2.Synthesis and in vitro antibacterial activity of 7-(4-alkoxyimino-3-methyl-3-methylaminopiperidin-1-yl)quinolones.

To explore new agents of quinolone derivatives with high antibacterial activity, 7-(4-alkoxyimino-3-methyl-3-methylaminopiperidin-1-yl)quinolones were designed and synthesized, and their activity against gram-positive and gram-negative strains was tested in vitro. Sixteen target compounds were obtained. Their structures were established by 1H NMR, HRMS and X-ray crystallographic analysis. Compounds 14k and 14m-14o show good antibacterial activity against the tested five gram-positive strains and five gram-negative strains (MIC: 0.25-16 micromg x mL(-1)), of which the most active compound 14o is 8-fold more potent than levofloxacin against S. pneumoniae (MIC: 4 microg x mL(-1)), and comparable to levofloxacin against S. aureus, S. epidermidis, E. faecalis and E. coli (MIC: 0.25-1 microg x mL(-1)), but generally less potent than gemifloxacin.
3.Effects of Naofucong grain on brain NMDA receptor in experimental dementia mice

ObjectiveTo observe effects of Naofucong grain on learning and memory in cerebral ischemic mice, and identify the mechanisms on N-methyl-D-aspartate (NMDA) receptor.MethodsExperimental dementia mice were induced by ischemia reperfusion injury, and treated with Naofucong grain or Dangguishaoyao powder (positive control medicine). Learning and memory were tested in the Morris water maze. Activities of NMDA receptor in brain cortex and hippocampus were detected ResultsIncubation periods of mice treated with Naofucong grain were significantly shorter than that of ischemic model mice (P<0.01) Activities of NMDA receptor in brain cortex and hippocampus showed no difference between Naofucong grain treated mice and normal mice (P>0.05)Activities of NMDA receptor in brain cortex and hippocampus in Naofucong grain treated mice were significantly lower than that in ischemic model mice(P<0.01) ConclusionThe improvement of Naofucong grain in the memory and learning in cerebral ischemic mice may be related to the inhibition of the activity of NMDA receptor.
4.Studies on the protective effect of drug serum taken from rats administered with Naofucong in PC12 cell ischemic model

ObjectiveTo examine effects of traditional Chinese herbs Naofucong grain on PC12 cells injured by H2O2 in vitro MethodsRats random divided into 2 groups: Naofucong grain treating group (administered with Naofucong grain) and control group (administered with distilled water) After 3 days administration, serums were obtained and added to cultured PC12 cells which were under different concentrations of H2O2 Morphological examinations in PC12 cells were detected by MTT measurement ResultsMTT measurement showed that Naofucong grain administered rats serums significantly increased the survival number of PC12 cells in different concentrations of H2O2.Conclusion Naofucong grain may have obvious protective effects on PC12 cells injured by H2O2.
5.Cloning of the Coding Region and 3′Non-coding Region of Calmodulin 2 in Guinea Pig

Objective To clone the coding region and 3′non?coding region of calmodulin 2(CaM2)in guinea pig,to provide the genetic informa?tion for studying the gene function of Calmodulin 2. Methods Total RNA was extracted from heart tissue of guinea pig,the coding region and 3′non?coding region of CaM2 were amplified by RT?PCR and 3′?RACE PCR methods,and the recombinant plasmid was constructed by inserting cDNA of the coding region and 3′non?coding region of CaM2 into the cloning vector by genetic engineering technology followed by DNA sequencing and se?quence analysis. Results The cloned coding region of CaM2 was 450 bp,and the 3′non?coding region of CaM2 was 660 bp. The amino acid se?quences of the coding region of CaM2 was consistent with those of other CaM subtypes,and the 3′non?coding region of CaM2 had low homology with those of other subtypes. Conclusion The cloning of CaM2 coding region and 3′non?coding region in guinea pig was the foundation for further study on the gene function of CaM2 and its role in related diseases.

Pseudogenes have long been regarded as fossilized,nonfunctional genomes that are evidence of human evolution.However,in recent years,pseudogenes have been found to play an important role in both physiological and pathological processes,especially in tumors,regulating occurrence and development of tumors in parental gene dependence and parental gene independence,including liver cancer.In this paper,we summarized research progress of pseudogenes and their roles in proliferation,apoptosis,inva-sion and metastasis of liver cancer cells,providing new ideas for treatment of liver cancer.

8.Rhinoplasty of unilateral cleft lip.

OBJECTIVETo investigate the technique for unilateral cleft lip nasal deformities.
METHODSThrough the traditional columella margin incision, the alar cartilage and the lateral nasal cartilage were exposed. After the mucosa between the two cartilages was divided, a mucosa-cartilage flap was developed and anchored, moving the lateral crus upwardly and medially, correcting the nasal deformity.
RESULTS52 cases of primary nasal deformities and 18 cases of secondary nasal deformities were corrected with this technique. Satisfactory results were achieved.
CONCLUSIONThis technique is recommendable, which is more likely to restore the physiological characters of the nose than the conventional technique.

9.The influence of Matrine on apoptosis of fibroblasts and the expression of apoptotic modulation related protein in hypertrophic scar of rabbit ear.

OBJECTIVETo investigate the effects of Matrine on apoptosis of fibroblasts and the expression of apoptotic modulation related protein in the hypertrophic scar.
METHODSHypertrophic scar was produced on the ear of 24 New Zealand white rabbits, which were employed as the model, and were randomly and equally divided into control (CC) and Matrine (M) groups (12 in each group). Matrine (50 g/L) was injected into the ear scar in M group and with normal saline in C group once every four days. At 2, 4, 6, 8 and 12 weeks after the injection, the apoptotic fibroblast count in the scar was determined by TUNEL method, and the expressions of apoptosis related modulation proteins p53, bcl-2, bax were detected by immunohistochemistry method.
RESULTSThe apoptotic fibroblast count was much larger in M group than that in C group at all test time points (P < 0.05). Furthermore, the bax expression was increased and that of p53 and bcl-2 was decreased significantly in M group. In adding, the scar became flat in M group.
CONCLUSIONMatrine might obviously enhance the fibroblast apoptosis in rabbit ear hypertrophic scar, and up-regulate the expression of apoptosis related modulation protein bax and down-regulate the expression of p53 and Bcl-2.

10.A virtual deformable mandible model used for reconstruction computer aided design of large mandibular defects

Objective To establish a three-dimensional virtual deformable mandible model used for individual reconstruction design of large mandibular defect.Methods A virtual deformable mandible model has been established by a 3D animation software.The model could be used for preoperative reconstruction design of large mandibular defects cases.According to the temporomandibular joint fossa position,maxillary dental arch,the normal relationship of cranio-maxillofacial profile,and the morphology of the residual segments of mandible,the virtual mandible model could be scaled and adjusted and a virtual mandible with individual features was obtained.Three normal skulls have been used to validate the adjustment ability of the virtual deformable mandible model.The preoperative reconstruction design process of 1 typical large mandibular defect case was demonstrated.Results The deformation matching ability of the virtual deformable mandible model was very good.The registration between the design model and the orignal mandible was over 90%.The design effect of the large mandiblar defect case was satisfied.Conclusions Virtual deformable mandible model is a new feasible method to aid preoperative reconstruction design of large mandibular defects.
