1.Short-term efficacy of nucleos (t)ide analogues in patients with hepatitis B virus-related chronic liver failure: a Meta-analysis

Objective To evaluate the efficacy of nucleos(t)ide analogues in patients with hepatitis B virus(HBV)-related chronic liver failure through Meta-analysis.Methods Literature search was done in Medline, PubMed, China National Knowledge Infrastracture(CNKI), Wanfang Database and China Biology Medicine(CBM)for papers published from 1990 to 2010 in Chinese or English, as well as the references in the relevant literature complemented by retrospective and manual literature search. Only literatures fully matching the inclusion and exclusion criteria could be included in this study. RavMan 5.0 statistical software was utilized for Meta-analysis. Then the sensitivity and publication bias about the data were measured by forest pl0t, funnel plot and fail-safe number. Results Total 1240 patients from 20 controlled clinical trials reported in Chinese and English papers were included in this study. The indicators of therapeutic efficacy were as below: HBV DNA undetectable rate relative risk(RR)=3. 37, 95%CI: 2. 20, 5. 16(Z=5. 60, P<0. 01), prothrombin activity(PTA)weighted mean difference(WMD)=34.70, 95%CI: 25.62, 43.79(Z=7.49, P<0.01), albumin(Alb)WMD=4.73, 95% CI: 2.95, 6.51(Z=5.21, P<0.01), alanine aminotransferase(ALT)WMD=42.58, 95%CI: -59.74, -25.41(Z=4.86, P<0.01), total bilirubin(Tbil)WMD=-150.95, 95%CI: -199.29,-102.62(Z=6.12, P<0.01), mortality RR=0. 55, 95% CI: 0. 48, 0. 64(Z=8. 09, P<0. 01). Conclusion Nucleos(t)ide analogues treatment could significantly increase HBV DNA undetectable rate, improve PTA and Alb, decrease levels of ALT and Tbil, which further reduces the short-term mortality of the patients with HBV-related chronic liver failure.

4.Effects of Heterocyclic Oleanolic Acid Derivatives-Aspirin Conjugates on Inhibition of Serotonin Biosynthesis and Increase of Bone Formation

Objective To explore the effects of the heterocyclic oleanolic acid derivatives-aspirin conjugates on inhibition of serotonin biosynthesis and increase of bone formation.Methods The inhibition rate,content and mRNA expression of heterocyclic oleanolic acid derivatives-aspirin conjugates on tryptophan hydroxylase 1 (TPH-1),which was the principal enzyme in the biosynthesis of serotonin,were tested by HPLC,ELISA kit and real-time PCR,respectively.Osteoporosis model was established by ovariectomy (OVX).The rats were randomly divided into conjugate groups,parathyroid hormone group,model control group and sham operation group.Serum and gut serotonin levels were tested by HPLC after 35 days of administration,and the antiosteoporosis activity of the heterocyclic oleanolic acid derivatives-aspirin conjugates was evaluated by using osteoblast-like cells isolated from murine calvaria by MTT assay.Results The preliminary biological results showed that heterocyclic oleanolic acid derivatives-aspirin conjugates displayed a dose-dependent suppression on TPH-1 in RBL-2H3,and the inhibition rate was 20.4%-92.5%.Specifically,conjugate 7 at 10 μmol·L-1 concentration showed the greatest inhibition rate on TPH-1 (92.5%).Content of TPH-1 was 51 ng mL-1,significantly lower than that in the control group (216 ng·mL-1),as well as the TPH-1 mRNA.Compared with the sham operation group,levels of serum and gut serotonin were increased in OVX group,while they were significanlty decreased in conjugate groups,and reduced significantly in conjugate 7 group (232 and 155 ng·mL-1) as compared with OVX group (1 050 and 783 ng·mL-1);Moreover,heterocyclic oleanolic acid derivatives-aspirin conjugates could increase the bone formation.Conclusion This study provides information and basis for development of novel,efficient and harmfulless oleanolic acid derivatives with anti-osteoporosis.
5.Comparative study of cisplatin plus gemcitabine versus cisplatin plus vinorelbine in patients with advanced stage non-small cell lung cancer

