1.Effect of Jianwei Yuyang Granules on expression of epidermal growth factor receptor in gastric mucosa of gastric ulcer patients

OBJECTIVE: To observe the effect of Jianwei Yuyang Granules on the expression of epidermal growth factor receptor (EGFR) in the gastric mucosa of gastric ulcer patients. METHODS: Sixty gastric ulcer patients (final diagnosis by gastroscope) were randomly divided into Jianwei Yuyang Granules treated group (JWYY group, n=30) and western medicine treated group (control group, n=30). Ten patients without gastric mucosa lesion were treated as normal control group. The expression of EGFR in the mucosa was tested by immunohistochemistry and RT-PCR. RESULTS: Expression of EGFR and EGFR mRNA was not observed in patients without gastric mucosa lesion, and slight expression was observed in the edge of ulcer region of the gastric ulcer patients. The expression of EGFR and EGFR mRNA was increased in JWYY group and control group. CONCLUSION: Jianwei Yuyang Granules enhances the expression of EGFR in the mucosa of gastric ulcer patients to prevent peptic ulcer recurrence.
2.Pathological and clinical observation of preoperative selective arterial infusion chemotherapy of colorectal cancer

Objective To study the effect of preoperative selective arterial infusion chemotherapy (PSAIC) on large intestine cancer. Methods 63 patients with colorectal cancer underwent PSAIC with 5 fluorouracil,mitomycin and E adriamycin; the changes of clinical manifestation and pathology were observed and analyzed. Results (1)clinical manifestation:abdominalgia relieved in 18 patients and abdominal distention relieved in 16 patients.Improvement of hematochezia was found in 7 patients.In addition, of 13 patients with partial ileus, the clinical symptoms relieved in 9 patients. (2) pathology: there were karyopyknosis,karyorrhexis,coagulation and necrosis of cytoplasm in cancer cells. infiltration of inflammatory cells, edema and fibroelastosis in mesenchyne of cancer tissue. Proliferous intima and thrombus were also observed in the vessels. Most of these changes were moderate,and marked changes could also be seen in necrosis of cytoplasm and vessel intima proliferation. Conclusion PSAIC can control the focus of primary disease and the micrometastatic foci as well as improve the clinical symptoms, such as intestinal obstruction, and hematochezia, so PASIC is helpful to subsequent operation,and can improve the survival rate of patients with colorectal cancer.
3.Effects of Jianwei Yuyang Granule on TGF-?_1 Expression during the Healing Process of Rat Gastric Ulcer Induced by Acetic Acid

Objective To study the effects of Jianwei Yuyang granule on TGF-? 1 expression in the healing process of rat gastric ulcer induced by acetic acid. Methods Animal model of gastric ulcer in rat was set up with the modified Okabe's method, and the TGF-? 1 expression in the gastric ulcerated tissues was detected by immunohistochemical technique. Results TGF-? 1 expression in gastric ulcerated tissues during the healing process in Jianwei Yuyang group significantly increased compared with the normal, model and famotidine treatment groups. Conclusion TGF-? 1 expression in gastric ulcerated tissues induced by acetic acid during the healing process significantly increased,especially in Jianwei Yuyang group. Jianwei Yuyang granule may accelerate gastric ulcer healing by increasing TGF-? 1 expression in gastric ulcerated tissues.
4.The Effect of Jianweiyuyang Granule on Gastric Ulcer and Expression of VEGF and PDGF in Rat

Objective To study the effect of Jianweiyuyang(JWYY) granule on gastric ulcer of rats and its effective mechanism. Methods The rat model of gastric ulcer was set up by Okeba's method with some modification, and the expression of vascular endothelial growth factor (VEGF) and platelets-derived growth factor (PDGF) in the gastric ulcer tissues was examined by immunohistochemical method. Results JWYY granule could elevate the expression of both VEGF and PDGF, obviously decrease the ulcer index, increase the thickness of gastric mucosa, and promote the healing of gastric ulcer. Conclusion The results indicated that Jianweiyuyang granule can increase the expression of VEGF and PDGF, which may be one of the mechanism of its promoting the healing of the gastric ulcer.
5.Functional study of hepatocytes cultured in sandwich configuration

Objective To develop a new culture system in which hepatocytes can restore in-vivo like function. Methods Sandwich configuration was used for the culture of hepatocytes, and protein synthesis as well as P 450-b ,albumin mRNA expression by in situ hybridization was studied. ResultsStable and gradually increasing protein synthesis and high level of P 450-b , albumin mRNA expression were observed. In contrast, protein secretion of hepatocytes cultured in conventional single collagen culture system was low and decreased until approximately zero seven days after, and mRNA expression of P 450-b ,albumin was low. Conclusions Sandwich configuration in which cell polarity of hepatocytes is reestablished like in vivo situation.
6.Experimental Observation of the Specific Immune Response Basis About the Relationship Between the Liver-qi Stagnation Syndrome And the Spleen Deficiency Syndrome

