1.Comparison study of emergency laparoscopic appendectomy and open appendectomy

ObjectiveTo compare the efficacy of laparoscopic appendectomy(LA) and open appendectomy (OA) in treating acute appendicitis.MethodsThe clinical data of eighty patients admitted with acute appendicitis given LA or OA were setrospectively studied and the recovery condition after LA or OA of the two groups were compared.ResultsTime of gastrointestinal functional recovery and hospital day of the LA group were obviously shorter than the OA group[(15.1±7.2)d vs (28.0 ± 11.3)d,(4.2 ±1.9)d vs (5.5 ±3.2)d,all P <0.05].The rate of postoperative complications after LA was fewer too.ConclusionLA had advantages of faster function recovery,shorter hospital day and fewer complications,which had higher clinical application value.
2.Cyr61 expression influences cancer cell proliferation and apoptosis via PI3K pathway in human ovarian carcinoma cells

Objective To investigate the relationship between cysteine-rich protein 61 ( Cyr61 ) and phosphatidylinositol 3-kinase ( PI3K ) signal pathway on cell proliferation and apoptotic in human ovarian carcinoma cells.Methods Recombinant human Cyr61 (rhCyr61) was pretreated with ovarian carcinoma cells.The expression of Cyr61 protein was detected by confocal spectral microscopy.Then treated the ovarian carcinoma cells with PI3K transduction inhibitors (LY294002) for 24 hours.Cell apoptosis was detected by flow cytometry (FCM).Cell viability was determined by methyl thiazolyl tetrazolium (MTT) method.The mRNA expressions of Cyr61,the protein levels of protein kinase B ( PKB),phospho-PKB and Cyr61 were assaved by real time-PCR and western blot analysis,respectively.Results The Cyr61 and phospho-PKB protein expression in two ovarian carcinoma cells (OV2008 and OVCAR-3 ) were increased in rhCyr61pretreated group.The decreasing of cell apoptosis [ ( 1.4 ±0.9)%,(2.1 ± 1.0)% ] and increasing of cell proliferation [ ( 124.0 ± 1.8)%,( 133.0 ±2.2)% ] was detected in the same time,compared with negative control group,there were significant difference ( P < 0.05 ).After exposed to LY294002 for 24 hours,the apoptosis rate of OV2008 and OVCAR-3 in pretreated with rhCyr61 group exposed to LY294002 were (21.1 ± 1.6)% and (26.4 ± 1.5 )%,respectively.Cells viability [ (59.0 ± 2.3 )%,(51.0 ± 2.0)% ]was also significantly decreased in OV2008 and OVCAR-3 pretreated with rhCyr61 cells.Meanwhile,the mRNA expressions of Cyr61 (3.2 ± 0.8,6.2 ± 1.1 ) and the protein levels of phospho-PKB and Cyr61 were greatly decreased.Compared with negative control group,there were significant difference in OV2008 and OVCAR-3 cells (all P < 0.0l ).Conclusions The activation of PI3K intracellular signaling pathways may lead to up-regulation of Cyr61 expression.Block PI3K signal pathway could significantly inhibit the expression of Cyr61,and may promote the apoptotic effects and inhibit the cell growth of ovarian carcinoma cells.
3.Content Determination of Baicalin in Chang'an Capsule by RP-HPLC

OBJECTIVE:To establish the method for determination of baicalin in Chang’an capsule by RP-HPLC. METHODS:HPLC was performed on C 18 column with methanol-0.07%phosphoric acid solution(44∶56)as mobile phase at a flow rate of lml/min and the temperature of column kept at room temperature.The UV detection wavelength was280nm.RESULTS:The linear range of baicalin was sample size0.1054?g~1.0540?g(r=0.9996),the average recovery was97.44%(RSD=2.92%,n=5).CONCLUSION:The method was simple,accurate and fast,and can be used for the determination of baicalin.
4.Experimental study of range verification of proton radiotherapy using off-line PET/CT

Objective:To test the usefulness of PET-range verification (RV) method for proton radiation accuracy verification in poly (methyl methacrylate) (PMMA) phantom using off-line PET/CT scanning.Methods:Proton irradiation dose of 2 Gy and 4 Gy were delivered in PMMA phantom. Given the difference of clinical target volume (CTV), 7 subgroups with different depth (5.0, 7.5, 10.0, 12.5, 15.0, 17.5, 20.0 cm) were set for each dose (14 radiation plans or radiation fields). PET/CT scan was performed 10 min after irradiation of 48-221 MeV proton beam. A co-registration between CT from treatment planning system and PET/CT was performed, as well as the smoothing and normalization of PET/CT data. The region of interest (ROI) and profile lines were drawn with the Raystation PET-RV software. The predictive induced radioactivity and the measured induced radioactivity profile lines were analyzed to evaluate the Δ R50, namely, the error at the position corresponding to 50% of the maximum predictive induced radioactivity at the end of both curves. Results:The size of each ROI was 5.0 cm×5.0 cm×2.5 cm. Profile lines were evenly distributed with the interval of 3 mm, and totally 289 pairs of profile lines were drew. The 2 Gy- and 4 Gy-dose groups yielded similar mean depth errors (Δ R50 between 1 mm and -1 mm with a standard deviation <1 mm). Conclusions:The off-line PET/CT scanning of PMMA phantom reveals a good agreement between predicted and measured PET data, with error of ±1 mm. The PET-RV method can be extended to clinical cases′ verification in human body treatment with further investigation.
5.Analysis of high risk factors for fever after hysterectomy in patients with uterine fibroids

Objective :
To explore the risk factors of fever after hysterectomy in patients with uterine fibroids,and establish a nomogram prediction model to provide theoretical guidance for clinical diagnosis and treatment.
Methods :
The data of patients who underwent hysterectomy due to uterine fibroids were collected and randomly divided into training group and validation group at 6 ∶ 4.Univariate and multivariate analyses were performed by χ2 test and Lo- gistic regression model analysis.The nomogram was established based on the results of multivariate analysis,the calibration curve and receiver operating characteristic,curve were used to evaluate the prediction ability and accura- cy of the nomogram.The decision curve reflected the clinical application value of the model.
Results :
Among 264 patients in the training group,33 patients had fever,and the postoperative disease rate was 12. 5%.The results of univariate and multivariate analysis showed that anemia,preoperative curettage,operation scope and operation time were independent risk factors for fever after hysterectomy in patients with uterine fibroids.The internal and external calibration curves had a good fit with the actual observation results,with an average error of 0. 031 and 0. 025,re- spectively.The area under receiver operating characteristic curve of the internal and external verification of the no- mogramare 0. 788 and 0. 712,respectively,reflecting the good predictive ability of the model.The decision curve showed that the model had good clinical benefit in a certain threshold range.
Conclusion
Based on multivariate a- nalysis,the independent risk factors of fever after hysterectomy in patients with uterine fibroids,including anemia, preoperative curettage,operation scope and operation time.In addition,we established a nomogram with good pre- dictive ability and accuracy,which is helpful for clinicians to timely manage fever after hysterectomy in patients
with uterine fibroids and guide clinical individualized treatment.
