2.Isolation and antitumor activity in vitro of three kinds of bufadienolides

AIM: To isolate three kinds of bufadienolides composition from Venenum bufonis in order to investigate their antitumor activity in vitro.METHODS: Alcohol extraction and isolation by Silica gel colume were employed in this study.Their antitumor activities in vitro were evaluated by MTT colorimetric method.RESULTS: With such methods,three kinds of bufadienolides,including Bufalin,cinobufagin,and resibufogenin with purity above 90% were prepared.The in-vitro results showed that three kinds of bufadienolides had prominent inhibition effect on human lung cancer A549 cells,human breast cancer MDA-MB-435 cells in the range of 0.001 ?g/mL ~ 100 ?g/mL.CONCLUSION: Isolation by Silica gel colume can be used to separate three kinds of bufadienolides from Venenum bufonis and its constituents has prominent anti-cancer effects.

4.Extraction of Banxiaxiexin Decoction

AIM: To optimize the extraction process for Banxiaxiexin Decoction (Rhizoma pinelliae, Radix Scutellariae, Rhizoma coptidis, Radix Ginsenp, Rhizoma zinpiberis, Fruents Jujubae, and Radix Glycyrrhizae). METHODS: The content of berberine、baicalin and total solid in extract liquor were determined by orthogonal design and single factor experiment in combination with glycyrrhizic acid content and identification of Rhizoma zingiberis, Radix Ginseng and Rhizoma Pinelliae. RESULTS: The extracting was arrived at in the condition of adding eighteen times of 70% alcohol as much as crude drug and refluxing 2 times, 2h and 1h, respectively. CONCLUSION: The extraction is stable and feasible.
5.Clinical observation of FOLFOX4 protocol for advanced colorectal cancer

Objective To evaluate the efficacy and toxicity of the protocol of FOLFOX4 for advanced colorectal cancer. Methods 27 patients received FOLFOX4oxaliplatin 85 mg/m2 as a 2-hour infusion on day 1 and a 2-hour infusion of LV (200 mg?m-2?d-1) followed by a 5-Fu bolus (400 mg?m-2?d-1) and 22-hour infusion (600 mg?m-2?d-1) for 2 consecutive days every 2 weeks. Four courses were carried out with an interval of one month. Results The total effective rate was 44.44 %, CR(3.70 %), PR(40.74 %). Median survival of all patients was 10.0 months. Mean Survival was 11.5 months. One year survival rate was 30.02 %. Median duration of 12 effective patients were 5.3 months. Objective To discuss the clinical effects of treatment of femoral intertrochanteric fracture by DHS internal fixation. Methods 42 cases with femoral intertrochanteric fracture were treated by DHS internal fixation. According to AO classification, there were 16 cases of type A1,14 A2,and 12 A3. Results 36 patients were followed up for 6 to 18 months, with an average of 10 months. Their results were satisfactory. Conclusion Treatment of femoral intertrochanteric fractures with DHS internal fixation is an effective method.
7.Meta-analysis on the efficacy and safety of R-CHOP chemotherapy for the treatment of low and moderate malignant B cell non-Hodgkin lymphoma

Objective To comprehensively evaluate the curative effect and adverse effects of rituximab plus cyclophosphamide, vincristine, doxoruhicin and prednisone(R-CHOP) chemotherapy and CHOP chemotherapy alone on the treatment for low and moderate malignant B cell non-Hodgkin lymphoma (NHL). Methods By the application of the systematic review method of Cochrance International Collaboration, the world-wide randomized controlled trials (RCT) on the comparison between different curative effects of R-CHOP and CHOP chemotherapy alone on the treatment low and moderate malignant B cell NHL was collected and the study results was evaluated systematically. Results Seven RCT studies involving 1569 patients and had no heterogeneity between themselves (χ=5.31,P=0.50). The baseline of patients characteristics was comparable in all the studies. By comparing complete response (CR) rate and adverse effects through fixed effect analysis model, the results showed that R-CHOP was better than CHOP chemotherapy on the treatment for low and moderate malignant B cell NHL(OR =2.22, 95 %CI 1.72-2.85, P <0.000 01), and adverse events of R-CHOP had no significant difference compared with CHOP chemotherapy alone (P>0.05). Conclusion With good curative effect on the treatment low and moderate malignant B cell NHL and without obvious differences from the CHOP chemotherapy alone in adverse effects, R-CHOP chemotherapy should be recommended as the best treatment method for low and moderate malignant B cell NHL And much more well-designed clinical RCT should be made to further prove its clinical effect.
8.Effect of HiHi and HiLo on the Lipid Metabolism in Rats

