1.The effect of topotecan on lung cancer cell cycle and the mechanism of topotecan-induced apoptosis

AIM To study the effect of Topotecan(TPT) on cell cycle and apoptosis of hunman lung cancer SPC A 1 cells and the role of Caspase 3 and bcl 2 involved in TPT induced apoptosis. METHODS Cells were incubated with TPT(5,10,15 and 20 mg?L -1 ) for 24 h. Apoptosis was detected by TUNEL staining, electronic microscope and DNA agarose gel electropheresis. Flow cytometer was used to analyse cell cycle changes and to detect apoptotic rates and bcl 2 expression of cells treated with TPT or TPT and 50 ?mol?L -1 Ac DEVD CHO(specific Caspase 3 inhibitor). RESULTS Apoptosis of cells treated with TPT(5 mg?L -1 ) was not detected, but apoptosis rates increased with the increase of TPT concentration and there was significant difference between each concentration group( P
2.Treatment of NH_3~-burned patients complicated with inhalation injury

Objective: To investigate the management of NH~-_3 burned cases with inhalation injury. Methods:The clinic treatments of 15 NH~-_3 burned cases complicated with inhalation injury in his hospital between January 1998 and April 2004 were analyzed retrospectively.His management included reasonable fluid therapy,prompt and efficient usage of antibiotics ,tracheotomy after diagnosis,mechanical ventilation,bronchoalveolar lavage using fibrobrochoscope after the shock period and short-time usage of glucocorticoid. Results: Of the 15 cases,14 patients were cured and 1 patient was improved without any death.The cure rate was 93.3%. Conclusion: All of the managements should be performed as quickly as possible.Paying much attention to prevent the complications could improve the cure rate.Tracheotomy and bronchoalveolar lavage in the early stage are vital to patients.
3.Expression of E-cadherin and ?/? Catenins and their Relationship with the Invasiveness of Pituitary Adenoma

Objective To explore the relationship between the expression of E-cadherin catenin complex(CCC) and the invasiveness of pituitary adenoma.Methods The expession of E-cadherin catenin complex was determined by immunohistochemistry in 45 cases of human pituitary adenomas by random sampling,including invasive adenoma 27 cases,noninvasive adenoma 18 cases.The relativity of their expression with hormone-producing,pituitary apoplexy and necrosis or cystoid change,tumor diameter was analyzed.Results The invasive group had a significantly lower expression of E-cad and ?-cat than that of noninvasive group(?2=8.551,P0.05).Moreover,the expression of E-cad was significantly lower in endocrine inactive group than that in endocrine active group(?2 =5.020,P
4.Pharmaceutical Care for One Case of Natural Killer/T-cell Lymphoma Complicated with Diabetes

OBJECTIVE:To explore the mode,methods and role of clinical pharmacists providing pharmaceutical care for pa-tients with natural killer/T-cell lymphoma (NK/TL) complicated with diabetes. METHODS:Clinical pharmacists participated in SMILE chemotherapy plan for a NK/TL patients with diabetes mellitus,evaluated potential chemotherapy risk during first chemo-therapy,implemented individual medication monitoring and put forward suggestion. It included insulin injection 4 U,ivgtt,to con-trol the increase of blood glucose induced by dexamethasone;sodium bicarbonate 40 ml,ivgtt,bid to alkalize urine;allopurinol 0.1 g,po,tid to inhibit uric acid;fluid infusion 4 000 ml/d to prevent renal failure;mesna 0.4 g/time to protect urinary passage;monitorring potential allergic reaction and arrest of bone marrow;providing medication education on the application of calcium foli-nate in SMILE chemotherapy regimen. RESULTS:Physicians adopted the suggestions of clinical pharmacist,and chemotherapy was carried out smoothly. No ADR was found,and patients manifested good medication compliance. CONCLUSIONS:Clinical pharmacists participate in chemotherapy,evaluate potential risk and provide individual medication monitoring,which optimize ther-apy plan,reduce medication risk and guarantee the safety of drug use.
5.Construction of training evaluation system for laboratory animal practitioners

