1.Cloning of rat neuroglobulin gene and construction of its eukaryotic expression vector

Objective To clone rat neuroglobulin (NGB) gene and construct its eukaryotic expression vector. Methods The total RNA was extracted from Wistar rat brain and the full length cDNA encoding NGB was obtained by RT-PCR. After the sequence was confirmed by sequencing and BLAST, it was inserted in the eukaryotic expression vector pCDNA3.1 (+), then the sequence and reading frame were confirmed by two restriction endonucleases and sequencing. Results The NGB gene was cloned with four bases mutated and its eukaryotic expression vector was constructed. Conclusion NGB expressed in Wistar rat brain. NGB gene was successfully cloned and inserted in eukaryotic expression vector.
2.Protective role of recombinant plasmid pCDNA3.1(+)/Ngb during focal cerebral ischemia of rat brain

Objective To observe the protective effect of recombinant plasmid pCDNA3.1(+)/Ngb during focal cerebral ischemia in rat brain.Methods Fifty-four male Wistar rats were randomly divided into three groups:normal saline(NS)control group,plasmid control group,and recombinant neuroglobulin group.NS,plasmid pCDNA3.1(+)and recombinant plasmid pCDNA3.1(+)/Ngb were respectively injected into two sites of the rat cerebra1 cortex 24 hours before induction of neocortical focal ischemia by occlusion of the right middle cerebral artery for 24 hours.The condition of local ischemic damage,expression of bcl-2 and the apoptosis in neural cells were confirmed by staining with 2% 2.3.5-triphenyltetrazolium chloride,in-site cell apoptosis detection,indirect immunofluorescent staining and Western blotting,respectively.Results The extent of cerebral infarction tissue and the apoptosis cells in the pCDNA3.1(+)/Ngb group were significantly reduced than those in other control groups(P
3.Effects-of nuclear factor-kappa B in the progress of acute pancreatitis

The cascade release of cytokine the critical aggravation factor for acute pancreatitis,induces the multi-ple organ dysfunction syndrome (MODS) and death at last. Nuclear factor-kappa B as a number of regulation of the nuclear factor can specific bind with a variety of gene promoter sites κB site, which cause inflammatory reaction, apoptosis, pathophysiologic reaction. The specific inhibitor of nuclear factor-kappa B can inhibit the activity of nu-clear factor-kappaB, inhibit the body's inflammatory response, and improve the prognosis of acute pancreatitis; it may be a potential target site for acute pancreatitis therapy. However, the other researches also found that inhibition of nuclear facter-κB could significantly increase the aggravation of acute pancreatitis. Accordingly, nuclear factor-kappa B research possess the important rank about the body's inflammatory respanse.
4.Children with urinary fistula after hypospadias: causes and repair (report of 33 cases)

Objective To study the causes of congenital hypospadias fistula and the repair methods,summarize surgical experience,and improve the success rate of fistula repair.Method s From January 2001 to February 2011,33 cases of urinary fistula after hypospadias urethroplasty were collected,the average age being 6.2 years (3 to 13 years).A total of 38 fistula were found,17 with fistula diameter of less than 3 mm,16 with fistula diameter of 3 - 10 mm,5 greater than 10 mm.Ligation embedding method were used,Y-V flap coverage method,a continuous inverted fistula suture,Thiersch method,Denis-Browne method and the U shaped flap covering method were used for multi-urinary fistula repair.Among the 33 cases,successful repair was found in 30 cases,with normal urination during flollow- up,good penile appearance,the success rate being 90.9%.Results The causes of hypospadias fistula after surgery in children are:vascularized free when properly drawn,resulting in poor blood supply to the flap; urine drainage was not smooth,resulting in premature suture wounds soaked in urine,causing infection; distal urethral stricture,high pressure within the urethra during urination,causing anastomotic dehiscence; suture materials of poor quality; technical immaturity.Conclusions The repair of urinary fistula after hypospadias in children has a variety of surgical procedures,choice of which should be based on fistula location,size,quantity and local conditions.

Objective] Calrify the inherent laws of the appearance of Neijing’s viscera-state theory of spleen, enhancing the cognition of life view of traditional Chinese medicine’s ability. [Methods]Using the genetic methods to discuss the physiological function, physiological characteristics and physiognomy of spleen, as well as social, cultural, philosophical background and method of thinking construction. [Results] Under the guidance of the overall concept and from the pre-Qin rank system, cultural thoughts of Confucianist and Taoists, Yin-Yang five elements philosophy doctrine,by using the analogism methods and observation methods of “ estimating interior by taking charge of exterior” and the opinion of middlebrow and harmony, construct the basic frame of viscera-state theory of spleen.[Conclusion] The genetic method of TCM is an important way to deepen the level of Chinese medicine theory.

