1.Preparation and Characterization of Andrographolide Phospholipid Compound

OBJECTIVE:To prepare andrographolide phospholipid compound,optimize its formulation technology and charac-terizeit. METHODS:Andrographolide phospholipid compound was prepared with solvent method. Using compound rate as index, the effects of reaction solvent,reaction time,reaction temperature,drug concentration,the ratio of reactants and other single fac-tors on compound rate were investigated. The ratio of reactants,reaction temperature and drug concentration were optimized by or-thogonal test and validated. The dissolubilities of andrographolide,andrographolide phospholipid physical mixture and androgra-pholide phospholipid compound in water,soybean oil and Lauroglycol FCC were compared and characterized with UV and DSC. RESULTS:The optimal preparation technology was tetrahydrofuran as reaction solvent,reaction time of 4 h,reaction temperature of 50 ℃,drug concentration of 10 mg/ml,ratio of andrographolide to phospholipid 1:3. The compound rate of andrographolide phospholipid compound was 83.2%(n=3). Compared with raw material and physical mixture,phospholipid compound significant-ly improved the dissolubility of andrographolide in water,soybean oil and Lauroglycol FCC. UV and DSC showed that compound had been prepared. CONCLUSIONS:Andrographolide phospholipid compound is prepared successfully.

3.The relation between interleukin-10 promoter-1082 and -819 sites gene polymorphism with irritable bowel syndrome

Objective To explore the relation between IL-10 promoter region gene polymorphism and irritable bowel syndrome (IBS).Methods By polymerase chain reaction combined with restrition fragment length polymophism (PCR-RFLP),gene type of IL-10 promoter -1082 and -819 sites in 313 IBS patients and 281 controls was analyzed.Results The distribution of IL-10-1082 and-819 allele frequencies in IBS group,control group and total was in accordance with Hardy-Weinberg equilibrium law.The frequency of IL-10-819 T allele in diarrhea subtype (79.8%) and mixed IBS subgroup (77.1%) was significantly higher than that in control group (65.7%).There were no significant differences in IL-10-1082 A/G allele frequency between each subtypes and control group (P>0.05),however there was statistically difference between diarrhea subtype and mixed IBS subgroup (P<0.05).The frequency of-819 T/T genotype in IBS group (51.1 % )was significantly higher than that of control group (40.2%),the frequency of C/T genotype was significantly lower than that of control group,and the difference was statistically significant (all P<0.05).The IL-10-819 T/T allele frequency of all IBS subtypes was significantly higher than that of control group; however C/T allele genotype frequency of all IBS subtypes was significantly lower than that of control group,and the difference was statistically significant (all P < 0.05). There was no significant difference of C/C allele genotype between subtypes (P<0.05).There was no significant difference of -1082 allele genotype between IBS group and control group (P>0.05).The frequency of -1082 A/A genotype in diarrhea subtype of IBS patients (93.3%) was significantly higher than that of mixed IBS subtype (82.4%),while the frequency of A/G genotype was lower than that of mixed IBS subtype,and the difference was statistically significant (all P < 0.05 ); there was no significant difference between other IBS subtypes and control group (P>0.05).Conclusion IL-10-819 promoter T/Tgenotype may be related to IBS pathogenesis.
4.Studies on analgesic effect and toxicity of Weifuqitong Capsule

AIM: To study the analgesic effect and toxicity of Weifu qitong Capsule (Rhizoma Corydalis, cortex Magnoliae Officinalis, Radix Aucklandiae, etc.) to provide experimental data for its clinical application. METHODS: The analgesic effect of Weifu qitong Capsule was assessed by hot-plate test and writhing test, its acute and subacute toxicity studies were also conducted. RESULTS: Taking high dose of Weifu qitong Capsule, after 4.5 h (6.249 g/kg) WFQC could increase pain threshold in hot-plate test. Taking low, middle or high dose of WFQC after 2 h, WFQC could significantly extend writhing latent period and decrease writhing frequency (P
5.The efficacy evaluation of multiplane operations in obstructive sleep apnea-hypopnea syndrome.

OBJECTIVE:
To evaluate the therapeutic effect of multi-plane operations in treating obstructive sleep apnea-hypopnea syndrome (OSAHS).
METHOD:
One hundred and fifteen patients with OSAHS diagnosed by polysomnography were treated with uvuplopalatopharyngoplasty. Eighteen of them were treated combining with nasal septal construction. Twenty six patients also received nasal septal construction and partial inferior turbinectomy. One patients also received Genioglossus advancement and partial resection of corpus linguae. Five patients also received partial resection of corpus linguae. Some patients with the nasal disease and/or the lingual hypertrophy; AHI > 40 and/or BMI > 30 are received tracheotomy before general anaesthesia.
RESULT:
According to the postoperative follow-up 43 patients, were cured, 46 patients were improved, 26 patients were invalid, the effective power was 77.4%.
CONCLUSION
The operative effective power were increased by multi-plane operations in OSAHS patients. The serious complication were prevented through tracheotomy before general anaesthesia in multi-plane operations of severe OSAHS.

