1.Cloning of the human β-globin MAR and construction of the pCAT-MAR expression vector

Objective To clone the human β-globin matrix attachment region(MAR) and construct the mammalian animal expression vector pCAT-MAR, which contains the MAR and CAT reporter gene.Methods The human genomic DNA was extracted through phenol/chloroform and precipitated with ethanol, followed the MAR was amplified through PCR using the primers designed according to the GenBank sequence. After identified by agarose gel electrophoresis, sequenced and analyzed by the software, the PCR products were cut with restriction enzymes and ligated into the pCAT3-control vector to construct the pCAT-MAR vector. Results About 770bp band appeared in the agarose gel electrophoresis, the similarly compared with the published MAR sequence was 99.9%, the DNA fragment has the MAR typical features. The pCAT-MAR vector was demonstrated to be right through digestion with the corresponding enzymes and agarose gel electrophoresis. Conclusion The humanβ-globin MAR is cloned through PCR, and the expression vector pCAT-MAR containing the MAR is successfully constructed.
2.Relationship between dynamic pulse pressure and renal damage in elderly patients with essential hypertension

60 mm Hg (PP1 group) or 40 mm Hg≤PP≤60 mm Hg (PP2 group),1 mm Hg =0.133 kPa.Ambulatory blood pressure monitoring,the serum and urine ?_2-MG,urine mAlb,blood BUN and Cr were measured in both groups,the blood pressure,serum and urine ?_2-MG,urine mAlb,blood BUN and Cr were compared between two groups. Results Compared with PP2 group,the PP1 group showed higher levels of renal function indexes. Conclusion Renal damage is related with pulse pressure in senile essential hypertension.
3.Study on the effect of web-based teaching, focused on the subject of history of nursing

Objective To evaluate the effect of web-based teaching focused on the subject of history of nursing in the course of Introduction to Nursing. Method The 133 undergraduates in Class 2005 were randomly assigned to web-based teaching group and traditional teaching group focused on the subject of history of nursing in accordance with the same teach-ing objectives. The teaching outcomes were evaluated by a closed-book exam and questionnaire investigation. Results The students' performance in the exam was higher in the web-based teaching group than that of traditional teaching group(P<0.01). There were significant differences in the self-rated improvement of recognition and skills domains (P<0.05),while no significant difference in the affective domain (P=0.181). As a whole,the students' acceptance to web-based teaching was higher than the traditional teaching. Conclusion The web-based teaching can achieve good teaching outeomes,which can embody "student-centered" principle,improve the students' learning interest and initiative,and meet the teaching objectives in line with the re-quirements of quality education.
4.The investigation of present situation of nursing education in our country from the perspective of educate

Objective The main purpose of this study was to discuss the current situation of our nursing education from the perspective of educate,and to provide enlightenment for the higher nursing education reform.Methods The current situation of the development of nursing education questionnaire was adopted to conduct the survey.Results Nursing education in our country faced with the problems such as imperfection of the education concept,lack of humanistic curriculum,less practice classes,theory was divorced from practice,single evaluation tools of teaching.Conclusions Our country should carry on the reform of education mode,build international nursing personnel training system,improve the training quality and competition of nursing personnel.
5.Making suggestions of nursing postgraduates' training mode reform through the state of their core competence

Objective To make suggestions of reform of nursing postgraduates' training mode according to the state of their core competence. Methods 297 nursing postgraduates were investigated using the assessment form of nursing postgraduate's core competence, the questionnaire of training mode and general conditions. Results Nursing postgraduate's core competence were not good, the four lowest scored ability respectively were English ability, scientific research ability, teaching ability, grasping ability of professional knowledge and skill. Conclusions In order to promote the teaching quality, we should reform the training mode from the following aspects: to make clear training target; to make clear professional direction, formulate and carry out training program effectively; to increase the courses related with profession; to reinforce the study of public English and professional English ; to improve the ability of scientific research; to strengthen and perfect the teaching practice; to consolidate the basic knowledge,the basic theory and the basic skill of nursing; to build a unified and normal evaluation system.
6.Chronic Toxic Effect of Herbicide Mixture on Mice Hepatic Function and Ultrastructure

Objective To observe the chronic hepatotoxicity of the commonly used herbicide mixture in the mice. Methods Low doses herbicide mixture diluted in mice drinking water. The parameters related to hepatic function and ultrastructural changes of the hepatocytes were examined after 18 weeks and 24 weeks of treatment. Results No changes was observed in every parameter after 18 weeks of treatment compared with the control group. After 24-week of treatment, the electron microscope revealed the ultrastructural changes in the hepatocytes, rough endoplasmic reticulum showed dilatation, mitochondria showed matrix muddy with distortion and disappearance of inner crista and the serum transaminase were higher than those in the control group(P
7.Effects of circulating hormones, cell cation transport and cyclic nucleotides on left ventricular hypertrophy in essential hypertension

