2.Immunometabolism and regulatory T cells

Recent studies have shown that immune cells play an important role in the development of non-immune diseases.Regulatory T cells (Treg) are a group of T cell subsets which have immunoregulatory and immunosuppressive functions.Abnormalities in the number and function of Treg cells may cause immune imbalance, which leads to the development of diseases.Immunometabolic disorders may in turn affect the generation, proliferation and function of Treg cells.Clarification of the relationship and interaction between immunometabolism and Treg cells will make it possible to treat metabolic diseases with cellular immunotherapy.
3.Comparative analysis of upper ureteral stones treated with retroperitoneoscopic ureterolithotomy and percutaneous nephrolithotomy

To explore an appropriate treatment for patients with upper ureteral stone (> 15 mm in size) by comparing the therapeutic outcomes for those undergoing retroperitoneoscopic ureterolithotomy (RPUL) and percutaneous nephrolithotomy (PCNL).A total of 62 patients with large upper ureteral stone were divided randomly into 2 groups.RPUL was performed in group A (n =30) and PCNL in group B (n =32).No significant inter-group difference existed in age,gender,stone location and size,operative duration or surgical success rate (P > 0.05).Blood loss volume,rate of postoperative analgesic usage,time of hospital stay and incidence of postoperative complications in group A were significantly lower than those of group B (P < 0.05).Thus RPUL offers more advantages and better efficacies.
4.The effect of community comprehensive in prevention and treatment of hypertension

Objective To analyze the clinical effect of community comprehensive intervention in the prevention and treatment of hypertension.Methods A total of 160 cases of patients were randomly divided into two groups,the control group (80 cases)were treated with conventional antihypertensive drug treatment;the observation group 80 cases of patients were treated with comprehensive community intervention on the basis of treatment of the control group.Two groups of patients before and after the intervention of blood pressure changes were observed and recorded,treatment effect and medication compliance were comparative analysized.Results The total effective rate of patients in the observation group was 86.25%,which was significantly higher than that in the control group (57.5%),the difference was statistically significant (χ2 =20.44,P <0.05).DBP,SBP after intervention of patients in the observation group were significantly lower than those before intervention and in the control group after the intervention,the differences were statistically significant (t =18.48,6.06,all P <0.05).Conclusion The effect of community comprehensive intervention on hypertension patients is significant,it can effectively control the patient's blood pressure level,improve the quality of life of patients,which is worth promoting.
5.Dosimetry of endovascular irradiation using beta emitters in the prevention of restenosis

Objective To give a dose reference to the researcher who study prevention of restenosis using endovascular brachytherapy Methods The dose distributions around a 3 mm diam by 20 mm long balloon filled with 90 Y , 32 P and 186 Re separately have been calculated by a computer program based on a dose point kernel Results The peak value of the absorbed dose is found inside the balloon, the inflexion of the dose curve at the endothelial surface of the vessel In the vessel tissue the dose decreases with distance in an approximately exponential manner The lower the nuclide energy is, the faster the dose falls with distance, and the longer the region of uniform dose distribution along vessel wall is The dose rate at the surface of the vessel wall is approximately 74, 71, and 45 mGy/min per mCi/ml, with the dose rate decreasing to 53%, 48% and 28% at 0 5 mm for 90 Y , 32 P and 186 Re separately Conclusion The fast falling of the dose in an approximately exponential manner in the vessel tissue makes for focusing ? radionuclide energy to the vessel wall target and reducing the injury of the normal tissue around the vessel, and especially 186 Re with lower energy has a more obvious advantage in this aspect
6.Study on change of thymus in different clinical period of EAE mice

Objective:To investigate the change of thymus in different period of EAE mice. Methods: The thymic index and thymocyte number at 0,10,15,20 days after EAE induced were recorded. The percent of thymic apoptosis and CD4+CD44+T cells in spleen was acquired by Flow Cytometry. The content of p53 and Bcl-2 gene was detected by PCR and electrophoretic technique. Results:The thymic index and thymocyte number correspondingly reduced at 0,10,15 days after EAE induced. Apoptotic rate of thymocyte increased at 10,15 days after EAE induced,highest at 10 days. The percent of CD4+CD44+T cells also increased at 10,15 days after EAE induced, highest at 10 days. The content of p53 increased, while Bcl-2 reduced at 10 days after EAE induced. Conclusion:The atrophy of thymus is most serious at 15 days after induction;while the apoptotic percent is highest at 10 days after induction,which have a relationship with regulation of p53 and Bcl-2. The change of thymus in EAE mice closely related with EAE attack,and the recovery of thymus precede EAE.

