Annexin A5 is a member of Ca 2+-dependent phospholipid binding proteins family .It exists in histocytes . And because of its unique structure , Annexin A5 plays an important role in many critical functions such as cell in-side/outside anticoagulation and pro-apoptosis .Moreover , Annexin A5 has certain relevance with many diseases such as systemic lupus erythematosus ,tumour and other diseases .
2.Investigation of blood collection and supply in Chengdu from 2000 to 2006

Objective To investigate the blood collection and supply in Chengdu,from 2000 to 2006,and also to analyze its development tendency.Methods The volume of blood collection,blood component preparation,the positive rate of blood screening,the volume of clinical consumption,as well as the population statistics and bed numbers in medical institutions and hospitals,from 2000 to 2006,were reviewed.Correlation and coefficient of regression were used for the statistical analysis.Results The annual average rate of growth(AARG) of blood collection and supply is 17.74%.The rate of RBC separation increased from 27.4%% to 84.55%,and the AARG of platelet apheresis product was 38.00%.Blood components had become much more popular than whole blood in clinical application.There was a good linearity correlation between the blood provision and the population statistics(?=0.997,P
3.Effect of ligustrazine on the human scavenger receptor-AI expressed in Xenopus oocytes

Aim To investigate and evaluate the effect of ligustrazine on the expression of scavenger receptor type AI, the human ?-interferon activation site (GAS) elements regulatory systems of SR-AI expression wasfirst established in the Xenopus oocytes. Methods A plasmid, in which the human SR-AI gene was constructed downstream from the GAS elements, was microinjected into the nucleus of Xenopus oocytes. The oocytes then were cultured in the smooth muscle cell conditioned medium (SCM), In which the cell were cultured with the treatment of oxLDL for 2 days. In the other groups, the oocytes were treated with SCM containing the nature medicines ligustrazine, and the positive control drug ?-interferon respectively. Results The results demonstrated that SCM could upregulate the expression of SR-AI in this system, this action was suppressed by ligustrazine and ?-interferon. Conclusion Similar to the action of ?-interferon, ligustrazine can prevent the over-expression of SR-A induced by oxLDL, via their effect on the GAS elements regulatory pathway.
4.Study of Dicliptera Chinensis Polysaccharide in Counteracting Liver Injury Induced by Antituberculosis Drugs

Objective To study the therapeutic effect and mechanism of Dicliptera chinensis polysaccharide ( DCP) on liver injury induced by antituberculosis drugs. Methods Sixty mice were randomly divided into six groups, namely normal control group, model group, glucurolactone group (in the dosage of 200 mg·kg-1·d-1), and high-, middle- and low-dose DCP groups ( in the dosage of 600, 400, 200 mg·kg-1·d-1, respectively). Except for the normal control group, the rats in the other groups were given intragastric administration of isoniazid and rifampicin ( 100 mg/kg) to induce liver injury model, and were simultaneously treated with corresponding agents, once a day. On the experiment day 30, the blood and liver tissue were sampled. The serum levels of alanine aminotransferase ( ALT) , aspartate aminotransferase ( AST) , alkaline phosphatase ( AKP) and microsomal nitric oxide ( NO) were detected by biochemical method. The contents of tumor necrosis factor alpha (TNF-α) and interleukin 6 (IL-6) in liver tissue were determined by enzyme-linked immunosorbent assay ( ELISA) , and the hepatic histopathology was observed after HE staining. Results In DCP groups, the hepatic pathological changes of the mice were improved, the number of the inflammatory cells was reduced, and the activities of serum ALT, AST and AKP as well as the contents of hepatic TNF-α, IL-6 and NO were reduced ( P<0.05 or P<0.01 compared with those in the model group). Conclusion Dicliptera chinensis polysaccharide is effective for liver injury induced by antituberculosis drug, and the mechanism may be associated with its anti-inflammatory action.
5.Effect of sodium hyaluronate on malondialdehyde and superoxide dismutase levels in the synovial fluid of patients with knee osteoarthritis

