Objective: To investigate the effect of a long-term CpG-ODN stimulation on the maturation of murine bone-marrow derived dendritic cells (BMDCs). Methods: Murine bone-marrow cells were cultured in GM-CSF alone or with CpG-ODN for 7 d or for last 36 h (days 6, 7). Cell phenotypes and antigens uptake by BMDCs were analyzed by flow cytometry. Cytokines released by BMDCs were detected by ELISA. The antigen presenting capability by BMDCs was evaluated by mixed lymphocyte responses. Results:Compared to those of the short-term CpG-ODN stimulation group, the expression of MHCⅡ, CD86, CD40, and secretion of IL-12(p70) by BMDCs in long-term stimulation group were not increased. The phagocytosis of FITC-OVA by BMDCs in long-term CpG-ODN stimulation group was strengthened, but the activation of allogenic and homogenic lymphocyte cells proliferation was impaired. Conclusion:Long-term CpG-ODN stimulation can suppress the maturation of DCs, which may explain the low adaptive immunity in sepsis patients.

In this paper, the bacterial community structure in the gills and intestines of Penaeus chinensis and Metapenaeus ensis were investigated using a culture-independent method of 16S rDNA PCR-DGGE fingerprinting. It was proved that there are abundant and various bacteria in the gills and intestines of these prawns. The bacterial community in the gills and intestines of the same prawn or different kinds of prawns were obviously different based on the clustering analysis of DGGE fingerprints. On the other hand, the same bacteria were found in different kinds of prawns. This was the first time to aim to set up a method to reveal the bacterial community in prawn based on 16S rDNA PCR-DGGE fingerprinting which is of great value for the studies of the relationship between prawn and environmental microorganism as well as the related diseases.

The association between serum fetuin A and diabetic nephropathy (DN) in patients with type 2 diabetes mellitus(T2DM) was investigated.Serum levels of fetuin-A and adiponectin were measured by ELISA and the clinical data were collected in 101 T2DM patients with normal albuminuria (NA group),87 patients with microalbuminuria (MA group),44 patients with overt albuminuria (OA group),and 130 normal control subjects (NC group).Fetuin-A levels increased progressively from NC,NA,MA,to OA groups, and there were significant differences among groups(P<0.05 or P<0.01).Fetuin-A level in patients was positively associated with duration of illness, urinary albumin to creatinine ratio (ACR),fasting blood glucose,2-hour postprandial blood glucose, and C-reactive protein(CRP,P<0.05 or P<0.01),but was negatively associated with age and adiponectin (all P< 0.01).Multiple stepwise regression analysis indicated that duration of diabetes,fetuin-A,CRP,and adiponectin were independent determinants for ACR (all P < 0.01).Fetuin-A may be involved in the development and pathogenesis of DN by proinflammatory effects and repressing adiponectin production.

Objective To find noninvasive estimation of partial pressure of carbon dioxide in artery (PaCO2) by measuring the end-tidal CO2 partial pressure (PETCO2) in elderly patients with respiratory failure with chronic obstructive pulmonary disease (COPD). Methods All the 30 acutely exacerbated COPD subjects received routine clinical treatment including bronchodilators, mucolytics, glucocorticosteroid, antibiotics and oxygen therapy for 5-7 days, and part of them received noninvasive positive-pressure ventilation (NIPPV) treatment concurrently. They were both tested by eupnea method and prolonged expiratory method before and after treatment. Results Before treatment, PET CO2 (Q) (end-tidal CO2 pressure with eupnea, (50.72±8.93) mm Hg wassignificantly lower than PaCO2 (F=38.73, P<0.01 ). Yet, PETCO2(P) (end-tidal CO2 pressure with prolong expiration) was (70.35±8.91) mm Hg and PaCO2 was (71.25±9.08) mm Hg. There was no significant difference between PETCO2 (P) and PaCO2 (P>0.05). The similar results were found after treatment. By linear regression analysis, PetCO2(P) was remarkably positive correlated with PaCO2 before and after treatment (r=0.96 and 0.97, respectively, P<0.01). According to TABG,PCO2(C) which was calculated by the average expiratory time of fitting curve was (71.78±9.04)mm Hg. And there was no significant difference between PCO2 (C) and PaCO2 (P>0.05);Thesimilar results were founcl after treatment. By linear regression analysis, PCO2 (C) was remarkably positively correlated with PaCO2 (r=0.97 and 0.98, respectively, P<0.01) before and after treatment. Conclusions In COPD patients with type Ⅱ respiratory failure, conventional PETCO2 (Q) is significantly lower than PaCO2. Yet, PETCO2 (P) could exactly estimate PaCO2 and is appropriate to its dynamic monitoring. And PCO2 (C) which is calculated by mode Boltzmann on eupnea curve also obtain similar results.
