1.Effect of Nicergoline for chronic cerebrovascular insufficiency: a meta analysis

Objective To systematically review the efficacy and safety of nicergoline on chronic cerebrovascular insufficiency (CCVI).Methods Databases including the Cochrane Library,PubMed,CNKI,VIP and WanFang Date were electronically searched for relevant randomized controlled trails (RCTs) which studied the effectiveness and safety of Nicergoline on chronic cerebrovascular insufficiency.Two reviewers independently screened literature according to the inclusion and exclusion criteria,extracted data,and assessed the methodological quality.Metaanalysis was performed by using RevMan 5.2 software.Results Totally 9 studies involving 1 030 patients were included.The meta-analysis results showed that,compared with conventional treatment alone,the Nicergoline group was superior in improving the velocity of middle cerebral artery(MCA) in both systolic(MD=16.43,95%CI:10.39-22.46) and the late diastolic (MD=11.48,95%CI:5.34-17.63).Besides,Nicergoline significantly improved the pulse index (PI) of MCA (MD=-0.37,95%CI:-0.48--0.26).Likewise,it obviously improved the velocity in both systolic (MD=11.23,95%CI:6.10-16.35),the late diastolic (MD=8.80,95%CI:5.20-12.40) and the PI of vertebral artery (VA) (MD=-0.38,95%CI:-0.47--0.28).The remission rate of the nicergolinegroup was higher than that of the control group (OR=3.93,95% CI:2.66-5.81).Drug-related adverse reactions were not reported in included studies.Conclusions Nicergoline shows a certain efficacy on CCVI without obvious adverse reactions.
2.Immune Response to Respiratory Syncytial Virus Infection in Ovalbumin Sensitized Mice

Objective To observe the lung inflammation and the expression of T - helper cell related cytokines in respiratory syncytial virus(RSV) infected Balb/c mice pre - sensitized with ovalbumin(OVA). Methods Mice were randomly divided into two series: nonsen-sitized and sensitized animal model with nebulized OVA once a day for 10 days. Each of the series was divided again into two groups: control group,RSV infected group with nasal draping plus nebulizing. All the mice were sacrificed 5 days later of infection. Viral isolation of lung organization was performed in each group. Pulmonary pathological detection and the levels of interleukin - 4(IL-4), interferon-? (IFN- ?) mRNA were evaluated by RT- PCR. Results The RSV were found only in RSV infected groups. The Balb/c mice developed typical interstitial pneumonia after RSV infection. When the mice pre- sensitized were infected with RSV, the pulmonary inflammation, lymphocyte and eosinophils infiltration and cell - collar peribronchiles were more severe in lung organization and bronchiole than those in the single RSV infection group. In the lung organization of control group, there were no mRNA expression of IL - 4 and IFN - ?. The mRNA of IFN -? was expressed in the lung organization of RSV infected groups while had not showed mRNA of IL- 4. In the OVA+ RSV group the mRNA expression of IL - 4 was obvious, while the expresses of IFN -? scarcely. Conclusions Pulmonary inflammation is more severe in the infection after OVA sensitization. RSV infection alone results in a Th1 - like cytokine response, while the infection after OVA sensitization results in Th2 - like response.
3.Inhibitory effect of VEGFR-3-targeted siRNA against adherence and invasion of human colon cancer cells

Objective:To construct a small interfering RNA(siRNA)expression vector(psiRNA-VEGFR-3)targeting vascular endothelial growth factor receptor 3(VEGFR-3)and to investigate the effects of VEGFR-3 siRNA on the adherence and invasion of human colon cancer cells.Methods:A siRNA expression vector(psiRNA-VEGFR-3)targeting VEGFR-3 were constructed and was used to transfect LoVo cells via lipofectamine 2000.The mRNA and protein expression of VEGFR-3 were examined after transfection by reverse transcriptase polymerase chain reaction(RT-PCR)and Western blotting,respectively.The tumor adhesion ability was detected by cell-matrix adhesion experiment and the invasion ability of tumor cells was evaluated by millicell chamber model.Results:The VEGFR-3 siRNA expression vector was successfully constructed.The expression of VEGFR-3 mRNA and protein was inhibited after psiRNA-VEGFR-3 transfection.Seventy-two hours after psiRNA-VEGFR-3 transfection,Western blotting assay showed that the expression of VEGFR-3 protein was decreased from(1.26?0.19)to(0.39 s0.12)(P

5.Whole-genome expression analysis of synovial fibroblasts in rheumatoid arthritis