BACKGROUND:Intra-articular injection of sodium hyaluronate is an effective method for the treatment of knee osteoarthritis, with significant effect and less adverse reactions, but the mechanism is unclear. OBJECTIVE:Through testing the malondialdehyde and superoxide dismutase levels in the synovial fluid of knee osteoarthritis before and after injection of sodium hyaluronate, to evaluate the clinical efficacy of sodium hyaluronate in the treatment of knee osteoarthritis. METHODS:Thirty-seven patients with knee osteoarthritis (40 knees) were enroled and divided into mild (n=10, 10 knees), moderate (n=17, 18 knees), and severe (n=10, 12 knees) groups according to the Japan's knee osteoarthritis indications. Patients were subjected to intra-articular injection of 25 mg sodium hyaluronate, once a week for 5 weeks. The levels of malondialdehyde and superoxide dismutase in the synovial fluid before and 4 weeks after treatment were detected, and then clinical effects were evaluated based on the clinical scores according to the Japan’s knee osteoarthritis indications. RESULTS AND CONCLUSION: The indication rating results of the mild and moderate groups were decreased significantly 4 weeks after injection (P < 0.05), but there were no significant difference in the severe group before and after treatment. The malondialdehyde level in the synovial fluid was decreased obviously in the three groups at 4 weeks after injection (P < 0.05), while the level of superoxide dismutase was increased remarkably (P < 0.05). These findings indicate that sodium hyaluronate can treat knee osteoarthritis by reducing the malondialdehyde level and increasing superoxide dismutase level in the synovial fluid, but this method is more suitable for treatment of mild to moderate knee osteoarthritis.
6.Application of TG-ROC analytical method in detection reagent quality evaluation

Objective To compare the relationship between the enzyme‐linked immunosorbent assay(ELISA) reagent and West‐ern blot(WB) confirmation reagent for analyzing the quality lever of human T‐cell lymphotropic virus(HTLV) detection reagent . Methods The WB confirmation reagent was used to detect anti‐HTLV antibody in 156 human serum samples of ELISA prelimina‐ry screening positive .The ELISA cut‐off value(optimal value) was selected by using the two‐graph receiver operating characteristics (TG‐ROC) analytical method .The two‐by‐two table analysis was constructed to analyze the consistency of results detected by the two methods ,moreover the McNemar test was used to evaluate the consistency of detection results .The quality level of HTLV de‐tection reagent was comprehensively evaluated .Results Among 156 serum samples of ELISA preliminary screening positive ,only 40 samples were positive by the WB confirmation ,and other 116 samples were negative .The sensitivity and specificity of ELISA de‐tection reagent obtained by TG‐ROC analysis were 97 .5% and 45 .7% respectively ,the TG‐ROC test also indicated that the detec‐tion results had significant difference between ELISA and WB(P<0 .05) .By adjusting the cut‐off value ,the sensitivity and specific‐ity of ELISA were increased to 88 .8% (parametric method) .In the comparison of the parametric method and the non‐parametric method ,the obtained areas under the curve(AUC) was 0 .923 5(parametric method) ,their results were basically consistent .Conclu‐sion Although above results indicate that the detection results of ELISA reagent are different from those of WB ,but adjusting the cut off value can increase its sensitivity and specificity ,thus increases the reliability of diagnosis result .

In recent years,the incidence of type 2 diabetes mellitus(T2DM)keeps increasing in China,and seriously affects people's health.The M1/M2 polarization of macrophages is unbalanced in patients with T2DM,which is manifested by the increase of pro-inflammatory M1 macrophages and the decrease of anti-inflammatory M2 macrophages.Macrophage polarization imbalance can cause insulin resistance or damage pancreatic islets β cells through various ways.Adjusting the polarization of macrophages plays a therapeutic role in T2DM.This article reviews the research progress on the relationship between macrophage polarization and T2DM.
8.Screening specific minimum amino acid sequence triggering immunity to enterovirus 71

