China Pharmacy 2015;(22):3086-3088
OBJECTIVE:To investigate the feasibility of mouse models establishment with kidney-positive deficiency with anti-sense oligonucleotide-loaded biodegradable polylactic acid microspheres. METHODS:Three-week old mice were randomized into groups A,B,C,D and E,with 6 mice in each group, where the mice in group A were given subcutaneous injection 0.2 ml nor-mal saline, and those in groups B-E were given subcutaneous injection 0.1,0.2,0.4 and 0.8 mg each mice of antisense oligonucle-otide-loaded biodegradable polylactic acid microspheres. After the administration, the weights and activities of mice in all groups were observed;and one month later, their livers, adrenal glands and brains were collected to detect the expression of glucocorti-coid receptor (GR) by real-time quantitative polymerase chain reaction (RT-PCR). RESULTS:All mice except those in group A were found to have kidney-positive deficiency symptoms such as developmental delay, light weight, less activities and gathering to-gether. Compared to group A,other groups showed lower GR expressions in liver, adrenal gland and brain. There was statistically difference (P<0.05). CONCLUSIONS:Antisense oligonucleotide-loaded biodegradable polylactic acid microspheres can inhibit GR expression, and the mice whose GR expression is inhibited show symptoms of kidney-positive deficiency, which provides a re-ference for the establishment of mouse models with kidney-positive deficiency.

International Eye Science 2014;(6):1089-1091
AIM:To analyze relevancy between the changes of the retinal thicknesses of children's anisometropic amblyopic macular at Area A1 before and at the end of amplyopia cure and amplyopia cure effect.
METHODS: Through randomly selecting 30 cases ( 60 eyes) of anisometropic amblyopic children as test group and 29 cases ( 58 eyes ) of children with normal vision after the vision correction as control group, the changes of the retinal thicknesses of macular of the test group and the control group at Area A1 by OCT-3 was observed dynamically and analyzed.
RESULTS: Compared with first diagnosis, the comparative difference of retinal thicknesses of macular at Area A1 in control group at 3, 6mo has statistical significance. The test group was divided into amblyopia group and non-amblyopia group. There has statistical significance for the comparative difference between the retinal thicknesses of macular centre at 3, 6mo and the first diagnosis in the non - amblyopia group, the amblyopia group was divided into the invalid cure group, progress group and the cured group according to amblyopic cure effect. Compared with first diagnosis, there is no statistical significance of macular at Area A1 in invalid cure group and progress group at 3, 6mo, and in the cured group at 3mo, but in the cured group at 6mo has statistical significance.
CONCLUSION: There is a certain relationship between the changes of retinal thicknesses of macular of children with anisometropic amblyopic at Area A1 before and at the end of amblyopia treatment and amblyopia cure effect. OCT is a kind of objective auxiliary test method and provides new clues for amblyopia research.

Chinese Journal of Blood Transfusion 2002;0(05):-
Objective To establish a method for detection of RhD(+) red blood cells mixed in D-negative blood by flow cytometry(FCM).Method RhD(+) and RhD(-) RBCs were mixed according to predefined ratios.Cells were indirectly labeled, with IgG anti-D labeled as the first antibody, and FITC-anti-IgG F(ab')2 as the second antibody. The percentage of RhD(+) RBCs was determined by FCM,and the best dosage of IgG anti-D was also defined. The ratio of red cells in the two groups, measured by FCM,was compared with the actual ratio. The consistency of method was also evaluated. Results The effective dosage of IgG anti-D was 1∶4,and 50?l/1?106 cells. When the actual percentages of RhD(+) cell among RhD(-) cells were 2.5%-0.312%,the correlation coefficient between the percentages measured by FCM and the actual percentages was 0.987.The same tubes, containing 10% and 2.5% RhD(+) RBCs,were each tested for 10 times, and their coefficient of variation were 3.4%,and 4.9%,respectively.Conclusion The method of quantifying the RhD(+) RBCs in D-negative blood by FCM is feasible and repeatable, which deserves a further clinical application.

Objective To investigate the standardized treatment of 33 children with thalassemia and their family financial burden registered in Bao'an district, Shenzhen city, and to provide basic information for formulating health policy for the government. Methods In 2009, preliminary investigations on 39 registered families with thalassemia children were conducted by telephone, and a household survey was made to collect treatment and economic status by questionnaire on 33 children. Results Among 33 cases of thalassemia children, 21 cases(63.7%) were severe anemia, 5 cases( 15.1%) in need of care or special care, and 25 cases(75.8%) were difficult or unable to maintain standardized treatment. The average family monthly income and expenditure was (4060 ± 2002) and (4926 ± 2991) yuan, respectively. The average monthly treatment costs were (2665 ± 1872) yuan, and the average debt amounted to (64 600 ± 53 940) yuan. Fifteen families[60.0%(15/25)] would reduce the times of blood transfusions or iron transpirations when they encountered revenue deficiency. Conclusions The heavy economic burdens on families with children thalassemia result in inadequate or interrupted treatment on sick children and affect their survival and quality of life, which should be taken more attention and social care.
