1.Study on apoptosis of K562 cells induced by tumor-bearing herb serum of selenium-protein polysaccharide extracted from Qinba selenium-mushroom(秦巴硒菇)

Objective: To explore the effect of selenium-protein polysaccharide extracted from Qinba (selenium-mushroom)(秦巴硒菇) on apoptosis of K562 cells.Methods: Tumor-bearing mice model was established,and selenium-protein polysaccharide extracted from Qinba selenium-mushroom contained-herb serum was prepared by the method of serum pharmacology.K562 cells were conventionally cultured.Growth inhibition rate of tumor cells was detected by methyl thiazolyl tetrazolium(MTT) method.Morphological changes of apoptotic cell were observed under a fluorescent microscope.Cell apoptosis was observed by DNA electrophoresis.Caspase3 relative gene was measured by colorimetry.Results: Selenium-protein(polysaccharide) extracted from Qinba selenium-mushroom could significantly inhibit the growth of K562 cells,there was a remarkably positive correlation between drug concentration,time and inhibitory rate.Apoptotic phenomenon was certained via morphological examination and DNA electrophoresis.Compared with control group,caspase3 gene was markedly upregulated.Conclusion: Selenium-protein(polysaccharide) extracted from Qinba selenium-mushroom could induce apoptosis of K562 cells.The mechanism may be related to upregulation of caspase3.
2.Effect of Total Flavonoids of Hedysarum Polybotry on Inducing Differentiation of Leukemic Cells

Objective:To investigate the effect and mechanism of total flavonoids of Hedysarum polybotry on induction of differentiation in human leukemia HL-60 cells. Method After the treatment of HL-60 cells with total flavonoids of Hedysarum polybotry, the cell differentiation was detected with NBT reduction method. Cell cycle, CD11b and C-fos were analysed by the flow cytometry. Result The positive rate of NBT reduction and the expression of CD11b were significantly increased. Similar, the expression of C-fos gene was upregulated. The growth of HL-60 cells was arrested at G0/G1 and G2/M phase. Conclusion Total flavonoids of Hedysarum polybotry could induce differentiation of HL-60 cells. Its molecular mechanism might be related to the modulation of gene expressions associated with the proliferation and differentiation, which leads to the inhibition of DNA synthesis.
3.ITS Sequence Analysis of Angelica from Different Source in Gansu Province

Objective To establish the basis of molecular systematics of Angelica from different areas of Gansu province so as to offer molecular basis of identification Gansu-produced Angelica with geoherbalism. Methods The ITS gene of Angelica was obtained by PCR and sequenced to be analyzed by DNAStar software to calculate the genetic distance among different specimens. Results The ITS sequence of Angelica between Min County and Zhang County of Gansu Province shows trivial differences with the range of 0.000 0~0.008 3. All 5 Min County-produced Angelica specimens have same bases at 586 site with "C" and 589 site with "T" with Zhang County-produced Angelica DG9 specimen, whereas the 586 site and 589 site bases of the other 3 Zhang County-produced Angelica specimens are "T" and "G" respectively. Conclusion The ITS sequence characteristics can be recognized as effective molecular marker of Angelica, and both 586 site base and 589 site base of the ITS sequence of Angelica can be recognized as identification bases of Min County-produced Angelica.
4.Effects of Jiuxieling Granules on Inflammatory Lesion of Colon Mucous Membrane in Rats with Ulcerative Colitis

Objective To observe the effects of Jiuxieling granules on rats with ulcerative colitis and explore its mechanism.Methods Ulcerative colitis models were established by immune method combined with local stimulation of 5% acetic acid.Wistar rats were randomly divided into control,model,positive control and low,middle,high dose Jiuxieling granules group.Rats in positive control group were treated with sulfasalazine,and those in three treatment groups were treated with low,middle and high dosage of Jiuxieling granules respectively for 14 days continuously.24 hours after last administration,the rats were killed to obtain colon mucous membrane to test GSH-Px,SOD,MDA,NO,tNOS and iNOS concentration of the homogenate of colon mucous membrane.Results Jiuxieling granules could markedly improve the levels of GSH-Px and SOD activity,whereas decrease the levels of MDA,NO,TNOS and iNOS.Conclusion Jiuxieling granules exert its marked curative effects on rats of ulcerative colonitis by resisting oxygen free radical lesion,inhibiting the generation of NO and NOS,and enhancing the activity of antioxidase.
5.Preparation of ?-elemene solid lipid nanoparticles by film-ultrasonic wave dissolving techniques

Objective To prepare ?-elemene solid lipid nanoparticles and investigate their particle size diameter and entrapment efficiency.Methods The ?-elemene solid lipid nanoparticles were prepared by film ultrasonic wave dissolving techniques.And the optimum formula was selected through orthogonal design test according to the entrapment efficiency.Results The optimizing technique was ?-elemene(20 ?L),stearic acid(90 mg),lecithin(90 mg),Tween80(2.5 %,5 mL) and Poloxamer 188(2.5 %,5 mL).Conclusion The technique of preparing ?-elemene by film-ultrasonic wave dissolving technique is feasible.

Objective To observe the effects of hedysari polysaccharide (HPS) on myocardial fibrosis and the expression of MMP-2/TIMP-2 in model mice of diabetic cardiomyopathy; To discuss the mechanism of action of prevention and treatment of myocardial fibrosis in diabetes.Methods Sixty mice were randomly divided into model group, rosiglitazone group and HPS high-, mediume- and low-dose groups. The normal group was 12 non-transgenic male BKS.Cg-Dock7m+/+Leprdb/JNjumice with the same age. Each group was given relevant medicine for gavage, for 8 weeks. Blood glucose of mice before and after medication 2, 4, 6, and 8 weeks was detected. The levels of MMP-2, MMP-2 and MMP-9 in myocardium were measured by Masson staining. The protein expressions of MMP-2 and TIMP-1 in myocardium were detected by Western blot.Results Compared with the model group, the blood glucose of HPS (high- and medium dose) groups and rosiglitazone group decreased significantly. Masson staining showed that the green fibers in the model group significantly increased and rosiglitazone group and HPS high-dose group decreased compared with the model group. Western blot showed that the expressions of MMP-2 in model group and MMP-2/TIMP-2 ratio were declined significantly, while the expression of MMP-2 was increased and TIMP-2 was decreased significantly, and the ratio of MMP-2/TIMP-2 increased in rosiglitazone group and HPS high- and medium-dose group.Conclusion HPS may reduce the degree of myocardial fibrosis in model mice with diabetic cardiomyopathy. The therapeutic effect of HPS may be to relieve myocardial fibrosis in model mice by increasing the ratio of MMP-2/TIMP-2.

Pseudogenes have long been regarded as fossilized,nonfunctional genomes that are evidence of human evolution.However,in recent years,pseudogenes have been found to play an important role in both physiological and pathological processes,especially in tumors,regulating occurrence and development of tumors in parental gene dependence and parental gene independence,including liver cancer.In this paper,we summarized research progress of pseudogenes and their roles in proliferation,apoptosis,inva-sion and metastasis of liver cancer cells,providing new ideas for treatment of liver cancer.
