1.Combined use of docetaxel and bone marrow mesenchymal stem cells in human hepatoma cell line SMMC-7721

BACKGROUND:Docetaxel, a cel cycle specific anti-tumor drug, is a drug that is used primarily for treating breast cancer; however, its efficacy is low when used for treatment of cancer not sensitive to radiotherapy. Bone marrow mesenchymal stemcels have been shown to strengthen the effects of tumor-specific targeting chemotherapy drugs. OBJECTIVE:To investigate the effects of docetaxel combined with bone marrow mesenchymal stem cels (BMSCs) on human hepatoma cel line SMMC-7721. METHODS:BMSC cels were culturedin vitro. The logarithmic growth of SMMC-7721 hepatoma cels were randomly divided into blank control group, BMSCs group and combined treatment group (combined treatment of BMSCs and docetaxel). SMMC-7721 cel cycle was detected usingflow cytometry. Cel growth rate of SMMC-7721 was determined by MTT assay. mRNA and protein expressions of tumor suppressor genes PTEN and p53 were detected by reverse transcription polymerase chain reaction and western blot assay. RESULTS AND CONCLUSION:Combined treatment of docetaxel and BMSCs inhibited SMMC-7721 cell proliferation. Compared with the blank control group, the number of cells at the G0/G1 phase was significantly increased in BMSCs group and combined treatment group. The cell growth rate of SMMC-7721 was significantly inhibited in BMSCs group compared with the blank control group, and that was further inhibited in combined treatment group (P< 0.05). mRNA and protein expressionof PTENandp53 were significantly increased in combined treatment group compared with BMSCs group and blank control group (P< 0.05). Our results suggest that BMSCs inhibit the growth of SMMC-7721 cells,andcombined use of docetaxel and BMSCs strengthensthe antitumor effect of BMSCs.
2.Clinical and genetic analysis of a pedigree with acne inversa

Objective To detect γ-secretase gene mutations in a large Chinese pedigree with acne inversa (AI).Methods Clinical evaluation was carried out in a large pedigree with AI through field investigation.Peripheral blood samples were obtained from 17 family members (11 affected and 6 unaffected) and 100 unrelated healthy human controls.DNA was extracted from the blood samples,and PCR was performed to amplify all the coding regions of PSEN 1,PSENEN and NCSTN genes followed by DNA sequencing analysis.Results There were 67 members over 5 generations in this family,of whom,25 (13 males and 12 females) were affected by AI.AI was inherited in an autosomal dominant manner in this family.Skin lesions were mainly distributed on the neck,back,chest and buttocks,and occasionally in subaxillary regions.DNA sequencing revealed a novel missense mutation,c.1258C> T (p.Q420XP),in the exon 11 of the NCSTN gene in 11 affected family members,which leads to a substitution of glutamine by a premature termination codon at amino acid 420 (p.Q420X).The mutation was undetected in either the unaffected members or the unrelated healthy controls,and had not been registered in the single nucleotide polymorphism (SNP) database in National Center for Biotechnology Information.Conclusions There is a novel heterozygous missense mutation,c.1258C > T in the exon 11 of the NCSTN gene,which may be the molecular basis of pathogenesis of AI in this family.
3.Unbalance and involvement of Th17/Tr cells in patients with cervical cancer

Objective To detect the levels of Th17 and regulatory T(Tr)cells in Imripheral blood mononuclear and tumor tissue from patients with cervical cancer and analyze the relationship hetween Th17 and Tr cells in cervical cancer progression.In addition.the significance of the Th17/Tr cells ratio in cervical cancer pathogenesis was discussed.Methods The expression levels of Th17 and Tr cells were determined by flow cytometry from 32 patients with cervical cancer and 15 health people.The mechanism of involvement of Th17 and Tr cells proportionality in cervical cancer pathogenesis and the correlation between Th17 and Trwas assessed by bivariate correlation analysis.Results The expression levels of both Th17 and Tr in patients were higher than control groups,especially in late stage patients Th17 and Tr proportionality lower than early group,and there was a negative correlation between them.Conclusion Th17 and Tr cells proportionality may be involve in the development of cervical cancer so as to provide novel strategies for tumor immunotherapy.

