1.Comparison of the effects of single emotional stimulus and complex emotional stimuli on the rat model of liver depression syndrome

Objective To compare the effects of single and complex emotional stimuli on animal model of liver depression syndrome,in order to find out an animal model of liver depression syndrome with more similar clinical manifestations.Methods Based on the previous screening work,different methods,i.e.tail clamping,limb binding,putting a cangue on the neck,were used respectively,or using the three methods in combination to establish rat models of liver depression syndrome.A comparison table of human and rat liver depression syndrome was prepared,a mark sheet of evaluation criteria was made,general condition was observed and body weight was measured,and positive response to drugs and changes of hormonal levels were evaluated to identify whether the models were established successfully.Results Rats in the control group grew well and showed no abnormalities.There were lots of abnormal phenomena in the model group especially in the complex emotional stimulus modeling group.They were maniac with acute stress or depressive with chronic stress,had dry hair,small and less feces,many of the rats had hair loss,red eyes and shortness of breath symptom.They showed a significantly lower body weight gain and higher levels of CRH in hypothalamus and plasma ACTH and CORT.The manifestations were much more severe in the complex-emotional stimuli-induced model group than that in the single-stimulus prepared model groups.The drug-treated group showed considerable alleviation of the symptoms and improvement of the parameters closing to normal levels.Conclutions Establishing the rat model of stagnation of liver depression syndrome by complex emotional stimuli modeling method has good stability,with a short preparation period and high success rate,and presenting a better consistence with clinical liver stagnation of depression syndrome,so as to provide a suitable tool for TCM research.
2.Determination of alkaloid from combination of Rhizoma Coptidis and Fructus Evodiae by TLCS

AIM: To investigate the changes of alkaloid in Rhizorrm Coptidis and Fructus Evodiae before and after combination. METHODS: The content of berberine hydrochloride,palmatine hydrochloride,jatrorrhizine hydrochloride in Rhizoma Coptidis and Fructus Evodiae before and after combination were determined by TLCS.RESULTS : After combination of Rhizoma Coptidis and Fructus Evodiae,contents of berberine hydrochioride,palmatine hydrochloride,jatrorrhizine hydrochloride significantly reduced,among which the reduction of berberine hydrochloride was more obvious.CONCLUSION: In decoction,Fructus Evodiae appears to restrict the alkaloid from Rhizoma Coptidis in order to lower the side effect of Rhizoma Coptidis.
3.Optimization of the establishment protocol of a mouse model of MBA/TPA-induced skin cancer

Objective To induce skin cancer in BALB/c mice using DMBA as initiator and TPA as tumor promot-er. Through optimizing the doses and frequencies of DMBA administration to establish a stable skin cancer model with less time and causing less skin damage. Methods Shaving the back of mice to expose a piece of skin around 2 cm × 2 cm. The mice were divided into a blank control group and four treatment groups randomly. These four groups were given 1, 2, 4, 7 times 100μg/100μL DMBA/acetone, respectively, in the first week, and twice 4μg/100μL TPA/acetone per week in the next 11 weeks. The body weight changes, time of tumor formation and number of tumors formed were recorded during the experiment. The mice were sacrificed at 12th week and samples of tumor tissue and adjacent normal skin tissue were taken for pathological examination using HE staining. Results Tumors were observed at the 7th week in the group with once DMBA administration in the first week and at the 4th week in the group with twice DMBA administration in the first week. Skin cancers were formed also in the group with 4?time DMBA administration in the first week, however, with signif?icantly more severe skin damages. The mice receiving DMBA everyday in the first week died at the 3th week. Conclusions The best induction protocol for skin cancer in BALB/c mice should be twice DMBA in the first week followed by twice TPA
4.Differences of serum uric acid levels between Uygur and Han people who underwent physical examination and its correlation with lipid metabolism

Objective To investigate the relationship between uric acid levels and lipid metabolism in Uyghur and Han people who underwent physical examination.Methods From February 2012 to February 2013,8 197 blood samples were collected for the tests of serum uric acid and some other relevant lipid metabolism indicators in the First Affiliated Hospital of Xinjiang Medical Uni-versity.Results The uric acid level of Han was higher than that of Uyghur people(P <0.05),the levels of uric acid has gradually increased with the growing age both in the Uygur and Han people.The uric acid and high-density lipoprotein and apolipoprotein-A showed negative correlation between Uyghur and Han(P <0.05),however,the uric acid and other factors showed positive correla-tion(P <0.05).Conclusion There is ethnic differences in the level of uric acid and the prevalence of hyperuricemia in people who underwent physical examination of Urumqi,there are correlations between uric acid levels and the lipid metabolism.

