1.Mismatch repair in hereditary nonpolyposis colorectal cancer

Hereditary nonpolyposis colorectal cancer as a eolorectal cancer is of most hereditary characters.With developing of the molecular genetics on detection technology,mismatch repair gene has already been detected by multi-approach,and been applied to screen hereditary nonpolyposis eolorectal cancer patients.In this artical,we mainly discuss the correlated study progression between hereditary nonpolyposis eolorectal cancer and several mismatch repair genes such as MSH2,MLH1,MSH6,PMS1,PMS2,MLH3 and EXO1.
2.Preliminary study on self-emulsifying drug delivery system of tanshinone

Objective To screen the formulation for tanshinone self-emulsifying drug delivery system (SEDDS) and evaluate its stability.Methods The optimal tanshinone SEDDS formulation was established through solubility experiment,emulsion examination,fully emulsified time,droplet size determination,and pseudo-ternary diagram drawing.The content and stability were evaluated by HPLC assay under the condition of illumination,high and low temperature.Results The oil phase,surfactant and co-surfactant in the optimal tanshinone SEDDS formulation were ethyl oleate,TX10,Tween 80,and isopropyl alcohol (60∶84∶21∶35).Conclusion The acquired formulation of tanshinone SEDDS is stable in the dark and room temperature.
3.Establishment of Guinea Pig Models of Allergic Rhinitis with Kidney-yang De ficiency

Objective To find a new method for the establishment of allergic rhinitis(AR) models with kidney-yang deficiency.Methods Fourty-five Hartely guinea pigs were randomized into 4 groups: hydroxycarbamide+ ovalbumin(OV); predisone+ OV;hydroxycarbamide+OV+Shenqi decoction;normal control).The general behavior and the body weight of guinea pigs were observed before and after treatment.After 3 weeks,the reaction of guinea pigs to histamine nasal drops was observed and then all animals were sacrificed to examine the pathological changes of nasal septum mucosa.Result The new model induced by hydroxycarbamide and OV was proved to be set up successfully: the manifestations of kidneyyang deficiency in the new model were more representative than those induced by predisone and OV,and the time for modeling was shorter.Conclusion Using hydr oxycarbamide and OV to set up kedney-yang deficiency AR models is an new feasible method.The result of this study will provide evidence for the further research of kidney-yang deficiency AR.
4.Comparison of EEG complexity between rats under awaking and anesthesia

Objective To compare the EEG complexity between rats under awaking and differ-ent depth of anesthesia via analyzing sample entropy and fractal dimension.Methods Sixteen SD rats were intraperitoneally injected with urethane twice,first with 500 mg/kg and second with 800 mg/kg one hour later.The scalp EEG was collected in stage of awaking (W),light anesthesia (LA)and heavy anesthesia (HA).The sample entropy (SampEn)and fractal dimension (FD)were computed by MATLAB.The characteristic values were denoised by linear dynamic system method during the whole process.Results The value of SampEn and FD gradually dropped from awaking to heavy anes-thesia.The SampEn and FD in W was significantly higher than the value in LA or in HA (P <0.05). The SampEn and FD in HA was significantly lower than that in LA (P < 0.05 ).Conclusion The SampEn and FD of EEG could be used to monitor the depth of anesthesia.
5.Study on the present situation and the influential factors of self-care ability among elderly hypertensive patients in Hengyang community

Objective To explore the present situation of self-care ability among elderly hypertensive patients in the community and its influential factors.Methods 310 elderly hypertensive patients in Hengyang community were studied with a demographic questionnaire,ESCA and SSRS.Results The total mean scores of self-care ability were (105.43±19.25).68.71% of the elderly hypertensive patients were at medium level and 29.35% of them were at high level.Statistically significant relations were found between self-care ability and the following condition factors:education level,marital status,monthly income,obtaining health education status,social support,subjective support,availability,objective support and the severity of hypertension.There were no significant relations with sex and occupations.The results of multiple regression showed that the main factors ranged from high to low including the severity of hypertension,education level,monthly income,subjective support,obtaining health education status and marital status.Conclusions Community health nurses should aim directly at the influential factors,take effective measures and improve their self-care ability.
6.Role of acid-sensing ion channel 1a in global cerebral ischemia-reperfusion injury in rats

