1.Variations of the functions of T helper lymphocyte subsets in peripheral blood of children with mycoplasma pneumoniae pneumonia in acute phase

Objective To observe the variations of the functional states of T helper cells(Th) in peripheral blood of children with mycoplasma pneumoniae pneumonia(MPP) in acute phase,and to explore their roles in the pathogenesis of MPP.Methods Peripheral blood mononuclear cells(PBMC) of 40 MPP children were stimulated with PMA and ionomycin and cultured in vitro for 48 hours.The levels of IL-4,IFN-?,and TGF-?1 in the supernatants of PBMC were detected by ELISA.Results The levels of IFN-? and the ratio of IFN-?/IL-4 of MPP children in acute phase were significantly lower than those of the controls(P0.05).Conclusions The function of Th2 relatively prevails over Th1 in MPP children.The protective or regulative function of Th3 in vivo increases,but the protective regulation of TGF-?1 is insufficient,so the imbalance of Th1/Th2 can not be corrected.The variation of the functional states of T helper cells in acute phase has no relationship with or without the extrapulmonary damage.
2.Correlation of postoperative serf-efficacy and quality of life for patients with breast cancer

Objective To know the relationship of self-efficacy and quality of life of patients with breast cancer after postoperative chemotherapy, and then carry out reference to clinical nursing cares. Methods Interviewed 115 patients with breast cancer after postoperative chemotherapy by questionnaire to know their conditon about self-efficacy and quality of life, and then analysis the relationship between them. Results The general score of quality of life of patients with breast cancer was 105.62±6.68, which was significant lower than the normal model. There was significant correlation of self-effi-cacy and quality of life of patients with breast cancer. Conchusions Related experts should emphasis the postoperative management of patients with breast cancer, and then promote their self-efflcacy and quality of life.
3.Detection of K-ras mutation in DNA extracted from the plasma of patients with colorectal cancer

Objective The purpose of this study was to determine the value of K-ras mutation in DNA extracted from the plasma as clinical indicator of colorectal cancer. Methods Point mutation at codon 12 of K-ras gene was assayed by polymerase chain reaction of sequence-specific primers in DNA extracted from the plasma and tumors from 32 patients with colorectal cancer. The mutation was further confirmed by dideoxy-mediated chain-termination method of DNA sequencing. Results Fourteen patients (44%) had a codon 12 K-ras mutation within their primary tumors and identical mutations were found in the plasma DNA of 13 patients (93%). Mutant DNA was not detected in the plasma specimens of 18 patients whose tumors tested negative for K-ras alterations or in 5 healthy control subjects. Conclusion Preliminary results suggest the detection of K-ras mutations in circulating DNA extracted from the plasma specimens may have some clinical uti- lity in the detection of colorectal cancer.
4.Influence of health behavior intervention on coping style and hope level in lung cancer patients undergoing chemotherapy

Objective To study the influence of health behavior intervention on coping style and hope level in lung cancer patients undergoing chemotherapy. Methods 60 lung cancer patients undergoing chemotherapy were divided into the intervention group and the control group with 30 patients in each group. Two groups all had an operation, carried on the normal nursing care and health education. After operation, the intervention group accepted health behavior intervention from the beginning of the first period chemotherapy. After the third period chemotherapy, coping style and hope level of the two groups were measured. Results After intervention, positive coping style "confrontation" had a higher score. Negative coping style such as obviation and acception got lower scores in the intervention group compared with the control group. In addition to "keep an intimate relation(I)",the rest dimensions of hope level showed significant differences as well. Conclusions Applying the related theories of health behavior to build up mental nursing intervention mode has a significant meaning on lung cancer patients undergoing chemotherapy. It can increase patients' positive coping style and reduce negative aspects. At the same time, hope levels of lung cancer patients also have an improvement.

7.Use of rats mesenchymal stem cells modified with mHCN2 gene to create biologic pacemakers.

The possibility of rats mesenchymal stem cells (MSCs) modified with murine hyperpolarization-activated cyclic nucleotide-gated 2 (mHCN2) gene as biological pacemakers in vitro was studied. The cultured MSCs were transfected with pIRES2-EGFP plasmid carrying enhanced green fluorescent protein (EGFP) gene and mHCN2 gene. The identification using restriction enzyme and sequencing indicated that the mHCN2 gene was inserted to the pIRES2-EGFP. Green fluorescence could be seen in MSCs after transfection for 24-48 h. The expression of mHCN2 mRNA and protein in the transfected cells was detected by RT-PCR and Western blot, and the quantity of mHCN2 mRNA and protein expression in transfected MSCs was 5.31 times and 7.55 times higher than that of the non-transfected MSCs respectively (P<0.05, P<0.05). I(HCN2) was recorded by whole-cell patch clamp method. The effect of Cs(+), a specific blocker of pacemaker current, was measured after perfusion by patch clamp. The results of inward current indicated that there was no inward current recording in non-transfected MSCs and a large voltage-dependent inward and Cs(+)-sensitive current activated on hyperpolarizations presented in the transfected MSCs. I(HCN2) was fully activated around -140 mV with an activation threshold of -60 mV. The midpoint (V(50)) was -95.1+/-0.9 mV (n=9). The study demonstrates that mHCN2 mRNA and protein can be expressed and the currents of HCN2 channels can be detected in genetically modified MSCs. The gene-modified MSCs present a novel method for pacemaker genes into the heart or other electrical syncytia.
8.Factors related to use of emergency medical services in patients with acute stroke

