Objective To study the sensitivity and reliability of rat skin anaphylactoid test method, detect and evaluate the anaphylactoid reaction of traditional Chinese medecine injection. Methods The condition of rat skin anaphylactoid test was optimized by studying the influencing factors of sensitivity and reliability of test with C48/80 as a tool drug, Tween80, endotoxin, China cobra toxin, trichosanthin injection, Tanreqing injection and Xuesaitong injection were investigated. Results The best conditions of rat skin anaphylactoid test was as follows:intrademal inject the drug with insulin syringe, 50-100μL per point, immediately inject 0.5%Evans blue dye 1 mL though caudal vein, 15 min later, kill the rat by carotid artery bleeding, clip dorsal skin to do the test. With this method, Tween80, endotoxin, China cobra toxin and trichosanthin injection all can induce blue stain in rat skin. Tanreqing injection showed no blue stain at the clinical dose. Xuesaitong injection although can induce blue stain in rat skin at the clinical dose, but the results cannot exclude the interference of its pharmacological function. Conclusion The method is simple with short test cycle, less dose of test drug, high detection sensitivity and good reproducibility, but some drug can show false positive result due to its own property.

Objective To simulate a finite element model for biomechanical analysis of mandible chin blast injury and analyze the mechanism of maxillofacial blast injury.Methods The three-dimensional element model of human mandible blast injury was established to simulate the dynamic process of injury to the mandible chin.Von Mises stress and effective strain were evaluated in biomechanical study of the simulation results.Results The dynamic damage process of human mandible chin blast injury was simulated successfully.In the condition of 1 000 mg and 3 cm,the Von Mises stress and effective were maximum at condylar neck region (9.1 × 106 Pa,0.62 × 10-3ε),were second at mandibular angle region (6.1 × 106 Pa,0.42 × 10-3ε),and minimum at mental foramen region (6.1 × 106 Pa,0.39 × 10-3ε).Blast distance rather than blast equivalent produced more effect on the mechanical parameters and damage degree.Conclusions Von Mises stress and effective strain can be applied to the evaluation of bone tissue damage.The finite element method is effective in simulating mandible blast injury and can provide a new thought and approach to clinical treatment of oral and maxillofacial blast injury.

[Objective] To explore the feasibility and advantages of blood microdialysis technique for the pharmacokinetic study of sinomenine. [Methods] In this study, rats served as the experimental subjects and sinomenine was administered to the rats by intravenous injection. With microdialysis technique for blood sampling, the blood concentration of sinomenine was determined by high performance liquid chromatography (HPLC) and the associated pharmacokinetic parameters were analyzed by 3p97 program. [Results] The pharmacokinetics process of sinomenine in rats presented as the two-compartment model. The half-life of distribution phase was 10.98min and that of elimination phase was 44.71 min. [Conclusion] It is feasible to study pharmacokinetic parameters of sinomenine by using blood microdialysis technique. In comparison with the traditional technique, blood microdialysis technique is superior to the examination of drug blood concentration .

AIM The proofs of evidence-based medicine of Qingkailing Injection's effect on viral hepatitis are lack in spite of the wide use of Qingkailing Injection in medical practice.This study aims to use Meta-analysis to evaluate its clinical effect and safety.METHODS Such databases as CNKI,VIP and Wanfang were searched for randomized controlled trials related to Qingkailing Injection in treatment of viral hepatitis,which were published officially between 1996--2016.Revman 5.3 software was used to conduct Meta-analysis for the effective rate,rate of adverse reaction,the decrease rate of ALT,AST and TBIL of Qingkailing Injection.Meanwhile,subgroup analysis was conducted to the clinical effect of doses [＜ 0.5 mL/(kg · d),0.5-0.67 mL/(kg · d),＞0.67 mL/(kg · d)],treatment courses (≤ 14 d,＞ 14 d),manufacturers (Beijing University of Chinese Medicine Pharmaceutical Factory,Shenwei Pharmaceutical Company,Shanxi Taihang Pharmaceutical Company),year (1996 to 2002,2003 to 2009,2010 to 2016).RESULTS Fourteen studies were included according to the criterion.The results of Meta-analysis showed that the total effective rate,the decrease rate of ALT,AST and TBIL in Qingkailing Injection group were significantly higher than those in the control group (P ＜ 0.05).Whereas,no significiant differences were found between Qingkailing Injection group and the control group of its rate of adverse reaction (P ＞ 0.05).Subgroup-analysis showed that the effective rate of Qingkailing in doses subgroup,treatment courses subgroup,manufacturers subgroup and year subgroup were significantly higher than those of the control group.CONCLUSION Qingkailing Injection has obvious benefit in the treatment of viral hepatitis,which can reduce the activity of ALT and AST,also can reduce the content of TBIL.But we should pay attention to the prevention of allergic reaction in the clinical use.

