1.Establishment of a rat model for orthotopic allograft kidney transplantation

Objective:To establish a feasible and stable rat model of kidney transplantation. Methods:SD and Wistar rats were used as the donors and the recipients. with the hellp of extradural catheter,the donor inferior vena was anastomosed end-to-end with the recipient left renal vein. An end-to-side anastomosis of abdominal aorta was operated. The donor ureter with ureterocystic flap was conducted to the recipient’s bladder. Results:A modified rat model of kidney transplantation was successfully established. The average total operation time was(130?20) min. The average time of warm ischemia and cold ischemia was less than 20 s and 50 min respectively. The success rate of operation reached about 90.77%. Conclusion:This feasible model of kidney transplantation is feasible and stable.

Objective:To assess the relation and clinical significance between p57 kip2 protein and the biological behavior of bladder transitional cell carcinoma(BTCC).Methods:The expression of p57 kip2 protein and PCNA was detected in 50 cases of BTCC,14 normal bladder mucosa by immunohistochemical method.Results:Positive rate of p57kip2 protein in BTCC was 46%,which was significant lower than 78.6% in normal bladder muscosa.( ? 2=4.660, P 0.05);positive rate of PCNA in BTCC was 60 %(30/50),which was significantly higher than 0%(0/14) in normal bladder mucosa( ? 2=15.812, P
3.Expressions of HSP70 and HSP90? in human bladder transitional cell carcinoma and the clinical significance

Objective To investigate the expression of heat-shock protein 70 (HSP70) and HSP90? in bladder transitional cell carcinoma (BTCC) and its clinical significance in the malignancy of BTCC. Methods Immunohistochemical method was used to detect HSP70, HSP90?, and proliferating cell nuclear antigen (PCNA) in 50 cases of BTCC and 14 cases of normal bladder muscosa served as the controls. Results The positive expression rates of HSP70 and HSP90? in BTCC were 56% (28/50) and 66% (33/50), respectively. They were significantly correlated with the pathological grade, clinical stages, and prognosis. The expressions of HSP70 and HSP90? were significantly correlated to the expression of PCNA. Conclusion Expressions of HSP70 and HSP90? are closely associated with the differentiation of BTCC and its depth of invasion, which may play an important role in the genesis and development of BTCC. HSP70 and HSP90? can be used as a useful molecular marker for prognosis of BTCC.
4.Comparison Study on the Associated Gene Expression in Thymus-dependent Lymphocyte of COPD Patients with Pulmonary Qi Deficiency Syndrome and Pulmonary Yin Deficiency Syndrome

Objective To study the associated gene expression in thymus-dependent lymopholyte of the COPD patients with pulmonary qi deficiency syndrome(PQDS)and pulmonary yin deficiency syndrome (PYDS)by gene chips.Methods Collect peripheral blood of patients with PQDS and PYDS(as experimental samples)and of nomal man(as control samples).By Ficoll method,peripheral blood lymphocyte was collected,then thymus-dependent lymphocyte were extracted and purified by flow cytometry.Total RNA were extracted by one-step technique and purified.Then,synthesis double strand cDNA template from total RNA,transcription of cRNA probe with biotin labeling,subsequently,cRNA sample were fragmented. The gene chip(Human Genome U133 Plus 2.0 Array,38500 genes)was hybridized and scanned.Then fluorescent signal value of gene expressing was obtained,and differential expression genes were sifted. Results There were 45 genes expressed differently among PQDS and normal man,including up-regulated 41 and down-regulated 4;32 genes expressed differently between PYDS and normal man,including up-regulated 19 and down-regulated 13;5 up-regulated genes expressed differently between PQDS and PYDS with normal man.Conclusion Gene chip can be applied to study gene expression profiles effectively and to screen PQDS and PYDS associated genes.

