1.Prokaryotic Expression of eBCMA-Fc Fusion Protein, a Soluble BLyS Receptor, with Computer-aided Molecular Design in E. coli

B cell maturation antigen (BCMA) is a receptor of B lymphocyte stimulator (BLyS). Human IgG1Fc fusion proteins with the extracellular domain of BCMA(eBCMA), also called decoy receptors, have beenused as a potential BLyS antagonists to block BLyS activities. In order to design novel BLyS antagonistpeptides, computer-aided homologue modeling was used to construct an eBCMA-Fc fusion protein based on thecrystal structures of BCMA and Fc fragmant. To ensure the activity of eBCMA not to be interfered by Fcfusion, the root mean square distance (RMSD) for eBCMA and Fc were calculated to be 0.036 nm and 0.064nm, respectively, based on molecular docking modeling. An eBCMA-Fc fusion gene was constructed andintroduced into E. coli for expression. As expected, the purified 36 kD eBCMA-Fc fusion protein was able tobind BLyS in vitro at a dosage-dependent manner and demonstrated an anti-proliferative activity induced byBLyS in Daudi cells. The results have provided useful information on the evaluation of computer modeling andthe in vitro biological activity for the design of potential BLyS antagonist peptides.
2.Astaxanthin inhibits sodium azide-induced cytotoxicity in hepatocyte L-02 cells probably by H+ transferring function.

This study is to investigate the protective effect of astaxanthin against injured hepatocyte L-02 cells induced by sodium azide (NaN3) and reveal the possible mechanisms. Hepatocyte L-02 cells were exposed to 100 mmol.L-1 NaN3 with various concentrations of astaxanthin pre-incubated, then the cell viability was measured by MTT method; The level of reactive oxygen species (ROS) was determined by DCFH-DA method; The changes of mitochondrial membrane potential (MMP) and apoptosis ratio were detected by JC-1 method and Annexin V-FITC/PI double stain method, respectively. Results showed that after cells were exposed to 100 mmol.L-1 NaN3 for 3 hours, the cell viability significantly decreased; ROS level and the percentage of late phase apoptosis increased obviously; MMP was also declined. When cells were pretreated with astaxanthin, the cell damage and late phase apoptosis ratio reduced and MMP was maintained. However, the level of ROS showed insignificant decrease (P>0.05). The beneficial concentration of astaxanthin in improving cell viability and MMP was not in a dose dependent manner and the most effective of which was 0.10 nmol.L-1 (P<0.01). In order to reveal its possible non-antioxidant mechanism, mitochondrial membrane was imitated and H+ transferring function of astaxanthin was also detected by bilayer lipid membrane (BLM) method. Results showed that 2.0% astaxanthin could transfer H+ efficiently. These suggested the mechanisms of astaxanthin in protection of hepatocyte L-02 cells not via its ROS quenching capability but via its H+ transferring function, which improved the mitochondrial function and had the sequence biology effects.
4.Improvement and Application of Drug Procurement Management Module in Our Hospital

OBJECTIVE:To improve the drug turnover rate and the work efficiency of drug storehouse,and to reduce the cost of drug storage. METHODS:Based on ABC dynamic classification,original drug procurement management module in HIS was im-proved. Scientific,accurate and reasonable drug procurement plan was generated by the system automatically through setting pro-curement proportion coefficient. Drug procurement by type was achieved through adding classification scanning function,based on distribution mode and drug property classification. Related index were compared before and after improvement to evaluate the ef-fects of the improvement. RESULTS:Compared with before improvement,average monthly inventory amount of drugs in our hos-pital reduced by 6.44%,turnover rate increased by 47.13%(increasing from 4.18% to 6.15%),and the time for developing pro-curement plan decreased by 75.00%(decreasing from 8 h to 2 h). CONCLUSIONS:The improvement of drug procurement man-agement module effectively reduce drug inventory and improve turnover rate of drugs and work efficiency.
5.The value of ankylosing spondylitis disease activity score in evaluating disease activity of ankylosing spondylitis

Objective To investigate the value of ankylosing spondylitis(AS)disease activity score (ASDAS)and the Bath AS disease activity index(BASDAI)in evaluating disease activity in AS.Methods ASDAS and BASDAI were used to evaluate disease activity in patients with AS,taking the patients' global scores,physician global scores and treatment decision as the “gold standard” for disease activity.According to the “gold standard”,the patients were divided into low and high disease activity groups.Statistical analysis included descriptive statistics,linear correlation and regression and ROC curve.Results A total of 227 patients with AS were included.ASDAS and BASDAI showed good correlation with disease activity as reflected by the patients' global score[r for BASDAI,ASDAS based on ESR(ASDAS-ESR)and ASDAS based on C-reactive protein(ASDAS-CRP)were 0.713,0.698 and 0.725,respectively,P =0.000]and the physician global score(r for BASDAI,ASDAS-ESR and ASDAS-CRP were 0.771,0.782,0.847,respectively,P =0.000).Both scores showed good discriminative ability for high and low disease activity states.ASDAS-CRP had the highest sensitivity.Conclusion ASDAS is a promising tool for evaluating disease activity in AS patients.
6.The diagnostic value of musculoskeletal ultrasound in gouty arthritis

