1.Expression of RET gene in cervical squamous cell carcinoma

Objective To explore the expression of RET in cervical squamous carcinoma tissues and evaluate its relationship with cervical squamous carcinoma clinical pathological indexes and its prognostic value. Methods The expression and distribution of RET in normal cervical tissues ,CINⅠ,CINⅡ& CINⅢ and cervical squamous carcinomas tissues were detected by immunohistochemistry in wax blocks. Clinical data and follow-up data are integrated to analyze its relationship with clinical pathological factors and prognosis value. Results The positive rate of RET protein in cervical squamous carcinomas tissues is higher than in other tissues.The positive rate was related to FIGO stage and lymph node metastasis(P<0.05),and not related with the age of patients or the degree of tumor differentiation(P>0.05). The result of survival analysis with Kaplan-Meier method showed poorer disease-free survival time in the patients with high expression of RET (P < 0.05). Prognostic analysis of patients with cervical cancer through COX proportional hazard regression model suggested that RET expression was an independent prognostic factor. Conclusions RET has been involved in the progression ,FIGO stage and metastasis of cervical squamous cell carcinomas.
2.Study on chromatographic fingerprint of sarcandra glabra (Thunb.) by microwave-assisted extraction coupled to HPLC/DAD

Microwave-assisted extraction (MAE) was used for extraction of effective components of sarcandra glabra (Thunb.), and then chromatographic fingerprint of sarcandra glabra (Thunb.) was studied by high performance liquid chromatography/diode array detector (HPLC/DAD). The conditions of MAE were optimized by an orthogonal experiment, and then the authentication and validation of the chromatographic fingerprint were conducted. Nine peaks were identified as common peaks in the fingerprint chromatograms, and isofraxidin was considered as a reference compound and quantified. Relative standard deviations of retention time and peak area of each component were less than 3% and 8%, respectively. Similarity and difference analysis were conducted by use of PCA and relation coefficient. Twenty batches of sarcandra glabra (Thunb.) samples from two different producing areas could be classified into two different groups in the PCA model. The results showed that MAE-HPLC/DAD method was simple, efficient and stable for the study of complex chromatographic fingerprint of sarcandra glabra (Thunb.), which could provide more reliable and precise information for quality evaluation.
3.Expression and clinical significance of serum microRNA-100 in gastric cancer patients

Objective: To investigate the expression of microRNA-100 (miR-100) in the serum of gastric cancer patients and its clinical significance. Methods: At The First Affiliated Hospital of Bengbu Medical College, 40 gastric cancer patients who underwent surgery with complete follow-up data and 40 healthy controls were selected as research subjects. Total miRNA was isolated from the se-rum samples of the cancer patients. A stable and sensitive detection method for the absolute quantification of miR-100 was established. The serum levels of miR-100 in the patients and healthy controls were tested according to this novel method. Differences of the miR-100 expression in the serum samples of the patients and healthy controls were analyzed using statistical analysis. The correlation between miR-100 expression levels and the clinicopathological features of gastric cancer was also analyzed. Results: The expression level of miR-100 in the serum was significantly higher in cancer patients[(2.78±1.92) fmol/L]than in the healthy controls[(0.19± 0.15) fmol/L, P<0.01]. The receiver operating characteristic curve of miR-100 indicated that serum miR-100 has satisfactory specificity and sensitivity for the diagnosis of gastric cancer (AUC=0.985). Further analytical results indicated that no significant differences were found among the expression levels of serum miR-100 with respect to the gender, age, tumor diameter, infiltrating depth, differentiation degree, frequency of lymph-node metastasis, and TNM stage (P>0.05). Conclusion: Serum miR-100 testing may be helpful in the diag-nosis of gastric cancer.
4.The clinical application of the open supraclavicular approach in thyroidectomy

Objective: To analyze the clinical application of the open supraclavicular approach in thyroidectomy. Methods: The clinical practicability of open supraclavicular thyroidectomy was explored by comparing the traditional anterior low arc incision thyroidectomy procedure with open supraclavicular thyroidectomy in terms of patients'aesthetic satisfaction, effectiveness of the operation, operation time, and so on. Result: Twenty-two cases of open supraclavicular thyroidectomy (group B) had better aesthetic satisfaction than 29 cases of traditional incision thyroidectomy (group A)(P<0.05), and had the same operative effect with traditional incision. Open supraclavicular thyroidectomy is associated with good aesthetic satisfaction, and has the same effect as the traditional incision does. Conclusions: Open supraclavicular thyroidectomy has good clinical value for benign thyroid tumors and some malignant tumors that require unilateral lobectomy, and even for tumors larger than the incision diameter. It has good aesthetic value while ensuring the curative effect of surgery.
5.Risk of Immunomodulatory Drugs in the Treatment of Deep Venous Thrombosis in Multiple Myeloma :Systematic Evaluation and Meta-analysis

OBJECTIVE: To evaluate immunomodulatory drugs (IMiDs) in the treatment of deep venous thrombosis (DVT) in patients with multiple myeloma (MM) systematically, and to provide reference for safe drug use in clinic. METHODS: Retrieved and collected randomized controlled trials (RCTs) about the risk of DVT in MM patients treated with IMiDs from PubMed, Web of Science, Cochrane library, CJFD, Wanfang database, VIP, www.ClinicalTrials.gov during database and Dec. 31, 2018. Meta-analysis was conducted for the incidence and relative risk of DVT (RR) by using Stata 12.0 statistical software. Evidence was evaluated and graded by using GRADE system. RESULTS: A total of 11 RCTs were included, involving 3 365 patients (including 3 drugs). Results of Meta-analysis showed that the incidence of DVT was 7.3% [95%CI (4.5%, 10.2%)] during IMiDs in the treatment of MM. Compared with conventional chemotherapy, IMiDs had a higher risk of DVT in MM patients [RR=3.57,95%CI(2.42,5.27), P<0.01]. Subgroup analysis in different treatment stage showed that after IMiDs treatment for MM patients at induction stage, the risk of DVT increased by 386% compared with conventional chemotherapy plan [RR=4.86, 95%CI (2.85, 8.30), P<0.01], which evidence was moderate. Compared with conventional chemotherapy plan, there was no significant difference in the risk of DVT among MM patients treated with IMiDs at maintenance stage [RR=2.40, 95%CI (0.70, 8.27), P=0.16] and relapse stage [RR=2.01, 95%CI (0.74, 5.46), P=0.17]. The incidence of severe DVT caused by thalidomide and lenalidomide were 11% [95%CI (9%, 13%)] and 3% [95%CI (2%, 4%)]. CONCLUSIONS: The current evidence suggests that patients with MM treated with IMiDs are at a high risk of serious DVT, and clinical medication should be cautious.