7.Effect of Atorvastatin on expression of vascular endothelia growth factor in rabbit avascular necrosis models of the femoral head

BACKGROUND: Studies have confirmed that Atorvastatin drugs can increase the number of endothelial progenitor cells significantly in vitro, as well as the content of vascular endothelial growth factor(VEGF). OBJECTIVE: To investigate the effect of Atorvastatin on VEGF expression in necrotic femoral heads of rabbits. DESIGN, TIME AND SETTING: The randomized controlled animal experiment was performed at the Department of Orthopedic Surgery, Renmin Hospital of Wuhan University, from September 2007 to November 2008. MATERIALS: Forty-five male and female healthy New Zealand white rabbits weighing 2.5-3.5 kg were randomly divided into normal control group, model control group and AtorvastaUn group, 15 rabbits in each group. METHODS: Nitrogen refrigeration was used to develop femoral head necrosis models of rabbits in the model control and Atorvastatin groups. Two weeks after modeling, the animals in the Atorvastatin group were administered intragastically with Atorvastatin, normal control and model control group were treated with the same volume of normal saline. MAIN OUTCOME MEASURES: Each five rabbits were sacrificed at the 4th, 8th, and 12th weeks respectively for general observation, X-ray and histological observation. VEGF protein expression was assessed by immunohistochemistry method and VEGF mRNA level was assessed by reverse transcription - polymerase chain reaction method. RESULTS: The VEGF protein and mRNA levels in the model control and Atorvastatin groups were obviously lower than those in the normal control group, while the VEGF protein and mRNA levels in the Atorvastatin group were much higher than those in the model control group at the 8th and 12th weeks alter the treatment with Atorvastatin (P < 0.05). CONCLUSION: Atorvastatin can significantly upregulate the expression of VEGF, which is probably an effective clinical treatment to avascular necrosis of the femoral head.
8.Influence of up-regulated Foxp3 expression on regulatory T cells function and kidney transplantation chronic rejection reaction in rat model

Objective To explore the influence of up-regulated Foxp3 on Treg function and kidney transplantation chronic rejection reaction in rat model.Method The kidney transplantation chronic rejection reaction rat model was established.The F344 kidney was transplanted to Lewis rats,and retroviruses highly expressing Foxp3 were constructed.The Banff 97 hierarchical diagnostic criteria were used to diagnose chronic renal allograft nephropathy (CAN).The rat models were divided into three groups by random number table.In experimental group,the pSCV-BsdRFP-FoxP3 retroviruses were injected into the rats via the tail vein after operation.In negative control group,the pSCV-BsdRFP retroviruses were injected into the rats via the tail vein after operation.In blank group,the normal saline was injected into the rats via the tail vein after operation.Enzyme-linked immunosorbent assay (ELISA) was used to detect the changes of interleukin-10 (IL-10) and tumor necrosis factor-β (TGF-β) immediate,1,2,3,and 4 weeks after operation.The rats were killed at 4th week after operation,and kidney tissues were taken out for pathological examination.Result The pathological changes of CAN were observed at 4th week.The typical chronic rejection change was seen at 12th week.The levels of IL-10 and TGF-β were increased,and reached the peak at 3rd week.The levels of IL-10 and TGF-β in experimental group were higher than in negative control group and blank group at 1st,2nd,3rd,and 4th week.At 4th week,obviously different degrees of intimal thickening,and mild hyperplasia of interstitial fibers,glomerular sclerosis and infiltration with lymphocytes and plasma cells were observed in the three groups.In the experimental group,the lesions were mildest,and apparent neointimal hyperplasia was found.Conclusion pSCV-BsdRFP FoxP3 retroviruses can reduce the kidney transplantation chronic rejection reaction in rat model,and have the potential treatment effect.
9.Effect ofXiao-Qing-LongDecoction on AngⅡ, ALD, Na+-k+-ATPase Contents and Expressions of AT1 and AT2 mRNA in Cor pulmonale Rats

