1.Effect of Puerarin on Insulin Resistance in Gestational Diabetic Mellitus Rats

Objective To observe the effect of puerarin injection (PI) on insulin resistance in gestational diabetic mellitus(GDM) rats. Methods Rats with gestation for 5 days received onedose injection of STZ 35 mg/kg into subaortic caudal vein. After injection for 72 hours, rats with blood sugar over 13.5 mmol/L indicated the modeling being successful. Then the model rats were divided into treatment group (TG, using Puerarin), westernmedicine control group(WCG, using rosiglitazone) and model group (MG, using 0.9 % NaCl), 10 rats in each group .Ten normal gestational rats were used as normal control group (NCG, using 0.9 % NaCl).Two weeks later, the changes of blood sugar, Creactive protein (CRP) and tumor necrosis factor- ? With the improvement of China's population aging and health care consciousness, female pelvic floor dysfunction gets increasing attention in society. Modern TCM scholars conclude TCM doctors’ theories about etiology and pathogenesis of pelvic floor dysfunction, summarized as debility of Chong Ren meridians and inability to lift, and then they advocated invigorating qi and elevating yang, with a purpose to protect pelvic floor stability. This article conducted relevant discussion on the significance of rehabilitation of pelvic floor dysfunction based on TCM adjusting qi activity.
3.Qin Jiajia’s Treatment of Premature Ovarian Failure by Mainly Reinforcing Kidney

Objective] To summarize teacher Qin’s clinical experience in treating premature ovarian failure by mainly reinforcing kidney. [Method] Collect the clinical cases, and analyze the etiology and pathogenesis, syndrome differentiation and treatment of premature ovarian failure for teacher Qin ’s. [Result] Teacher Qin thought that this disease was mainly due to kidney deficiency, especially kidney-yin deficiency, and accompanied by liver depression. This disease can be divided into kidney-yin deficiency, kidney-yang deficiency, deficiency of kidney-yin and yang, deficiency of kidney and liver depression. Reinforcing kidney-yin for kidney-yin deficiency; reinforcing kidney-yang for kidney-yang deficiency; reinforcing kidney-yin and yang for deficiency of kidney-yin and yang; tonifying the kidney essence and soothing liver-qi stagnation for deficiency of kidney and liver depression. [Conclusion] Treatment of premature ovarian failure is based on tonifying kidney, supplement on soothing liver, while some change therapies, should combine disease with syndrome.
4.Experimental Study on Gukang in Improving Bone Mineral Density of Rats with Osteoporosis

Objective To observe the effect of Chinese medicine of Gukang(GK) on bone mineral density(BMD) in osteoporosis rats.Methods Seventy-two female SD rats were randomized into 6 groups: pseudo-operation group,model group,nilestriol group(in the dose of 1mg/kg),and high-,middle-and low-dose GK groups(9.6,4.8 and 2.4 g?kg-1?d-1 respectively).Rat models of osteoporosis were induced by removal of bilateral ovaries.Three months after operation,the rats were given the corresponding medicine according the experimental design.After treatment for 3 months,in-vivo BMD as well as the in-vitro BMD in the isolated left femur and tibia was detected with dual energy X-ray bone densitometer.Results The in-vivo general BMD and lumbar BMD of the model group were decreased(P

BACKGROUND: 5-azacytidine (5-Aza) has been frequently used to induce bone marrow mesenchymal stem cells (BMSCs)differentiation into cardiomyocyte.OBJECTIVE: To observe expression of cardiomyocyte-related receptors in cardiomyogenic differentiation of rat BMSCs.METHODS: BMSCs of passage three were assigned to four groups: group Ⅰ: L-DMEM solution alone was replaced; Ⅱ:L-DMEM solution was replaced after induction of 100 mg/L AST+5 μmol/L 5-Aza for 24 hours; group Ⅲ: L-DMEM solution wasreplaced after induction of 10 μmol/L 5-Aza for 24 hours; and group Ⅳ: L-DMEM solution was replaced after induction of 5 μmol/L5-Aza for 24 hours. Culture medium was replaced every 3 days in each group. Differentiated cells were identified after 30 days ofinduction.RESULTS AND CONCLUSION: Expression of cardiomyocyte specific proteins Nkx2.5, cTnT and Desmin was detected in groupsⅢ, Ⅳ and Ⅱ after induction compared with group Ⅰ , with significant differences (P < 0.01). The amount of cTnT and Desminexpression expression was significantly higher in groups Ⅱ and Ⅲ compared with group Ⅳ (P < 0.01). The level of Nkx2.5expression was significantly higher in groups Ⅱ (P < 0.01) and Ⅲ (P < 0.05) compared with group Ⅳ. No Nkx2.5, cTnT andDesmin espression was detected in group Ⅰ. After induction for 2 weeks, cells with spontaneous contractility were observed ingroups Ⅱ and Ⅲ, indicating differentiation towards cardiomyocyte after induction. Results demonstrated that induction effectswere similar between 100 mg/L AST+5 μmol/L 5-Aza and 10 μmol/L 5-Aza. This may contribute to cytoprotective effects of AST,which can promote vascular endothelial cell proliferation, enhance celss tolerance to 5-Aza-induced cytotoxicity and upregulatecardiac-specific protein expression.
6.Analysis of ultrasonic diagnosis and misdiagnosis of breast fibroadenoma

