BACKGROUND: Cryotherapy of acute soft tissue injury has been widely used in clinical practice.OBJECTIVE: To observe the histological changes and treatment effect of different cryotherapies on the rats' acute damage of soft tissue. METHODS: Neonatal Wistar rats were randomized to normal, model, intermittent cryotherapy and continuous cryotherapy groups. Models of acute damage of soft tissue were established in model, intermittent cryotherapy and continuous cryotherapy groups. In intermittent cryotherapy group, the injury was treated by intermittent cryotherapy with ice bag at 4 °C; in the continuous cryotherapy group, the injury was treated by continuous cryotherapy with ice bag at 4 °C; the model group was not treated. Histological changes were observed at 48 hours. Injury degree was evaluated using injury symptom index.RESULTS AND CONCLUSION: Compared with model group, the scores of injury symptom index and histology were lower, interleukin-1β expression was reduced, and transforming growth factor-β1 (TGF-β1) expression was increased in intermittent cryotherapy and continuous cryotherapy groups (P < 0.05). Compared with intermittent cryotherapy group, the scores of injury symptom index and histology were reduced (P < 0.05), interleukin-1β expression was reduced (P < 0.05), and TGF-β1 expression was increased in continuous cryotherapy group (P < 0.05). Results demonstrated that cryotherapy can cure the acute damage of soft tissue by reducing interleukin-1β expression and raising TGF-β1 expression. Continuous cryotherapy is superior over intermittent cryotherapy.

Acta Nutrimenta Sinica 1956;0(02):-
The thiamine and nicotinic acid content of the husked rice (whole rice) samples from 9 representative varieties of paddy collected from Jiangsu and Zhejiang provinces were analyzed, and the effect of degree of milling on the content of these two vitamins were studied. The paddy samples were husked with wooden mill and the husked rice samples were milled with laboratory rice mill to 5 grades, namely, "96", "94", "92", "90" and "88" percent extraction (by 96% extraction we mean 96 grams of finished rice were derived from 100 grams of husked rice), and the thiamine and nicotinic acid content of each sample were determined. It is found that the thiamine and nicotinic acid content of the rice of different variety may vary considerably, and both were decreased with the increase of degree of milling. Generally, above the "94" grade, the loss of thiamine is higher than that of nicotinic acid, while down from the "92" grade, less thiamine and more nicotinic acid were lost. The rate of decrease in nicotinic acid content is relatively irregular as compared with that of thiamine, and different varieties are affected differently by the milling process.
5.Impact of AMPKgamma silencing on AMPK activation and intracellular lipids regulation.
Acta Pharmaceutica Sinica 2013;48(6):842-7
The study is aimed to confirm the silencing efficiency of the vector in human hepatocellular liver carcinoma cell line (HepG2), and observe effects of AMPKgamma silencing on the AMPK stimulating activity and lipid synthesis of cordycepin (CCS), a natural product with known AMPK activating function. The downregulating efficacy of siRNAs on AMPKgamma expression was confirmed in our previous study. The double stranded shRNA Oligo was ligated to lentivirus vector and verified by sequencing. The lentiviral which can effectively inhibited protein expression levels of AMPKgamma was selected by Western blotting, and the regulation of CCS on protein expression of AMPKgamma and p-AMPK in AMPKgamma silence cells were detected by Western blotting analysis. The lipid accumulation in cells was observed by Oil-Red O stain and cells were collected for the estimation of cholesterol (TC), triglyceride (TG). The results showed that the lentiviral vector carrying a shRNA targeting the AMPKgamma gene was successfully constructed. Western blotting analysis confirmed that GR085 had the highest interfering efficiency. Treatment with CCS can significantly increase the levels of phospho-AMPK in normal cells, and the level of TC, TG was reduced, but in AMPKgamma silence cells the effects of CCS on AMPK activation and lipid synthesis were almost completely abolished without changing the expression levels of total AMPK or AMPKgamma protein. In conclusion, the AMPKgamma gene may be related to AMPK activation and intracellular lipids regulation by CCS.

Cancer Research and Clinic 2009;21(6):377-379
Objective To study the abnormal expression of P-AKT in follicular thyroid carcinoma and its relationship with angiogenesis. Methods MVD and expressions of P-AKT, VEGF in 40 specimens of FTC tissues were detected by immunohistochemical PV6000 method. Results The expression rate of P-AKT in FTC was higher than that of non-FTC tissues. The result was of statistical significance (P<0.001). There were correlations between the MVD of FTC tissue and the expression of P-AKT and VEGF (P<0.05). The MVD of P-AKT positive FTC tissue was higher than that of negative tissues (P<0.05). The MVD of VEGF positive FTC tissue was also higher than that of negative tissues(P<0.05). There were correlations between the expression of P-AKT and VEGF(P<0.05). Conclusion The hyperexpression of P-AKT played an important role in the secondary deposits and development of human follicular thyroid carcinoma. AKT/VEGF pathway may exist in FTC, and AKT is the key factor in the pathway which regulated the expression of VEGF.