Objective:To screen the neutralizing epitope of enterovirus 71 (EV71) and determine the specific minimum amino acid sequence that triggers immunity for providing a theoretical basis for the development of synthetic peptide vaccines.Methods:EV71 neutralizing antibody-specific binding clones were panned and sequenced using a phage display random 12-peptide library to obtain the key sequences of neutralizing epitopes. A series of peptides containing the key sequences with N-terminal acetylation (AC) and C-terminal linking to Keyhole limpet hemocyanin (KLH) were synthesized. Serum samples were collected after immunizing mice with the modified peptides. Then the immunogenicity of the peptides and the neutralizing activity of serum samples were analyzed by Western blot, ELISA and neutralization test.Results:After three rounds of panning, cloning and sequencing, KQEKDL was identified as the key motif. The serum samples collected from the mice immunized with the modified series of peptides containing key motifs had different degrees of binding ability to EV71 and VP1 protein. The serum samples of mice immunized the synthetic peptide containing only the minimum key motif (AC-KQEKDL-KLH) had the strongest response to the other three peptides and EV71 and the highest neutralizing titer.Conclusions:The EV71 neutralizing epitope was successfully screened using the phage display random peptide library. The key motif of KQEKDL might be the specific minimum amino acid sequence that triggered the immune system. This study provides a theoretical basis for better understanding the immune response mechanism, evaluating the immunogenicity of the antigens and further research and development of polypeptide vaccines.
9.Analysis of the Regularity of Chinese Patent Medicine Prescriptions for the Qi Deficiency and Blood Stasis Syndrome

OBJECTIVE:To investigate the regularity of Chinese patent medicine prescriptions for Qi deficiency and blood sta-sis syndrome (QDBSS),and to provide reference for studying QDBSS diagnosis standard and new TCM for related symptoms. METHODS:Chinese patent medicine for QDBSS were collected from Chinese Pharmacopoeia (2015 edition) and Chinese Mate-ria Medica Preparation(1993 version). The regularity of Chinese patent medicine prescriptions and symptom association rule were analyzed by using TCM inheritance auxiliary system (V2.5). RESULTS:Among the 171 symptoms,chest tightness,shortness of breath,chest pain,fatigue and palpitation were common symptoms,and three groups of symptoms were included as chest tight-ness-shortness of breath-chest pain-palpitation-irregularly pulse;mouth and eye deviation-numbness of hands and feet-hemiple-gia-stiff tongue-spasm;hypomenorrhea-irregular menstruation-abdominal mass-dysmenorrheal. One hundred and thirty-one Chinese patent medicines included 304 ingredients,top 5 in the list of frequency were Astragalus membranaceus,Salvia militiorrhiza,An-gelica sinensis,Ligusticum chuanxiong,Panax ginseng. A. membranaceus- A. sinensis,L. chuanxiong-A. sinensis,S. militiorrhi-za-A. membranaceus,L. chuanxiong- A. membranaceus and L. chuanxiong-S. militiorrhiza were frequently used herbal combina-tions. A. membranaceus-A. sinensis-L. chuanxiong-S. militiorrhiza-P. ginseng-Carthamus tinctorius-Panax notoginseng was the core combination. A new prescription for QDBSS was formed (P. ginseng-Radix Paeoniae Rubra-borneol-Gastrodia elata-artificial moschus-Myristica fragrans). CONCLUSIONS:QDBSS has common symptoms which exist in many kinds of diseases,such as chest tightness,chest pain and shortness of breath;it also has the unique symptoms among various diseases. Invigorating Qi and promoting blood circulation is the basic treatment for the QDBSS. It is of significance to pay attention to"supplement by passing, combination of pass and supplement"and combine regulating Qi-flowing method with enriching blood method or aromatic warming method for QDBSS therapy according to the differ-ence of Zhengxie shengshuai.
10. Effects of live attenuated polio vaccine on the immunogenicity of live attenuated rotavirus vaccine

Objective:
To evaluate whether simultaneous vaccination with live attenuated polio vaccine affects the immunogenicity of live attenuated rotavirus (RV) vaccine.
Methods:
Rotarix produced by GlaxoSmithKline was used as the research object. Two doses of Rotarix were orally administered on day 0 and month 1, and oral live attenuated polio vaccine (OPV) was administered on day 0, month 1 and month 2 according to the national vaccination plan. Healthy infants aged 6 to 16 weeks were randomly divided into two groups: interval vaccination group (Rotarix and OPV were vaccinated on different days) and simultaneous vaccination group (Rotarix and OPV were vaccinated on the same day). Serum samples were collected on day 0, month 2 and month 12, and serum RV-IgA was measured by enzyme linked immunosorbent assay. Statistical analysis was performed to evaluate whether there were statistical differences in the seroconversion rate and level distribution of RV-IgA between the two groups.
Results:
The seroconversion rate of serum RV-IgA in month 2 was 73.84% in the interval vaccination and 63.95% in the simultaneous vaccination group, and the difference between them was statistically significant (