6.Hairy root culture optimization and aucubin medical composition production of Eucommia ulmoides.

OBJECTIVETo introduce the hairy root of Eucommia ulmoides, and screen put the excellent roots.
METHODHairy roots were obtained by infecting the different explants of E. ulmoides. Aucubin content was detected by HPLC.
RESULTThe best transformation condition was embryonic axis infected by twenty minutes bacillus water dip-dye and three-days co-culture. Liquid culture was better than solid culture, and advantageous to accumulating of biomass and medical contents of hairy root. The E1 was the excellent roots.
CONCLUSIONHairy root of E. ulmoides will be useful for the production of active components in aucubin.

7. Three-dimensional echocardiography combined with myocardial contrast echocardiography for assessment of myocardial dysfunction in patients with type 2 diabetes mellitus

Objective: To explore the value of real-time three-dimensional echocardiography (RT-3DE) combined with myocardial contrast echocardiography (MCE) in evaluating myocardial dysfunction in type 2 diabetes mellitus (T2DM) patients. Methods: Totally 58 T2DM patients were divided into group (T2DM isolated ) and group Ⅱ (T2DM associated with microvascular complications, each n=29). Group C included 32 normal individuals. The standard deviation (Tmsv16-SD, Tmsv12-SD, Tmsv6-SD) and the maximum time difference (Tmsv16-Dif, Tmsv12-Dif, Tmsv6-Dif) of the time to the minimum systolic volume of left ventricle were measured with RT-3DE. MCE was performed to obtain perfusion measurement of each segment of ventricular wall, including the acoustic intensity (A), microvascular flow velocity (β) and A•β. Results: There were statistically significant differences in all synchronize measurements except Tmsv6-Dif between group Ⅱ and group C (all P<0.05). Tmsv12-Dif, Tmsv12-SD, Tmsv12-Dif% and Tmsv12-SD% of group prolonged comparing with group C (all P<0.05). All the parameters of A, β and A•β showed a gradually decreasing trend in group C, group and group Ⅱ, while those in group Ⅱ were lower than group C (all P<0.05). The parameters of β and A•β in group Ⅱ were lower than those in group (both P<0.05). Compared with group C, the values of β and A•β in group decreased (both P<0.05). There was no significant difference of the same measurement in segments among three groups (all P>0.05). Conclusion: Left ventricular desynchrony can be observed in T2DM patients with or without microvascular complications. Myocardial perfusion reduce in T2DM patients presenting as diffuse damage, which aggravated with microvascular complications in other organs.
8.A 180-day mortality predictive score based on frailty syndrome in elderly patients with sepsis: a Logistic regression analysis model

