1.Experimental Study of Percutaneous Absorption of Neostigmine Cream in Vitro

OBJECTIVE:To study the in vitro percutaneous absorption of neostigmine cream.METHODS:Using isolated mouse skin as permeation barrier,the influence of different concentrations of azone on percutaneous permeation of neostigmine was studied.RESULTS:Azone could obviously enhance the percutaneous permeation of neostigmine,and the accumulative permeation quantity of neostigmine cream containing2%and5%azone increased7.17%and11.87%respectively compared with that of cream without azone.CONCLUSION:The percutaneous permeability of neostigmine cream is satisfactory and the best concentration of azone in enhancing permeation seems to be5%.
2.Determination of the Serum Concentration of Buspirone by HPLC in Rabbits

OBJECTIVE:To establish a HPLC method for determination of the concentration of Buspirone in serum.MET_ HODS:The chromatographic system consisted of ZORBA SB-C 18 column and mobile phase of methanol-acetonitrile-water-0.1mol/L NaAC buffer solution(pH4.5)(10∶38∶51∶1),with a detection wavelength of237nm,and the content was calcu?lated by peak area internal standard method.RESULTS:The linear range of Buspirone was1~80ng/ml,r=0.9979.CONCL_ USION:This method is simple and accurate for determination of Buspirone in serum.
3.Investigation of the 7-year System Medical Students' Extracurricular Reading and its Enlightenment

After investigating conditions of 210 seven-year system medical students' extracurricular reading,we discovered that most medical students were rather interested in extracurricular reading,but there existed certain problems in the aspects of the arrangement of time and choice of books etc,which not only lowered the quantity of the student' extracurricular reading,but was disadvantageous to the effective display of the humanities education.Therefore,it was necessary to enhance the seven-year system medical students' extracurricular reading.
4.Evidence-based medicine proof and development trends of first-line treatment for advanced hepatocellular carcinoma

Tyrosine kinase inhibitors (TKIs) are used as the primary first-line treatment for advanced hepatocellular carcinoma, and the combination of immune checkpoint inhibitors (ICIs) and angiogenesis inhibitors have also been recommended as first-line treatment recently. For hepatocellular carcinoma patients with portal vein tumor thrombus, hepatic artery infusion chemotherapy (HAIC) is supported by progressively more evidence in improvement in the overall survival benefit. Based on relevant literatures and combined with clinical practices, the authors investigate the clinical application and development trends of TKIs, ICIs and HAIC in the first-line treatment for advanced hepatocellular carcinoma.
5.Curative effect of interference current and direct current pharmaceutical iontophoresis in patients with retrograde gonarthritis

Objective To observe and analyse the efficacy of retrograde gonarthritis with interference current and drug-iontophoresis direct current. Methods 185 patients with retrograde gonarthritis were randomly divided into treatment group 92 cases and control group 93 cases. The patients in treatment group underwent interference current and drug-iontophoresis direct current and patients in control group performed modulated medium frequency drug-ionto-phoresis direct current for two treatment courses. Results The comprehensive score of various kind of symptom such as pain、swelling、flexion、walking ability, up-down stair ability in treatment group significantly improved than that of control group (P < 0.01). Conclusion The interference current and drug-iontophoresis direct current is one of effec-tive and safe measurement in treating arthritis with main manifestation of pain and soft tissue disease.
6.The effects of H.pylori and its crude extracted proteins on isolated rabbit parietal cells acid secretion

Objective To explore the effects of H.pylori and crude extracted proteins secreted by H.pylori(broth culture filtrate protein,BCF-P)on acid secretion from isolated rabbit parietal cells.Methods Parietal cells from rabbit gastric mucosa were isolated and enriched with digestion and elutriation.H.pylori(NCTC 11637,CagA+ VacA+)were grown in liquid broth culture and BCF-P was precipitated with ammonium sulfate.The vacuolation activity of BCF-P was evaluated with neutral red dye uptake test in HeLa cell.Isolated parietal cells were incubated with H.pylori(bacteria/cell=100∶1)for 2 h and 16 h,or BCF-P(100μg/ml)for 1 h and 12 h.Acid secretion from parietal cells was studied using 14C-aminopyrine(14C-AP)accumulation indirectly and H+-K+ ATPase α subunit mRNA expression was assessed using RT-PCR.Results (1)BCF-P containing vacuolating cytotoxin(VacA)with vacuolation activity on HeLa cells had positive result on neutral red uptake test.(2)The basal expression of H+-K+ ATPase α subunit mRNA could be detected in isolated parietal cells and 14C-AP accumulation was significantly increased in response to the stimulation of histamine with different concentrations for 30 min(P<0.05).These results indicated that the isolated parietal cells retain relative intact acid secretion function.(3)The histamine(1.0×104 mol/L)stimulated acid secretion was inhibited sustainedly in response to H.pylori by 81% at 2 h and by 94% at 16 h(P<0.05).However,H+-K+ ATPase α subunit mRNA expression was up-regulated in tlle acute period(2 h)and was down-regulated in the chronic period (16 h)by H.pylori(P<0.05).(4)BCF-P significantly inhibited the histamine-stimulated acid secretion by 24% at 1 h and by 58% at 12 h(P<0.05),and this inhibition was accompanied by the down-regulated expression of H+-K+ATPase α subunit mRNA.Conclusions Intact H.pylori and VacA secreted by H.pylori could directly inhibit histamine-stimulated acid secretion from parietal cells and this inhibition may be mediated by the down-regulated H+-K+ ATPase expression.
7.Experimental study on Helicobacter pylori-related gastric carcinogenesis mediated by interleukin-1β

