Objective To screen proteins binding with interferon?(IFN?)from human hepatic cDNA libraty by yeast-two hybrid technique.Methods The IFN?gene was amplified by polymerase chain reaction(PCR)and constructed into pGBKT7 vector as the bait plasmid in yeast-two hybrid system3,pGBKT7-IFN?was then transfected into yeast AH109.The transfected yeast were mated with yeast Y187 containing liver cDNA library plasmid in 2?YPDA medium.Diploid yeast was plated on synthetic dropout nutrient medium(SD/-Trp Leu-His-Ade)and synthetic dropout nutrient medium(SD/-Trp-Leu-His-Ade)containing X-?-gal for selecting.After plasmid extracting and en- zyme cutting analysis,the blue colonies were performed sequence analysis,the results were analyzed by bioinformatics.Results IFN?gene was successfully cloned and expressed in yeast cells.Thirty- four positive colonies were obtained using yeast-two hybrid technique.After sequence analysis,eight clones were found may have a binding effect with IFN protein.Conclusions IFN?genes was success- ful cloned and eight proteins that could bind with IFN?protein were also screened.

ObjectiveTo investigate the relationship between tumor necrosis factor (TNF) α308,TNF-β252 genotypes and serum TNF-α and TNF-β levels in patients with gastric cancer (GC).MethodsA total of 57 pathological diagnosed GC patients were collected,of which 49 cases were from Zhongnan Hospital of Wuhan University and 8 cases were from Tumor Hospital of Hubei Province.Another 18 age and sex matched healthy controls were enrolled at the same time.The TNF-α308 and TNF -β252 polymorphism was genotyped by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP).The serum TNF-α and TNF-β levels of 57 GC patients and 18 healthy controls were measured by ELISA.The difference of TNF serum level in different TNF genotypes of GC and the difference between GC patients and the healthy controls was analyzed.Its relationship with clinical pathological characters was also analyzed.Results With TNF-α308 genotype,GA were 6 cases and GG were 51 cases.With TNF-β252 genotype,GG were 17 cases,GA and AA each were 20 cases.The serum TNF-α level of GC patients was significant higher than that of healthy controls (median 445 × 10-3 μg/L vs 5 × 10-3 μg/L,P＜0.05),and the serum level of each TNF-α308 andTNF-β 252 genotypes was significant higher than that of healthy controls (P＜0.05).However there was no statistical significance in TNF -β level compared with healthy controls (P＞0.05).In addition,the serum TNF-α levels of the TNF-α308G/TNF-β252G and TNF-α308G/TNF β252A haplotypes in GC patients were significant higher than those of the healthy controls (P＜0.05),and the increase like serum TNF-α level was associated with the patients'age and lymph node metastasis (P＜0.05).The TNF-β level in patients with TNF-α308A and TNF-β252G high-risk haplotypes showed a significant relation with smoking history (P＜ 0.05). Conclusions Serum TNF-α level of GC patients was significantly higher,however there was no significant association between the increase and TNF-α308 and TNF-β252 genotypes.The serum TNF-α levels of TNF-α308G/TNF-β252G and TNF-α308G/TNF-β252A haplotypes in GC patients were significant higher,and associated with the patients'age and lympb node metastasis.It was indicated that TNF haplotypes may have certain impact on the TNF expression and clinical subtypes in GC.

Objective To investigate the influential factors of systemic lupus erythematosus(SLE) activity during pregnancy and their relationship with pregnancy outcome.Methods A retrospective analysis of the clinical history of 66 pregnant women with SLE from 1991 to 2005 was carried out.Results(1) Those patients with unstable status progestation,patients being newly diagnosed with SLE during pregnancy or patients irregularly using prednisone became active during pregnancy.The disease was active in 32 cases(the active group)and inactive in 34 cases(the inactive group).(2)Obstetric complications in the active group included:9 cases of preeclampsia,13 cases of fetal growth restriction(FGR),7 cases of therapeutic abortion and 15 cases of premature labor;and the corresponding numbers in the inactive group were 1,5,1 and 4, respectively.All the numbers were significantly different between the two groups(P