AIM: To explore the differences of efficacy and side-effects in advanced stage non-small cell lung cancer (NSCLC) patients treated with gemcitabine plus cisplatin or vinorelbine plus cisplatin. METHODS: Eligible patients were randomly assigned to GP (gemcitabine+cisplatin) group or NP (vinorelbine plus cisplatin) group. In GP group, 36 evaluable patients were treated with gemcitabine 1000 mg?m~ -2 IV on day 1 and 8 and cisplatin 75 mg?m~ -2 IV which was divided into 1-3 days dosing, in a 21 days per cycle manner. In NP group, 30 evaluable patients were treated with vinorelbine 25 mg?m~ -2 IV on day 1 and 8 and cisplatin 80 mg?m~ -2 IV which was divided into 1-3 days dosing, 21 days per cycle. All the patients at least received two cycles therapy. The response rate, median survival time (MST), l year survival, and side-effects were observed. RESULTS: The response rates were 41.6 % vs 36.7 %; MST were 10.3 months vs 9.6 months; 1 year survival rate were 44.4 % vs 40.0 %(P= 0.33 ) in GP group and NP group, respectively. 6.Study on the Preparation of PEG-SOD Enteric-coated Capsuls

OBJECTIVE:To study the method of preparing PEG-SOD enteric-coated capsuls and quality standard.ME_ THODS:Orthogonal design was used to select the optimal conditions for preparing PEG-SOD enteric-coated capsuls and the relevant quality standard of PEG-SOD enteric-coated capsuls was established according to the China Pharmacopoeia(2000). RESULTS:The optimal conditions were as follows:the content of PEG-SOD per capsule was40000IU;the weight ratio of stabilizer lactose was2.5%;filler was carboxymethyl starch;drying process was at60℃for20min.The preparation was ac?corded with the relevant quality standard.CONCLUSION:The technic of preparation is feasible,the method of quality control is reliable and the quality of preparation is stable.
7.The application of extended supraglottic horizontal partial laryngectomy in advanced laryngeal carcinoma and vallecula carcinoma.

OBJECTIVE:
To discuss the function recovery and treatment effect of extended supraglottic orizontal-partial laryngectomy in the treatment of advanced laryngeal carcinoma and vallecula carcinoma.
METHOD:
Forty-four patients who received extended supraglottic orizontal partial laryngectomy were followed up on the survival, breath function, pronunciation and swallowing function.
RESULT:
The 2-year, 3-year survival rates of this group were 72.7% and 71.4%, respectively. There were no recurrence cases in this group. All cases of death were due to cervical lymph node metastasis or recurrent cervical lymph node metastasis after cervical lymph node dissection. The extractive rate of tracheostomy tube was 97.7%. Forty-two patients own good pronunciations. Two patients who received partial arytenoid cartilage resection own weaker pronunciations. After practising, all the patients can eat without the nasal feeding tube.
CONCLUSION
Extended supraglottic orizontalpartial laryngectomy give complete resection of the primary lesion. Patients who received extended supraglottic orizontalpartial laryngectomy have good breath, swallowing and pronunciation function.

10.Analysis of protoscoleces-specific antigens from Echinococcus granulosus with proteomics combined with Western blot.

OBJECTIVETo establish and optimize the proteomic analysis of protoscoleces-specific antigens from Echinococcus granulosus. To provide a foundation for identifying specific antigens in the soluble proteins of E. granulosus protoscoleces for further research.
METHODSBrood capsules were collected aseptically from fertile E. granulosus cysts from the livers of an infected patient. The fertile E. granulosus cysts were fractured, and protoscoleces were collected by centrifugation. The soluble proteins of protoscoleces were acquired using the 2D Quant kit according to the manufacturer's instructions. We employed two-dimensional electrophoresis (2-DE) combined with immunoblot assay (Western blot) to analyze the soluble components of E. granulosus protoscoleces antigens. The 2-DE and immunoblot maps obtained were analyzed with PDQuest 8.0 image analysis software.
RESULTSAbout 233 soluble protein spots were identified with Coomassie-stained gels. Most of the proteins had a molecular weight of 16,000 Da to 117,000 Da, and an isoelectric point value of 3.0 to 10.0. 2-DE immunoblot was conducted and 57 specific antigen spots were observed, among which 23 spots were identified.
CONCLUSION2-DE combined with Western blot is the key to successful proteomic analysis and presents a new possibility for searching the specific E. granulosus protoscoleces antigens.