Objective To investigate the role of acid-sensing ion channel 1a(ASIC1a) in global cerebral ischemia-reperfusion injury in rats.Methods Forty male SD rats weighing 250-300 g were randomly divided into 4 groups (n =10 each): sham operation group (group S),cerebral ischemia-reperfusion group (group I/R),solvent control group (group SC) and group PcTX1 (a ASIC1 a blocker,group P).Global cerebral ischemia-reperfusion was induced by four-vessel occlusion.PcTX1(500 ng/ml)6 μl or solvent 6 μl was injected into the crerbral ventricular at the begining of reperfusion in groups P and SC respectively.The rats were sacrificed at 24 h of reperfusion,and then the hippocampi were removed for determination of Caspase-3,Bcl-2 and Bax protein expression and microscopic examination.Results Compared with group S,the expression of Caspase-3,Bcl-2 and Bax protein was up-regulated in groups I/R,SC and P (P < 0.05).Compared with group I/R,the expression of Caspase-3 and Bax was down-regulated,and the expression of Bcl-2 was up-regulated in group P ( P < 0.05).There was no significant difference in Caspase-3,Bcl-2 and Bax protein expression between groups I/R and SC (P > 0.05).The histopathologic damage was ameliorated in group P as compared with group I/R.Conclusion ASIC1a can induce global cerebral ischemia-reperfusion injury in rats by up-regulating Caspase-3 and Bax expression,and down-regulating Bcl-2 expression and inducing apoptosis.
7.Effects of different doses of PcTx1 on global cerebral ischemia-reperfusion injury in rats

ObjectiveTo explore the effects of different doses of PcTx1,a specific blocker of acid-sensing ion channel 1a,on global cerebral ischemia/repedfusion (I/R) injury in rats,MethodsSixty adult male Sprague Dawley rats (weighing 250-300 g) were randomly divided into 6 grups ( n =10 each):sham operation group (group S),I/R group,different doses of PcTx1 ( 10 ng/ml,group P1 ; 25 ng/ml,group P2 ; 50 ng/ml,group P3 ;and 500 ng/ml,group P4 ) groups.Global cerebral ischemia was induced by the modified procedure of Pulsinelli 4-vessel occlusion.In groups P1,P2,P3 and P4,different doses of PcTx1 ( 10,25,50 and 500 ng/ml),6 μl each,were respectively injected into the lateral cerebral ventricle at the initiation of reperfusion,while equal volume of double distilled water was injected instead in group I/R.Six rats in each group were sacrificed at 24 h of reperfusion,and the brains were immediately removed,Thereafter,the contents of malondialdehyde (MDA),reduced glutathione (GSH) and ritric oxide (NO),the activities of constitutive NO synthase (eNOS) snd inducible NO synthase (iNOS) were detected in hippocampus.Four rats in each group were sacrificed at 72 h of reperfusion,and hematoxylin and eosin staining was used to observe the pathomorphological changes of the hippocampal neurons.ResultsCompared with group S,the other groups showed decreases in the contents of GSH,while increases in the contents of MDA and NO and the activities of cNOS and iNOS ( P < 0.05 or 0.01 ).The contents of GSH increased,while the contents of MDA and NO and the activities of cNOS and iNOS decreased in groups P2,P3 and P4 compared with group I/R ( P < 0.05 or 0.01).Compared with group P1,the contents of GSH increased,the contents of MDA and the activities of cNOS decreased in groups P2,P3 and P4,and the contents of NO and the activities of iNOS decreased in groups P3 and P4 ( P < 0.05 or 0.01 ).Compared with group P2,the activities of iNOS decreased in groups P3 and P4(P < 0.05 or 0.01).The damage to neurons in hippocampal CAI was severe in groups I/R and P1,but it was attenuated in groups P3 and P4.ConclusionPcTx1 25,50 and 500 ng/ml (6 μl)injected into lateral cerebral ventricle can attenuate global cerebral I/R injury in rats,and the dose 50 ng/ml (6 μl) is more suitable.
8.Performance of transfection with a complex plasmid encoding green fluorescent protein tagged cathepsin D in researches on chronic photodamaged fibroblasts