Because of the changed metabolic behaviors of cancer cells, tumor cells uptake a corresponding larger amount of glucose in physiological condition when compared with normal cells. And they were prone to metabolize glucose for generating energy in anaerobic glycolysis ways in order to grow quickly. Anaerobic glycolysis consumes more glucose than aerobic way when the same amount of energy is obtained, which also results in large demand of glucose in tumor cells. This review briefly describes therapy methods related to characteristic mentioned above, and summarizes the research progress of drugs, diagnostic reagents and carriers conjugated with glucose, glucose derivatives or other kinds of sugars for cancer targeting. Furthermore, typically relative research reports from 2012 till now were listed and analyzed.

Objective To probe the value of ultrasound in detection of malignant foci among multiple thyroid nodules. Methods Ultrasonograms of 83 patients with pathologically confirmed thyroid cancer combined with benign nodules were analyzed.Results Among 83 patients of thyroid carcinoma, 89 malignant and 87 benign nodules were detected with ultrasound. Significant differences of two-dimensional ultrasounic signs including low echoes, border fuzzy, micro-calcification and rear echo attenuation were found bteween malignant and benign nodules (P<0.01). There were significant differences in the grade Ⅱ, Ⅲ blood flow between malignant and benign ones (P<0.01). Of 89 malignant nodules, small thyroid carcinoma with diameter ≤1 cm accounted for 52.81% (47/89), and ultrasound diagnostic rate of all cancer nodules was 67.42% (60/89).Conclusion Ultrasounic characteristics including low echoes, border fuzzy, micro-calcification, rear echo attenuation and internal rich blood flow contribute to differentiate the thyroid microcarcinoma from multinodules.In particular, the microcarcinoma should not be ignored.

Objective To investigate whether fasting obestatin level is different in patients with impaired glucose tolerance or type 2 diabetes, and to explore the association between obestatin and lipid metabolism. Methods Eighty-four subjects without known diabetes were divided into three groups: normal glucose tolerance(NGT), impaired glucose tolerance (IGT) and type 2 diabetes (DM) Plasma obestatin levels were measured with a radioimmunoassay. The relationship between fasting obestatin levels and metabolic parameters was also analyzed. Results Fasting obestatin levels were lower in DM group [(2.82±0.78)ng/ml] and IGT group [(3.25±0.29)ng/ml] than in NGT group[(3.55±0.57) ng/ml, P＜0.01]. Triglycerides and low density lipoprotein cholesterol levels gradually increased among the three groups (P＜0.05). Multiple linear regression analysis revealed fasting obestatin level was independently associated with waist-to-hip ratio, triglyeride and low density lipoprotein cholesterol. The regression equation was obestatin=6.953-3.412×W/H-0.175×TG-0.123×LDL-C. Conclusions The decreased obestatin may be associated with IGR and T2DM, and obestatin level may be associated with lipid metabolism.

Objective To introduce and evaluate a new surgical technique about concealed penis.Methods The incision was chosen on the outer lamina of the bilateral sides of penis.The excision of fibrous cord of penis dartos and suprapubic fat pad and skin fixation on the penis root were done to the patients.For the severe patients,the superficial layer of the suspensory ligament of penis must be excised.Thirty-nine cases were treated with this new technique,22 cases with Devine technique,and 23 cases with modified Shiraki technique and clinical comparative research was carried out.Results Penis of 39 cases were found 2 to 3 centimeters pro-pubic projection.Within 3 to 36 months of follow-up,no obvious reduction of penis was found and the erection function was normal.By new evaluat standard of surgical efficacy,the clinical efficacy of new technique,Devine′s and modified Shiraki′s technique were 92.31%(36/39 cases),77.27%(17/22 cases) and 78.26%(18/23 cases),respectively and the efficacy rate in new technigue was significcmthy higher than that of Devines and modified Shirak′s groups(Pa