Objective To investigate the inhibitory effects of IL-18 gene on HCC growth in vivo. MethodsThe recombinant adenovirus vector containing IL-18 gene was constructed and cotransfected into 293 cells together with EcoT22 I-digested Ad5 DNA-TPC, the recombinant adenoviruses were generated, and injected into a rat model bearing HCC. Results The recombinant adenovirus vector containing IL-18 gene inhibited the proliferation of HCC cell line CBRH 3. 8.Diagnosis and surgical treatment for primary presacral tumors

Objective To investigate the clinical characteristics and surgical treatment for primary presacral tumors.Methods The clinical data of 42 patients of primary presacral tumors from January 2013 to May 2015 were analysed retrospectively.Results Of the 42 patients,16 cases were asymptomatic while 26 patients had discomfort at the sacral or abdominal region,or difficulty in urinating or defecation.90% of the cases were digital rectum examination (DRE) positive.Among the 42 patients 36 cases underwent surgical treatment,1 case underwent radiotherapy,5 cases refused surgical treatment.Among those receiving surgical resection,28 cases had trans-abdominal surgery and 4 cases had trans-sacral surgery,while 3 cases had trans-abdominal & trans-sacral surgery,1 case had trans-abdominal and perineal surgery.Tumors were completely resected in 31 cases,and palliatively resected in 5 cases.3 cases suffered from intra-operative presacral hemorrhage.1 case with delayed hemorrhage required surgical intervention.2 cases from incision infection recovered after wound disinfection and dressing.3 cases had postoperative hip or leg numbness;1 case with high fever was cured by intensive antibiotics treatment.Conclusion The low incidence of presacral tumors makes early detection difficult.A diagnosis can be obtained by a positive DRE combined with CT or MRI results.Resection is a therapy of choice after biopsies.
9.Molecular Identification of Processed Medicinal Insects Chinese polyphaga Based on Cytb Gene

OBJECTIVE:To provide new identification method for processed medicinal material Chinese polyphaga(Eupolyph-aga sinensis,Steleophaga plancyi) and their adulterants by establishing molecular identification method based on Cytb genes. METHODS:The total DNA of Chinese polyphaga and their adulterants was extracted using modified saturation sodium chloride method. The Cytb genes of all samples were amplified with PCR using general primers REVCB2H and REVCBJ. The phylogenetic tree of all samples was constructed with Neighbor-Joining(NJ)method using MEGA 5.1 software. The sequences of the Cytb gene of all sampled were compared by using DNAMAN sofetware. The difference between genuine product and their adulterants were analyzed,and the specific primers Esin-F and Esin-R were designed for molecular identification in different regions. RESULTS:DNA extracted from processed medicinal insects was successful to amplify Cytb gene segments. The phylogenetic tree of all sam-ples was consistent with their genetic relationship. A fragment was amplified only from genuine product but not from other adulter-ants with the designed specific primers Esin-F and Esin-R. CONCLUSIONS:DNA extraction method from processed Chinese polyphaga and their adulterants have been established. Designed specific primers are highly specific to genuine product Chinese polyphaga,and can be used for the identification of Chinese polyphaga and their adulterants.
10.Synthesis of podo-and epipodo-11C-podophyllotoxin and their biodistribution in mouse bearing EMT6 tumor

Objective To synthesize podo-and epipodo-podophyllotoxin labeled with 11C and investigate their biodistribution in mice bearing EMT6 tumor by microPET.Methods 11 C-podo-or epipodopodophyllotoxin was synthesized by 11 C-CH3-Triflate mixed with 4'-methyl-demethmyl-podophyllotoxin (podo-and epipodo-) and purified using HPLC.The radiochemical purity (RCP) was analyzed by HPLC.Thirty mice bearing EMT6 tumor were divided into 2 groups and injected with 3.7 MBq 11C-podo-or epipodo-podophyllotoxin.The biodistribution of 11C-podo-or epipodo-podophyllotoxin was evaluated with microPET.The ID%/g was calculated.Paired t test was used to analyze the data.Results The yields of 11C-podo-and epipodo-podophyllotoxin were both >90%.The RCP was >99%.The biodistribution of 11C-podo-and epipodo-podophyllotoxin was similar with slow blood clearance and high uptake in abdomen.The tumor uptake of 11C-epipodophyllotoxin and 11C-podophyllotoxin at 30 min was (3.63±0.98) %ID/g and (3.16±0.27) %ID/g,respectively.The uptake ratio of tumor to blood and to muscle was 0.68 vs 0.62 and 1.52 vs 1.22,respectively.There was no significant difference between the tumor uptake of the two agents (t=0.47,P>0.05).The result of microPET imaging was consistent with that of mice experiment.Conclusion 11C-podophyllotoxin has limited clinical value for tumor imaging.