Objective To evaluate the performance of transfection with a complex plasmid encoding green fluorescent protein tagged CatD (GFP-CatD)in researches on chronic photodamaged fibroblasts. Methods Human dermal fibroblasts (HSFs)were irradiated with ultraviolet A (UVA)at 25 J/cm2 once a day for 21 consecutive days to establish a chronic photodamaged cell model. A plasmid encoding GFP-CatD was constructed and transfected into some chronic photodamaged fibroblasts (experimental group). The photodamaged HSFs receiving no treatment served as the blank control group, and those transfected with the negative plasmid encoding GFP only as the negative control group. After additional culture, fluorescence microscopy and Western-blot analysis were performed to observe and measure the expression of GFP-CatD in HSFs respectively, flow cytometry and methyl thiazolyl tetrazolium (MTT)assay to evaluate the apoptosis and proliferation of chronic photodamaged fibroblasts respectively. Results Fluorescence microscopy showed the expression of GFP-CatD in cytoplasm of chronic photodamaged fibroblasts at 96 hours after transfection with the GFP-CatD-encoding plasmid. Western-blot analysis revealed that the expression of CatD in the experimental group was 1.28 times that in the blank control group. There were no significant differences in the apoptosis rate(4.29% ± 1.30%vs. 3.03% ± 1.70% , P > 0.05)or proliferative rate (45.20% ± 4.70% vs. 43.60 ± 3.90% , P > 0.05)between the experimental group and blank control group. Conclusion CatD could be traced in chronic photodamaged fibroblasts with no changes in biological activity or cell cycle after transfection with the GFP-CatD-encoding complex plasmid.
9.Role of mitochondrial permeability transition pore of hippocampai neurons in process of hydrogen-rich saline attenuating global cerebral ischemia-reperfusion injury in rats

Objective To investigate the role of mitochondrial permeability transition pore (mPTP) of hippocampal neurons in process of hydrogen-rich saline attenuating global cerebral ischemia-reperfusion (I/R) injury in rats.Methods Seventy-two male Sprague Dawley rats,weighing 250-300 g,were randomly divided into six groups ( n =12 each):sham operation group (group S),cerebral ischemia-reperfusion group (group IR),normal saline group (group NS),hydrogen-rich saline group (group H),atractyloside group (group A) and hydrogen-rich saline + atractyloside group (group HA).Global cerebral I/R injury was produced by four-vessel occlusion method.Bilateral vertebral arteries were cauterized.Then bilateral common carotid arteries were occluded for 15min and followed by reperfusion.In groups H and HA,hydrogen-rich saline 5 ml/kg was injected intraperitoneally immediately after reperfusion,while equal volume of normal saline was injected in the other four groups.The rats in groups A and HA received intracerebroventricular injection of atractyloside 15 μl 10 min before reperfusion,while groups NS and H received intracerebroventricular injection of equal volume of normal saline.After the neurological behavior was evaluated at 24 h of reperfusion,8 rats in each group were sacrificed and the hippocampi were immediately isolated and homogenized followed by density gradient centrifugation.The opening degree of mPTP was assayed with spectrophotometry and the mitochondrial membrane potential (MMP) was detected with Rhodamine 123 method.Four rats in each group were killed at 72 h of reperfusion and the brains were removed for microscopic examination of the area CA1 of the hippocampus and determination of the number of normal pyramidal neurons.Results Compared with group S,the neurological behavior was compromised,MMP was decreased and mPTP opening degree was enhanced in the other five groups ( P < 0.05).The neurological behavior was better,MMP was increased and mPTP opening degree was decreased in groups H and HA as compared with group IR ( P < 0.05).Compared with group H,the neurological behavior was compromised,MMP was decreased and mPTP opening degree was enhanced in group HA ( P < 0.05).Compared with group IR,the number of normal pyramidal neurons at 72 h of reperfusion in the CA1 region of the hippocampus was higher in group HA ( P <0.05).The injury of the CA1 region of the hippocampus at 72 h of reperfusion was attenuated in group H as compared with groups IR,NS,A and HA.Conclusion Hydrogen-rich saline can attenuate global cerebral I/R injury throngh inhibiting the mPTP opening and reducing the dissipation of MMP,thus maintaining the mitochondrial function.
10. Application of whole body exercise quality assessment in prognosis evaluation of asphyxia children

Neonatal asphyxia is a clinical disease in which newborns are unable to establish regular and effective respiration after birth due to various factors, resulting in hypoxia, acidosis and hypercapnia.Severe cases can cause damage to the nervous system of children, mental retardation, cerebral palsy, and even death.The quality assessment of systemic exercise(general movements, GMs)is a simple, non-invasive and easy to implement technique for early diagnosis of neurodevelopmental disorders in children.Although GMs quality assessment has been widely used in the assessment of nervous system injury after asphyxia resuscitation at home and abroad, there has not been a systematic introduction to its pathogenesis, specific manifestations and the correlation between them.Therefore, this paper systematically reviewed the above problems combined with the latest research at home and abroad, in order to provide help for clinical work.