Objective To investigate the possible roles of hemodynamics, circulating hormones, cell cation transport and cyclic nucleotides in left ventricular hypertrophy (LVH) of essential hypertension (EH) in vivo. Methods Blood pressure, total peripheral resistance (TPR) and plasma norepinephrine (NE), endothelin (ET), angiotensin Ⅱ (AngⅡ), endogenous digitalis like substances (EDLS) and atrial natriuretic peptide (ANP) were determined, along with the measurements of lymphocytic membrane ion pumps, cations and cyclic nucleotides in 45 EH patients with LVH (12 concentric hypertrophy, 33 eccentric hypertrophy), 30 normal subjects and 45 EH patients without LVH (as double concurrent controls). Multiple stepwise regression analysis was computed using left ventricular mass index (LVMI) as dependent variable and 22 parameters as independent variables. Results (1) In EH patients with LVH, increases were seen in systolic blood pressure, mean arterial pressure (MAP) and NE, ET, AngⅡ, EDLS and ANP as well as lymphocytic Na +, Ca 2+ , cAMP and cAMP/cGMP ratio, while Na + K + ATPase and Ca 2+ ATPase activities, K + and Mg 2+ decreased. (2) Five hormones were related respectively to ion pumps, Na +, Ca 2+ and cyclic nucleotides. (3) LVMI were correlated with NE, ET, AngⅡ and EDLS, Ca 2+ ATPase, Na +, cAMP and cAMP/cGMP ratio. (4) The regression equation revealed that NE, ET, Ca 2+ ATPase, Mg 2+ and cAMP were the independent factors affecting LVH. (5) TPR and AngⅡ were higher, K + was lower in concentric LVH than those in eccentric LVH. Conclusion The changes of hemodynamics, circulating hormones, cell cation transport and cyclic nucleotides, or their interaction may be involved in the pathogenesis of LVH in EH. NE, ET. Ca 2+ ATPase, Mg 2+ and cAMP seem to play more important parts in LVH . TPR , whereas AngⅡ and K + may relate to cocentric LVH.
8.EXPERIMENTAL STUDIES ON THE ANITUMOR EFFECT OF OYSTER EXTRACT

Balb/c mice with HAC murine hepatic cancer were fed with oyster extract at a daily dose of 0. 2 ml (20 mg) for ten successive days. We found that the tumor-caused decreased immunological indicarors including total T cell number, the ratio of helper T cells, the transformation strength induced by mitogens, and NK activity increased markedly. Compared with the control, mice fed with oyster extract showed prolonged survive and their tumor weight reduced significantly. When mice earring human colon cancer were fed with oyster extract at the same dose for 15 days, the tumor size reduced to 44. 6% of the control tumors. The possiblity that oyster extract inhibits tumor growth by enhencing host immune function, especially killer cell activity, was discussed.
9.Detection of specific proteins in serum of acute stage patients with SARS and its clinical application

Objective To observe the changes of specific proteins in serum of acute stage patients with SARS and its clinical application. Methods The acute stage patients with SARS group included 42 cases who had clinical symptoms less than five days. In normal group: 30 healthy adults were as controls. Tests for specific proteins were immunoglobin G ( IgG ) , immunoglobin A ( IgA) , immunoglobin M ( IgM) , complement C3( C3),complement C4(C4),C-reaction protein (CRP), al acid glycoprotein( a1-AG) , a1 antitrypsin(a1-AT) ,transferrin(TRF), ceruloplasmin( CER) in sera by Ratenephelometer. Results The level of CRP, a1-AG, a1-AT in sera of patients with SARS were higher than that in control group ( P 0. 05). Conclusion It suggested that specific proteins in serum of acute stage patients with SARS change in a particular way. There were some help for the diagnosis of the phlogistic degree of acute stage patients with SARS by detecting CRP, a1-AG and TRF simultaneously.
10.Study on the relationship of social support and life quality of life of liver transplantation patients

Objective To study the relationship of social support and quality of life of liver transplantation patients, investigating the effective measures to improve their life quality. Methods Questionnaires were filled in by 90 liver transplantation patients and a descriptive study was used. Results Positive correlation was found between social support and life quality of liver transplantation patients. Conclusion Social support was related to the life quality of liver transplantation patients. Nurses should pay attention to the effect of social system to improve their life quality.
