·AlM:To study the MRG15 (death factor related gene) in age-related cataract ( ARC ) , differential expression of normal lens epithelial cells.
· METHODS: Forty mature healthy female SD rats were randomly divided into study group and control group, 20 rats in each group, the study group were ovariectomized, low concentration of naphthalene long interval of administration, the establishment of perimenopausal ARC model, the control group of conventional farming.ln the subtractive hybridization cloning by MRG15, and make the probe of the cDNA fragment using digoxigenin labeled. Access to the two groups of rats anterior lens capsule after slicing, and then through the differential expression in situ hybridization clear lens epithelial cells.
· RESULTS: The cloned MRG15 through BamH1, EcoR1 enzyme digestion and agarose gel electrophoresis, available for 639bp long cDNA fragment. GeneBank display contrast, their homology was 99.0%. ln situ hybridization, ARC patients and normal lens epithelial cells were observed in the expression of MRG15.Study group the percentage of positive cells compared with control group, showed a significant difference (P<0.05). lntegral index study group and the control group compared with, was significantly difference (P<0.05). · CONCLUSlON: ARC, MRG15 in normal lens epithelial cells expressed ARC, and compared with the normal expression of lens epithelial cells, which may produce inhibitory effects are associated with MRG15 transcription in human lens epithelial cells in the part of key genes, by reducing the lens epithelial cell fu nction, make its appear aging, and the formation of cataract, clinical response to this should further study.

OBJECTIVETo investigate the effects of recombinant human growth hormone on serum lipid in aged male patients with chronic heart failure (CHF).

METHODSEighty seven patients with chronic heart failure(> or = 60 years old) were randomly divided into 2 groups: the CHF control group (n = 46) who received regular therapy and the CHF experimental group (n = 41) who received regular therapy and recombinant human growth hormone. The treatment would be continued for 3 months. Another group was normal control group (n = 10). The detection of serum growth hormone (GH), insulin-like growth factor (IGF-1), total cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C), high density lipoprotein cholesterol (HDL-C) was carried out before and after treatment in the participants.

RESULTSBefore treatment, the levels of GH and IGF-1 were not significantly different among groups. After treatment, the levels of GH (0.71 +/- 0.34 vs 0.96 +/- 0.48) and IGF-1 (95.64 +/- 21.11 vs 111.64 +/- 23.14)in CHF experimental group were higher than those before the treatment. In CHF control group, the levels of GH(0.81 +/- 0.32 vs 0.79 +/- 0.29) and IGF-1 (97.82 +/- 19.74 vs 99.65 +/- 20.11) had no significant change after the treatment. After treatment, the levels of GH (0.96 +/- 0.48 vs 0.79 +/- 0.29) and IGF-1 (111.64 +/- 23.14 vs 99.65 +/- 20.11) in CHF experimental group were higher compared with that of CHF control group. Before treatment, the serum levels of LDL-C, HDL-C, TC and TG had no significant difference among groups. After treatment,the levels of LDL-C (2.11 +/- 0.82 vs 1.76 +/- 0.51) and TC (3.78 +/- 1.34 vs 3.21 +/- 1.17) in CHF experimental group were lower than those before the treatment. However, the levels of HDL-C (1.10 +/- 0.31 vs 0.99 +/- 0.28)and TG (1. 89 +/- 1.07 vs 1.66 +/- 0.95) had no significant change after the treatment compared with before treatment. In CHF control group, the serum lipid levels had no significant change after the treatment.

CONCLUSIONAs the treatment of rhGH for aged male patients with chronic heart failure, GH influences lipid metabolism, which reduces the level of LDL-C, TC. However GH has no effects on the serum HDL-C and TG level. With the treatment of rhGH for long-term, lipid metabolism should be paid attention,and the treatment for blood lipid reduction should be adjusted in time.

This paper aims to analyze Professor Guo Rongjuan's medication experience on insomnia therapy based on the Traditional Chinese Medicine (TCM) Inheritance Support Plat. First, TCM formulae prescribed by Professor Guo for insomnia therapy were collected from the TCM Inheritance Support Plat. Next, unsupervised data mining algorithms, including apriori, modified mutual-information, and entropy clustering of complex system were applied to obtain the frequencies for different herbs and identify the association rules among the herbs. Accordingly, we can gain new insights into Professor Guo's medication experience on insomnia therapy. Based on analysis of 3 084 formulae, we determined the frequencies for herbs in the formulae and identified the association rules among these herbs. At last, 41 core combinations and 7 new formulae were obtained. The identified medication experience conform with Professor Guo's views on the etiology and pathogenesis of insomnia: "pathogenic fire derived from stagnation of liver-QI (Gan Yu Hua Huo)" is the core pathogenesis of insomnia; "liver stagnation and spleen deficiency" and "chronic illness transferred to kidney" are the main features for insomnia. The TCM Inheritance Support Plat is of great practical value for mining clinical experience of famous TCM doctors.

The immunoregulatory effect of TLSF_(JM) on the levels of IP3,Ca~(2+) in activated T cells andthe expression of Fos protein was investigated by ABC immunohistochemistry technique and Fu-ra-2/AM labelled T cells and anion-exchange chromatography techniques in this paper.The re-sults showed that TLSF_(JM) can the same time,it can strongly inhibit the levels of IP3、Ca~(2+) in ac-tivated T cells.

