1.Influence of hypothermia environment on treatment of limb gunshot fracture with internal fixation in sheep

[Objective]To compare the influence of hypothermia environment on healing of limb gunshot fracture in sheep with debridement and internal plate fixation at different time after wounded.[Method]Thirty sheep were divided into normal environment group(NE group) and hypothermia environment group(HE group) at random.They were kept in 26℃ and -25℃ environment respectively for 3 h,and then one side of hind limb was made fracture at middle part of the tibia by a 7.7 type handgun.The wound track was debrided at 6,12 and 24 h after wounded.The fracture was fixed by plate respectively.The healing of the wound track and fracture were observed and compared between groups.[Result]At 24 h after wounded,infection of the wound track occurred in 2 cases,and osteomylitis occurred in one by X-ray and in two by pathological examination in NE group.The healing of the wound track and fracture were excellent in HE group,and no osteomyelitis was observed by X-ray and pathological examination in HE group.The earlier the management started,the better the wound and fracture healed.[Conclusion]The hypothermia environment benefit the healing of the wound and the fracture after debrided and internal fixation.It is suggested that the management of gunshot fracture in hypothermia environment can make the time-limit of debridement and internal fixation prolonged.
2.Effect of rivaroxaban on prevention from deep venous thrombosis after hip replacement surgery

Objective To investigate the effect of rivaroxaban on prevention from deep venous thrombosis after hip replacement surgery .Methods 80 patients with lower limb parallel arthroplasty were randomly divided into observation group and control group from January 2014 to January 2015.40 cases in each group.Control group was given aspirin treatment, observation group was given rivaroxaban treatment, indicators changes were followed up and recorded.Results PT, APTT values of observation group one week after treatment were (35.79 ±2.16),(15.84 ±2.11) s, which were better than control group (45.64 ±2.12),(18.72 ±2.02) s (P<0.05).TT, D-dimer of observation group(13.48 ±2.10)s,(2.34 ±0.33)g/L after a week were better,than control group (16.38 ±1.79)s, (1.42 ±0.21)g/L (P<0.05) respectively.Conclusion Effect of rivaroxaban in prevent deep venous thrombosis after hip replacement surgery is exact without obvious adverses reaction, it is worthy of further research and application.
3.The blind area in routine gastric lavage and its clinical significance

Objective To observe the blind area in routine gastric lavage and evaluate its significance. Methods Each of 10 healthy volunteers drank 60 ml water containing 185 MBq 99mTc. Images of mouth, esophagus and stomach of all subjects were collected from anterior position with gamma camera, and intraoral and intraesophageal radiocounts were monitored with " Region of Interesting" technique at 0, 0.5, 1,2,3 and 4 hours. Ratios of the counts at subsequent time points vs the count of 0 hour were calculated. For other 13 volunteers, 100 ml, 200 ml, 300 ml and 400 ml normal saline containing 25% compound meglucamine diatrizoate were drunk and injected into stomachs through stomach tube respectively. After that, X-ray chest films were taken at the dorsal and left lateral decubitus positions to observe the oral and e-sophageal developments. Results The images of mouths and esophagus of 10 subjects could be identified more than 4 hours after the administration of 99mTc. The ratios of oral counts and esophageal counts at 0. 5 ,1, 2, 3 and 4 hrs were 30. 7% , 10. 5% , 6. 3% , 4. 7% , 3. 7% , and 18. 6% , 4. 4% , 3. 5% , 3. 0% ,2. 7% respectively. The ratios of both oral and esophageal counts were 49. 3% , 14. 9% , 9. 8% , 7. 7% and 6.4%. When the liquid was drunk, full oral and esophageal developments could be identified. While the liquid was injected through the tube, no development was found. Conclusions Liquid drug may remain in mouth and esophagus for a long period of time after administration. Routine gastric lavage through nasogastric tube can not clean the oral and esophageal drug residues. There are oral and esophageal blind areas when routine gastric lavage is employed.
4.The experience of diagnosis and treatment of primary ureteral polyps:a report of 27 cases

