1.PROTEIN EXPRESSION AND PHOSPHORYLATION OF MITOGEN ACTIVATED PROTEIN KINASES IN MURINE KUPFFER CELLS INDUCED BY LIPOPOLYSACCHARIDE(LPS)

To elucidate the signal transduction mechanism of systemic inflammatory response syndrome induced by LPS,Western blot analysis was used to assay the protein expression and phosphorylation levels of ERK, JNK,and p38 MAPK in murine KC which were stimulated by LPS. It was found that LPS treatment resulted in no significant change in their protein expression, but a rapid transient increase in their phosphorylation levels, which peaked at 30, 45,and 20 minutes,respectively; The values returned to mear baseline within 2 hours. Kinase phosphorylation levels of these three MAPKs induced by LPS were found to be dose dependent in a range of 10 pg/ml to 100ng/ml. No phosphorylation was observed in unstimulated cells. Among them, p38 MAPK shoned fastest and highest phosphorylation levels. The results show that LPS can markedly activate ERK, JNK,and p38 MAPK in KC, and p38 MAPK may play a more important role in LPS induced kupffer cell activation than ERR and JNK.

Objective To study the effects of three kinds of endoscopic therapy on gastric varices. Methods In this study, the efficacies of endoscopic band ligation ( EVL) , endoscopic injection of tissue adhesives (histoacryl) and sclerotic agent (EIS) for gastric varices were assessed. Forty consecutive, non-randomized patients with gastric varices were prospectively studied. EVL was done for 11 cases, Histoacryl 15 cases, and EIS 14 cases, to control and prevent relapse of bleeding. 3.Effect of Health Education Intervention on the Recovery of First Onset Schizophrenia

ObjectiveTo investigate the short-term effect of health education intervention on the recovery of new schizophrenia patients. Methods82 patients were randomly divided into observation group, in which patients accepted routine antipsychotic medication, general nursing and system health education intervention, and control group, in which patients accepted antipsychotic medication and general nursing. Brief psychiatric Rating Scale (BPRS) and Positive And Negative Syndrome Scale (PANSS) were used to assess the effects. ResultsThere was no difference in the score of every factor before intervention (P>0.05), but it became different after intervention (P<0.01 or P<0.05). ConclusionHealth education intervention can improve the effect on schizophrenia.
4.Transcranial approach for resecting spheno-orbital meningiomas

Objective To explore the effect and safety of transcranial approach for spheno-orhital meningioma. Design Retro- spective case series. Participants Thirty-two patients being operated with transcranial approach. Twenty-four cases were meningothelial meningiomas, 3 cases were fibrous meningiomas, 1 case was psammomatous meningioma, 2 cases were atypital meningiomas, 2 case were malignant meningiomas. Methods All patients underwent frontal-temporal craniotomy, the involved sphenoid wing bone and peri- orbit were removed to prevent recurrence. The superior orbital fissure and optic canal were decompressed, the dural and periorbital de- feet were repaired by autogenous temporal fascia or artificial dura. Main Outcome Measures Preoperative and postoperative exoph- thalmus and eyeball movement, the extent of tumor resection, the ratio of recurrence. Results The extent of tumor resection: 8 cases were Simpson gradeⅡ, 20 cases Simpson gradeⅢ, 4 cases Simpson grade IV. After surgery, proptosis were improved in all patients, ophthalmoplegia was found in 6 eases. There was no operation-related death or other significant complication. Tumor recurred in 6 cas- es. Conclusions Adequate exposure of the tumor and bony decompression of the cranial nerves can result from transcranial approach, all the involved bone should be removed in order to prevent recurrence. This approach is relatively safe and the ptoptosis are improved significantly. Complete surgical resection is difficult because of the involvement of the orbital apex, superior orbital fissure and cav- ernous sinus.

6.Current development of scaffold design and fabrication in bone tissue engineering

With the development of tissue engineering,the bone tissue engineering is developing rapidly. Engineering hone typically uses an artificial extracellular matrix (scaffold),osteoblasts or progenitors,and os- teoinductive factors which promote cell attachment,differentiation,and mineralized bone formation.Highly porous scaffolds play a critical role in cell seeding,proliferation,and new 3D-tissue formation.A variety of biodegradable polymer materials and scaffolding fabrication techniques for engineering bone have been developed over the past decade.This article reviews the scaffold design and fabrication methods tor engineering bone to analyze their ad- vantages and limitations.Various architectural parameters of scaffolds important for bone tissue engineering (e.g. porosity,pore size,interconnectivity) are discussed.
7.Relative and Absolute Quantification-Expression Analysis of CsSAMDC Gene As a Case