This article was aimed to study the effect ofXiao-Qing-LongDecoction (XQLD) on plasma AngⅡ, ALD, Na+-k+-ATPase content, and plasma AT1 and AT2 mRNA expressions in Cor pulmonale rats, in order to further explore the mechanism of ventilating lung qi for diuresis. A total of 60 Wistar rats were randomly divided into the normal group, model group and XQLD, with 20 rats in each group. The enzyme-linked immunosorbent assay (ELISA) method was used to determine the AngⅡ, ALD and Na+-k+-ATPase content. The fluorescence quantitative PCR was used to detect the AT1 and AT2 mRNA expression. The results showed that compared with the normal group, the AngⅡ, ALD and Na+-k+-ATPase contents in the model group were significantly increased (P< 0.05, orP< 0.01). Compared with the model group, the AngⅡ, ALD and Na+-k+-ATPase contents in the XQLD group were obviously decreased (P< 0.05). Compared with the normal group, AT1 mRNA expression was increased; and AT2 mRNA expression was decreased in the model group (P<0.05, orP< 0.01). Compared with the model group, AT1 mRNA expression was decreased; and AT2 mRNA expression was increased in the XQLD group (P < 0.01). It was concluded that XQLD can effectively regulate the AT1 and AT2 mRNA expressions, influence ALD content to ventilate lungqi for dieresis.
10.Effects of Liuwei Buqi capsules on CD4+CD25+forkhead box protein 3+regulatory T cells,helper T cells in patients with chronic obstructive pulmonary disease

Objective To observe the effects of Liuwei Buqi capsules on CD4+CD25+forkhead box protein 3+(Foxp3+)regulatory T cells(Tregs),helper T cells(Ths)in patients with chronic obstructive pulmonary disease (COPD). Methods Eighty COPD patients admitted into the First Affiliated Hospital of Anhui University of Traditional Chinese Medicine from January 2012 to May 2013 were randomly divided into a traditional Chinese medicine (TCM)experimental group(40 cases)and a western medicine control group(40 cases);there were 40 research volunteers enrolled as healthy controls. After admission,both the TCM experimental group and western medicine control group were given conventional western medicine comprehensive treatment. Additionally,in the TCM experimental group,Liuwei Buqi capsules were given orally once 3 pills(0.4 g/pill)and twice a day,while in the western medicine control group nifedipine was given,once 10 mg and 3 times a day,30 days constituting one therapeutic course for both groups. BODE evaluation means that the body mass index(B),the degree of airflow obstruction(O),the scores of dyspnea(D)and excercise capacity(E)of COPD patients were evaluated. A spirometer was used to observe the changes in the patient's pulmonary function to detect the dyspnea score. The γ-interferon(IFN-γ),interleukin (IL-4 and IL-17)in serum were detected by enzyme-linked immunosorbent assay(ELISA). The expression of Treg of peripheral blood was detected by flow cytometry. Results Compared with healthy control group,lung function parameters,IL-4,CD4+CD25+Treg and CD4+CD25+Foxp3+Treg of peripheral blood were increased,while dyspnea score,BODE score,the expressions of IFN-γ,Th1/Th2,IL-17 in serum were decreased significantly in the COPD group(P<0.05 or P<0.01). After treatment,compared with those before treatment in the two groups, the forced expiratory volume in 1 second(FEV1),FEV1/forced vital capacity(FVC),IL-4,CD4+CD25+Treg, CD4+CD25+Foxp3+ Treg were all increased,while dyspnea score,BODE score,IFN-γ,Th1/Th2,IL-17 were all decreased(P<0.05 or P<0.01),and the peak expiratory flow(PEF)was decreased in western medicine control group and increased in TCM experimental group. After treatment,the comparisons of ratio of FEV1/FVC〔(78.12±14.96)%vs.(67.52±10.39)%〕,BODE score(1.07±0.72 vs. 1.77±0.74),IL-17(μg/L:40.80±8.97 vs. 48.22±6.51) Th1/Th2(1.05±0.23 vs. 1.42±0.21)and CD4+CD25+Foxp3+ Treg〔(6.61±2.26)% vs.(5.25±2.03)%〕between the two groups were all of statistical significant difference(all P<0.05). Conclusion Liuwei Buqi capsules can improve COPD symptoms by up-regulating the expressions of CD4+CD25+Foxp3+ Treg,IL-4,and down-regulating the expressions of IFN-γ,IL-17 to correct the balance of Th1/Th2 in patients with COPD.