Objective To explore the diagnosis ,treatment methods and clinical features of primary ureteral polyps.Methods Clinical data of 27 cases with primary ureteral polyps were analyzed retrospectively .The clinical features and treatment of this disease were analyzed .Results 7 cases were treated with polypectomy ,10 cases with basement fulguration by ureteroscopy operation or polyps-removing by ureter forceps ,4 cases with distal ureter resec-tion and termino-terminal anastomosis ,4 cases with lesions ureter resection and bilateral ureteric reimplantaion and 2 cases with nephrectomy due to nonfunctioning kidney .All cases were confirmed to be primary ureter polyp by pathology.All cases were followed up for 6-12 months with no recurrence and canceration ,only 1 case had ureteral stenosis.Conclusion Primary ureteral polyps is a benign disease with rare malignancy .The primary and effective treatment method is surgery which has few complications and good effect .
5.Osteogenic differentiation of adipose-derived stem cells transfected by lentivirus vector carrying LIM mineralization protein-1 and hypoxia-inducible factor-1alpha

BACKGROUND:LIM mineralization protein-1 (LMP-1) and hypoxia-inducible factor-1α (HIF-1α) as intracelular proteins can induce osteogenic differentiation and promote angiogenesis, respectively. Therefore, their combination is of great significance for effectively inducing the osteogenic differentiation of adipose-derived stem cels.
OBJECTIVE:To study the osteogenic differentiation of adipose-derived stem cels transfected by lentivirus vector carrying LMP-1 and HIF-1α.
METHODS:Reverse transcription-PCR technology was employed to clone LMP-1 and HIF-1α genes, and thegenes were cloned to lentivirus vectors pLVX-EF1α-DsRed-Hyg and pLVX-EF1α-IRES2-AcGFP1 to construct main lentiviral vectors pLVX-EF1α-DsRed-Hyg-RLMP-1 and pLVX-EF1α-IRES2-AcGFP1-RHIF-1α. Then, Lenti-X 293T cels were transfected with main vectors pLVX-EF1α-DsRed-Hyg-RLMP-1 and pLVX-EF1α-IRES2-AcGFP1-RHIF-1α, packaging plasmid and coated plasmid. After that, lentiviral vectors were packaged to transfect adipose-derived stem cels from rats that were obtained by tissue explants culture and enzyme digestion methods. At 3, 7, 14 days after transfection, reverse transcription-PCR technology was adopted to detect the expression of osteogeic genes, such as bone morphogenetic protein 2, Runx-2, alkaline phosphatase, osteocalcin as wel as to detect the expression of vascular endothelial growth factor.
RESULTS AND CONCLUSION:Lentiviral vectors pLVX-EF1α-DsRed-Hyg-RLMP-1 and pLVX-EF1α-IRES2-AcGFP1-RHIF-1α were effectively transfected into adipose-derived stem cels. Reverse transcription-PCR results showed that from the 7th day to the 14th day after lentivirus transfection, bone morphogenetic protein 2, Runx-2, alkaline phosphatase and osteocalcin al over-expressed. These findings indicate that the combination of LMP-1 and HIF-1α can enhance the osteogenic activity of adipose-derived stem cels.
6.Cloning of P1 and P3 promoters of human insulin-like growth factorⅡ gene

AIM: To clone P1 and P3 promoters of the human insulin-like growth factor Ⅱ(IGF-Ⅱ) gene. METHODS: According to the complete DNA sequence of IGF-Ⅱ gene, the nested primer PCR was performed for amplifying P1 and P3 promoter fragments of the gene from human L-02 cell line.These PCR products were analyzed by agarose gel electrophoresis, and cloned by using TOPO TA Cloning kit. The positive clones containing P1 and P3 fragments were selected and confirmed by sequencing.RESULTS: The DNA sequences of P1 and P3 promoters cloned were accordant with GenBank data. CONCLUSION: In this study P1 and P3 promoters of the IGF-II gene were cloned successfully.
7.Effect of Tripterygium wilfordii combined with methotrexate on short-term effect of rheumatoid arthritis and the levels of IL-6, IL-10 and TNF- in peripheral blood

