1.Proliferation and migration of breast cancer cells inhibited by miRNA-16 in vitro Bu Deyong , Jia Hongyan

Objective To investigate the expression of miRNA-16 (miR-16) in breast cancer patients and its effect on the proliferation and migration of breast cancer cells. Methods Polymerase chain reaction (PCR) was used to detect the expression of miR-16 in 30 breast cancer patients. miR-16 mimics was transfected to MDA-MB-231 and MCF-7 cell, and CCK-8 as well as Transwell assay was applied to detect the effect of miR-16 on cell proliferation and migration of MDA-MB-231 and MCF-7 cells. Results Among 30 breast cancer patients, the expression of miR-16 was down-regulated in 23 cases. Cell proliferation and migration of MDA-MB-231 and MCF-7 cells were inhibited significantly after transfection of miR-16 mimics. Conclusion miR-16 is down-regulated in breast cancer patients. miR-16 inhibits significantly the cell proliferation and migration of breast cancer cells in vitro.
2.The effects of rosiglitazone on neointima and inflammatory factors after carotid artery balloon injury of rats

Objective To investigate the effect of rosiglitazone on neointima and inflammatory factor after carotid artery injury of rats. Methods A total of 60 SD rats were divided randomly into 3 groups: the control group, the surgery group and the resiglitazone therapeutic group. The left common carotid arteries were injured by balloon in the surgery group and the therapeutic group. Rosiglitazone was administrated intragastricly to therapeutic group. The plasma tumor necrosis factor-? (TNF-?) and interleukin-6 (IL-6) levels were detected with radioimmunoassay at 4h, 1 and 7d after the operation. After 2 weeks, the injured vessels were stained by HE and immunity histochemistry method,then observed with microscope. Neointimal area (NIA), internal elastic lamina area (IELA), stenosis index (SI) and optical density value of protein expression of NF-?B were calculated. Results Rosiglitazone inhibited the neointima of injured vessels, decreased the protein expression of NF-?B and reduced plasma TNF-? and IL-6 levels. Conclusion Rosiglitazone can inhibit the inflammatory reaction of injured vessels and lighten the restenosis of injured vessels.
3.Analgesic efficacy of methylene blue with ropivacaine on anorectal disease postoperation

Between January 2011 and December 2011,86 patients undergoing operation for anorectal disease were randomized in a double-blind manner to receive a mixed injection of methylene blue,ropivacaine and sodium chloride (observation group,n =43) or traditional analgesic methods (control group,n =43).Patients in observation group had less visual analog scale (VAS) scores of pain at 6-48 h postoperation than control group (P <0.05).The scores had no significant difference at 72 h postoperation between two groups (P > 0.05).The total postoperative complications (8,19%) and hospitalization duration [(5.3 ± 2.2) days] in observation group were significantly less than those [15,35 % and (6.4 ±1.7) days] in control group (P < 0.05).A mixed injection of methylene blue and ropivacaine intraoperatively can relieve the postoperative pain of patients with anorectal disease and reduce the incidence of postoperative complications and the duration of hospitalization.
4.Mechanism of Clinical Cross Allergy Reaction of β-lactam Antibiotics and Introduction of Related Treat-ment Processes in USA

OBJECTIVE:To investigate the mechanism of cross allergy reaction during the application of β-lactam antibiotics, and to provide reference for rational drug use in clinic. METHODS:Based on study experience of author in UIC and its affiliated hospital during advanced study,according to the experience of drug use safety management in patients allergic to β-lactam antibiot-ics from Rush University Medical Center,the mechanism of cross allergy reaction during the application of β-lactam antibiotics was summarized,and the disposal procedure for patients allergic to β-lactam antibiotics in the Affiliated Hospital of UIC was intro-duced. RESULTS:The principal reason for cross allergy reaction induced by β-lactam antibiotics were same or similar side chains between drugs. Cross allergy reaction occurred when IgE recognized these side chains. The disposal procedure for patients allergic to β-lactam antibiotics in the Affiliated Hospital of UIC included that the indication of β-lactam use was evaluated;standard penicil-lin skin testing according to evaluation results,anti-infection treatment by Grade challenge β-lactam antibiotics and course and rap-id drug tolerance induction. CONCLUSIONS:The disposal method for patients allergic to β-lactam antibiotics in the Affiliated Hos-pital of UIC can provide new thought for domestic clinical pharmacists in rational drug use among the patiens with reported aller-gies to special group as pregnant women,children.
5."Effect of ""brisk walking"" on male elderly with hypertension in community-based health centers"

Objective · To explore the effects of brisk walking on blood pressure, body mass index (BMI), and autonomic nerve activity of male elderly with hypertension in community-based health centers in Shanghai. Methods · A total of 630 elderly men received brisk walking treatment (>10000 steps/d) voluntarily, who were selected from six community-based health centers in Pudong New Area of Shanghai. Among them, 210 people stick to training for 12 weeks. According to whether suffering from hypertension, the people were assigned to hypertension intervened group (intervention group, n=110) and normal intervened group (control group 1, n=100). Blood pressure, BMI, heart rate and autonomic nerve activity before and after the training were compared. Results · After 12 weeks of training, systolic blood pressure (SBP), diastolic blood pressure (DBP) and sympathetic nerve activity of intervention group were significantly lower than those before the training (P<0.05). SBP was also significantly lower than control group 2 (P<0.05). However, heart rate, BMI, parasympathetic nerve activity and baroreceptor reflex sensitivity did not change significantly after the training (P>0.05). Conclusion · Brisk walking treatment lasting for 12 weeks can decrease blood pressure and sympathetic nerve activity in elderly men with hypertension. It is worthy to be popularized in the community.
6.Myofascial pain syndrome treated with sparrow-pecking moxibustion at trigger points: a randomized controlled trial.