Objective To prepare and explore the pharmacokinetic parameters for ~ 99 Tc~ m -ASON-EGF in healthy rabbits. Methods ~ 99 Tc~ m -ASON-EGF was prepared according to previous methods and its changes of concentration in blood were measured by radioactivity counts per minute. The experimental data were dealt with by 3p97 software and its true compartment model was estimated by AIC value, R~ 2 value, the 1/c and F test. Subsequently, its half-life of distribution (T_ 1/2 ?), half-life of elimination (T_ 1/2 ?), central compartment volume of distribution (Vc), total apparent volume of distribution (Vd) and total rate of clearance (CL) were calculated by the software. Finally, the binding rate of plasma protein was determined by trichloroacetic acid precipitation. Results The best model of ~ 99 Tc~ m -ASON-EGF in vivo was two-compartment model and its T_ 1/2 ?, T_ 1/2 ?, Vc, Vd and CL were 5.28 min, 89.23 min, 67.8 ml, 915.6 ml and 7.1 ml/min respectively. After being incubated with fresh plasma for 1.5 h, its binding rate was 10.69%. Conclusion Its process of transportation in healthy rabbits is fitted to two-compartment model and the pharmacokinetic properties are desirable.

Objective Finite element numerical simulation technique was applied to simulate the dynamic projectile injury process of human chin in different injury conditions and the mechanism of injury was discussed by using biomechanical analysis . Methods The 3D finite element model of human mandible projective injury was established to simulate the dynamic projectile inju‐ry process of human chin in different injury conditions (high ,medium and low speeds) ,and the simulation results were used to com‐parative analysis of biomechanics .Results The dynamic damage process of human chin projectile injury was simulated successfully in different injury conditions ,and the more serious injury of mandible was caused by faster speed .Conclusion The finite element method can simulate the projectile injury of mandible effectively ,and can provide a new thought and method for basic research and clinical treatment of oral and maxillofacial war injury .

Objective To develop a three-dimensional element model of pig mandible impact injury and test the simulation results in an attempt to determine the feasibility and reliability of finite element numerical simulation method used in the maxillofacial impact injury.Methods CT data was used to develop a three-dimensional finite element model of pig mandible impact injury,and the dynamic process of impact injury was simulated.The simulation results were compared with the animal experiment and had energy check to validate the reliability and feasibility of the modeling and simulation methods.Results The three-dimensional finite element model was established successfully,containing 61,512 hexahedrons,5,450 tetrahedrons,4,030 trihedrons,and 67,159 nodes.The simulation process was realistic,and the simulation results showed no statistical difference with the animal experiment with regard to strain,acceleration,and other biomechanical properties (P ＞ 0.05).The simulated damage shape had a high similarity with animal specimens,and the result of energy check also complied with energy conservation law.Conclusion Finite element method is effective to simulate the dynamic process of mandible impact which ensures a correct and reliable model and simulation,and thus can be used to analyze the mechanism of maxillofacial impact injury.

Objective The simulation of the human mandible injury was carried out by using the finite element simulation technology ,and the biomechanical analysis of simulation results was developed to explore the mechanism of injuries .Methods The Chinese Visible Human digital data were used to establish the three-dimensional element model of mandible injuries ,and the dynam-ic processes of human mandible injuries in different conditions were simulated ,and the biomechanical analysis were carried out by u-sing the Von Mises stress and effective strain .Results The three-dimensional element model of mandible injuries was established , the dynamic damage and fracture of human mandible were simulated successfully ,the mandibular angle and condylar were the predi-lection parts of high-stress ,high-strain and fractures .Conclusion The Von Mises stress and effective strain can be used to predict and judge the bone tissue injuries ,the finite element method can simulate the impact injuries of mandible effectively ,and the simula-ted results can provide guidance and reference for basic research and clinical treatment of oral and maxillofacial injuries .