5.Analysis of Marketing Conception in State-Owned and Private-Funded Hospital Management

Objective To compare the differences between state-owned hospitals and private-funded hospitals in Shanghai on the introduction and application of hospital marketing conception. Methods Questionnaire and interviews were conducted to reflect understandings in marketing conception,advertisements,hospital brand,customers' sense of satisfaction,etc. Results State-owned hospitals were less concerned in marketing conception compared with their private counterparts,even less sufficient in marketing input or management. Conclusion The state-owned hospitals should update their conception and enhance their ability of marketing management.While the private hospitals need to maintain the moderation of marketing and deal with the relationship among marketing,profit and development properly.
6. Two-photon in vivo imaging of blood brain barrier injury in the ultra-early stage of cerebral ischemic stroke

Objective: To evaluate the effect of two-photon in vivo imaging on detecting the blood brain barrier (BBB) injury in the ultra-early stage of cerebral ischemic stroke. Methods: Twelve clean grade C57BL/6 healthy male mice aged 8-12 weeks were randomly divided into Sham group and middle cerebral artery occlusion (MCAO) group,which were sham operated or middle cerebral artery occluded,respectively. After 60 min of ischemia,MCAO mice were treated with reperfusion for 30-60 min after the suture being removed. The vessels and the neutrophils of mice in the two groups were labeled intravenously with Alexa-Fluora-488 conjugated dextran and rhodamine 6G,respectively. The integrity of BBB was detected by intravenous injection of tetramethylrhodamine-5-maleimide (TMR). Before and after the stroke,the real-time changes of the fluorescence intensity of the inside and outside cerebral vessels of mice in the MCAO group were observed by two-photon fluorescence microscopy. Results: The fluorescence intensity of TMR in the external cerebrovascular of mice in the MACO group was significantly increased within 30-60 min after stroke (P=0.000),suggesting there existed tracer leakage. Compared with the Sham group,the movement of neutrophils in the blood vessels of mice in the MACO group was significantly slowed down (P=0.000). Conclusion: Two-photon in vivo imaging can be used to detect the BBB injury in the ultra-early stage of cerebral ischemic stroke,which provides a certain reference value for clinical application.
7. Relationship between high serum uric acid level and short-term progression of albuminuria in hospitalized diabetic patients

Objective: To explore the risk factors associated with short-term progression of albuminuria in the patients with type 2 diabetes mellitus (T2DM). Methods: The retrospective cohort study design was performed to consecutively recruit 210 hospitalized T2DM patients who met the inclusion criteria, including 102 males and 108 females from April 2010 to April 2015. The data of the patients twice in hospital (as baseline and follow-up information, respectively), including general data, biochemical examinations and medication were collected. Multivariate Logistic analysis was used to explore the potential risk factors for baseline albuminuria and short-term progression of albuminuria. Results: Serum uric acid levels were significantly higher in the patients with macroalbuminuria ( ≥ 300 mg/24 h) than those in the patients without albuminuria (< 30 mg/24 h) at baseline (P=0.002). Among the 207 patients (excluding 3 samples with missing data) during the second hospitalization, 51 patients (24.6%) had progress in albuminuria and 156 patients (75.4%) had none. Serum uric acid levels were significantly higher in the progression group than those in the non-progression group (P=0.009). Multivariate Logistic regression analysis showed that high serum uric acid level was an independent risk factor for progression of albuminuria in T2DM patients (OR=1.349, 95%CI 1.014-1.793, P=0.040). Conclusion: The high serum uric acid level may be an independent risk factor for short-term progression of albuminuria, and early intervention with hyperuricemia might delay the progression of diabetic kidney disease in T2DM patients.
8. Evaluation of the effect of using the new PRISM IV to assess the condition of children with mechanical ventilation

Objective • To evaluate the application effect of the newly updated pediatric critical illness score PRISM on assessing the condition of children with mechanical ventilation after congenital heart disease (CHD). Methods • T test, area under ROC curve and Hosmer-Lemeshow goodness of fit test were used to assess the performance of PRISM . The performance of PRISM was also compared with PRISM III and specific severity of illness scoring system for CHD. Results • 230 children with CHD were included. Reliability among evaluators was good. The discriminatory power (AUC>0.75) and calibration capability (P>0.05) of PRISM , PRISM III and specific score for CHD were tolerable. The results of PRISM evaluation were similar to specific scores for CHDT0-4 and CHDT0-12 (P>0.05). Conclusion • PRISM has been updated to shorten the assessment time and has certain advantages over other tools. It is recommended that nursing staff take it in the prognosis prediction system for critically ill children based on clinical status and practical needs, to detect high-risk children early, and to intervene early to reduce the complications and mortality of children.
9.Preliminary study on immunosuppressive effects evaluation of biomaterial.

In this paper, an immunosuppression model of immunotoxicity built through applying immunosuppressive agent-cyclophosphamide. Subsequently the changes of some assessment indexes including total amount of lymphocytes and concentration of cytokine TNF-alpha in peripheral blood were observed and were used to evaluate immunotoxicity induced by Medical Heat Vulcanizing Silicone Rubber. The final results showed no immunosuppressive effect caused by this material. The study provide effective and sensitive detection technique for evaluation of medical devices and biomaterials' immunotoxicity.
