1.Role of tumor suppressor gene PTEN in apoptosis of isolated ectopic endometrial cells induced by gestrinone

Objective To investigate the role of tumor suppressor gene PTEN(phosphatase and tension homologue deleted on chromosome 10,PTEN)in apoptosis of ectopic endometrial cells induced by gestrinone. Methods Ectopic endometrium cells obtained from endometriosis were cultured and exposed to gestrinone in different concentrations. The inhibition of the cells during 48 hours was determined by MTT assay, and the cell growth curve was plotted. Then the morphological changes in cells treated with gestrinone for 24h were observed with transmission electron microscope, and the apoptosis rate, cell cycle of the cells and PTEN expression were assessed with flow cytometry analysis (FCM) at the same time. Results Gestrinone in different concentrations could inhibit the growth and proliferation of ectopic endometrium cells in a does and time dependent manner, and the cell growth curve was changed accordingly. After 24 hours exposure to gestrinone in the concentrations of 10 -6 to10 -4 mol/L, some apoptotic changes were observed in the cells under transmission electron microscope. FCM showed that after the exposure to gestrinone, the apoptotic rate of ectopic endometrial cells was 1.3% in 10 6 mol/L group and 15.0% in 10 -4 mol/L group, indicating that there was significantly increase in apoptosis when compared with the control group. At the same time the level of PTEN expression was also increased. Conclusion Gestrinone can significantly inhibit the growth and proliferation of ectopic endometrial cells in endometriosis, and this effects seems to be related to an increase in PTEN expression.
2.Effects of methoxychlor on placenta of gravid rats and their progeny

Objective To investigate the effects of exposure of gravid rats to the persistent organic pollutants (POPs), methoxychlor (MXC), on the placenta and their progeny during gestation periods. Methods According to different dosage levels of MXC [16, 32, 64 and 0 mg/(kg?d)], forty female SD rats aged 3-month were randomly divided into low-dosage group, mid-dosage group, high-dosage group and control group, with 10 animals for each. All the rats received intraperitoneal injection of MXC for 20 days. The influence of MXC on placenta of gravid rats and their progeny were observed in all aspects. 3.Hospital Infection among Surgical System Inpatients

OBJECTIVE To study the hospital infection among surgical system inpatients in order to work out more effective management plans concerning hospital infection.METHODS Totally 5963 surgical patients from Dec 2000 to Nov 2002 were analyzed by means of the retrospective study.RESULTS Among them there were hospital infections in 243 cases.The surgical hospital infection rate was 4.08% and the neurosurgical hospital infection rate was 5.37% which was the highest among all departments.The main part of infection mostly located in lower respiratory tract(21.30%),then were higher respiratory and urinary tracts(9.88%),incision site(10.70%),gastrointestinal tract(9.05%) and the others(6.17%).The infection rate in male patients was higher than in the females.The sex ratio was 2.2∶1.The infection rate was 4.67%.The highest infection rate after admission in hospital and at 31 to 60 days from January to March(4.95%) as well as from October to December(4.58%).The bacteriological rate was 28.40%,lower than the rate of≥50.00% demanded by the Ministry of Health.CONCLUSIONS The hospital infection in surgical patients is closely related to the departments,sex,duration of hospital stay,season and invasive clinical procedures.
4.Effect of gestrinone on growth and apoptosis in isolated ectopic endometrium cells in vitro

Objective To investigate the effects of gestrinone on growth and apoptosis, as well as the expression of phosphatase and tension homologue deleted on chromosome 10(PTEN) in isolated ectopic endometrium cells in vitro and the underlying mechanisms. Methods Ectopic endometrium cells were cultured and exposed to gestrinone of different doses of 0, 10 -6 and 10 -4 mol/L respectively. The inhibition of the cells during 48 hours was determined by methylthiazolyl tetrazolium (MTT) assay, and the cell growth curve was made. Gestrinone was administered to the cells and at 24 hours the morphological changes were observed by transmission electron microscopy and the apoptosis rate, cell cycle and PTEN expression were monitored by flow cytometry (FCM) at the same time. Results Gestrinone at different concentrations could inhibit the growth and proliferation of ectopic endometrium cells in a dose- and time-dependent manner. 5.Effects of curcumin on activity of NR2B and NR1 in spinal dorsal horn in a rat model of diabetic neuropathic pain