Objective:To establish a 180-day mortality predictive score based on frailty syndrome in elderly sepsis patients [elderly sepsis score (ESS)].Methods:A prospective study for sepsis patients aged 60 years and above who were admitted to a medical intensive care unit of the General Hospital of Southern Theatre Command from January 1st, 2018 to December 31st, 2018 was conducted. Univariate analysis was performed on 19 independent variables including gender, age, body mass index (BMI), tumor, charlson comorbidity index (CCI), activity of daily living (ADL), instrumental activity of daily living (IADL), mini-mental state examination (MMSE), geriatric depression scale (GDS), clinical frail scale (CFS), sequential organ failure assessment (SOFA), Glasgow coma scale (GCS), acute physiology and chronic health evaluation (APACHEⅡ, APACHEⅣ), modified NUTRIC score (MNS), multiple drug resistance (MDR), mechanical ventilation (MV), continuous renal replacement therapy (CRRT) and palliative care. Continuous independent variables were converted into classified variables. Multivariate binary regression analysis of risk factors was conducted to screen independent risk factors which affecting 180-day mortality in elderly sepsis patients. Then a 180-day mortality predictive score was established, and the discrimination of the mortality of patients using CFS, SOFA, GCS, APACHEⅡ, APACHEⅣ, MNS scores were compared.Results:A total of 257 patients were enrolled, with a 180-day mortality of 60.7%. Univariate analysis showed that age, tumor, CCI, ADL, IADL, MMSE, CFS, SOFA, GCS, APACHEⅡ, APACHEⅣ, MNS, MDR, MV, CRRT, palliative care were risk factors of 180-day mortality in elderly sepsis patients [age: odds ratio ( OR) = 1.027, 95% confidence interval (95% CI) was 1.005-1.050, P = 0.018; tumor: OR =2.001, 95% CI was 1.022-3.920, P = 0.043; CCI: OR = 1.193, 95% CI was 1.064-1.339, P = 0.003; ADL: OR = 0.851, 95% CI was 0.772-0.940, P = 0.001; IADL: OR = 0.894, 95% CI was 0.826-0.967, P = 0.005; MMSE: OR = 0.962, 95% CI was 0.937-0.988, P = 0.004; CFS: OR = 1.303, 95% CI was 1.089-1.558, P = 0.004; SOFA: OR = 1.112, 95% CI was 1.038-1.191, P = 0.003; GCS: OR = 0.918, 95% CI was 0.863-0.977, P = 0.007; APACHEⅡ: OR = 1.098, 95% CI was 1.053-1.145, P < 0.001; APACHEⅣ: OR = 1.032, 95% CI was 1.020-1.044, P < 0.001; MNS: OR = 1.315, 95% CI was 1.159-1.493, P < 0.001; MDR: OR = 2.029, 95% CI was 1.197-3.437, P = 0.009; MV: OR = 6.408, 95% CI was 3.480-11.798, P < 0.001, CRRT: OR = 2.744, 95% CI was 1.529-4.923, P = 0.001, palliative care: OR = 5.760, 95% CI was 2.177-15.245, P < 0.001]. By binary regression analysis, CFS stratification ( OR = 1.934, 95% CI was 1.267-2.953, P = 0.002), MV ( OR = 4.531, 95% CI was 2.376-8.644, P < 0.001), CRRT ( OR = 2.471, 95% CI was 1.285-4.752, P = 0.007), palliative care ( OR = 6.169, 95% CI was 2.173-17.515, P = 0.001) were independent risk factors of 180-day mortality in elderly patients with sepsis. The model of "ESS = 0.660×CFS stratification+1.511×MV+0.905×CRRT+1.820×palliative care" was established. Receiver operating characteristic curve (ROC curve) analysis showed that the area under the ROC curve (AUC) for predicting 180-day mortality by ESS was 0.785 (95% CI was 0.730-0.834, P < 0.001). When the best cut-off value was 2.2 points, its sensitivity was 78.9%, specificity was 70.3%, the positive predictive value was 80.4%, and the negative predictive value was 68.3%. Simplified ESS was defined as "0.5×CFS stratification+1.5×MV+1×CRRT+2×palliative care". ROC curve analysis showed that AUC for predicting 180-day mortality by simplified ESS was 0.784 (95% CI was 0.729-0.833, P < 0.001). When the best cut-off value was 2.0 points, sensitivity was 76.9%, specificity was 70.3%, the positive predictive value was 80.0%, and the negative predictive value was 66.4%. Compared with CFS, SOFA, GCS, APACHEⅡ, APACHEⅣ and MNS, ESS had a significant difference in discriminating 180-day mortality in elderly patients with sepsis (AUC was 0.785 vs. 0.607, 0.607, 0.600, 0.664, 0.702, 0.657, 95% CI: 0.730-0.734 vs. 0.537-0.678, 0.537-0.677, 0.529-0.671, 0.598-0.730, 0.638-0.766, 0.590-0.725, all P < 0.05). Conclusions:CFS, MV, CRRT, and palliative care are independent risk factors of 180-day mortality in elderly patients with sepsis. We established ESS based on these risk factors. The ESS model has good discrimination and can be used as a reference and assessment tool for prediction and treatment guidance in elderly patients with sepsis.
9.The application of three-dimensional breath-hold gradient and spin-echo sequence in the MR cholangiopancreatography