Aim To study the effects of HiHi and HiLo on the lipid metabolism in rats through blood lipids and fatty acid oxidation in gastrocnemius of rats. Methods Thirty male SD rats were selected by means of adaptive training, and then divided into 3 groups randomly: living low-training low group (LoLo), living high-training high group (HiHi), and living high-training low group (HiLo). The rats ran on a treadmill 60 min a day at the speed of 35 m/min under normoxic condition or 30 m/min under hypoxic condition (13.6% of ambient FIO_2), 6 days a week for 4 weeks. The samples from blood and gastrocnemius were removed 24 hours after the last training by the end of 4-week experiment. TC, TG, HDL and LDL were tested with a full-automatic biochemical analyzer. LPL, leptin and AD were measured using ELISA. Real-time quantitative PCR was adopted to test the expression of PPARa and CPT-1 mRNA in rats' gastrocnemius. Results Compared to the LoLo, TC and TG decreased significantly (P0.05) and LPL and AD increased significantly (P<0.01) in HiHi.' PPARa mRNA and CPT-1 mRNA expressions in gastrocnemius were significantly higher in HiHi(P<0.05) than in LoLo, and significant descent of HDL(P<0.05) and CPT-1 mRNA expression(P<0.01) revealed in HiLo, as compared to the LoLo. Compared to the HiLo, HDL(P<0.05), and LPL, AD and CPT-1 mRNA expression(P<0.01) enhanced in HiHi. Conclusion (l) HiHi was more superior in regulation of blood lipids than LoLo due to increasing of LPL, and could facilitate fatty acid oxidation because of enhancement of CPT-1 mRNA, AD and PPARa mRNA. (2) Comparing to the normoxic training, HiLo had no beneficial effect on the blood lipid metabolism and decreased the fatty acid oxidation in gastrocnemius.(3 )HiHi was more superior in the effeet of HDL thah HiLo due to increasing of LPL, and could facilitate fatty acid oxidation because of enhancement of AD and CPT-1 mRNA.
9.Impact of RNAi targeting survivin gene on tumor growth, apoptosis and radiosensitivity in nude mice xenograft of human cervical carcinoma