Objective To evaluate the effects of curcumin on the activity of NR2B and NR1 in the spinal dorsal horn in a rat model of diabetic neuropathic pain (DNP).Methods Diabetes mellitus was induced by high-sucrose and high-fat diet and intraperitoneal streptozotocin 35 mg/kg,then confirmed by fasting blood glucose level ≥ 16.7 mmol/L 3 days later in male Sprague-Dawley rats.DNP was confirmed by the mechanical paw withdrawal threshold (MWT) and thermal paw withdrawal latency (TWL) measured on day 14 after streptozotocin administration < 80% of the baseline value.The rats were then randomly divided into 3 groups (n =27 each) using a random number table:DNP,DNP+ curcumin group (group DCur)and DNP + solvent control group (group DSC).Curcumin 100 mg· kg-1 · d-1 and corn oil 4 ml· kg-1 · d-1 were injected intraperitoneally for 14 consecutive days starting from 14 days after administration of streptozotocin in DCur and DSC groups,respectively.Another 27 normal male Sprague-Dawley rats served as control group (group C) and were fed with normal forage.At 3,7 and 14 days after curcumin injection,MWT and TWL were measured and the lumbar segments (L4-6) of the spinal cord were removed.The expression of pTyr1472-NR2B and pSer896-NR1 in the spinal dorsal horn was determined by immunohistochemistry and Western blot.Results Compared with group C,MWT was significantly decreased,TWL was shortened,and the expression of pTyr1472-NR2B was up-regulated at each time point in group DNP.Compared with group DNP,MWT was significantly increased,and TWL was prolonged at 7 days after curcumin injection,and the expression of pTyr1472-NR2B was down-regulated at 3 days after curcumin injection in group DCur.There was no significant difference in each parameter between DNP and DSC groups,and in the expression of pSer896-NR1 between the four groups.Conclusion The mechanism by which curcumin mitigates neuropathic pain in type 2 diabetic rats may be related to inhibition of up-regulation of pTyr1472-NR2B expression,while unrelated to pSer896-NR1 expression in the spinal dorsal horn.
6.The relationship between operation scores and the expression of caveolin-1 protein in the cortex of mice

Objective To explore the relationship between operation scores and the expression of caveolin-1 ( Cav-1 ) protein in the cortex of mice.Methods Male Kunming mice were used and divided into two groups (excellent group and bad group) according to the training scores after 4 days' Y-maze learning training.The expression of cav-1 protein in cortex and hippocampus of the mice with different scores were measured through Western blot technology.Results Cav-1 protein expression of excellent group( Cav-1/β-actin was 5.71 + 1.11 ) in the cortex was significantly higher than that of the bad group ( Cav-1/β-actin was 1.69 + 0.20) (P < 0.01 ).While there was no significant difference in the two groups in the expression of Cav-1 protein in hippocampus(P > 0.05 ).Conclusion The operation scores have distinct correlation with the expression of Cav-1 protein in the cortex of mice.
7.Effect of placental growth factor gene silencing on migration and invasion of human pancreatic carcinoma cell line PANC1

Objective To explore the effect of inhibiting placental growth factor ( PIGF ) by small interfering RNA ( siRNA) on migration, invasion and chemoresistance of human pancreatic cancer cell line PANC1.Methods Three specific siRNAs targeting PIGF (siRNA-PIGF) were designed.PANC1 cells were transfected with siRNA-PIGF by liposome transfection using untransfected cells as blank controls and nonspecific siRNA ( siRNA-NC) transfected cells as negative controls .The PIGF mRNA and protein expression was examined by real-time RT-PCR and ELISA.MTT method was used to assess the inhibition rate of chemotherapeutic reagents on cell proliferation .The abilities of migration and invasion were evaluated by Transwell assay.Results The inhibition rate of PIGF mRNA in PANC1 cells transfected by 3 siRNA-PIGF were (64.38 ±8.92)%, (70.48 ±7.72)% and (81.25 ±6.02)%, which was lowest in siRNA-PIGF-3 transfected cells.The expression of PIGF mRNA in PANC1 cells were decreased by (63.72 ±8.20)%at 24 h after siRNA-PIGF transfection compared with siRNA-NC transfected cells;and the level of PIGF protein in the supernatant of cultured PANC1 cells was lowered by (42.92 ±1.34)% compared with siRNA-NC transfected cells and by (46.25 ±3.64)% compared with untransfected cells at 48h after transfection, which all had significant difference .There was no statistical difference between untransfected and siRNA-NC transfected cells.After 3 ng/L gemicitabine treatment , the inhibition rate of cell proliferation in siRNA-PIGF group was even higher than that in siRNA-NC and untransfected group [(44.35 ±5.05)% vs(34.29 ±3.60)% and (31.01 ±1.08)%;both P<0.05], and no significant difference was not observed after 5-FU and adriamycin treatment.In migration and invasion assay , the number of transmembrane cells from siRNA-PIGF group was 38.1%and 28.2%of that from siRNA-NC group and 40.8% and 36.2% of that from untransfected group , which had statistical difference (all P<0.05).Conclusions PIGF silencing could significantly suppress the migration and invasion of PANC 1 cells and improve the sensitivity to gemicitabine .
8.Current state of secondary prevention medication for patients with high risk of recurrent ischemic stroke in Changzhou city