Objective To explore the atypical ultrasonic appearances of breast fibroadenoma and analyze the cause of mis diagnosis.Methods A total of 493 lesions in 485 patients of breast fibroadenoma detected by ultrasound examination andconfirmed by pathology were retrospectively analyzed.Atypical appearances were analyzed and misdiagnosis rate were cal-culated.Statistical methods were taken to compare the misdiagnosis rates in lesions with various atypical appearances andthose with none or single atypical appearance.Misdiagnosis rates in lesions with different sizes and pathological types wereanalyzed statically.Then misdiagnosing causes were analyzed.Results A total of 404 lesions were diagnosed correctly,in-cluding 99 lesions with atypical appearances,and the other 89 lesions were misdiagnosed,which showed atypical appear-ances.The misdiagnosis rate of lesions with ≥2 atypical features was higher than that with none or single atypical feature(x2 =256.40,P< 0.05).Large lesions (maximum diameter> 3 am) showed higher misdiagnosis rates than small ones(maximum diameter ≤3 cm,x2=6.73,P<0.05),and complex fibroadenoma lesions also showed higher misdiagnosisrate than simple ones (x2 =188.01,P<0.05).Conclusion The lesions with various atypical appearances,large size andcomplex fibroadenoma in pathology are easy to be misdiagnosed.
7.Effects of tonifying kidney yin and yang on expressions of TNF-α, IFN-γin peripheral blood and apoptosis of granulosa cells in chemotherapy-in-duced premature ovarian failure rats

AIM:To treat the chemotherapy-induced premature ovarian failure ( POF) rats with the methods of tonifying kidney yin and yang by observing the indicator changes in apoptosis of ovarian granulosa cells and related cyto -kines before and after medication .METHODS:SD rats ( n=36) were chronically given intraperitoneal injection of cyclo-phosphamide ( CTX) for 15 d in order to establish the animal model of POF .The rats were randomly divided into control group, CTX group, CTX+kidney yin group and CTX +kidney yang group.The levels of tumor necrosis factor alpha (TNF-α) and interferon gamma ( IFN-γ) in the peripheral blood were detected by ELISA .The apoptosis of ovarian granulosa cells was observed by TUNEL method .The levels of apoptosis related proteins were determined by Western blot .RE-SULTS:After modeling , the levels of TNF-αand IFN-γin the peripheral blood were significantly increased .The protein expression of Bax was up-regulated and Bcl-2 was down-regulated .The apoptosis of ovarian granulosa cells was significantly induced with a deep staining .After treatment with tonifying kidney yin and yang , the levels of TNF-αand IFN-γin the pe-ripheral blood were decreased , the protein expression of Bax was down-regulated and Bcl-2 was up-regulated, and the apop-tosis of ovarian granulosa cells was reduced .CONCLUSION:The reserve function of the ovarian can be recovered by pre-scriptions of tonifying the kidney yin and yang via regulating the expression of TNF -α, IFN-γ, Bax and Bcl-2, and inhibi-ting the apoptosis of ovarian granulosa cells .

10.Analysis of risk factors for urinary tract infection in pregnant women with acute renal colic after indwelling ureteral D-J tube

Objective:To investigate the risk factors related to urinary tract infection after indwelling ureteral D-J tube in pregnant women with acute renal colic.Methods:Clinical data of 109 pregnant women with acute renal colic undergoing indwelling ureteral D-J tube in emergency department of Taizhou Hospital or Enze Hospital from January 2008 to December 2019 were retrospectively analyzed. There were 31 cases (28.4%) with urinary tract infection after discharge and before delivery (infection group) and 78 cases without infection (non-infected group). Univariate and multivariate logistic regression analyses were used to evaluate the risk factors of urinary tract infection after ureteral D-J tube insertion.Results:In 109 patients the average age was 28 (25,33) years, the average gestational age was 26 (21,31)weeks, and the average body mass index (BMI) was 24.84 (22.60,27.43) kg/m 2. Compared with the non-infected group, the infection group had a higher rate of positive urine culture [38.7%(12/31) vs. 15.4%(12/78), χ2=6.56, P=0.010] and diabetes [45.2%(14/31) vs. 11.5%(9/78), χ2=13.86, P<0.001], and a lower gestational age [23(20,28) vs.27(21,32) weeks, Mann-Whitney U test, P=0.006]. Multivariate analysis showed that diabetes( OR=7.739,95 %CI:2.579-23.223, P<0.001), positive urine culture( OR=3.249,95 %CI:1.131-9.330, P=0.029), and gestational age( OR=0.201,95 %CI:0.042-0.955, P=0.044) were independent risk factors for urinary tract infection in pregnant women with acute renal colic after ureteral D-J tube insertion. Conclusion:History of diabetes, gestational age, and positive preoperative urine culture are risk factors for urinary tract infection after ureteral D-J tube insertion in pregnant women with acute renal colic.