Objective Laboratory animal practitionres need further management through technical and capacity training.Methods Relying on the Laboratory Animal Center of University and its highly qualified faculty, a high-quality training base for laboratory animal practitioners has been set up.Results Based on the relevant laws and regulations about experimental animals, appropriate training outlines, different training books and courses were prepared and organized for different trainees.Focusing on the detailed and exact experiments, training base has put emphasis on the practical ability and operational skills.Conclusions How to regulate the training for laboratory animal practitioner by means of base training is discussed and tried out in this paper.
6.Early rehabilitation for children with brain impair syndrome and cerebral palsy in hospital and in family

ObjectiveTo compare the early rehabilitation of child with brain impair syndrome and cerebral palsy executed in hospital and in family.Methods180 children of 0~2 years old with brain impair syndrome and cerebral palsy were divided into group A and group B randomly. The children of 0~6 months old were in group A1 and group B1, while that of 6 months~2 years old were in group A2 and group B2, correspondingly. Children in group A accepted early formal strengthen rehabilitation training in hospital, and that in group B accept family rehabilitation through guidance. Development quotient (DQ) were assess with Japan Yuanchengsi infant analysis growth scale half a year after.ResultsThe DQ of group A showed higher than that of group B(P<0.01), and DQ of group A1 was higher than DQ of group A2(P<0.05).ConclusionEarly rehabilitation training in hospital is more effective on early cerebral palsy.
8.Inhibition of mda-7/IL-24 recombinant adenovirus on proliferation of small cell lung cancer NCI-H446 cells

Objective To prepare recombinant mda-7/IL-24 adenovirus to study its function on the proliferation of small cell lung cancer NCI-H446 cells. Methods According to the manufacturer's instructions of AdEasy vector system, mda-7/IL-24 cDNA was subcloned into the adenoviral shuttle vector pAdTrack-CMV. The efficient recombination of adenoviral backbone vector pAdEasy-1 and pAdTrack-CMV-mda-7/IL-24 was achieved in bacteria E. coli BJ 5183. The recombinant adenoviral vector pAd-mda-7/IL-24 linearized by Pac Ⅰ was transfected into HEK293 cells with lipofectamine 2000. To generate higher titer viral stocks, the amplification of recombinant adenovirus was accomplished in packing cells. Viral titers were measured by tissue culture infectious dose 50 (TCID_ 50 ) method. Ad.mda-7/IL-24 was identified by PCR. The expression of pAd-mda-7/IL-24 in NCI-H446 cells was detected by Western-blot analysis. The function of Ad.mda-7/IL-24 on the proliferation of NCI-H446 cells was assayed by MTT after cells were infected by 50 pfu/ml adenovirus. Results The recombinant adenoviral shutter vector pAdTrack-CMV-mda-7/IL-24 and recombinant adenoviral vector pAd-mda-7/IL-24 were constructed successfully as identified by sequence analysis. PCR assay showed that adenovirus Ad.mda-7/IL-24 contained mda-7/IL-24 cDNA. After amplification in packing cell HEK293, the titer of virus was 2?10~ 10 pfu/ml measured by TCID_ 50 assay. Western-blot results identified that MDA-7/IL-24 could be expressed in NCI-H446 cells. After infected by 50 pfu/ml adenovirus, the proliferation of NCI-H446 cells was inhibited by 21.37% with MTT method. Conclusion Ad.mda-7/IL-24 can inhibit the growth of NCI-H446 cell obviously. This result lays foundation to study its function mechanism and to apply it in gene therapy of the small cell lung cancer.

Objective: To assess the therapeutic effect and safety of Gongmo Kang capsule (GKC) in treating metrorrhagia (with blood stasis syndrome) caused by drug - induced abortion. Methods: A randomized double - blind controlled design was adopted. One hundred and five patients were treated with GKC, and other 104 patients in the control group with Gongxue Ning capsule (GNC). Results: In GKC group, the remarkably effective rate was 73. 33%, the total effective rate was 85. 70% ; the remarkably effective rate for improving syndromes was 80.00% and the total effective rate was 88.57% ; the mean bleeding period was (4.46 ? 1.71) days.