Objective To survey the significance of specific immune response in the relationship between Sagnation the Liver-qi syndrome and the spleen Deficiency syndrome.Methods The rats with the Liver-qi Stagnation syndrome and the Spleen Deficiency syndrome were respectively corresponding and crossely treated with Radix Bupleuric dispersing the stagnated Liver-qi powder Decolion of Four Noble Drugs. Their T, B lymphocyte activity and specific immune response indexes changes (such as IgG, IgM in the serum) were respectively observed by lymphocyte transformation test and immunoturbidimetry. Results There were no notable changes about the IgG level among all kinds of groups. The IgM levels in the Liver-qi Stagnation syndrome and the Spleen Deficiency syndrome decreased, especially in the Liver-qi Stagnation syndrome (P0 05), Decolion of Four Noble Drugs could increase the IgM level of the patients with the Liver-qi Stagnation syndrome and the Spleen Deficiency syndrome (P0 05); The T lymphocyte transformation rate of the Spleen Deficiency syndrome obviously decreased, and Decolion of Four Noble Drugs could recover it to the normal level (P0 05). The rate of B lymphocyte transformation the Liver-qi Stagnation syndrome's also showed the increasing tenderly (P0 05). Conclusions The relationship between the Liver-qi Stagnation syndrome and the Spleen Deficiency syndrome could occur in the phase of antigen presentation, especially in the phase of the T H cell activation stimulated by antigen. Radix Bupleuric dispersing the stagnated Liver-qi powder could enhance T, B lymphocyte activities by reverse the Liver-qi Stagnation syndrome,and Decolion of Four Noble Drugs could enhance T lymphocyte by improving the Spleen Deficiency syndrome.
7.Expression of Th Differentiation Signaling Transduction Protein Calcineurin Affected by Jianpi and Shugan Prescription

To explore the mechanism of Jianpi(splenic tonic) and Shugan(dispersing the stagnated liver-qi) prescription regulating immunity by testing the expression of the differentiation signaling transduction protein,calcineurin.Methods We prepared the Sijunzitang and Chaihushugansan prescription serum,take the Ova as antigen induction and B cell as presenting cell,intervened culturing the CD 4 +T cells for 24 hours, and then examine the expression of calcineurin by RT-PCR,Western-blotting.Results The expression of calcineurin was increased in the stimulated Th cells, and the Sijunzitang and Chaihushugansan prescription serum can reduce it significantly,the Chaihushugansan also improved the expression deficit of calcineurin.Conclusions Jianpi and Shugan prescription can regulate the balance of immunity through affecting the expression of signaling transduction proteins at the Th activation and differentiation stage,The regulating the flow of qi prescription is superiority to supplementing qi one.in treatment of the Th2-mediated autoimmune disease.
8.Protective Effect of KangAiZengXiao Decoction on Immunological Deficit Rats With Implanted Tumor

Objective To study the effect mechanism of anticancer synergic (KangAiZengXiao,KAZX) decoction on gastric cancer in immunologic deficit rats with implanted tumor. Methods Forty rats were divided into normal control group(n=10), model group(n=15) and KAZX treating group(n=15). Model group and KAZX group were subcutaneously injected axillary the left upper limb with 0 5ml gastri cancer cells,MGC-803 cell(1?10 7/ml) after irradiated by 60 Co(dose:10Gy). Then the three groups were treated with KAZX decoction or distilled water every day, respectively. After three weeks, the rats were sacrificed,and blood WBC, the weight of spleen, activity of NK cell and transformation efficiency of lymphocyte were also detected.Results The rate of loaded tumor in KAZX group was significantly lower than in model group(20% vs 66 7%,P
9.The Influence of Ascites in Severe Acute Pancreatitis (SAP) on the Viability and Function of Peritoneal Macrophages

Objective To observe the influence of ascites in severe acute pancreatitis (SAP) on the function and viability of peritoneal macrophages in order to investigate the role of peritoneal macrophages in pathophysiological alteration and secondary pancreatic infection in SAP. Methods After the ascites of SAP models treated peritoneal macrophages for 1,3,6,12 and 24 hours in vitro, neutral red phagocytosis, cell viability and TNF secretion of peritoneal macrophages were determined respectively. Results The phagocytosis, cell viability and TNF secretion of macrophage all decreased with the treating time prolonged in the tests. Conclusions The ascites of SAP decreased the phagocytosis, viability and TNF secretion of peritoneal macrophages, and was one of the factors to promote secondary pancreatic and peripancreatic infection as well as bacterial translocation of gut.
10.Minimally invasive and open reduction plate fixation for proximal humerus fractures:range of motion of the shoulder joint

BACKGROUND:The conservative treatment of humeral fracture is difficult to achieve a good reduction. Minimaly invasive percutaneous plate fixation has been widely used, and has good repair results, but the specific mechanism of action is not clear.
OBJECTIVE:To compare the repair effect of different fixation methods on proximal humerus fractures.
METHODS: From August 2011 to October 2014, we selected 96 patients with proximal humerus fractures from the Shougang Hospital of Peking University. These patients were equaly divided into two groups according to the principle of random draw. Patients in the control group were treated with open reduction and conventional surgery fixation. Patients in the treatment group received minimaly invasive percutaneous plate fixation. Operation time, intraoperative blood loss, incision length and postoperative hospital stay were recorded in both groups. At 8 weeks after treatment, patients received radiography to identify the reduction. Range of motion of the shoulder joint was scored. Within 8 weeks after treatment, the occurrence of complications was observed and compared in both groups.
RESULTS AND CONCLUSION:Intraoperative blood loss, incision length and postoperative hospital stay were significantly less in the treatment group than in the control group (P < 0.05). No significant difference in operation time was found between the two groups (P > 0.05). At 8 weeks after treatment, the excelent and good rate of reduction was significantly higher in the treatment group than in the control group (98%, 81%,P < 0.05). Flexion, abduction, external rotation and internal rotation scores were significantly higher in the treatment group than in the control group (P < 0.05). Bone compartment syndrome, vascular injury, infection and bleeding were significantly lower in the treatment group than in the control group (P < 0.05). These findings confirm that compared with open reduction and internal fixation, minimaly invasive percutaneous plate fixation of proximal humerus fractures has less impact on the body, can promote reduction of the fracture, has few complications, and contributes to the recovery of shoulder function.
