time were all significantly shorter than those in the control group (P<0.05); after 3 courses of treatment, in the two groups, children temperature, sore throat, tonsil fester and tonsil swollen main symptom scores were all significantly decreased (P<0.05), but the observation group decreased more obviously (all P<0.05). After treatment, C-reactive protein, white blood cell count, neutrophils of the two groups all significantly reduced (P < 0.05), but the observation group decreased more obviously (all P<0.05). Conclusion Using Kaihoujian spray to treat acute suppurative tonsillitis could significantly improve the clinical symptoms and signs, whose clinical curative effect is good, and has no adverse reactions.
10. Role of Cdc 42 gene in the suppression of filopodia formation induced by non-steroidal anti-inflammatory drugs in breast cancer cells
Tumor 2011;31(2):93-98
Objective: To investigate the effect of cell division cycle 42 (Cdc 42) gene on the formation of filopodia suppressed by non-steroidal anti-inflammatory drugs (NSAIDs), and to explore the probable signal transduction pathway involving Cdc 42 in the inhibition effects on migration and invasion abilities of breast cancer cells induced by NSAIDs. Methods: MCF-7 cells were divided into four groups: blank control group, NS-398 (100 μmol/L)-treated group, cyclooxygenase-2 (Cox-2) small interfering RNA (siRNA)-transfected group and the negative siRNA-transfected group. The expressions of Cox-2 and Cdc42 mRNAs were detected by real-time fluorogentic quantitative PCR(RFQ-PCR), and the expressions of Cox-2 and Cdc42 proteins were measured by Western blotting. Actin-tracker Green fluorescent probe was used to examined the morphology of filopodias in MCF-7 cells. The invasion ability of MCF-7 cells was detected by using the Martrigel-coated Transwell. Results: The filopodias in MCF-7 cells disappeared in the NS-398-treated group, and the invasion ability of MCF-7 cells was also significantly decreased in this group compared with that in the blank control group (P<0.05). There was no difference in the expression level of Cox-2 mRNA or protein between the blank control group and the NS-398-treated group (P>0.05), and the difference was also not seen in the expression of Cdc42 mRNA or protein between these two groups (P>0.05). The active-Cdc42 level was significantly decreased in the NS-398-treated group compared with that in the blank control group (P<0.05). The filopodias in the Cox-2 siRNA-transfected group disappeared, and the invasion ability of MCF-7 cells transfected with Cox-2 siRNA was significantly decreased compared with that in the negative siRNA-transfected group (P<0.05). The active-Cdc42 level was also significantly decreased in MCF-7 cells transfected with Cox-2 siRNA compared with those transfected with negative siRNA (P<0.05). Conclusion: NSAIDs can suppress the invasion ability of breast cancer cells. This effect may be achieved by inhibiting the activity of Cox-2, decreasing the expression level of active-Cdc42, and suppressing the formation of filopodias.