Objective To investigate the application of secondary prevention medication for patients with high risk of recurrent ischemic stroke in Changzhou city,analyze the reasons for decreased medication compliance,and evaluate the current secondary prevention medication.Methods We investigated 300 consecutive hospitalized patients with acute non-cardiogenic and ischemic stroke high risk.High risk of recurrent stroke was defined as ESSEN Stroke Risk Score (ESRS) ≥3.Different ESRS scales consisting of different parameters were analyzed.All of the patients received standard secondary prevention of ischemic stroke at discharge.After three months and a year follow up,antiplatelet therapy,therapy of risk factors (hypertension and diabetes mellitus),lipid lowering therapy,and medication compliance were investigated.Results Except for age (x2 =126.54,P =0.000) and previous cerebral ischemic stroke or transient ischemic attack (TIA) (x2 =21.27,P =0.000),there were no significant differences in other risk factors (hypertension,diabetes,previous myocardial infarction,heart diseases,smoke) in patients with different ESRS scale scores (all P> 0.05).Antiplatelet therapy utilization was 98.3% (295/300),antihypertensive and antidiabetic drug use rates were 95.0%(255/268) and 100%(72/72),statin use rate reached to 99% (297/300) at discharge.After three months follow up,medication compliance in hypertension and diabetes mellitus therapy was the best [88.1%(222/252)and 86.2% (56/65)],followed by aspirin [82.0% (228/278)],and clopidogrel [6.1% (17/278)].The medication compliance in lipid lowering therapy was the worst [60.1% (167/278)].After a year follow-up versus the previous three-month follow-up,the medication compliance in hypertension and diabetes mellitus therapy was increased,but had no significant difference [89.9 % (220/245) vs.88.1% (222/252),93.4%(57/61)vs.86.2%(56/65),P>0.05],and the medication compliances inantiplatelet therapy with aspirin and clopidogrel,and lipid lowering therapy were increased significantly [93.2% (245/263)vs.82.0% (228/278),30.8(81/263) vs.6.1% (17/278),88.9% (234/263) vs.60.1% (167/278),all P<0.01].The increment of use rate was higher in clopidogrel therapy than in aspirin therapy.Conclusions The secondary prevention medication achieves certain efficacies in patients with high risk of recurrent ischemic stroke in changzhou city.Long term follow-up and good communication between doctor and patient can effectively improve the medication compliance in secondary prevention,and can increase the use rate of antiplatelet therapy in patients with high risk of recurrent ischemic stroke.
9.Efficacy of Drugs with Different Mechanisms of Action on Functional Dyspepsia:A Comparative Study

Background:Oryz-Aspergillus Enzyme and Pancreatin Tablet is a double-deck digestive enzyme pellet containing Oryz-Aspergillus enzyme and pancreatin that has been widely used for treatment of functional dyspepsia (FD)in clinical practice. However,there is no randomized controlled trial focusing on the efficacy of this agent versus other drugs used for treatment of FD such as prokinetics and proton pump inhibitors (PPI). Aims:To compare the efficacy and safety among Oryz-Aspergillus Enzyme and Pancreatin Tablet,Domperidone Tablet and Esomeprazole Magnesium Enteric-coated Tablet, three drugs with different mechanisms of action on FD,in Chinese population. Methods:A total of 82 Helicobacter pylori-negative outpatients fulfilling the diagnostic criteria of FD in Rome Ⅲ were recruited from Nov. 2015 to Jun. 2016 at the First Affiliated Hospital of Zhejiang Chinese Medical University. These patients were randomly allocated into 3 groups,and received Oryz-Aspergillus Enzyme and Pancreatin Tablet (group A),Domperidone Tablet (group B)and Esomeprazole Magnesium Enteric-coated Tablet (group C)orally for 4 weeks,respectively. The improvement of dyspeptic symptoms and adverse events were observed and recorded. Results:After 4 weeks treatment,the overall efficacies for global symptoms in group A,group B and group C were 93. 1%,88. 9% and 69. 2%,respectively,statistically significant difference was existed among the three groups (P < 0. 05). Domperidone Tablet was effective for postprandial fullness and early satiety;Esomeprazole Magnesium Enteric-coated Tablet was sensitive for epigastric pain,epigastric burning,and belching and regurgitation;the efficacies of Oryz-Aspergillus Enzyme and Pancreatin Tablet for all five dyspeptic symptoms were in between. No adverse events were observed during treatment course. Conclusions:Digestive enzymes,prokinetics and PPI have different sensitive symptoms and optimal indications for treatment of FD. The overall efficacy of Oryz-Aspergillus Enzyme and Pancreatin Tablet is superior to that of prokinetics and PPI.
10.Application of multi-copies in expression of smaller peptides: a review.

The technology of genetic engineering has been widely used to express macromolecules such as enzymes. However, it is difficult to detect and purify the micromolecules such as small peptides, because of their instability and degradability. Construction of multi-copy recombinant expression plasmid can be achieved by inserting multiple target genes or expression cassette containing target genes with the same orientation into expression vector. This is effective to increase the expression level of small peptides. In this article we described four methods in order to provide some optional methods and ideas for the expression of active small peptides.
