Fifteen min after adding nimodipine 50 ?mol/L, the contractility of the left atrial muscle decreased from 100 % of control to 55.5 +4.2 o/o and its inhibitory effect was equal to the effect of verapamil (25 ?mol/L ) . Nim at the same concentration inhibited strongly the positive staircase phenomenon but could not reverse it into the negative staircase phenomenon as verapamil 25 ?mol/L did. The same concentration of Nim suppressed significantly the post rest contraction. On the contrary, the post rest contraction was not affected by verapamil at 25 ?mol/L. In addition, Nim 50 ?mol/L reversed or prevented significantly the ouabain-induced arrhythmias in isolated left atria. These results suggest that the negative ino-trbpism of Nim may be related not only to the inhibition of Ca2 + influx to the cells and also to the decrease of the intercellular Ca2+ release and that Nim might present the possibility in treating arrhythmias or other heart disease
5. Cloning and purification of paired immunoglobin-like type 2 receptor beta and preparation of its polyclonal antibody

Objective: To express and purify paired immunoglobin-like type 2 receptor beta (PILRβ) and prepare the polyclonal antibody against its protein. Methods: The PILRβ cDNA was amplified from the total RNAs extracted from human peripheral blood cells by RT-PCR and was cloned into pET-32a vector. The vector harboring PILRβ gene was then expressed in E. coli by IPTG induction and the product was purified by Ni2+-NTA affinity chromatography. Polyclonal antibody was prepared by immunizing rabbits with the purified protein. The titer and specificity of the antibody were examined by ELISA and Western blot, respectively. Results: Highly purified PILRβ protein (Mr 42 000) was obtained. The prepared polyclonal antibody was highly specific and had a titer of 1:10 000. Conclusion: PILRβ gene is successfully cloned and the purified PILRβ protein is expressed. Polyclonal antibody against PILRβ(with high titer and specificity) is successfully obtained, which provides a basis for further study on PILRβ.

Tibetan medicineLiu-Wei Mu-Xiang(LWMX) pill, a traditional compound medicine, has special therapeutic effects in the treatment of gastrointestinal anabrosis, especially chronic gastric ulcer. It has been widely applied in the clinics in Tibetan region. Nevertheless, the current standards for quality control are insufficient and incomplete, which makes it difficult to ensure its quality. This review focused on two closely related perspectives, which were the effective material basis and quality control, to summarize and review the recent advancement in relevant studies. This review also pointed out that the burning questions for Tibetan medicine standards were the origin confusion and“different medicines with the same name”. It offered meaningful suggestions for the consideration to improve its quality standards.
10.Silencing of Bcl-2 gene expression by siRNA transfection inhibits the protective effect of fluvastatin against cell apoptosis in human aortic endothelial cells

Objective To study the protective effect of fluvastatin,one of the HMG-CoA reductase inhibitors (statins),against oxygen radical-induced oxidative damages in human aortic endothelial cell,and the role of Bcl-2 in this protection.Methods Human aortic endothelial cells with or without Bcl-2 siRNA transfection were subjected to 1-100 nM of fluvastatin and 100 la hydrogen peroxide for 24 hours.Bcl-2 mRNA and protein expression were measured by Taqman quantitative PCR and Western blotting.Cell apoptosis was measured by normal and fluorescent microscopy and Cell Death Detection ELISA.Results In the Bcl-2-expressed cells,fluvastatin significantly reversed hydrogen peroxide-induced microscopic apoptosis and apoptotic DNA fragmentation,which were accompanied by a markedly upregulation of Bcl-2 expression by fluvastatin.However,the endothelial protection by fluvastatin was completely lost in Bcl-2 siRNA transfected cells.Conclusion Fluvastatin protects human endothelial cells against oxygen radical-induced cell apoptosis in vitro,and this protection seemed to be mediated in a Bcl-2 dependent pathway.(J Geriatr Cardil 12008;5:33-38)