This experiment was based on a full-length SAMDC sequence from the drought stress induced cDNA library of Cleistogenes songorica.Two methods,absolute quantification and relative quantification,were used to analyze the gene expression under different tissue and drought stress treatment by real-time quantitaive PCR experiments and were compared each other,while CsSAMDC gene was used as a case.Leaves and roots tissue were sampled from the plants at 0,4,6,8,and 10 d of drought stress and 1 and 4 d after rewatering.The 2-CT method was used to analyze the relative changes in gene expression from quantitative real-time PCR experiments.A standard curve was generated by the purified plasmid DNA.The primer concentration in the realtime PCR was optimized.All absolute quatification data were normalized by the normalization factor from the most stable house-keeping genes in C.songorica.Relative quantification results using 2-CT method showed that CsSAMDC transcripts increased 5.93?0.71 fold in roots,down regulated 0.62?0.13 fold in leaves,under drought stress.Absolute quantification results showed that CsSAMDC transcripts up-regulated in roots significantly under drought stress as 1ong as 8 to 1 0 days after drought stress,while down-regulated in both roots and leaves after drought stress within 6 days and 1 to 4 days after rewatering,compared with control.CsSAMDC transcipt showed similar tendency of gene expression compared by using relative and absolute quantification methods.

Objective To observe the therapeutic effect of acupuncture plus oral taking Chinese medicine compared with that of oral taking Chinese medicine only for the treatment of insomnia.Methods Totally 64 patients were randomized into treatment group(33 cases)and control group(31 cases).The treatment group was treated by acupuncture with filiform needle[Baihui(GV 20),Sishen- cong(EX-HN1),Shenting(GV 24),Neiguan(PC 6),Shenmai(BL 62)and Zhaohai(KI 6)being the main points] plus oral taking Chinese medicine.The control group was treated by oral taking Chinese medicine only.All 64 cases were examined by SPIEGEL scale. Aim To investigate the effect of ultra low molecular weight heparin(ULMWH)to protect primarily cultured rat cortical neurons from the deleterious effects of the neurotoxicant glutamate (Glu)and explore the related mechanism.Methods Cortical neurons of fetal rats were cultured carefully in vitro and treated with 100 ?mol?L-1 Glu so that the protective effects of ULMWH were observed thoroughly. And then the viability of cortical neurons, morphological change together with the number of apoptotic neurons,and the concentration of intracellular free Ca2+([Ca2+]i)were measured by MTT assay, Hoechst33258 staining and Fura-2/AM double wavelength fluoremetry, respectively.Results Brief exposure of cultures to 100 ?mol?L-1Glu led to extensive neuronal death and rapid increase of [Ca2+]i.Pretreatment with ULMWH over the concentration range of 0.01~1 mg?L-1 significantly inhibited the Glu-induced neuronal cell death assessed by a MTT assay and the number of apoptotic nuclei, evidenced by Hoechst 33258 staining.Glu-induced elevation of[Ca2+]i was decreased by pretreatment with 1 mg?L-1 ULMWH, which was indicated by Fura-2/AM not only in assay medium containing Ca2+ but also in Ca2+-free assay medium.Conclusions Those results suggest that ULMWH protects the cultured neurons from Glu-induced neurotoxicity, which may be attributed to its alleviating intracellular free Ca2+ overload via suppressing intracellular Ca2+ release from internal stores induced by Glu.
10.Detecting KRAS,EGFR mutations in nonsmall-cell lung cancer/colorectal cancer patients by high resolution melting curve analysis

ObjectiveTo evaluate the feasibility of detecting the mutations of KRAS,EGFR in nonsmall-cell lung cancer and colorectal cancer patients with high resolution melting curve analysis. MethodsAt first,the mutations in KRAS exon 2 and EGFR exons 18 to 21 of 5 patients with non-small cell lung cancer and 5 patients with colorectal cancer were detected by high resolution melting curve analysis.Then the results were confirmed with direct sequencing. ResultsBy high resolution melting curve analysis,1 of 10 patients was found to carry EGFR 19 mutation who was harboring 2236-2250del mutation.By direct sequencing,consistent results of the patients with mutations orwithout mutations were got. Conclusion s The new high resolution melting curve analysis was more efficient,more convenient than direct sequencing.Moreover,it was a low-cost test,which was suitable for clinic test.
