2.CONSTRUCTION OF THE PLASMID pKChiA AND pMchiA CARRYING CHITINASE GENE AND THEIR EXPRESSION IN E.coli

A 1.8 kb HinfI fragment carrying the chitinase gene(chiA) from Serratia marcescens was isolated from the plasmid pLCHIA and was inserted into the SmaI site downstream of the strong Ptac promoter in the expression vector pKK223-3,yielding the plasmid pKChiA.The 2.1 kb PtacChiA fusion fragment was excised by BamHI from the plasmid pKChiA and was inserted into the single BamHI site present within the plasmid pMC71A,thus generating the plasmid pMChiA.High levels of chitinase were produced by the E.coli strains HB101 and JM105 carrying the plasmid pKChiA or the plasmid pMChiA,with the amount of production were higher 1~3 fold than that produced by the strains carrying the plasmid pLCHIA.
3.Retroperitoneal Versus Transperitoneal Laparoscopic Partial Nephrectomy:A Systematic Review and Meta-analysis

Objective To review published literatures comparing the safety and effectiveness of retroperitoneal laparoscopic partial nephrectomy (RLPN) with transperitoneal laparoscopic partial nephrectomy (TLPN) and provide reference for clinical work. Methods The search strategy was performed to identify relevant papers from the Cochrane Central Register of Controlled Trials, EMBASE, MEDLINE, Google Scholar, China Hospital Knowledge Database, Wangfang Chinese Periodical Database, and VIP Chinese Periodical Database. All papers comparing RLPN with TLPN were included from 2000 to 2015.Two to three reviewers independently screened, evaluated, and extracted the included papers. A Meta-analysis was executed by using Review Manager 5.3 software. The interesting outcomes were tumor size, operating time, estimated blood loss, warm ischaemia time, length of hospital stay, positive margin rate, open conversion rate, overall complication rate, and recurrence rate. Results The literature search obtained 378 papers, then 10 of them were ultimately met the inclusion criteria and included in the systematic review. Finally, 6 of the 10 papers were included in the Meta-analysis. RLPN had significantly less operating time [P= 0.01, mean difference (MD)=-33.68, 95% confidence interval (CI) within (-60.35,-7.01)] and shorter length of hospital stay [P< 0.0001,MD=-1.47, 95%CI within (-2.18,-0.76)] than TLPN. Significant differences were not found between RLPN and TLPN in other outcomes. Conclusions RLPN may be equally safe and be faster than TLPN. Each center can choose a modality according to your own operating habits and experience.

6.Urgent management of quadriplegia following Sichuan earthquake under field condition

Objective:To explore the urgent management strategy for quadriplegia following China Wenchuan earthquake under filed condition.Methods:The 57-year-old male patient was sent to the earthquake relief team of Changzheng Hospital,Second Military Medical University.He was hit by heavy object during the Sichuan earthquake 6 days ago.Physical examination revealed cervical spinal cord injury accompanied by incomplete paralysis.The patient underwent cervical 3-7 laminectomy under general anesthesia.Results: After surgery,the muscle strength of lower limbs recovered to grade four from grade one;the muscle strength of upper limbs recovered to grade five from grade three.Sensory loss was found below the nipple level and totally recovered after operation.Conclusion:Early surgery for quadriplegia following earthquake trauma can timely relieve compression to the spinal cord and therefore improve prognosis of patients.Under field condition where facilities are not readily available,decompression of spinal cord can be achieved by lamineetomy via posterior approach.The key to successful operation is correct diagnosis and skillful manipulation.
7.Association between cervical curvature and cervical sympathetic symptoms.

OBJECTIVETo investigate the association between cervical curvature and cervical sympathetic symptoms.
METHODSThe clinical data of 318 patients with cervical spondylosis who underwent surgical treatment in our department between July 2003 and December 2007 were retrospectively analyzed. All patients were divided into group without sympathetic symptoms (n = 284) and group with sympathetic symptoms (n = 34) . The curvatures of both groups on cervical lateral radiographs were measured using Borden method and statistical analysis was performed.
RESULTSThe incidence of abnormal cervical curvature in group with cervical sympathetic symptoms were 67.6% (23/34), which was significantly higher than that in group without cervical sympathetic symptoms (50.7%, 144/284) (P < 0.05).
CONCLUSIONCervical curvature abnormality may be an independent factor that affects the cervical sympathetic symptoms.

8.The influence of acupuncture on Nestin expression in the cortex of hypoxia-ischemic neonatal rats

Objective To investigate the influence of acupuncture on Nestin expression in the cerebral cor- tex of hypoxia-ischemic neonatal rats.Methods A total of 72 Sprague-Dawley(SD)rats aged 7 days were random- ly divided into three groups:A normal group(n=24),the hypoxic-ischemic encepbalopathy(HIE)group(n=24), and an acupuncture+HIE group(n=24).The rats were sacrificed at time points including 3d,7d,14d and 28d af- ter insult,their brains were removed and sections were made.Any nerve stem cells(NSCs)in the cerebral cortex were detected using immunohistochemical staining of Nestin.Results Nestin-positive cells were found in the cere- bral cortex in the normal neonatal SD rats early after birth.After HI insult,the Nestin-positive cells had increased in the injured areas of the cortex and in contralateral mirror areas,especially around the injured areas.Peak Nestin ex- pression was 3 days after insult.In the acupuncture+HIE group,Nestin positive cells were significantly increased when compared with that in the HIE group,the peak was postponed,and the peak was longer than in the HIE group. The Nestin positive cells decreased,were seldom seen,and were not different among three groups 28 days after the HI insult.Conclusions Endogenous neural stem cells in the cerebral cortex were increased by acupuncture in hy- poxia-ischemic in neonatal rats,and acupuncture made the peak of Nestin expression longer.
9.Oligonucleotide array of genomic expression in complete androgenic hydafidiform mole

Objective To compare genomic expression differences between androgenic complete hydatidiform mole (AnCHM) and normal first trimester villi with similar gestation weeks,and search for potential adjuvant diagnostic molecular markers.Methods Short tandem repeat (STR) detection was used to identify AnCHM,human oligonucleotide array U133 Plus 2.0 was used to measure genomic expression differences between AnCHM and normal villi,and quantitative fluorescent RT-PCR was used to verify array of several genes.Results Nine of 11 histologically diagnosed complete hydatidiform moles were found to be AnCHM by means of STR,and the other 2 were biparental complete hydatidifonn mole (BiCHM). Compared with villi,oligonueleotide array showed 279 genes (0.72%,279/38 500) were over expressed and 1710 genes (4.44%,1710/38 500) under expressed in AnCHM.Bioinformatics analysis found that differentially expressed genes were involved in multiple biological processes and pathways.Changes of imprinting genes,growth hormone genes and chorionie somatomammotropin hormone genes were especially remarkable.Conclusions Pathogenesis of AnCHM is a complex process involving multiple genes and pathways.Altered expression of imprint genes may play important roles in the process.
10.Chemical constituents from the twigs and leaves of Harrisonia perforate.

This study was performed to investigate the chemical constituents in the twigs and leaves of Harrisonia perforate. Six compounds were isolated from the 95% EtOH extract of the twigs and leaves of Harrisonia perforate by silica gel, ODS, Sephadex LH-20 column chromatographies and preparative HPLC. On the basis of chemical properties and spectra data, these compounds were identified as harriperfin E (1), kihadanin A (2), kihadanin B (3), 6α-acetoxyobacunol acetate (4), gardaubryone C (5), and β-sitosterol methyl ether (6), respectively. Compound 1 is a new chromone, and compounds 2-6 are isolated from this plant for the first time.
