5.Experimental study of emodin inducing apoptosis of bile duct carcinoma cell QBC939
Journal of International Oncology 2012;39(4):316-320
Objective To investigate the effect and mechanism of emodin inducing the apoptosis of human bile duct carcinoma cell QBC939 in vitro. MethodsThe inhibition of cell proliferation was detected by MTT. The morphological changes of apoptotic cells were observed under fluorescence microscopy. The apoptosis rates and intracellular reactive oxygen species levels were detected by flow cytometry. The intracellular relative activities of caspase-9 and caspase-3 were tested through colorimetric method.ResultsEmodin inhibited cell proliferation of human bile duct carcinoma cell QBC939 in time-dose dependent fashion.Apoptotic cells displayed bright, nuclear presented divided leaves, debris and a set of edges under fluorescence microscopy.Treated with 30 μmol/L and 50 μmol/L emodin, the apoptosis rates of 24 hours were respectively 38.9% ± 9.07% and 67.09% ± 4.08% (P < 0.05).The intracellular reactive oxygen species levels after 30 minutes treatment were 1.65 ± 0.08 and 2.28 ± 0.04folds of the control group (P < 0.05). Emodin could activate caspase-9 and caspase-3,leading to elevations of their activities (P < 0.05).ConclusionEmodin inhibites cell proliferation of human bile duct carcinoma cell QBC939 through inducing apoptosis. The mechanism is associated with the elevated levels of reactive oxygen species and the activation of caspase-9 cells and caspase-3.
6.Research progress of side population cells
Journal of International Oncology 2011;38(8):579-582
In the occurrence and development of tumors, side population cells play an extremely important role. They have the characteristics of cancer stem cells, especially their potential of tumor originating, and stronger drug-resistance. The study of biological characteristics, sorting and training methods of side population cells and the relationship between side population cells and tumor drug-resistance may shed some light on the diagnosis and treatment of cancer.
7.Research progresses on the pathophysiology of hemorrhoidal diseases
Chinese Journal of Pathophysiology 2010;26(1):193-196
Various hypotheses have been put forward to explain the pathogenesis of hemorrhoidal diseases. However, the etiology and mechanisms of hemorrhoids are far from clear, and as a consequence the present therapeutic objective is just to relieve or abolish the symptoms of hemorrhoids. In this review, the recent research advances on the pathophysiological characteristics that are closely associated with hemorrhoids are analyzed and discussed, which include constipation, high anal resting pressure, anal mucosa damage, aging of the anal cushion supporting tissue, blood and vessel alterations, obstruction of microcirculation and biochemical changes.
8.Brain natriuretic peptide: a potential indicator of cardiomyogenesis after autologous mesenchymal stem cell transplantation?
Journal of Zhejiang University. Science. B 2006;7(9):766-768
We observed in a pilot study that there was a transient elevation of brain natriuretic peptide (BNP) level shortly after the transplantation in the patient with ischemic heart failure, which is unexplainable by the simultaneous increase of the cardiac output and six-minute walk distance. Similar findings were observed in the phase I trial. We postulated on the basis of the finding of Fukuda in vitro that this transient elevation of BNP level against the improvement of cardiac function and exercise capacity might indicate cardiomyogenesis in patients after mesenchymal stem cell transplantation. Further study is warranted to verify the hypothesis.

9.Randomized-controlled observation of naolibao in improvement of memory impairment in aged people with yin deficiency of liver and kidney
Chinese Journal of Tissue Engineering Research 2006;10(3):163-166
BACKGROUND: There are many reports on treatment of senile memory impairment with huperzine A, aniracetam, ginkgobiloba, etc. But, a strict double-blind and double-dummy design is deficit during experiment.OBJECTIVE: With cognition, symptoms of yin deficiency of liver and kidney and memory taken as observed indexes, double-blind and doubledummy method was used to observe the therapeutic effects of Chinese drug naolibao on senile memory impairmentof yin deficiency of liver and kidney. DESIGN: Prospective clinical experiment with randomized controlled double-blind and double-dummy method was designed. SETTING: Department of Neurology of Affiliated Hospital with Changchun College of Chinese Medicine, Department of Medicament of 2nd Clinical Hospital Affiliated to Jilin University.PARTICIPANTS: Totally 80 cases of memory impairment were from the clinic of Department of Neurology of affiliated Hospital with Changchun College of Chinese Medicine from April to December 2000. They were aged varied from 40 to 75 years, with mild non-constant poor memory, differentiated as yin deficiency of liver and kidney and diagnosed as questionable dementia in Chinese medicine, scored as 0 or 0.5 according to Clinical Dementia Assessment Scale (CDAS) and their integrals were > 17 (illiteracy) or > 20 (elementary-school educated) or > 23 (juniorschool educated) and < 30 according to MMSE (mini mental status examination).METHODS:The patients were randomized into experimental group (n=43) and the control (n=37). In experimental group, naolibao wan was administrated, composed of dihuang (Radix Rehmanniae), tusizi (SemenCuscutae), digupi (Cortex Lycii), yuanzhi (Radix Polygalae), shichangpu (Rhizoma Acori Graminei) and fuling (Poria), 4 pills/times, 3 times/day,and piracetan blank tablet was given, 2 tablets/time, 3 times/d. In the control, piracetan tablet 400 mg/tablet was prescribed, two tablets/time,3 times/day and naolibao blank pill was administrated, 4 pills/time,3 times/day. The course of treatment was 6 weeks. On observed spot, named base-line spot (before treatment) and terminal spot (after treatment), the integrals were examined and recorded on symptoms, physical signs and neuropsychology successively. Follow-up visit was carried on for the cases with remarkable effects. Therapeutic effects of syndromes in Chinese medicine were based on the evaluating criteria of Clinical Plan of Treatment with Naolibao Wan for Senile Memory Disturbance of Yin Deficiency of Liver and Kidney. In comparison of decreased symptomatic integral after treatment with average symptomatic integral before treatment, remarkable effect: ≥ 2/3, effect: 1/3-2/3, no effect: ≤ 1/3. The therapeutic effects on cognition and memory were assessed by the formula [(integral after treatment-integral before treatment)÷integral before treatment] ×100%. Clinical control: ≥ 85%, remarkable effect:≥ 50%, effect: ≥ 20% and no effect: ≥ 20%.MAIN OUTCOME MEASURES: ① Comparison before and after treatment of yin deficiency of liver and kidney in two groups. ② Comparison of integrals before and after treatment for yin deficiency of liver and kidney of two groups. ③ Comparison of evaluation of every index for memory in two groups. ④ Results of follow-up visits for the cases with remarkable effects. ⑤ Harmful accident and side effects.RESULTS: Totally 43 cases in experimental group and 37 cases in the control were all employed in result analysis. ① Comparison before and after treatment of yin deficiency of liver and kidney in two groups: The reduced score in differentiation of symptoms, improvement of severity of yin deficiency of liver and kidney and promoted therapeutic effects on syndrome in experimental group were superior to those in the control, indicating significant difference (P < 0.05-0.01). ② Comparison of integrals before and after treatment for yin deficiency of liver and kidney of two groups: The improvement of dream-disturbed sleep, tinnitus and deafness in experimental group was superior to that in the control, indicating significant difference (P < 0.05-0.01). ③ Comparison of evaluation of every index for memory in two groups: The improvement of immediate recall and retarded recall in experimental group was superior to that in the contol, indicating significant difference (P < 0.05-0.01). ④ Results of follow-up visits for the cases with remarkable effects: Three months later, the disease was not aggravated among the cases with remarkable effects in two groups.⑤ Harmful accident and side effects: After treatment, there was no abnormal alternation and harmful reaction for safe indexes compared with those before treatment, such as blood routine, urine routine, feces routine, liver function, kidney function, etc.CONCLUSION: Naolibao wan improves memory, immediate recall, recog nition, dream-disturbed sleep, tinnitus and deafness for aged people, which is superior to piracetan tablet and it is safe and reliable in treatment.

International Eye Science 2014;(6):1089-1091
AIM:To analyze relevancy between the changes of the retinal thicknesses of children's anisometropic amblyopic macular at Area A1 before and at the end of amplyopia cure and amplyopia cure effect.
METHODS: Through randomly selecting 30 cases ( 60 eyes) of anisometropic amblyopic children as test group and 29 cases ( 58 eyes ) of children with normal vision after the vision correction as control group, the changes of the retinal thicknesses of macular of the test group and the control group at Area A1 by OCT-3 was observed dynamically and analyzed.
RESULTS: Compared with first diagnosis, the comparative difference of retinal thicknesses of macular at Area A1 in control group at 3, 6mo has statistical significance. The test group was divided into amblyopia group and non-amblyopia group. There has statistical significance for the comparative difference between the retinal thicknesses of macular centre at 3, 6mo and the first diagnosis in the non - amblyopia group, the amblyopia group was divided into the invalid cure group, progress group and the cured group according to amblyopic cure effect. Compared with first diagnosis, there is no statistical significance of macular at Area A1 in invalid cure group and progress group at 3, 6mo, and in the cured group at 3mo, but in the cured group at 6mo has statistical significance.
CONCLUSION: There is a certain relationship between the changes of retinal thicknesses of macular of children with anisometropic amblyopic at Area A1 before and at the end of amblyopia treatment and amblyopia cure effect. OCT is a kind of objective auxiliary test method and provides new clues for amblyopia research.