Objective To observe the characteristic features of musculoskeletal ultrasound (MSUS) in gouty arthritis (GA) and to assess its value in GA diagnosis.Methods Patients with GA and non-GA who underwent MSUS examination in Chinese PLA General Hospital were enrolled in this study. The synovitis,tenosynovitis,soft tissue swelling,bone erosion,bursitis and ossification were measured by greyscale ultrasound,and the synovial vascularity was measured by power Doppler ultrasound to evaluate the degree of inflammation.Results The ultrasound images of GA were intended to have specific features like tophus,hyperechoic deposition on the surface of the articular hyaline cartilage ( double contour sign ),hyperechoic deposition in the synovium appearing as bright stippled foci,and bright strap in tendons with a significant difference when compared with non-GA group (P < 0.05).The percentage of tenosynovitis and bone erosion in non-GA group was higher than that in the GA group with a significant difference (P <0.05 ). Tophus,double contour sign,bright stippled foci or bright stippled foci in tendons together suggested gout with a specificity of 92.4% and a sensitivity of 60.3%,the AUC of ROC was 0.764 ( P <0.05).Conclusion Tophus,double contour sign,bright foci and bright strap detected by MSUS are of diagnostic value on GA and MSUS may be a promising tool in diagnosing GA.
7.CT Diagnosis of Primary Retroperitoneal Neoplasm

Objective To study CT diagnosis and differential diagnosis of primary retroperitoneal neoplasm(PRN).Methods CT data in 23 cases of PRN confirmed by pathology were retrospectively analysed.Results There were 11 results of pathology in 23 cases,of them,14 cases were mesenchymal tissue-origin(12 cases were malignant neoplasm),4 cases were nervous tissue-origin(all benign),4 cases were rudimental embryonal tissue-origin(3 cases were benign diseases and one case was malignant),the source of unknown-origin in one case was malignant neoplasm.Most of these PRN showed such common CT manifestations as other tumors in the same location,but they had own special CT features respectively.Conclusion PRN have many typies,CT features of PRN in combination with the history,age and location can improve the diagnosis of histological classification and the diagnosis of the nature of tumors.
8.The difference of ADM binding protein in human breast cancer cell line MCF-7/Wild and MCF-7/ADM Cell

Aim To study the mutidrugresistance mechanism of tumor cell by comparing the binding pro-tein with Adriamycin(ADM)in the MCF-7/Wild cell and MCF-7/ADM resistance cell.Methods The biotin-ADM compound was synthesized with the esterification-dehydration method,purified with HPLC and analyzed with mass spectrograph.The binding proteins against Adriamycin from MCF-7/ADM or MCF-7/Wild cell whose resistance index is above 200 were obtained respectively by the affinity screening magnetism bead method with biotin-ADM compound as a specific ligand.Some binding proteins with Adriamycin were identified with the MALDI-TOF-MS after isolating them with SDS-PAGE.Cell motor ability was compared in the migration experiment.Results The variability of biotin-ADM and ADM was negligible,while that IC50 of the two compounds to MCF-7/Wild was 0.796 ?mol?L-1 and 0.547 ?mol?L-1 respectively.The 20 types binding proteins with ADM were found from MCF-7/Wild cell,and 17 types binding proteins from MCF-7/ADM resistance cell.There was one protein band difference from the two cells on SDS-PAGE with silver staining.The same binding proteins on the two cells that identified with MALDI-TOF-MS were myosin,beta-actin,alpha-actin,keratin.MCF-7/W and MCF-7/ADM both migrated after being cultivated 72 h,with a faster migration of MCF-7/ADM.Conclusion There are different binding protein with ADM on the MCF-7/Wild cell and MCF-7/ADM resistance cell.The same binding protein with ADM on the two cells belongs to the cystoskeleton protein.The binding of the ADM with the cell cystoskeleton protein has some effect to the migration of the MCF-7/ADM.
9.Analysis of correlation between dry eye and diabetic retinopathy in type 2 diabetic patients

?AIM:To investigate the correlation between dry eye and different degrees of diabetic retinopathy ( DR ) in type 2 diabetic patients.?METHODS: In the cross-sectional study, 340 patients (340 eyes) with type 2 diabetes were enrolled. Tear film function tests including tear meniscus height, tear film breakup time ( BUT ) , fluorescein staining, Schirmer Ⅰtest were performed followed by surveying questionnaires about dry eye. Retinal status was evaluated by retinal color photography and indirect ophthalmoscopy exam with dilated pupils to evaluate DR and whether companied by macular edema.?RESULTS:The prevalence of dry eye was 49. 41%. The mean duration of diabetes in patients with dry eye was 11.15±7.07a, while 6.92±5.45a without dry eye(P<0.01). Dry eye had the positive correlation to the development of DR. The incidence of dry eye in people with mild nonproliferative diabetic retinopathy ( NPDR ) , moderate NPDR, severe NPDR and proliferative diabetic retinopathy (PDR) was 1. 097 times, 1. 724 times, 2. 86 times and 5. 43 times respectively, compared with people without DR. The occurrence of dry eye in people with macular edema increased by 3. 697 times compared with people without macular edema.?CONCLUSION: Dry eye was more prevalent in people with type 2 diabetes. The incidence of dry eye increased gradually with the occurrence and development of diabetic retinopathy.
10.The C825T polymorphism of GNB3 gene effects the response to 15-days-treatment of adalat

AIM To study the relationship between the response of hypertension patients to Adalat and the C825T polymorphism of the GNB3 gene. METHODS 554 hypertension subjects were treated with Adalat. Epidemiological and clinical data were collected. Genotypes of the GNB3 gene were measured by the PCR RFLP method. Linear and logistic regression were used to analyze the association between GNB3 genotypes and the response to Adalat. RESULTS The frequency of the 825T allele in our hypertensive group (48 4%) was slightly higher than that of the normal Chinese reported previously by Siffert W et al. (42%). The average SBP decrease after 15 days of Adalat treatment of subjects with the T allele was significantly less than that of the subjects with the CC genotype (?=-3 3, P= 0 03). Using logistic regression, the subjects with the T allele were less likely to reach good treatment result compared to those with the CC genotype (OR=0 33, P =0 02). CONCLUSION The GNB3 gene C825T polymorphism is related to the response to Adalat.
