1.The effects of basic fibroblast growth factor on the expression of heat shock protein 70 in rat's retina afterischemia/reperfusion injury

Objective To observe the effects of basic fibroblast growth factor (bFGF) on the expression of heat shock protein 70 (HSP70) in rat′s retina after ischemia/reperfusion injury. Methods The rat model of experimental retinal ischemia/ reperfusion injury was made by increasing the intraocular pressure. Tweenty-four Wistar rats were divided into normal (3 rats) and operation group (21 rats) randomly. The latter group was subdivided into group 0 hour, 4, 8, 12, 24, 48 and 72 hours after reperfusion, in which the left eyes of the rats were in the ischemia/reperfusion groups and the right ones were in the treatment groups (bFGF 2 ?g intracameral injection). The expression of HSP70 was observed by strept avidin-biotin complex (SABC) immunohistochemistry. Results No HSP70 positive cells were found in normal group; a few of HSP70 positive cells were found 0 hour after reperfusion [20.8?4.5)cells/mm 2], and increased gradually until reached the peak 24 hours later [(111.2?4.4) cells/mm 2] and then decreased gradually. Few HSP70 positive cells were found 72 hours after reperfusion. The amount of HSP70 positive cells increased in treatment group at all time courses, and the peak time was earlier and longer than that in ischemia group. HSP70 positive cells distributed extensively in retinal ganglion cell layer and inner nucleous layer. The difference of the amount of HSP70 positive cells between the two groups was significant ( P
4.Research for difference and sense of B cell epitope human hepatitis virus B genotype-B and genotype-C

Objective To study the difference and sense of B cell epitope of S protein HBV genotype B and genotype C. Methods Search for amino acid sequence of S protein of HBV genotype C and genotype B from GcnBank for international network and forecast B cell epitope of S protein of HBV genotype band genotype C by DNAStar Bio-software and by Kolaskear-tongaonaka method through the international network. Results There are five B cell epitopes about amino acid of S protein of HBV genotype B and three HBV genotype C and antigenic index (1. 0798) of amino acid of S protein of HBV genotype B is higher than that of HBV genotype C (1. 0729). Conclusion Human hepatitis B genotype C is easier to develop chronic hepatitis than that of Human hepatitis B genotype B, It may be related with the fact that there arc fewer B cell epitopes and lower antigenic index about S protein in hu-man hepatitis B genotype C than that in human hepatitis B genotype B.
5.Limit test for 5-hydroxymethylfurfural in Yueju Baohe pills

Objective: To establish an HPLC method to detect the limit of 5-hydroxymethylfurfural in Yueju Baohe pills.
Methods: A Zafex Acutfex PW-C18 column(4.6 mm×250 mm, 5 μm) was adopted with mobile phase consisted of acetonitrile-0.1% formic acid aqueous solution(2:98)at the flow rate of 1.0 mL·min-1. The column temperature was 40 ℃ and the detective wavelength was set at 284 nm.
Results: The linear range of 5-hydroxymethylfurfural was 0.491 9-122.98 μg·mL-1 (r=1.000). The average recovery(n=6) was 97.99% with RSD 0.61%. All of the RSDs for precision,repeatability and stability met the methodological requirements. According to the proposed limit, 17 of the 65 batches of samples did not meet the requirements, and the failure rate was 26.15%.
Conclusion: The established method is suitable for the limit test of 5-hydroxymethylfurfural in Yueju Baohe pills.There was a significant difference in the content of 65 batches of samples. It is necessary to investigate the limit of 5-hydroxymethylfurfural in Yueju Baohe pills.
6.Chromatography-efficacy relation study between HPLC fingerprints and allelopathic effect of Salvia miltiorrhiza aqueous solution on radish.

To explore the effective components represented by fingerprint contributed to allelopathic effect of different Salvia miltiorrhiza aqueous concentration on seeds and seedlings of radish, grey relational analysis was used to establish the chromatography-efficacy relation. The results show that 15 peaks devote high allelopathic contribution to radish seeds and seedlings. The study will provide a new concept for allelochemicals screening and study.

