1.Regulation of Hexokinase Activity by HB-EGF in Mesangial Cells
Journal of China Medical University 2001;30(2):91-93
Objective: Our aim was to study the concentrations and regulatory characteristics of heparin-binding epidermal growth factor-like factor (HB-EGF) and its functional consequences in inducing the activities of hexokinases (HKs) and net Glc utilization. Methods: The SV40MES13 cell line was used as cell model for this experiment to measure the total activity of Hks by using a standard G6PDH-coupled assay. The net Glc utilization was measured indirectly as Glc disappearance from the medium using the standard Glc oxidase-coupled reaction(Trinder). Results: The HB-EGF induced HKs activity in time- and dose-dependent manners and increased the net Glc utilization by SV40MES13 cells. Conclusion: The HB-EGF plays an important role in pathogenesis of diabetic nephropathy by inducing total HKs activity specifically and increases the net Glc utilization in mesangial cells.

Tyrosine phosphorylation is a key post-translational mechanism that regulates cellular processes and maintains homeostasis.Aberrant changes in tyrosine phosphorylation are often associated with disease states such as metabolicdisorders, cancer and cardiovascular disease.Protein tyrosine phosphatases (PTPs) are the enzyme family that regulates protein phosphorylation level of tyrosine residues in the cellular processes and signaling ways.So far, scientists have discovered 112 kinds of human PTPs.Among them,PTP1B is widely and clearly studied.Recently,as an enzyme that play a role in oncogenesis,PTP1B has been wildey studied by scientists. Here,we highlight the relationship between protein tyrosine phosphatase 1B and hematologic neoplasms.

OBJECTIVE TTo evaluate the clinical manifestations of the laryngeal amyloidosis and its laryngoscopic characteristics.METHODS The clinical data of 11 cases with laryngeal amyloidosis confirmed pathologically was studied retrospectively. RESULTS There were 4 males and 7 females in this study. The age of the patients ranged from 16 to 64 years old (average 43.36?4.16).The duration of symptoms was 4 months to 10 years (average 2.85?0.87 years).Hoarseness presented in 10 patients (90.90%) and dyspnea in 4 patients. The false vocal cord was involved in 8 patients, laryngeal ventricle in 6 patients, true vocal cord in 5 patients and subglottic area in 3 patients observed with fibrolaryngoscope. The appearance of the lesions was mainly described as waxy yellow or yellow-gray submucosal plaques and nodules without ulceration of the overlying mucosa. The laryngostroboscopy demonstrated that the wave of the mucosa and vibration of the involved vocal cords were adynamic or disappeared.CONCLUSION The chief complaint of the laryngeal amyloidosis was hoarseness. Laryngeal amyloidosis is a slowly progressive disease. The lesions commonly involve the false vocal cord, laryngeal ventricle, true vocal cord and subglottic area.

Objective To analyse protein tyrosine phosphatase 1B (PTP1B) gene mutation in myeloproliferative neoplasms (MPN).Methods DNA sequencing technology was used to detect DNA sequences of PTP1B in MPN patients (n =84) and normal controls (n =37).Results For Exon1-6,Exon9 and Exon10,84 cases of MPN patients and 37 cases of control group were not detected mutation. For EXON 8,18 of 84 MPN patients had Exon8 C/T heterozygous mutation and 10 of 37 normal controls were detected C/T heterozygous mutation. There was no significant difference between MPN patients and normal controls (x2 =0.453,P =0.501).Exon7 was detected in 38 MPN patients and 2 cases of patients were found C/T heterozygous mutation, while in the control group,1 case with G/C heterozygous mutation. All of the cases were not detected homozygous mutation. Conclusion Using DNA sequencing technology to detect gene mutations of PTP1B,there is no significant difference between MPN patients and normal controls.

A near infrared spectroscopy ( NIRS) method was used for rapid quality evaluation of ferulic acid content in chrysanthemum morifolium cv. ( Chuju) continuous cropping soil. Standard leverage, studentized residual and Mahalanobis distance were calculated to eliminate abnormal samples. After the initial near infrared spectrum was treated by two second derivative and Norris smoothing filter noise, 6000-4000 cm-1 wave number range and 7 factors were chosen for partial least squares ( PLS) calibration model. The results showed that good correlation was presented between the calibration set/validation set and the values determined by high performance liquid chromatography, and the calibration correlation coefficient ( Rc ) and validation correlation coefficient ( Rcv ) were 0. 9914 and 0. 9935, respectively. Root mean square error of calibration (RMSEC), root mean square error of validation (RMSEP) and root mean square error of cross-validation (RMSECV) were 0. 484, 0. 539 and 0. 615, respectively. This method was accurate, reliable, simple, rapid and nondestructive, and could be applied to the fast analysis for ferulic acid in continuous cropping soil.

Objective To study the effect of stigma maydis aqueous extract(SMAE) on glucagon in type 2 diabetic rats. Methods Type 2 diabetic male Sprague-Dawley rat mode was induced by high-fat and high-sugar dietary feeds and low dose streptozotocin(STZ). After 8 weeks of treatment, levels of fasting blood glucose(FBG) and glucagon(Glu) in serum were respectively assayed.Immunohistocmical study was adopted to analyze the expression of glucagon in islet cells. Results Compared with model control group,SMAE of middle and high dose remarkably reduced the level of FBG and Glu(P

OBJECTIVE To analyze the results of antimicrobial susceptibility for clinically isolated Corynebacterium striatum strains and investigate the distribution of minimal inhibitory concentration(MIC) of the antibiotics commonly used in clinic against C.striatum.METHODS C.striatum was identified with API Coryne System(bioMerieux,France).Antimicrobial susceptibility test was conducted by agar dilution method.RESULTS MIC90 of imipenem, vancomycin and rifampin against C.striatum was 8,0.5 and 2 ?g/ml,and that of amikacin, gentamicin, tobramycin, and minocycline was 8-16?g/ml,but MIC90 of the other antibiotics was 32-≥256 ?g/ml.The main source of 32 isolates was from bronchofibroscopic secretion.CONCLUSIONS The activity of vancomycin, rifampin and imipenem against C.striatum shows stable susceptibility, but of amikacin, gentamicin, tobramycin and minocycline shows inferiority. However, the susceptibility to other antibiotics is low.So vancomycin, rifampin and imipenem are the optimal antibiotics to treat the infections caused by C.striatum.

Objective To study the effect of minielctrocholecystectomy(MEC) for gallbladder stone. Methods The clinical data of 4,200 cases(1991-2000) undergoing MEC were analysed restrospectively; the results of different operotors for the MEC were compared. Results All 4,200 cases were cured. Of them, 41 cases(0.98%) had serious complications, including intraoperative bleeding in 4 cases(0.09%), biliary tract injury in 18(0.43%); stress ulcer in 8(0.19%), and residual stones of biliary tract in 11(0.26%). Conclusions MEC has the following advantages: less trauma, short operation time, fast postoperative recovery and lower expense. The different operotors on MEC have distinguish difference in the results. It is important to preven the complications of MEC.