Objective:To evaluate the clinical feasibility and image quality of three-dimensional breath-hold gradient and spin-echo (3D BH-GRASE) sequence in magnetic resonance cholangiopancreatography (MRCP).Methods:In this prospective study, 59 patients with clinically suspected pancreaticobiliary duct disease performed MRCP with both 3D BH-GRASE and 3D respiration-triggered turbo spin-echo (3D RT-TSE) sequences on 3.0 T scanner in the Affiliated Zhangjiagang Hospital of Soochow University from November 2017 to December 2018. The overall image quality was scored independently by 3 experienced radiologists based on the visibility of different anatomical features of the pancreaticobiliary duct. For comparing the 2 sequences, the statistical difference in scan time was assessed with a paired t test; while subjective scores, signal-to-noise ratios (SNR), contrast ratios (CR) and contrast noise ratios (CNR) were compared with Wilcoxon signed rank test. Results:The scan time of 3D BH-GRASE sequence was 16.4 s while that of 3D RT-TSE was (258.6±42.2) s. Their difference was statistically significant ( t=44.073, P<0.001), with the scan time for 3D BH-GRASE shortened by 94%. The overall quality scores of 3D BH-GRASE images were better than those of 3D RT-TSE ( Z=-6.595, P<0.001). There was no statistical difference ( P>0.05) in the scores regarding the visibility of the upper, middle and lower parts of common bile duct and the first and second branches of left and right hepatic ducts. For visualizing the bottom, body, neck and duct of gallbladder, the 3D BH-GRASE sequence received a higher score than the 3D RT-TSE sequence ( P<0.001). For displaying the proximal, middle and distal segments of main pancreatic duct, the 3D RT-TSE sequence was scored higher than the 3D BH-GRASE sequence ( P<0.05). There was no significant difference of SNR between the two sequences ( Z=0.403, P=0.687), whereas CR and CNR of 3D RT-TSE MRCP were better than those of 3D BH-GRASE MRCP ( Z=6.215, P<0.001 and Z=3.046, P=0.002, respectively). Conclusion:Under the prerequisite of ensuring image quality, a proper use of 3D BH-GRASE sequence can significantly shorten the scan time and thus greatly improve the working efficiency of MRCP examination.
10.Construction of conditionally replicative adenovirus vector carrying TRAIL gene and its mRNA and protein expressions induced by ionizing radiation

Objective To construct the conditionally replicative adenovirus vector pAd-Egr1-TRAIL-hTERT-E1A-E1Bp-E1B55K carrying early growth response gene-1 (Egr1)promoter and tumor necrosis factor related apoptosis inducing ligand (TRAIL)gene, and to observe the effects of the vector combined with 2 Gy irradiation on the TRAIL expression in MDA-MB-231 cells.Methods Egr-1 promotor sequence was cloned from pMD18 T-Egr1, TRAIL was constructed the downstream of Egr1 promoter, pShuttle-Egr1-TRAIL-hTERT-E1A-E1Bp-E1B55K (CRAd.pEgr1-TRAIL)was constructed,after the adenovirus vector was packaged successfully,MDA-MB-231 cells were infected with them and irradiated with X-rays.Real time PCR method and ELISA were used to detect the expression levels of TRAIL mRNA and protein, respectively. Six groups in the experiment were set up:control, 2 Gy,CRAd.p,CRAd.pEgr1-TRAIL,CRAd.p + 2 Gy and CRAd.pEgr1-TRAIL + 2 Gy. Results The recombinant adenovirus vector pAd-Egr1-TRAIL-hTERT-E1A-E1Bp-E1B55K was constructed and packaged successfully.The expression level of TRAIL mRNA in MDA-MB-231 cells transfected with the vector of 5 MOI for 24 h following 2.0 Gy X-rays irradiation began to increase and arrived to the top 8 h later in various groups,then declined.The expression level of TRAIL protein in MDA-MB-231 cells began to increase 6 h after irradiation and reached to the peak 24 h later,then declined 48 h later.There were significant differences in the expression levels of TRAIL protein between CRAd.pEgr1-TRAIL + 2.0 Gy and other groups at the same time point (P<0.01). Conclusion The recombinant adenovirus vector is obtained successfully, and the TRAIL mRNA and protein expression levels in MDA-MB-231 cells can be increased significantly by the vector combined with 2.0 Gy X-rays irradiation.
