1.Effect of epidural analgesia with different methods on intestinal peristalsis

Objective To explore the effect of PCEA on intestinal peristalsis.Methods 120 patients with ASA Ⅰ~Ⅱ lower abdominal or lower limbs surgery were randomly divided into four groups with 30 cases each and received PCEA at the end of operation sespectively.Group A were given 0.0002% fentanyl,0.2% Ropivacaine and 0.005 % Droperidol;0.01% morphine,0.2% Ropivacaine and 0.005% droperidol in group B;0.2% Ropivacaine in group C;0.9% saline in group D.Postoperative analgesia was provided by PCA pump with LCP model.Results Satisfactory analgesia in group A was 93%,97% in group B 47% in group C and 13% in group D.Both group A and B did not show any significant difference in the analgesic effects(P> 0.05).Satisfactory analgesia in group A and B were significantly better than in group C and D(P<0.05).The renewal of intestinal peristalsis were significantly faster in lower limbs surgery patients than lower abdominal patients(P< 0.01),but it did not show any significant difference in the same kinds of surgery in four groups (P>0.05).Conclusion Both group A and B can get a satisfied analgesia effect;intestinal peristalsis of lower limbs surgery patients renewed were shorter than in lower abdominal patients.

Learning modes based on the initiatives of students has become a new way in the development of medical students' quality.The second clinical medical college of Southern Medical University performed an improved teaching method:meliorated PBL based on TBL and taking clinical and scientific research ability as the core,which combined the advantages of PBL and TBL in view of the importance of clinical and scientific research ability for medical students.Practice proved that this teaching method was effective in improving comprehensive quality of medical students and cultivating more comprehensive talents for the society.
3.Comparative Identification of Aerial Roots of Ficus microcarpa Linn. f. and Ficus altissima Bl.

Objective To find out evidence for the identification of the aerial roots of Ficus microcarpa Linn. f. and F. altissima Bl.. Methods The fresh aerial roots of the two kinds of plants were collected. The appearance traits were observed by stereoscopy. The velamen surface, cross section of root tip, cross section of the posterior root and powder of two kinds of aerial roots were observed under microscope. Results The number of aerial roots of F. microcarpa Linn. f. was more than that of F. altissima Bl., while the diameter was less than that of F. altissima Bl.. Differences were shown in primary xylems of the transverse section of root tip and posterior root of the two kinds of aerial roots, there were 5-7 vascular bundles in F. microcarpa Linn. f. and 7-10 vascular bundles in F. altissima Bl.. As for the powder, the powder of F. microcarpa Linn. f. was yellowish brown, while that of F. altissima Bl. was reddish brown; F. altissima Bl. had more fibers and longer diameter than F. microcarpa Linn. f. and had cluster crystals in order. Conclusion The appearance and microstructure features showed by the results can be used to distinguish the aerial roots of F. microcarpa Linn. f. and F. altissima Bl..

Objective To observe the effects of estradiol-17? (E 2-17?) on the induction of experimental autoimmune thyroiditis (EAT). Methods Ten-week old female Wistar rats were ovariectomized (OVX). EAT of Wistar rats was induced by immunization with thyroglobulin (Tg) from SD rats in the presence of complete Freund′s adjuvant. Thyroiditis was pathologically evaluated by HE staining. E 2-17? and PRL levels were measured by RIA, anti-Tg antibody (TgAb) was detected by ELISA, and interferon (IFN)-?, interleukin (IL)-10 mRNA in thyroid were assayed by RT-PCR. Objective To investigate the influence of acupuncture on learning and memorial function and the expression of BDNF in prefrontal cortex of rats with sleep deprivation (SD), for providing theoretical evidence for treating sleep deprivation with acupuncture.Method Sixty Wistar rats were randomly divided into a control group, a model group, and an acupuncture group, 20 rats in each group. SD models were established by the modified multiple platform methods; the acupuncture group was intervened by acupuncture at Shenmen (HT 7); while the control group was left intact. Respectively after SD for 24 h and 72 h, the learning and memorial function was tested by using Morris water maze. Ten the rats were sacrificed to collect brain for detecting the expression of BDNF in prefrontal cortex via immunohistochemical method.Result The body weight, learning and memorial function, and the expression of BDNF in prefrontal cortex of the acupuncture group and model group were significantly different from those of the control group after SD for 24 h and 72 h (P<0.05). The learning and memorial function of the acupuncture group was significantly different from that of the model group after SD for 24 h and 72 h (P<0.05). The body weight and expression of BDNF in prefrontal cortex of the acupuncture group were significantly different from those of the model group after SD for 72 h (P<0.05).Conclusion Acupuncture can obviously improve the decline of the function of learning and memory of the SD rats, and also up-regulate the declined expression of BDNF in prefrontal cortex caused by SD.

Objective To obtain purified myosin-Vc (Myo5c) protein and prepare its polyclonal antibodies of mouse and rabbit. Methods The fragment encoding Myo5c specific protein (297 amino acids) was amplified with RT-PCR from human gastric mucosa. The product and the prokaryotic expression plasmid pQE-31 containing 6?His were used to construct pQE-31/Myo5c. The recombinant was transformed into E. coli JM109 and induced to express with IPTG. The objective product was purified. BALB/c mice and rabbits were immunized with the purified Myo5c protein and the antiserum was obtained. Titer and specificity of the polyclonal antibodies were determined by ELISA and Western blotting. Results pQE-31/Myo5c was successfully constructed. The fusion protein of Myo5c with molecular weight 42 000 was obtained and purified. The recombined protein showed immunogenicity. Mouse or rabbit antiserum with high level of specific antibodies for Myo5c was obtained. Indirect immunostaining and Western blotting analysis demonstrated that the antibodies could recognize native Myo5c protein. Conclusion Our results suggest that the prepared antibodies might be useful in studying the function of Myo5c in intracellular trafficking of endocytic vesicle, such as viruses.

OBJECTIVETo investigate the effect of functional blocking of endogenous miR-23a with a specific antisense oligonucleotide (ASO) on the proliferation and invasiveness of gastric adenocarcinoma cell line MGC803 in vitro.
METHODSA specific ASO targeting miR-23a, namely ASO-23a, was transfected into MGC803 cells to block endogenous miR-23a. The mRNA level of miR-23a in the transfected cells was detected with quantitative real-time PCR. The changes of cell proliferation following the transfection were detected with MTT assay and colony formation assay, and TUNEL assay and Transwell assay were employed to evaluate the changes in cell apoptosis and invasiveness, respectively.
RESULTSQuantitative real-time PCR demonstrated efficient functional blocking of endogenous miR-23a in MGC803 cells by ASO-23a. Suppression of miR-23a with ASO-23a obviously inhibited cell growth, colony formation and invasiveness of MGC803 cells and significantly enhanced the cell apoptosis.
CONCLUSIONASO-23a can efficiently block the function of endogenous miR-23a in MGC803 cells to inhibit cell proliferation and invasion and promote cell apoptosis.