Objective To analyze the histological changes of bone diseases and to investigate the noninvasive measurements for diagnosing renal osteodystrophy (ROD) in maintenance dialysis patients . Methods Ninety-one patients were selected to receive bone biopsy . The bone samples were stained with HE, toluidine blue and Masson, and were examined with light microscopy . The levels of immunoreactive parathyroid hormone (iPTH), osteoprotegerin (OPG),sRANKL and osteocalcin (OCN) were determined in the patients enrolled from 2004 to 2006 . The level of iPTH was measured by radioimmunoassay . OPG and sRANKL were measured by ELISA,and OCN was measured by chemiluminescence . Results The incidence of ROD in the maintenance patients was 100% . According to the histological appearance, 50 cases (54 .9%) were high turnover bone disease (secondary hyperparathyroid bone disease), 9 cases (9 .9%) were low turnover bone diseases(osteomalacia and adynamic bone disease), and 32 cases(35 .2% ) were mixed bone disease . The level of iPTH in patients with ROD was significantly increased compared with healthy controls . It was the lowest in low turnover bone diseases . There was no difference among three types of ROD . OPG level was significantly increased compared with healthy controls [(2176 .58±1576 .08) pmol/L vs (1310 .46±1254 .00) pmol/L, P＜0 .05] . The level in high turnover bone diseases was higher than that of the healthy controls [(2261 .85±1712 .22) pmol/L vs (1310 .46±1254 .00) pmol/L, P＜0 .05] . There was no difference among three types of ROD .sRANKL level in high turnover bone disease was significantly increased compared with healthy controls [(0 .328±0 .524)pmol/L vs (0 .084±0 .190) pmol/L, P＜0 .05] . OCN level was also higher than that of the healthy controls (P＜0 .05), and the OCN level in low turnover ROD was the lowest among three types of ROD . OCN level in mixed ROD was dramatically increased as compared to low turnover ROD [(226 .633±66 .455) pmol/L vs (193 .03±104 .269) pmol/L, P ＜0 .05] .Conclusions The histological changes of bone disease can be indicated by iPTH level, but the types of ROD can not be distinguished according to iPTH level neither be differentiated by the levels of OPG, sRANKL and OCN . Bone histomorphometry is still the golden standard for diagnosing renal osteodystrophy .

AIM:To demonstrate the inhibitoary effects of Longkai Granules(LKG) against experimental prostatic hyperplasia and evaluate its toxicity on animals taking the granules orally. METHODS: The prostate exponent,DNA content in prostate tissue、the activity of acid phosphatase in serum or the wet weight of spermatophores and testicles in normal immature mice and in the hyperplasia model mice induced by subcutaneous injecting testooslerone spropionate or by implanting of the urogenital sinus were determined after administrating of LKG intragastrically to the mice.The single maximum dosage of LKG in mice and its long-term(13 weeks) toxicity in Wistar rats and Beagle dogs in orally was evaluated. RESULTS: LKG could decrease the weights of prostates and DNA content in the tissue in the normal immature mice in the amount of 20 and 40 g/kg once a day.LKG,in the amount of both 10,20 and 40 g/kg for 10 days and 20 and 40 g/kg for 30 days,could inhibit the hyperplasia of ventral prostates in the model mice induced respectively by the injection of testooslerone spropionate and by implanting urogenital sinus.LKG,in the(amount) of 100 g/kg for 13 weeks to Wistar rats,would lead to prostatic atraphy in alight degree,and its epithelial cells change in shape from column to flat and prostatic cavity being small,which did not recover in 4 weeks after stopping administration of tested drug to the animals.The single maximum dosage by ig in mice was 200 g/kg.There was no significant toxicity reaction in rats in the amount of 10,40 and 100 g/kg for 13 weeks or in Beagle dogs in the amount of 12 and 60 g/kg for 13 weeks. CONCLUSION: LKG can inhibit the prostatic hyperplasia and shows no visible toxic reaction in animals orally.