Objective To investigate the effects of tripterygium glycosides combined with methotrexate in treatment of rheumatoid arthritis (RA) curative effect and peripheral blood of patients with IL-6, effects of IL-10 and TNF- alpha. Methods 80 patients with RA from February 2014 to August 2016 in our hospital were retrospectively analyzed. According to the different treatment methods by for the observation group, the control group (40 cases). To compare and analyze the curative effect before and after short-term treatment and treatment of patients in the 2 groups were the main clinical symptoms and signs of change and ESR, RF, CRP, IL-6, TNF- alpha, IL-10 index. Results The total effective rate of observation group (92.50%) was significantly higher than the control group (77.50%), the difference was statistically significant (P<0.05), 2 groups of patients before treatment of main clinical symptoms and signs, there was no significant difference, after treatment, clinical symptoms and signs of 2 groups of patients were compared with those before treatment was statistically significant to alleviate the differences (P<0.05), and the observation group of main clinical symptoms and signs were lower than the control group and the remission (down), the difference was statistically significant (P<0.05).Before ESR, the treatment of 2 patients with RF, CRP, TNF- IL-6, alpha, IL-10 index level, no statistical significance, the 2 groups after treatment in patients with ESR, RF, CRP, IL-6, TNF- alpha, IL-10 index level than before treatment with significant difference (P<0.05), and the observation group ESR, RF, CRP, TNF- alpha, IL-6 average index of water compared with the control group decreased significantly, IL-10 index increased significantly than the control group. The difference was statistically significant (P<0.05). Conclusion Tripterygium glycosides combined with methotrexate in treatment of rheumatoid arthritis And it can effectively inhibit the expression of peripheral serum L-6 and TNF- alpha, promote the expression of IL-10 and alleviate the inflammatory reaction

Objective To investiagate cell apoptosis and expressions of Bax,Bcl-2 andCaspase-3 in gambogic acid-treated colorectal cancer cells. Methods SW480/LOVO colorectal cancer cells were treated by gambogic acid. Cell Counting Kit-8 assay (CCK-8) was used to test cell proliferation. Microscopy was used to check the morphological changes. Immunofluorescence staining technique was used to detect cell apoptosis. Expressions of Bax,Bcl-2 and Caspase-3 protein were detected by Western blot assay. Results Gambogic acid inhibited the proliferation of SW480/LOVO in a dose and time-dependent manner. Gambogic acid could induce cell apoptosis. Gambogic acid increased expressions of Caspase-3 and Bax, increased the ratio of Bax/Bcl-2, and decreased Bcl-2 protein expression. Conclusion Gambogic acid can inhibit proliferation and induce apoptosis of SW480LOVO cells, with the mechanism of up-regulation of Bax/Bcl-2 and activation of Caspase-3.
9.The application of circulating tumor cells in early diagnosis of colorectal cancer metastasis

Objective To investigate the expression of circulating tumor cells (CTC) in peripheral blood of patients with different stages of colorectal cancer (CRC), and to evaluate its significance in early diagnosis of colorectal cancer metastasis. Methods Sixty patients with CRC (42 inⅡ-Ⅲstage and 18 in IV stage ) and 30 patients with benign rectal disease were recruited from January 2014 to December 2015. The CTC in peripheral blood was purified with Immunomagnetic Separation Technologie, and detected by immunofluorescence in situ hybridization (imFISH). The serum levels of CEA were detected by electrochemiluminescence method meanwhile. The correlation between CTC and CEA was analyzed. Results CTC positive rates in CRC patients were significantly higher than those in benign rectal disease controls. CTC positive rates inⅣstage were significantly higher than those inⅡ-Ⅲ stage. The expression of CTC was significantly correlated with CEA (r = 0.6652, P < 0.01). Conclusions The expression of CTC in CRC patients is significantly higher than that in benign rectal disease control group. It is closely related to clinical stages. Detection of peripheral blood CTC has important clinical significance in the early diagnosis of CRC metastasis.
10.The effects of premature atrial contractions on hemodynamics with coronary heart disease and hypertension without basic cardiovascular disease

Objective To explore the effects of premature atrial contractions on hemodynamics combined with coronary heart disease and hypertension .Methods According to the advance rate ,62 patients with premature atrial contractions were invided into 3 groups :< 20% group(n= 18) ,20% - 40% group(n= 20) and > 40% group(n= 24) .All of the patients were performed imped‐ance cardiography .The hemodynamics indexes at different times were compared ,and the variation rates in hemodynamics of the three groups combined with basic diseases were analyzed .Results Stroke volume(SV) ,stroke index(SI) ,aortic compliance(AC) , stroke work(SW) ,stroke work index(SWI) ,and heather index(HI) were smallest when premature atrial contractions ,which were middle when the first sinus beats before premature atrial contractions ,and largest when the first sinus beats after premature atrial contractions(P< 0 .05) .The variation rates in hemodynamics were smallest in < 20% group ,which were middle in 20% - 40%group ,and largest in > 40% group(P< 0 .05) .The percentage of patients with basic heart diseases was lowest in < 20% group , which was middle in 20% - 40% group ,and highest in > 40% group(P< 0 .05) .Conclusion The advance rate is relatively large in patients with basic cardiovascular disease .Premature atrial contractions has certain effects on hemodynamics ,and the effects are positively correlated with advance rate .