Background and purpose: Recently, survivin gene is one of the hot spots in tumor study. The purpose of the present study was to observe the impact of RNAi targeting survivin gene on tumor growth, apoptosis and radiosensitivity in nude mice xenograft of human cervical carcinoma. Methods: HeLa-s2, HeLa-NC, HeLa-U6 neo and HeLa cells were inoculated respectively in flank subcutaneous tissue of 24 female nude mice so as to establish xenograft models of human cervical carcinoma randomly. The tumor growth status was observed. The tumor volume was regularly measured and the tumor weight was investigated used to observe the impact of RNAi targeting survivin gene on tumor growth. The expression of survivin protein and FⅧRAg in tumor tissues were examined by immunohistochemistry SP method MVD was calculated. Cell apoptusis in tumor tissues was observed by HE staining,apoptotic index(AI) was quantified by TUNEL method. To observe the impact of RNAi targeting survivin gene on tumor radiosensitivity after irradiated, the tumor growth delay and tumor weight were investigated and cell apoptosis were examined by TUNEL method. Results: We established 4 groups of xenograft models of human cervical carcinoma.The tumor volume of HeLa-s2 group was obviously less than that of HeLa group at every checkpoint. The tumor weight of HeLa-s2 group was obviously lower than that of HeLa group, and they were(0.369±0.043)g and (1.150±0.136)g respectively(P<0.05).The tumor growth inhibitive rate of HeLa-s2 group was 67.9%. The expression of survivin protein FⅧ RAg examined by immunohistochemistry in tumor tissues of HeLa-s2 group decreased sharply and MVD went down to 23.4±3.1. Apoptotic cells increased in tumor tissues of HeLa-s2 group examined by HE staining and TUNEL method, AI was (22.74±1.4)%. The tumor volume of HeLa-s2 group was obviously less than that of HeLa group at every checkpoint after irradiation and the tumor weight of HeLa-s2 group was obviously lower than that of HeLa group, they were:(0.41±0.06)g and(1.38±0.29)g respectively(P<0.05). Cell apoptosis increased in tumor tissues of HeLa-s2 group.Compared with HeLa group, AI of HeLa-s2 group rose up notably, they were(30.06±0.98)% and (4.17±0.64)%(P<0.05).Conclusion: RNAi for survivin gone can reduce MVD in xenograft by inhibiting survivin protein expression, thus inhibit tumor growth and induce cell apoptosis, Survivin gene RNAi can also enhance tumor radiosensitivity.
10.The clinical significance of immune-related marker detection in idiopathic thrombocytopenic purpura

Objective To assess the clinical significance of detecting the immune markers in idiopathic thrombocytopenic purpura (ITP). Methods The frequencies of circulating B cells secreting platelet-specific antibody, platelet-specific antibody, the percentage of T lymphocyte subsets, the percentage of reticulated platelet and the level of thrombopoietin in 64 ITP patients and 31 healthy controls were measured with enzyme-linked immunospot assay (ELISPOT),modified monoclonal antibody immunobilization of platelet antigens assay (MAIPA), flow cytometry and sandwich enzyme-linked innnunosorbent assay respectively. Results Compared with the controls[1.3 ± 0. 5/105 peripheral blood mononuclear cell (PBMC), (0.33±0.06,0.41±0.03), (22.08±4.54)% and (8.19±2.46)%], the frequencies of circulating B cells secreting platelet-specific antibody (7.6±4.6/105 PBMC in acute ITP group, 5.3±3.0/105 PBMC in chronic ITP group), platelet-specific antibody (including the anti-GP Ⅱ b/Ⅲa antibody, anti-GP Ⅰ b/X antibody) (0.51 ±0.11, 0.48±0.06 in acute ITP group; 0.49±0.10,0.46±0.09 in chronic ITP group), the percentage of CD8+ T Lymphocyte (27.09±9.86 ) %, the percentage of reticulated platelet in ITP patients[the megakaryocyte cytosis group (24. 85 ± 19. 18)%, the normal megakaryocyte group (23.89±18.90)%]were significantly increased ( all P<0.05).The frequencies of circulating B cells secreting platelet-specific antibody in acute ITP patients were notably increased (P<0.05) compared to the chronic ITP patients. In T lymphocyte subsets, the percentage of CD3+T lymphocyte and CD4+ T lymphocyte and the ratio of CD4+/CD8+ in the patients with ITP[(60.88±14.59)%, (28.41±10.55)%, 1.18±0.59]were notably decreased than those in the healthy controls [(69.89±6.43)%, (35.38±5.05) %, 1.64±0.29, P<0.05]. There was no apparent difference of the level of thrombopoietin between ITP patients with megakaryocyte cytosis (72. 09 ± 41.64 ) and health controls (75.37± 26. 32, P > 0. 05 ), however, the level of thrombopoietin of ITP patients with normal megakaryocyte apparently increased (118.60±70.72, P<0.05). Conclusion Detecting the frequencies of circulating B cells secreting platelet-specific antibody, platelet-specific antibody, the percentage of T lymphocyte subsets, the percentage of reticulated platelet and the level of thrombepoietin in the patients with ITP may improve the diagnosis and guide clinical therapy.