7.EFFECT OF PROSTAGLANDIN E1 ON SIL-6R and ITSβ strands IN SERUM IN PATIENTS WITH CHRONIC SEVERE HEPATITIS

Objective To observe the effect of prostaglandin E1 on soluble interleukin-6 receptor(SIL-6R) and soluble interleukin-6 receptor β strands (sgp 130) in serum in patients with chronic severe hepatitis.Methods Sixteen patients with chronic severe hepatitis accepted therapy with prostaglandin E1.Serum levels of SIL-6R and sgp130 were detected by enzyme linked immunosorbent assay (ELISA) method before and after therapy.Results The content of SIL-6R and sgp 130 in serum were increased significantly (P<0.001) in patients with chronic severe hepatitis compared with normal control group and decreased obviously (P<0.01) after therapy with prostaglandin E1 compared with before therapy,and the rate of delth were decreased obviously (P<0.01) in PGE1 therapy group.Conclusion The levels of SIL-6R and sgp130 in serum can reflect the degree of liver damage in patients with chronic severe hepatitis and guide the assessment of prognosis;PGE1 can correct the immune disorder,attenuating hepatocyte inflammation and necrosis.
8.Quality standard for Shuwei Capsules

AIM: To study the quality standard for Shuwei Capsule (Resina Ferulae, Rhizoma Corydalis, Radix Aucklandiae, etc.). METHODS: Radix Aucklandiae, Rhizoma Cyperi were identified by TLC, and ferulic acid was determined by HPLC. RESULTS: TLC spots developed were fairly clear, and the blank test showed no interference. Ferulic acid showed a good linear relationship in the concentration range of 0.007~0.056?g and the average recovery was up to 98.51%, RSD was 2.64%. CONCLUSION: The method is simple with strong specificity and good reproducibility, and can be used as the quality control of this preparation.
9.The level and clinical significance of serum anti-lysosomal associated membrane protein-2 antibody in patients with anti-neutrophil cytoplasmic antibodies-associated vasculitis

Objective To explore the clinical.significance of serum anti-lysosomal associated membrane protein-2 (LAMP-2) antibody in the pathogenesis of anti-neutrophil cytoplasmic antibodies (ANCA)-associated vasculitis (AASV) by investigating the relationship of its levels and AASV.Methods Sera from twenty patients with AASV and twenty healthy controls were collected.Serum anti-LAMP-2 antibody was detected using commercial enzyme linked immunosorbent assay (ELISA) kits.Anti-LAMP-2 antibody levels between the two groups were assessed using the t test,the correlation between anti-LAMP-2 antibody levels and erythrocyte sedimentation rate (ESR),C-reactive protein (CRP) was assessed by Spearman's rank correlation test,the correlation between anti-LAMP-2 antibody levels and the Birmingham vasculitis activity score(BVAS),hemoglobin (Hb),albumin (Alb) was assessed by Pearson's rank correlation test.Results ① The serum level of anti-LAMP-2 antibody in patients with AASV [(3714±1446) pg/ml] was higher than that in the healthy controls [(174±43) pg/ml] (t=10.94,P<0.05).The serum level of Hb [(99±30) g/L] and Alb [(27±5) g/L]in patients with AASV was lower than that in healthy controls [(138±14) g/L,(44±3) g/L] (t=5.27,t=13.04,P>0.05).② The level of anti-LAMP-2 antibody in AASV was positively correlated with BVAS (r=0.669 9,P<0.05),and was not found elevated compared with ESR,CRP,Hb and Alb (P>0.05).Conclusion ① AntiLAMP-2 antibody is involved in the pathogenesis of AASV.② Anti-LAMP-2 antibody is correlated with the activity of AASV,it may be an indicator of AASV disease activity.
10.Effects of glutamine pretreatment on intestinal ischemia-reperfusion inju-ry in rats

AIM:To determine the effects of glutamine ( Gln) pretreatment on intestinal ischemia-reperfusion (I/R) injury in the rats.METHODS: Thirty male Wistar rats were randomly divided into 3 groups (n=10): sham group, I/R group and Gln pretreatment group .The rats in Gln pretreatment group were pretreated with 1 g· kg -1 · d-1 Gln by orogastric route for 7 d, the rats in the other 2 groups were pretreated with normal saline .Intestinal I/R was induced by 30-min occlusion of the superior mesenteric artery followed by 24 h of reperfusion .After the operation , the plasma endo-toxin, serum D-lactic acid, superoxide dismutase ( SOD) and malondialdehyde ( MDA) levels were measured .The intesti-nal mucosal injury was observed with HE staining and evaluated using Chiu 's scoring.RESULTS: Serum D-lactic acid, endotoxin level , MDA level and Chiu's score in I/R group were significantly higher than those in sham group and Gln group (all P<0.05).Serum SOD activity was significantly lower than that in sham group and Gln group (P<0.05).CON-CLUSION:Glutamine has a protective effect on the intestines during ischemia-reperfusion injury .The mechanism may be related to oxidative stress response .
