1.The effects of siRNA-mediated Plk1 depletion on COS-7 cells

Objective:To investigate the effects of siRNA-mediated Plk1 depletion on COS-7 cells and the therapeutic potential of Plk1 in cancer treatment.Methods:Small interfering RNA(siRNA) were used to specifically deplete Plk1 in COS-7 cells.RT-PCR and immol/Lunoblotting analysis were used to verify the inhibitory effect of siRNA against Plk1.Trypan blue exclusion assay and FACS analysis were conducted to determine the effects of Plk1 depletion on the growth and cell cycle profile of COS-7 cells.Results:Synthetic siRNA duplexes against Plk1 were introduced into COS-7 cells,which subsequently resulted in a significant inhibition in Plk1 expression in the cells.Cell growth assay indicated that siRNA-mediated Plk1 depletion significantly inhibited the proliferation of COS-7 cells.FACS analysis revealed that Plk1 depletion caused cell cycle arrest,resulting into large population of sub-G1 cells.Conclusion:siRNA-mediated Plk1 depletion significantly inhibited the proliferation of COS-7 cells suggesting the therapeutic potential of Plk1 in cancer treatment.

4.Depth of counseling process:exploring of a three-dimension model

Objective:To test Jiang Guangrong's(2005)three-dimension model on the depth of counseling process.Methods:In this process-outcome research,28 sessions in a university counseling center were videotaped and coded in terms of the model.Structure Equation Model(SEM)was used to examine the relations between the coding and counselors and clients' evaluation of the session depth.Results:(1)Clients and counselors evaluated differently on the session depth,and clients' evaluation was more positive than counselors' [(4.95±1.12)vs.(4.23±0.94),P<0.05];(2)The model was significantly correlated with counselors' session depth evaluation(path coefficient = 0.55,P<0.01),but not with the clients';(3)The session curves describing each dimension changing over time had different patterns in sessions of different depth level,through which more about the counseling process could be known.Conclusion:The three-dimension model on the depth of counseling process has got some empirical evidences.
5.Meta analysis on gene polymorphism of Matrix metalloproteinase-1,-7 in promoter regions and the susceptibility of cervical cancer

Objective To evaluate the association between genetic polymorphisms in promoter regions of matrix metalloprotein‐ase‐1 (MMP‐1)‐1607bp1G/2G ,matrix metalloproteinase‐7 (MM P‐7)‐181‐bpA /G and susceptibility of cervical cancer using Meta‐analysis .Methods Relevant studies that had relationships of the polymorphism in promoter regions of matrix metalloproteinase‐1 (MM P‐1)‐1607bp1G/2G ,matrix metalloproteinase‐7 (MMP‐7)‐181‐bpA /G and susceptibility of cervical cancer were collected as case‐control groups .RevMan5 .0 software was applied to analyze the odds ratios (OR) and 95% confidence intervals(CI) of the as‐sociation between polymorphisms and susceptibility of cervical cancer .Results Finally ,6 studies were selected ,containing 4 studies of MMP‐1 and 2 studies of MMP‐7 .For MMP‐1‐1607bp1G/2G ,OR was 0 .76 (95% CI = 0 .51 - 1 .13 ,P= 0 .17) under 1G/1G +1G/2G ∶ 2G/2G model ,OR was 0 .72 (95% CI = 0 .42 - 1 .23 ,P = 0 .42) under 1G/1G ∶ 2G/2G model ,OR was 0 .79(95% CI =0 .57 - 1 .09 ,P= 0 .15) under 1G ∶ 2G model ,it was concluded that there was no statistical significance between them .For MM P‐7‐181bpA /G ,OR was 0 .47 (95% CI = 0 .30 - 0 .74 ,P= 0 .001) under AA + AG ∶ GG model ,OR was 0 .43 (95% CI = 0 .26 - 0 .74 , P= 0 .002) under AA ∶ GG model ,OR was 0 .77(95% CI = 0 .62 - 0 .94 ,P= 0 .02) under A ∶ G model ,there were statistical signifi‐cance differences under all models .Conclusion Polymorphisms in promoter of MMP‐7‐118bpA/G are associated with the suscepti‐bility of cervical cancer and an allele is a protective factor .It is still not confirmed that significant association between the MM P‐1‐1607bp1G/2G and susceptibility to cervical cancer exists in current literature .

8.Recombinant AAV-IFNαinhibits HBV replication in vitro

Objective_To identify the anti-HBV efficiency of AAV-IFNαby infecting HepG2.2.15 cell line, and to develop a new strategy of curing hepatitis B.Methods_Firstly,recombinant vector was packaged into AAV by three plasmids co-transfection method;then AAV-IFNαinfected HepG2.2.15 cell line, and samples were collected on the 1st, 3rd,6th and 9th day;thirdly, The time-course of HBV DNA,HBsAg and HBeAg were detected by qPCR and ELISA.The proliferation of HepG2.2.15 cells detected by MTT.Results_Recombinant AAV-IFNαcan significantly inhibit HBV replication and expression in vitro because of the proliferation of HepG2.2.15 cells inhibited by AAV6-IFNα( P<0.05) .Conclusions_AAV-IFNαis a potential antiviral product for hepatitis B.

Acute promyelocytic leukemia (APL) is a subtype of acute myeloid leukemia,which is rare in children.The treatment of APL mainly includes anthracycline-based chemotherapy,all-trans retinoic acid,and arsenic trioxide,which allow for complete remission,improve the long time survival rate and reduce related toxicity as well.About the optimized treatment,quick breakthroughs have already reached,not only at home in the 80's at the end of the 90's,but also abroad,like National Comprehensive Cancer Network of the United States in 2006-2013.This review summarizes the progress in diagnosis and treatment of children with APL.
10.Clinical performance of mechanical versus bioprosthetic valves in patients aged 60-70 years

Objective To compare the mortality,valve-related morbidity and reoperation rate between mechanical prostheses and bioprostheses in patients aged 60 to 70 years.Methods The pre-operative characteristics of 119 patients who received mechanical prostheses and 181 those with bioprosthesis implantation in our department between January 2006 and December 2014 were analyzed,and their mortality,valve-related morbidity and reoperation rate were compared.Results Followup revealed mechanical prostheses had a lower all-cause mortality after mitral valve replacement than bioprostheses at 6 years after the operation (P=0.033),but there was no statistical difference in valve-related mortality (P=0.277).The complications after mechanical prosthesis replacement had embolism (8.4%) and bleeding (5.0%),bioprosthesis' those consisted of structural valvular deterioration (2.2%),nonstructural dysfunction (1.1%),thrombosis (2.8%),embolism (5.0%),bleeding (0.6%) and operated valvular endocarditis (1.1%).There was no statistically significant difference in total complication rate between the two groups (P=0.318).Biological group had 3 patients receiving reoperation in 8 years,while there was no reoperation in the mechanical group,but without statistical group difference.Conclusion Patients aged 60-70 years old could have a similar valve-related mortality,morbidity and reoperation rate after mitral valve replacement between mechanical and biological valves.