6.Atractylodes lancea and Its Geo-Varieties

The geo-herbalism of Cangzhu, Atractylodes lancea (Thunb.) DC. , was studied for itsstrict quality control. Samples of typical and representative populations of geo-herbal and non-geo herbalorigin were collected, examined and cultivated through field work. Volatile oils from samples of varioushabitats were determined by GC/MS, and their respective bacteriostatic minimum inhibitory concentration(MIC)determined. Results of the study showed that both the size of capitulum and cleft on the leaves var-ied greatly even in species of the same population. The main components in their volatile oil also variedgreatly with Maocangzhu (MCZ) contains atractylone (29.54%) and atractylodin (17.82%); that of Nan-cangzhu (NCZ) contains hinesol (61.86%) and β-eudesmol (19.79%), and Baicangzhu (BCZ) cantains βeudesmol or mixture of β-eudesmol and atractylone (over 30%). Volatile oils of speciaes from Luotina,Taibai, and Huanglong showed potent bacteriostatic activity against Streptococcus hemolyticus (SH), com-parable to penicillin or streptomycin. In conclusion, A. lancea is a polymorphic complex greatly influencedby geographic environment and a reasonable proposal is to name them by geo-herbal names such as: MCZas A. lancea var. maoshanensis Hu et Feng; NCZ as A. lancea subsp. luotianensis Hu et Feng; and BCZ asA. lancea var. chinensis (Bunge) Kitam other than independent species as A. chinensis Bunge. It was in-correct to say that MCZ has no atractylone in its volatile oil and therefore differs from A. macrocephalaKoidz. (Baizhu). Inhibitory activity of the volatile oils against SH and Pseudomonas aeruginosa (PA) arealso quite different due to the habitats and imply that the rhizome from Luotian may be used to cure infec-tion diseases caused by SH and PA besides their traditional uses.
7.Expression and transport function of epithelial sodium channel in alveolar epithelial type Ⅱ cells of hyperoxia-exposed neonatal rat

Objective To investigate the effect of hyperoxia on the expression and transport function of epithelial sodium channel (ENaC) in neonatal rat alveolar epithelial type Ⅱ(AT Ⅱ) cells.Methods AT Ⅱ cells were isolated from neonatal rats,and primarily cultured under hyperoxic or normoxic conditions.Western blot was applied to examine the ENaC expression,and the amiloride-sensitive Na + currents were recorded using the whole-cell patch clamp technique.Results Hyperoxia upregulate the expression of β-ENaC and γ-ENaC subunits in the neonatal rat ATⅡ cells(β-ENaC:1 d:0.43 ±0.06 vs0.32 ±0.04,P =0.047;2 d:0.73±0.06 vs 0.50±0.08,P =0.019;3 d:0.72 ±0.08 vs 0.52 ±0.06,P =0.027;γ-ENaC:1 d:0.64±0.05 vs0.53 ±0.05,P =0.044;2 d:0.76 ±0.03 vs 0.52 ±0.04,P =0.001 ;3 d:0.77 ±0.06 vs 0.61 ±0.05,P =0.025).In addition,the amiloride-sensitive Na+ currents in hyperoxia-exposed AT Ⅱ cells were also increased (1d:13.71 ±2.77 vs8.92±1.38,P<0.001;2d:29.12±11.03 vs 10.41 ±1.80,P<0.001),which was consistent with the upregulated expression of β-ENaC and γ-ENaC.However,the expression of α-ENaC was inhibited by hyperoxia to some extent (1 d:0.31 ± 0.05 vs 0.46 ± 0.05,P =0.025 ; 2 d:0.30 ±0.01 vs0.38±0.02,P=0.002;3d:0.37±0.06 vs 0.37 ± 0.08,P =0.983).Conclusion Hyperoxia enhanced the transport function of ENaC in neonatal rat AT Ⅱ cells.Dysfunctional transport of Na + may not be a key factor involving in pulmonary edema at the early stage of bronchopulmonary dysplasia.
10.Relationship between serum concentrations of IL-18, IL-10, IL-6 and acute coronary syndrome

AIM: To examine the relation between serum concentrations of interleukin-18, interleukin-10, interleukin-6 and acute coronary syndrome (ACS). METHODS: Serum concentrations of IL-18, IL-10, IL-6 were measured in 17 patients with acute myocardial infarction (AMI), 30 patients with unstable angina pectoris (UAP), 15 patients with stable angina pectoris (SAP) and 20 controls by enzyme-linked immunosorbent assay (ELISA) and radioimmunoassay (RIA).The relation between IL-18, IL-6 and IL-10 was compared. RESULTS: Serum concentrations of IL-18, IL-6 were significantly increased in the AMI and UAP groups in comparison with the SAP and control groups. Conversely, serum concentrations of IL-10 were significantly decreased in the AMI and UAP groups in comparison with the SAP and control groups. The correlation of concentrations of IL-18 and IL-6 had no significance; but the levels of IL-18 and IL-6 were negatively correlated with IL-10. CONCLUSION: Serum IL-18, IL-6 concentrations increase while serum IL-10 concentration decreases in patients with acute coronary syndromes. The inflammatory imbalance between IL-18, IL-6 and IL-10 may play an important role in the instability of atherosclerotic plaque.