OBJECTIVETo compare the efficacy difference in treatment of myofasical pain syndrome between sparrow-pecking moxibustion and acupuncture at trigger points so as to provide the reference of the effective therapeutic method for myofascial pain syndrome.
METHODSNinety patients were randomized into a sparrow-pecking moxibustion group and an acupuncture group, 45 cases in each one. The trigger points were selected in pain areas in the two groups. In the sparrow-pecking moxibustion group, the sparrow-pecking moxibustion was applied, 30 min in each time. In the acupuncture group, the filiform needles were inserted obliquely at 45 degrees and retained for 40 min in each treatment. The treatment was given once a day and 10 treatments made one session in the two groups. The short-form McGill pain questionnaire was used as the observation index, and the changes in pain rating index (PRI), present pain intensity (PPI) and visual analogue scale (VAS) before and after treatment were used for efficacy assessment.
RESULTSThe results of PRI, PPI and VAS after treatment were reduced apparently as compared with those before treatment in the sparrow-pecking moxibustion group and the acupuncture group (all P<0.001). The differences in PRI, PPI and VAS after treatment were not significant in comparison of the two groups (both P>0.05). The curative and remarkably effective rate was 80.0% (36/45) in the sparrow-pecking moxibustion group, which was better than 40.0% (18/45, P<0.001) in the acupuncture group.
CONCLUSIONSparrow-pecking moxibustion at trigger points achieves the superior efficacy on myofascial pain syndrome as compared with acupuncture at trigger points. This therapy is simpler in operation additionally.

7.Expression of microRNA-10a-5p in laryngeal epithelial premalignant lesions.

OBJECTIVETo investigate the expression of microRNA-10a-5p (miR-10a) in laryngeal epithelial premalignant lesions (LEPL) and to analyze the correlations of its dysregulation with clinicopathologic parameters of LEPL.
METHODSAccording to the WHO classification (2005), 94 cases of LEPL were grouped into mild dysplasia (MID), moderate dysplasia (MOD), severe dysplasia (SD) and carcinoma in situ ( CIS). The expression of miR-10a in 94 cases of LEPL was detected by quantitative real-time polymerase chain reaction (qRT-PCR), and correlated with the clinical and follow-up data of all LEPL patients.
RESULTSmiR-10a was down-regulated in LEPL with increasing grade of dysplasia. There was significantly statistical difference between low-risk ( MID + MOD) and high-risk ( SD + CIS) lesion groups (P = 0.03). The linear regression analysis showed that miR-10a was correlated with grade and gender of LEPL (P < 0.05).
CONCLUSIONSDown regulation of miR-10a may be a useful molecular marker for grading of LEPL and diagnosis of early laryngeal cancer.

8.Relationship between single nucleotide polymorphism of urokinase-type plasminogen activator gene and tongue carcinoma

Objective To determine association between tongue carcinoma and polymorphism of urokinase-type plasminogen activator (PLAU) gene.Methods PLAU genotypes of 97 patients with tongue carcinoma and 91 health controls were examined by the PCR-RFLP method.Statistical analyses included a chi-square test for homogeneity and logistic regression analysis.Results The polymorphism in PLAU gene was rs2227564 C/T.Logistic analyses indicated that compared with CT and TT genotypes,CC genotype was risk factor for development of tongue carcinoma (adjusted OR =1.281,95 % CI 1.098-2.577,P =0.037).Conclusion PLAU polymorphism may be associated with development of tongue carcinoma.

Since the biological characteristics of esophageal gastric junction (EGJ) cancer are different from those of gastric cancer and esophageal cancer, the choice of initial treatment is particularly important. This article introduces a case of locally advanced EGJ can-cer with single metastasis factor treated with preoperative radiochemotherapy in the Beijing Cancer Hospital. Through the multidisci-plinary team, we aim to achieve a better prognosis for this patient and propose new treatment practices for EGJ cancer.
10.Hyperalgesia induced by morphine on incision pain in rats

Objective To investigate the roles of different effective dosages of morphine on incision pain in rats.Methods Clean-degree male SD rats were randomly allocated to three groups(n=11): normal saline group(NS group),low-dose morphine group(LM group,0.6 mg/kg) and high-dose morphine group(HM group,6 mg/kg).After administration of morphine twice at 30 min interval,the incision pain models of rats were prepared according to the Brennan's method.Morphine was administered once again following the operation,while the NS group was administrated isovolumic normal saline.Then the mechanical threshold of rats was detected with von Frey filaments from the pre-operation to the 8th postoperative day,and the hyperalgesia induced by morphine on the praxiology level was explored.