Objective To investigate the potential mechanism of interleukin-1β(IL-1β)in H.pylori-related gastric carcinogenesis.Methods Human gastric cancer cell line(AGS),human gastric epithelial cell line(GES-1)and isolated parietal cells were treated with exogenous IL-1β(10 ng/ml)in the presence or absence of H.pylori(NCTC 11637).Cell proliferation and apoptosis were determined by MTT assay and flow cytometry,respectively. Expressions of cyclooxygenase-2(COX-2)mRNA and protein were examined using RT-PCR and flow cytometry,respectively.Acid secretion by parietal cells was tested using 14C-aminopyrine(14 C-AP) accumulation indirectly.H+/K+ ATPase α subunit mRNA expression was assessed using RT-PCR.Results The cell proliferation and H.pylori-related apoptosis in both GES-1 and AGS celL lines were stimulated and inhibited by IL-1β.IL-1β induced expression and upregulation of COX-2 mRNA in GES-1 and AGS cell lines,respectively.In addition,IL-1β continuously inhibited the ability of histamine-stimulated 14C-AP accumulation of isolated parietal cells accompanied by down-regulation of H+/K+ ATPase mRNA expression.Conclusions It suggests that IL-1β play an important role in H.pylori-related gastric carcinogenesis through two pathways:①to interfere gastric epithelial cell growth by up-regulating COX-2 expression,②to inhibit acid secretion from parietal cell by down-regulating H+/K+ ATPase expression.
8.Curative effect of cervical traction combined with low-cycle or intermediate frequency electrical current in cervical vertebrae

Objective To observe the curative effect of cervical traction combined with low-cycle or intermediate frequency electrical current in cervical vertebrae.Methods 171 patients with cervical vertebrae were randomly divided into the therapy group and control group,86 cases in therapy group were performed the curative measure of cervical traction and low-cycle,85 cases in control group underwent the curative measure of cervical traction and intermediate frequency electrical current.Both of the two groups carried out continuous therapy for 10 days.Results There was significant difference before and after treatment within each group in CASCS score(P < 0.01)and there was also significant difference after treatment between the two groups(P < 0.05),the effective rate of therapy groupwas significant higher than that of control group(P < 0.05).Conclusion The methods of cervical traction and low cycle or intermediate frequency electrical current has better curative effect in cervical vertebrae and cervical traction and low-cycle do much benefit in cervical vertebrae treatment.
9.Modern research progress regarding effect mechanism of acupuncture on post-traumatic stress disorder.

From the TCM theoretical basis and the evidences of acupoint selection and acupuncture and moxibustion methods regarding acupuncture for post-traumatic stress disorder (PTSD), a theoretical discussion is made in this article, also a review regarding the key mechanism of experiment researches on acupuncture for PTSD is made from aspects of neurobiology and brain functional imaging, etc., which could further clarify the effect mechanism of acupuncture on PTSD. It is found that PTSD is a kind of stress syndrome accompanied with a variety of abnormal mental symptom , and acupuncture has superior effect on PTSD. Based on these, the research progress and deficiency on current mechanism study of PTSD are reviewed, hoping to offer ideas for upcoming research and to serve the clinical practice better.

10.Study on in Vitro Percutaneous Permeability of Methylphenidate Cream

OBJECTIVE: To study in vitro percutaneous permeability of methylphenidate cream. METHODS: Isolated rat skin was taken as permeable barrier. The influence of different concentrations of azone(0%, 2%. 5% ) in methylphenidate cream on drug permeation was observed. RESULTS: Steady-state percutaneous flow(J ) of methylphenidate cream with 2% and 5% azone increased 27. 80% and 49. 05%. respectively. CONCLUSION: Methylphenidate cream will be a safe. effective and conve- nient new preparation.