8.Effects of uric acid in different concentrations on the proliferation of human umbilical vein endothelial cells in vitro

BACKGROUND:Recently, there is more and more attention on uric acid (UA), especially the correlation with cardiovascular disease in the filed of epidemiology; however, researches of the effect of UA on endothelial cells are lack based on cytology.OBJECTIVE:To explore the effects of UA in different concentrations on human umbilical vein endothelial cells line (ECV304), malondialdehyde (MDA), nitric oxide (NO) and 3-(4,5-dimethyl thiazole-2)-2, 5-diphenyl thiazolyl blue (MTT)in vitro.DESIGN: Randomized controlled study based on ECV304.5ETTING: Department of Endocrine, Xijing Hospital, the Fourth Military Medical University of Chinese PLA.MATERIALS:ECV304 was provided by Department of Immunology, the Fourth Military Medical University of Chinese PLA; DMEM Iow-glucose medium by Gibco Company, USA; bovine serum by Beijing Yuanheng Shengma Biotechnology Researching Institute; trypsin by Gibco Company, USA; MTT by Huamei Company; dimethyl sulfoxide (DMSO) analytical pure by Tianjin Bodi Chemical Engineering Co. Ltd.; UA by Sigma Company; NO kit and MDA kit by Nanjing Jiancheng Company; ordinary invert microscope and enzyme-linked immune detector IX70 invert microscope by Olympus, Japan;enzyme-linked immune detector by Eastern China Electron Tube Factory.METHODS: The experiment was carried out in Department of Endocrine and Burning, the Fourth Military Medical University of Chinese PLA from December 2003 to April 2004. Resuscitation, culture, regeneration and inoculation of endothelia cells were undertaken Cell Culture published by Situ et al. Endothelia cells were cultured with non-serum DMEM for 24hours so as to maintain synchronization at the phase of G0/G1 and divided into 4 groups, including control group, low-concentration UA group, moderate-concentration UA group and high-concentration UA group. Each group was divided into 3time points, including 24 hours, 48 hours and 72 hours with 8 samples in each time point. Samples were added with 5%serum medium containing 0 mmol/L, 0.1 mmol/L, 0.2 mmol/L and 0.4 mmol/L UA in control group, low-concentration UA group, moderate-concentration UA group and high-concentration UA group, respectively, and incubated in box with the volume fraction of 0.05 CO2 at 37 ℃. Twenty-four hours later, MDA and MTT were detected; additionally, MTT was detected once more after 48 and 72 hours.MAIN OUTCOME MEASURES: Proliferation of ECV304 and content of NO and MDA.RESULTS:With the increasing concentration of UA, content of MDA was decreased to (2.97±0.05),(2.89±0.09),(2.78±0.10) and (2.44±0.03) μmol/L, respectively; content of NO was (6.86±1.41), (12.5±2.7), (18.9±1.8) and (21.1±1.4) μ mol/L. Absorbencies of NO and MTT were increased and proliferation was increased remarkably at 48 hour.CONCLUSION: A which is characterized by anti-oxidation may promote proliferation of vascular endothelia cells and release of NO.
9.Value of ultrasonic diagnosis of congenital prepyloric diaphragm

Objective To evaluate the value of ultrasonic diagnosis of congenital prepyloric diaphragm. Methods Twelve cases were collected for the analysis of the characteristics of ultrasonography in diagnosis of congenital prepyloric diaphragm. Results Of the 12 cases of congenital prepyloric diaphragm, valvula in pyloric antrum could be observed in 5 cases, inclusive of 2 cases of complicating pachyntic stenosis of pyloric muscular layer. Other valvula in pyloric canal could not be found with ultrasonography. No pyloric muscular layer pachynsis was observed, but signs of pyloric obstruction such as gastric retention, slow or difficult transfer of gastric contents, gastro-esophageal reflux were found. Prepyloric diaphragm was confirmed surgically in all cases. Conclusion Ultrasonic diagnosis of congenital prepyloric diaphragm is of practical value in clinical practice and can provide reliable imaging data for the option of preoperative approaches.
10.Analysis on the effect of pantoprazole and omeprazole in the treatment of gastric ulcer

Objective To investigate and analyze the effect of pantoprazole and omeprazole in the treatment of gastric ulcer.Methods a total of 100 patients with gastric ulcer treated in Shizuishan First People's Hospital from January 2014 to June 2016 were selected and randomly divided into the control group and the experimental group, with 50 patients in each group.The control group was treated with omeprazole, and the experimental group was treated with pantoprazole.After taking one course of treatment, the clinical efficacy, treatment cost, eradication rate of Helicobacter pylori and recurrence rate of gastric ulcer were compared between the 2 groups.Results The total effective rate of the experimental group was 94%, which was significantly higher than that of the control group(72%), and the difference was statistically significant(P<0.05).After the corresponding treatment, the experimental group of 50 cases, Helicobacter pylori eradication rate was 72%, the recurrence rate of gastric ulcer was 12%.In the control group of 50 cases, the eradication rate of Helicobacter pylori was 52%, and the recurrence rate of gastric ulcer was 36%.As a result, the eradication rate of H.pylori in the experimental group was significantly higher than that in the control group.The cost of treatment in the control group was(499.78±74.81)yuan, and the treatment cost of the experimental group was(413.26±56.39)yuan.The treatment cost and gastric ulcer recurrence rate in the control group were significantly higher than those in the experimental group, with statistical difference(P<0.05).Conclusion Pantoprazole in the treatment of gastric ulcer is much better than omeprazole, high treatment efficiency, is the preferred drug for the treatment of gastric ulcer, after the treatment of gastric ulcer recurrence rate is low, the cost of treatment is low and high eradication rate, with the further promotion and application in clinic significance.
