Chinese Journal of Organ Transplantation 2010;31(2):105-108
Objective To examine the inhibitory effect of IL-2-PE40 on the mouse corneal allograft rejection. Methods A mouse corneal graft model was set up by using C57BL/6 mice as donors and Balb/c mice as recipients. In the treatment group, IL-2-PE40 (0.6 μg/g body weight) was intraperitoneally injected from the day of surgery every 12 h until rejection happened. In the control group, the equal volume of PS was injected intraperitoneally at the corresponding time points. The transplanted cornea was observed under slit-lamp twice a week and the transplanted corneal opacity and neovascularization were rated according to Horis grading standards. It led to the determination of rejection response. The survival of transplanted cornea was regarded to be stopped when the rejection occurred. The operated eyes were observed historically on the 10th, 15th, 25th and 35th day after the surgery, and the peripheral blood was collected for measurement of T cell subgroups and T lymphocyte colonies. Results The survival time of cornea in the treatment and control groups was (30.2±2.9) days and (15.1±2.1) days respectively. In the control group, rejection occurred on the 15th day after the surgery, CD4~+ cells started rise right after the surgery and increased most obviously on the 15th day [(63.9±4.0)%] and decreased afterwards. CD4~+ cells in the treatment group were increased slightly [(42.6±4.0)%] on the 15th day. The number of CD4~+ cells in the treatment group was obviously less than in the control group (P<0.01). No significant changes in CD8~+ cells were observed in both groups. The number of T lymphocyte colonies in the control group was increased at the beginning and dropped then. No obvious change was found in the treatment group. The number of T lymphocyte colonies in the treatment group was significantly less than in the control group (P<0.01). Conclusion IL-2-PE40 is a highly specific immunosuppressive agent. It can delay the development of corneal graft rejection and reduce the percentage of T-helper cells significantly. It also works to weaken the forming capacity of the peripheral T lymphocyte colonies.

Purpose To detect whether a 3243 point mutation existed in age related macular degeneration (AMD). Methods Twenty six cases of wet form AMD patients, ten cases of dry form AMD patients were selected, and compared with twenty nomal controls. After collecting anti coagulated blood samples, total cellular DNA were extracted and purified. Using polymerase chain reaction and restriction fragment long polymorphism techniques, the mtDNA A→G point mutation at position 3243 were detected. Results After cleaveded by restriction endonuclease Apa I, a 294 bp fragment remained only in all detected DNA samples including twenty six wet form AMD, and ten dry form AMD. No any other fragment appeared. The result showed that there was no A→G mutation at position 3243 found in AMD. Conclusion It is suggested that mtDNA 3243 point mutation due to maternal inheritance might be not concerned with both wet form AMD and dry form AMD.

Objective To investigate the effect of liraglutide on the expression of NF-kappa B (NF-κB), tumor necrosis factor-alpha (TNF-α) and interleukin-6 (IL-6) in renal tissue of insulin resistance rats. Method The rats were divided into 4 groups, control group ,high fat group ,liraglutide low dose group [100 μg/(kg· d)], liraglutide high dose group[100 μg/(kg·d)]. Detection of serum creatinine and urea nitrogen by automatic biochemical analyzer, and 24 h urine micro albumin was determined by immunoassay. The protein concentration of serum NF-κB,TNF-α and IL-6 was measured by ELISA. The expression of NF-κB,TNF-α and IL-6 mRNA in renal tissues was determined by RT-PCR. Results Compared with the control group, the serum creatinine, blood urea nitrogen, urinary protein, and the expression of NF-κB, TNF-α and IL-6 were significantly higher in high fat group, respectively (P < 0.05). The glomerular basement membrance thickening and foot process fusion were seen under electron microscope. Liraglutide high dose intervention can significantly reduce these changes. Conclusions Liraglutide can inhibit the expression of NF-κB,TNF-α and IL-6 in renal tissues of high fat induced insulin resistance rats,in a concentration dependent manner, which possibly improve renal function.

Objective To analyze the levels and speciation of arsenic metabolites in urine of rats treated with sodium arsenite and sodium arsenate in order to investigate the different aspects of metabolism between sodium arsenite and sodium arsenate, thus to understand further the basic data about relationship between it's metabolism and mechanism of toxicity. Methods Seventy Wistar rats, weighting 80-120 g,were divided into 7 groups of 10 each, such as normal control group, high, middle and low sodium arsenite group and high, middle and low sodium arsenate group. After the animals were fed for one month, the urine was collected by metabolic cage in 12 hours. Applying the high efficiency liquid chromatography and hydride genesis atomic fluorescence spectroscopy (HPLC-HGAFS),the levels and speciation of arsenic metabolites were determined in urine of rats. Meanwhile, the recovery rate of dimethyl arsinic acid(DMA) would be determined to estimate the degree of accuracy of results. Results The levels of iAs~(3+),iAs~(5+) and DMA in middle sodium arsenite group[(121.66±1.26),(10.26±2.68),(200.91±0.56) μg/L]were higher than the high sodium arsenite group[(113.20±0.75),(5.16±1.32),(147.70±μ0.77)μg/L,all P < 0.05]and low sodium arsenite group[(79.35±2.12),(5.13±2.25),(56.35±1.23)μg/L,all P < 0.05]. The levels of iAs~(3+) and DMA in middle sodium arsenate group[(315.81±1.69),(245.12±1.18)μg/L]were higher than the high sodium arsenate group[(85.03±0.56),(110.34±1.04)μg/L,all P< 0.05]and low sodium arsenate group[(22.97±2.67),(15.75±2.15)μg/L,all P < 0.05]. Compared with sodium arsenate group, the levels of iAs~(3+) and DMA in high and low sodium arsenite group were higher(all P < 0.05) ; and the levels of iAs~(3+) and DMA in middle sodium arsenite group were lower(all P < 0.05). Meanwhile, the average urinary recovery rate of DMA of rats in different sodium arsenite group were 94.80%-102.70%,and the average urinary recovery rate of DMA of rats in different sodium arsenate group were 95.33%-108.40%. Conclusion The speciation and levels of arsenic are influenced by the external exposure dose, and some distinction appeared in the metabolism and metabolic path between sodium arsenite and sodium arsenate in urine in vivo.

OBJECTIVETo study on the reliability of the Akagi line as a reference axis to guide for rotational alignment of the proximal tibial component in total knee arthroplasty (TKA) the rotational alignment reference bony landmarks of the proximal tibial component on magnetic resonance image (MRI) were measured.
METHODSFrom January 2010 to December 2013, 80 normal knees of Chinese volunteers including 35 males and 45 females with an average age of (35.4±6.1) years were reviewed. The images of the knees were obtained by MRI. The surgical epicondylar axis (STEA) was identified in the femoral transverse sections and then was projected to the side of tibia, forming the SETA'. A line connecting the medial border of the patellar tendon and the middle of the posterior cruciate ligament insertion (Akagi line) and its vertical line (AK), as well as a line connecting the medial 1/3 of the patellar tendon and the middle of the posterior cruciate ligament insertion and its vertical line (AP), were identified in the tibial transverse sections. The angles were measured between the AK, AP and STEA'.
RESULTSThe angle between AK and STEA' was (0.59±2.07)°, and there was no significant difference between the two lines (t=-2.54, P=0.13). The mean angle between AP and STEA' was (3.21±2.04)°, and there was a significant difference between the two lines (t=14.05, P<0.05). There was a significant difference between the AK and AP (t=-11.68, P<0.05).
CONCLUSIONThe reliability of the Akagi line as a reference axis to guide for rotational alignment of the proximal tibial component in TKA is good.