Objective To examine the availability of emergency medical services(EMS) for patients with acute stroke and to investigate influential factors affecting the preference of patients'to EMS.Methods Consecutive information of patients with acute stroke who presented to the emergency department of Zhongnan Hospital of Wuhan University from June 1, 2014 to December 31, 2015 were analyzed.Gender, age of patients, transport modality, risk factors in cerebrovascular diseases, initial symptoms, stroke types, onset and admission time were included to make analysis in retrospective study.The participants were divided into two groups based on the preference of patients, namely EMS group and self-transport group.Wilcoxon rank sum test or Chi-squared test was used to statistical analysis as appropriate.A multiple binary logistic regression was used to determine the relationship between various patient-related factors with probability whether patients chose EMS or not.Results Among the 240 patients, only 29.2%of them asked for the EMS at the onset of symptoms (EMS group, n=70), and the rest patients called for other services(self-transport group, n=170).Logistic regression analysis revealed that the patients with the symptom of altered consciousness or convulsion (95%CI:0.107-0.403,OR=0.208,P<0.01) were more likely to use EMS.The time consumed from onset to visit of patients with acute stroke to emergency physician was significantly shorter in EMS group (M, 60 min vs.180 min,P<0.01).Conclusion The symptom of altered consciousness or convulsion was the independent factor to determine whether patients with acute stroke preferred EMS or not.
9.Clinical Study on Live Lactobacillus Preparation in the Treatment of Postpartum Vagina Microenviroment Dysbiosis

Objective:To investigate the effect of live lactobacillus preparation in the treatment of postpartum vagina microenviro-ment dysbiosis. Methods:Totally 300 cases of postpartum women were selected in our hospital from February 2013 to November 2013, including those with normal delivery, cesarean section and vaginal delivery after lochia, and each was with 100 cases. The vaginal se-cretion was withdrawn from vagina and the microenvironment was assessed. The patients with dysbiosis were randomly divided into two groups, the observation group was treated with living lactobacillus preparation and the treatment group was without treatment. The effi-cacy and incidence of bacterial vaginosis(BV) were observed. Results:Totally 88 cases (29. 3%) occurred vaginal dysbiosis. There were significant differences in the dysbiosis incidence rate, pH, H2 O2 results, microscopic examination results and the distribution of flora between the two groups (P <0. 05). The effective rate of the observation group was 95. 5%, which was significantly higher than that of the control group (29. 5%). The incidence rate of BV was 4. 5% in the observation group, which was significantly lower than that in the control group [75%] (P<0. 05). Conclusion: The effect of the lactobacillus preparation in the treatment of postpartum vagina microenviroment dysbiosis is promising and safe.
10.Effects of Percutaneous Kyphoplasty on Osteoporosis Vertebral Compression Fractures in Elderly

Objective To observe the clinical effect and safety of percutaneous kyphoplasty (PKP) on osteoporosis vertebral compres-sion fractures (OVCF) in elderly. Methods From January, 2014 to May, 2015, 88 patients with OVCF treated with PKP were retrospective studied. Visual Analogue Scale (VAS) and Oswestry Dysfunction Index (ODI) were used to evaluate the pain and quality of life, and the ky-photic angle (Cobb's angle) and anterior vertebral body height were recorded before operation, one week after operation and the last fol-low-up. The cement leakage and its effect on nerve function were recorded. Results Bone cement leakage was found in eleven patients, in which two in pedicle, three in venous leakage, three in intervertebral disc and three in vertebral posterior. Nerve root symptoms appeared in one patient, who was treated with decompression. The scores of VAS and ODI, and the Cobb's angel were significantly lower (t>29.518, P<0.001), and the vertebral body height was significantly higher (t>35.173, P<0.001) one week after operation and the last follow-up than be-fore operation. Conclusion PKP is safe and effective for OVCF in elderly. It can efficiently reduce the pain, correct kyphotic deformity and improve the quality of life.