Objective　To discuss the clinical value of 18　F-FDG PET imaging in postoperative and postradiotherapeutic intracranial glioma during follow up study. Methods　18F-FDG PET imaging in 16 cases of postoperative and postradiotherapeutic intracranial glioma was compared with CT and/or MRI. Results　Contrast CT or MRI showed prominent irregular circular or nodular enhancement in 14 of 16 cases (64%),can not distinguished from postopertative changes, radioinjury, tumor residue or recurrence.Among them, 9 cases showed tumor residue or recurrence based on significantly increased FDG uptake, 5 were confirmed by pathologic study. The other 5 cases of cerebral necrosis and 2 cases with postoperative cerebral malacia demonstrated FDG uptake defects. Conclusions　18F-FDG PET imaging has significant dominance in characterizing lesions for differentiating residue or recurrent disease from radioinjury in intracranial glioma. Combined with CT and MRI can provide both anatomical and functional informations.

Objective To investigate the clinical value of FDG PET imaging in the follow up of nasopharyngeal carcinoma after radiotherapy. Methods Twelve patients with nasopharyngeal carcinoma underwent FDG PET, CT and/or MRI during the follow up of 12 18 months after radiotherapy. The results of FDG PET were double blind evaluated and compared with these of CT and MRI. The recurrent lesions in six of these patients were pathologically confirmed and six by CT scan. Results Nine of 12 patients did not show any recurrence by CT and MRI scans. However, FDG PET scans showed significantly increased FDG metabolism, indicating recurrence in 3 of these patients. In 2 patients, both CT/MRI and FDG PET indicated recurrence. The remaining one patient had radiotherapy induced cerebral necrosis. He was misdiagnozed as an intracranial recurrence by MRI, but FDG imaging was negative. Conclusions FDG PET imaging has an advantage in distinguishing the relapse or necrosis in the follow up of post radiotherapy nasopharyngeal carcinoma patients. PET, CT and MRI is able to give a detail both in morphological and functional changes of disease. They are especially useful in the follow up or the set up of treatment planning for recurrent nasopharyngeal carcinoma.

Objective To investigate the effect of a kind of tissue engineered bone implants on healing of segmental bone defects. Methods A 15 mm bone defect experimental model at the right radius of New Zealand white rabbit was made. 24 animals of this model were divided into three groups at random. PLGA scaffords loaded with 5 mg BMP and about 1?106 BMSCs were implanted in Group 1(n=10); PLGA scaffords loaded with 2.5 mg BMP and about 1?106 BMSCs were implanted in Group 2(n=7); PLGA scaffords loaded with 1 mg BMP and about 1?106 BMSCs were implanted in Group 3(n=7). The osteogenesis at the defect area was observed by regular roentgenography and X-ray analysis, histological changes at the bone defects at 4th, 8th, 12th weeks after operation were studied and the new bone formation was measured by image analysis. Biomechanical analysis was done 12 weeks after operation. Results A large quantity of callus was found in all the 3 groups 4 weeks after repair. At the 12th week, the complete bone healing rate was 7/8, 3/5, 3/5, respectively in Group 1, 2 and 3. Group 1 had the largest quantity of new bone measured by histopathological study and X-ray analysis, and at the 12th week, the medullary cavity in this group became united. The compress stiffness, torsion stiffness and the maximal load in Group 1 were better than those in other groups. Conclusion The tissue engineered bone contructed by PLGA scaffords that are loaded with 5 mg BMP and BMSCs is capable of repairing segmental bone defects.

AIM:It has been recently proved that either decreased number or impaired function of CD4+CD25+regulatory T cells(Treg)can lead to immune dysfunction.Deeper knowledge of CD4+CD25+Treg cells is crucial for a better understanding of the pathogenesis of many diseases,such as autoimmune diseases,graft-versus-host disease,and allergic disease.This study is aimed to investigate the correlation of CD4+CD25+Treg cells in patients with idiopathic thrombocytopenic purpura(ITP)and to analyze the possible immunopathogenesis. METHODS:The experiment was conducted in the laboratory of Institute of Hematology,Chinese Academy of Medical Sciences and Peking Union Medical College from June to September in 2007.After the patients and their relative signed informed consents, peripheral blood was obtained from ITP patients(n=27)and health people(n=15)admitted in the Hematology Hospital,Peking Union Medical College.All patients accorded with the standards of ITP diagnosis and curative effect.The amount of CD4+CD25+ Treg cells in peripheral blood of normal controls and ITP patients before and after therapy was detected by flow cytometry.For those 23 patients who received effective therapy,blood before and after therapy were both obtained,the mononuclear cells(MNCs)were isolated from peripheral blood by density gradient centrifugation,then reverse transcriptase polymerase chain reaction(RT-PCR) was used to analyze the expression of Foxp3 mRNA. RESULTS:The amount of CD4+CD25+Treg cells and the ratio of CD4+CD25+Treg to total CD4+T cells in peripheral blood of ITP patients were obviously decreased than those of the controls(P0.05),but the expression of Foxp3 mRNA in CD4+CD25+ Treg cells was significantly increased(P

One alkaline protease from Actinomucor elegans AS3.2778 was purified protein. The enzyme was purified using ammonium sulfate precipitation, ion exchange chromatography, hydrophobic chromatography and size exclusion chromatography method, and its properties were also investigated. The molecular weight of this enzyme is 32 kDa with SDS-PAGE method, optimum temperature is 60℃, optimum pH is 8.5 to 10.5, it is stable in the pH range of 6.0 to 9.0 at < 40℃ temperature, and being completely inhibited by the serine protease inhibitor, PMSF, indicated that it belongs to the serine protease family. Specificity test indicated this protease has extensive selectivity to peptide bones, especially to peptide bones composed of Leucine residue.