Objective To identify the crucial gene implicated in rheumatoid arthritis (RA) pathogenesis by comparing microarray-based gene expression profiles of synovial fibroblast in arthritis patients and that in control.Methods The public datasets were obtained from NCBI GEO and EBI ArrayExpress.The qualified microarray-based gene expression profiles were integrated and normalized using the method implemented in GeneSpring software.Furthermore,the differentially expressed genes (DEGs) were identified using significance analysis of microarrays (SAM) method.The online tool DAVID and STRING were applied to conduct the enrichment analysis and gene product interaction analysis respectively.Results There were two datasets that were qualified and analyzed in the present study.A total of 336 significant DEGs were identified by comparing the whole-genome gene expression profiles from synovial fibroblast of RA patients and control group.Among these DEGs,261 were significantly downregulated and 75 upregulated.About 13.6% of the downregulated genes were associated with extracellular matrix degradation.The COL9A3 and COL4A5,indispensable component of hyaline cartilage and basement membrane respectively,were significantly downregulated,as well as genes in WNT family,including WNT2,WNT11,and WNT16.In contrast,matrix metalloproteinase 13 (MMP13) was found to be significantly upregulated in RA patients.MMP13 is a matrix metallopeptidase that degrade extracellular matrix and hyaline cartilage,and it could possibly interact with other proteins to regulate morphogenesis.Conclusion Molecular mechanisms underlying RA pathogenesis were investigated by analyzing the public datasets.A few genes that associated with extracellular matrix degradation,construction and regulation,including MMP13,WNT2,WNT11,WNT16,COL9A3 and COL4A5,could be regarded as therapeutic targets in RA treatment.
6.The Value of Preserving Intercostobrachial Nerve during Modified Radical Mastectomy for Breast Neoplasm under Transverse Incision

Objective: To discuss the value of preserving ICBN during modified radical mastectomy for breast neoplasms. Methods: Sixty-three cases of breast carcinoma who underwent modified radical mastecto-my were studied. Of them, ICBN was completely preserved in 33 cases and was dissected in 30 cases. Sen-sory function of the medial upper arm of all patients was followed up after surgery. Results: No local recur-rence or metastasis was found among the 63 patients during the first, sixth, and twelveth month of follow-up period. Among the 33 cases with ICBN reserved, 29 cases (87.88%) had normal sensation of the skin in up-per medial arm and axilla, while 4 cases (12.12%) had paresthesia. Conclsion: Preserving ICBN can en-hance the quality of life of breast cancer patients after surgery.
7.Establishment of orthotopic implantation and spontaneous metastasis model of human ovary cancer in nude mice

Objective To establish a patient-like human ovary carcinoma/spontaneous metastasis model using orthotopic transplantation of histologically intact tumor tissue. Methods An highly metastatic ovarian tumor line (8910PM: human serum carcinoma of the ovary) previously grown substaneously was transplanted into the ovicapaule using microsurgery technique. Histologically intact human ovary tumor pieces gained from implantation site were passaged between ovicapsules for four generations. Results All mice developed ovary tumors and the metastatic rates were about 50 %. The tumors only metastasized to liver. The earliest appearance of metastasis was 14 days and the average survival period was 20.7 ±4.89 days. The microscopic appearance of the metastases was similar to the tumor observed in the substaneous xenografts and orthotopically transplanted.Chromosomes analysis exhibited the feature of human carcinoma and retained genetic stability during the processes of passage. Conclusion Orthotopic implantation provides a suitable micro-environment in which ovarian cancer can express its intrinsic clinically-relevant properties. This approach is relevant to the spontaneous development of ovarian cancer and is thought to be a useful model for studies of metastatic mechanism and therapy for ovary cancer.

BACKGROUND:Currently, what we know about exosomes is that it can be produced by a variety of cel s and transfer a variety of materials, producing subsequent function of regulation.
OBJECTIVE:To review the function of exosomes in mesenchymal stem cel differentiation, in order to provide reference for further in-depth study.
METHODS:With the key words of“exosome, mesenchymal stem cel , differentiation”in Chinese and English, respectively, a computer-based search was performed for articles published in CNKI, Medline and Embase databases from January 2001 to September 2016. After the initial screening, the reserved articles were further detailed, summarized and concluded.
RESULTS AND CONCLUSION:Exosomes are a kind of vesicles 40-100 nm in diameter, which can be secreted by a variety of cel types, containing functional products, such as functional protein, gene product, lipid and so on. As a bridge of adjacent cel s transferring functional products, exosomes are becoming an issue of concern in the microenvironment for cel interaction. In the differentiation of mesenchymal stem cel s, exosomes also play an indispensable role via pathway regulation.

10.EFFECT OF SELENIUM ON GENERATION OF SUPEROXIDERADICAL IN RAT HEART MITOCHONDRIA

Weanling rats were fed with a low-selenium diet (Se 0.009mg/kg, — Se group) and the diet supplemented with sodium selenite (Se 0. 232mg/kg,+ Se group) for 1, 2 and 3 months respectively. The generation rate of superoxide radical(O_2~-), activity of manganese dependent superoxide dismutase (Mn-SOD) in heart mitochondria and blood Se concentration from the rats were determined. The results showed that the mitochondrial O_2~- generation rate from — Se rats was significantly higher than those from + Se rats at 2 and 3 months. During all the feeding periods, no significant difference in mitochondrial Mn-SOD activity was found between the—Se and + Se groups,while the blood Se concentration in — Se group was significantly lower than those in + Se group at each feeding time and continually decreased in the 2 and 3 month. The results suggested that selenium could depress O_2~- generation in heart mitochondria.
